1.Content Determination of Sinomenine in Qihuang Capsule by HPLC

OBJECTIVE:To establish a method for the content determination of sinomenine in Qihuang capsule. METHODS:HPLC was performed on the column of welch C18-AQ with mobile phase of methanol-phosphate buffer(gradient elution) at flow rate of 1.0 ml/min,detection wavelength was 264 nm,column temperature was 30 ℃ and volume injection was 10 μl. RESULTS:The linear range of sinomenine was 0.200 3-10.016 0 μg(r=0.999 8),RSDs of precision,stability and reproducibility tests were lower than 1.0%,recovery was 98.80%-100.94%(RSD=0.79%,n=6). CONCLUSIONS:The method is simple,accurate and re-producible,and can be used for the content determination of sinomenine in Qihuang capsule.
2.Content Determination of Hordenine in Zang Medicine Herba Aconiti by HPLC

OBJECTIVE:To establish a method for the content determination of hordenine in Zang medicine Herba Aconiti. METHODS:HPLC was performed on the column of Gemini-NX C18 with mobile phase of acetonitrile-0.25 mol/l Ammonium ace-tate solution(ammonia water to adjust the pH to 9.5,gradient elution,at flow rate of 1 ml/min,the detection wavelength was 230 nm,the column temperature was 30 ℃,and the injection volume was 20 μl.RESULTS:The linear range of hordenine was 3.276-819μg/ml(r=0.999 5);RSDs of precision,stability and reproducibility tests were lower than 2.0%;recovery was 96.21%-104.04%(RSD=1.23%,n=9). CONCLUSIONS:The method is simple,stable and reproducible,and can be used for the content determi-nation of hordenine in Zang medicine Herba Aconiti.
3.Screening on Active Sites and the Best Compatibility of NewYouguiyin Prescription

This study was aimed to clarify the main active site and the best compatibility of the newYouguiyin prescription. The rat osteoblast-like cells was used as model cells. The investigation was made on the impact of different extraction parts of each herb and their compatibility on osteoblast proliferation. The results showed that the compatibility of butyl alcohol part fromDipsacus,Epimedium andCnidium had the most significant proliferation of osteoblast activity. It was concluded that the screening of active sites and the best compatibility of newYouguiyin prescription had laid the foundation for the development of subsequent innovation for traditional Chinese medicine (TCM).
4.Determination of Six Alkaloids in Crude and ProcessedRhizomaCoptidisby HPLC

An HPLC method to determine six alkaloids of the Coptidis for Chinese Pharmacopoeia of 2015 Edition was established through C18 column. The mobile phase was CH3CN-0.25 mol·L-1 NH4Ac (36:64) (containing 8 mmol·L-1 SDS and adjusting pH 9.3 with ammonia) at the flow rate of 1 mL·min-1, the detective wavelength was 270 nm and the column temperature was 35 oC. The linear ranges of jatrorrhizine hydrochloride, columbamine hydrochloride, epiberberine hydrochloride, coptisine hydrochloride, palmatine hydrochloride and berberine hydrochloride were 0.006 96-0.233, 0.004 75-0.152, 0.003 30-0.528, 0.006 31-1.010, 0.004 71-0.753, 0.017 8-2.884 μg·mL-1, respectively. The average recoveries were 99.65%, 98.59%, 98.49%, 98.66%, 98.64%, 98.63% and RSD were 0.03%、0.15%、0.21%、0.12%、0.28%、0.23%, respectively. The method is simple and accurate, and can be used to determine the contents of jatrorrhizine, columbamine, epiberberine, coptisine, palmatine and berberine in the Coptidis.
5.Difference of chemical constituents contained in Tibetan herb Jiadiranguo from different habitats by HPLC fingerprint.

OBJECTIVETo establish the HPLC fingerprint for Halenia elliptica herbs, a traditional Tibetan medicine, in order to study constituents contained in H. elliptica from different habitats and compare their differences.
METHODHPLC analysis was made on a Welchrom-C18 (4.6 mm x 250 mm, 5 microm) with water and acetonitrile as mobile phase. The wavelength was detected as 265 nm, the flow rate was 1.0 mL x min(-1), and the column temperature was 40 degrees C. The software for chromatographic fingerprint was applied to analyze the similarity. And principal component analysis was conducted.
RESULTTwelve common chromatographic peaks were identified by fingerprint, showing a low similarity in constituent and variety. The significant difference in the proportion between xanthones and aglycones in each batch of herbs indicated no notable correlation between constituent characteristics and geographic locations of habitats.
CONCLUSIONThe method is so simple, exclusive, stable and highly repeatable that it can provide reference for identification and quality assessment of H. elliptica herbs.

6.Quantitative determination of six coumarins in Angelica dahurica var. formosana by RRLC-UV.

OBJECTIVETo establish a RRLC-UV method for simultaneous determination of oxypeucedanin hydrate, bergapten, oxypeucedanin, imperatorin, cnidilin and oxypeucedanin in Angelica dahurica var. formosana.
METHODThe analysis was carried out on a Agilent Zorbax SB-C, (4.6 mm x 100 mm, 1.8 microm) column eluted with the mobile phases of methanol (A)-water (B) in gradient elution. The flow rate was 1.0 mL x min(-1), and the UV detection wavelength was set at 310 nm.
RESULTAll calibration curves of six coumarins showed good linearity regression within test ranges (r > 0.9998), and the overrall recoveries were in the range of 99.42%-101.4%, with RSD less than 2.4%.
CONCLUSIONThe method is simple, sensitive, reliable and reproducible which can be used for the quality study of Angelica dahurica var. formnosana.

7.Determination of xanthones in Tibetan herb Jiadiranguo (Herba Haleniae).

A HPLC method was developed for simultaneous determination of 1-hydroxy-2, 3, 4, 7-trimethoxyxanthone (1), 1-hydroxy-2,3, 7- trimethoxyxanthone (2), 1-hydroxy-2, 3, 4, 5-trimethoxyxanthone (3), and 1-hydroxy-2, 3, 5- trimethoxyxanthone (4) in Halenia elliptica. The analytical column was Welchrom C18 column (4.6 mm x 250 mm, 5 microm). The mobile phase was acetonitrile- water (43:57). The detection wavelength was 265 nm. The flow rate was 1 mL x min(-1) and the column temperature was set at 40 degrees C. There was good linearity between the peak areas and concentration at the ranges of 0.414-16.6, 1.73-69.6, 5.89-117, 3.01-120.5 mg x L(-1) for 1, 2, 3 and 4 respectively. The average recoveries (n = 6) of 1, 2, 3 and 4 were 102.5%, 100.5%, 97.9% and 101.2%. Those four xanthones in thirty samples of H. elliptica. were determined by this method. The method is simple, accurate, repeatable, which could be used for the quality evaluation of H. elliptica. The total content of those four xanthones in H. elliptica should not less than 1.80% by comprehensive analysis.

8.Simultaneous determination of six alkaloids in Coptis chinensis of different regions by RP-HPLC.

A reversed-phase HPLC method for simultaneous determination of gatrorrhizine, columbamine, epiberberine, coptisine, palmatine and berberine in Coptis chinensis was developed. Analysis was carried out on an Xtimate C18 column (4.6 mm x 250 mm, 5 microm) eluted with acetonitrile-30 mmol x L(-1) ammonium bicarbonate solution (including 0.7% ammonia and 0.1% triethylamine) by gradient elution. The detective wavelength was 270 nm, the column temperature was 30 degrees C, and the flow rate was 1.0 mL x min(-1). By the above method, the linear ranges of gatrorrhizine, columbamine, epiberberine, coptisine, palmatine and berberine were 0.85-16.96 (r = 0.9997), 1.25-24.96 (r = 0.999 5), 2.05-40.96 (r = 0.999 9), 3.65-72.96 (r = 0.999 9), 2.88-57.60 (r = 0. 999 8),13.25-264.96 mg x L(-1) (r = 0.999 6), respectively. The average recoveries (n = 6) of the six alkaloids were 101.6% (RSD 1.3%),102.5% (RSD 1.5%), 100.8% (RSD 1.9%),102. 6% (RSD 1.2%), 97.80% (RSD 1.3%), 99.01% (RSD 1.5%), respectively. The determined results demonstrate that there is a significant difference in the contents of six alkaloids and total alkaloids among the tested samples. The method is accurate, reliable and repeatable for simultaneous determination of gatrorrhizine, columbamine, epiberberine, coptisine, palmatine and berberine in C. chinensis.

9.Simultaneous determination of jatrorrhizine, palmatine, berberine, and obacunone in Phellodendri Amurensis Cortex by RP-HPLC.

OBJECTIVETo develop a HPLC method to determine the content of jatrorrhizine, palmatine, berberine, and obacunone in Phellodendri Amurensis Cortex simultaneously. MEHTOD: The separations were carried out at 25 degrees C on a Phenomenex Gemini C18 column (4.6 mm x 250 mm, 5 microm) eluted with acetonitril and water containing 0.1% phosphoric acid in gradient mode. The flow rate was 1.0 mL x min(-1), detection wavelengthes were 345 nm for jatrorrhizine , palmatine, berberine and 210 nm for obacunone.
RESULTThe average recoveries of jatrorrhizine, palmatine, berberine, and obacunone were 98.94%, 101.17%, 96.22% and 98.90%, respectively.
CONCLUSIONThe method is simple, accurate and repeatable, and can be used in content determination of jatrorrhizine, palmatine, berberine, and obacunone in Phellodendri Amurensis Cortex.

10.Dynamic accumulation of effective components and biomass of Coptis chinensis in Hongya county.

OBJECTIVETo study the dynamic accumulation of the effective components and biomass of Coptis chinensis, so to provide the experimental date of optimal harvest time for C. chinensis in Hongya county.
METHODThe samples of three to five years were gathered from the same field and time. The biomass was analyzed by weighed. The jatrorrhizine, columbamine, epiberberine, coptisine, palmatine and berberine in C. chinensis were analyzed by HPLC.
RESULTWith the increasing of years of growth, the rootstalk biomass of C. chinensis was increasing continuously. The biomass growth of four-year-old C. chinensis was the fastest in the year. From September to October was the fastest season of the growth of rootstalk. The dynamic accumulation in rootstalk C. chinensis had regularity in the certain extend. The contents of six alkaloids and all alkaloids in 4-year-old C. chinensis were more than that in 3-years-old and 5-year-old. The contents of six alkaloids were mostly highest in August. From July to December, there is no significant difference in the contents of columbamine, epiberberine, coptisine, palmatine, berberine and all alkaloids in 4-years-old C. chinensis.
CONCLUSIONAccording to the biomass and the accumulation pattern of the effective components in the C. chinensis, the optimal harvest time is from September to October of 4-year-old C. chinensis.
