Dietary essential amino acids are extracted in the first-pass metabolism by the small intestine.Besides incorporation into mucosal protein,they are also the fuels for the utilization by small intestinal mucosal cells and the precursors for intestinal synthesis of new amino acids and enormous biological important molecules such as glutathione,polyamines,etc.Thus,their gut metabolism is not only crucial for the maintenance of the integrity and function of intestinal mucosa,but also important for the regulation of whole body metabolism.
2.Quantitation of c-erbB-2 in patients with breast cancer by using fluorogenic quantitative polymerase chain reaction method

AIM: To establish a fluorogenic quantitative polymerase chain reaction (FQ-PCR) method for the routine examination of c-erbB-2 gene expression in breast cancer. METHODS: The c-erbB-2 standard gene was obtained by in vitro amplification of cloned c-erbB-2 fragment in plasmid PGEM-T easy vector. FQ-PCR product was detected by using a 7700 ABI PRISM sequence detector system and c-erbB-2 standard curve was obtained to quantity c-erbB-2 in unknown samples. RESULTS: “S” kinetics curve of FQ-PCR amplification was generated by relating the fluorescence signal intensity (△Rn) to the cycle number. The standard curve of c-erbB-2 was constructed by the linear relationship between the cycle threshold (ct) and the log of starting copy number. The high correlation (0.999) revealed the reliability of FQ-PCR. CONCLUSION: The FQ-PCR is a rapid, sensitive, reliable method for quantity of c-erbB-2 gene expression.
3.Association study of disrupted-in-schizophrenia-1 gene single nucleotide polymorphism with schizophrenia in Han Chinese population

ObjectiveTo analyze the association between single nucleotide polymorphisms (SNPs) in DISC1 gene.MethodsThe studied sample consisted of 528 patients with schizophrenia (264 males and 264 females) and 528 healthy controls (264 males and 264 females).Two function SNPs were selected and gcnotyped by Illumina Golden Gate assay.Genotype and allele frequencies were compared between patients and controls to assess the association to schizophrenia.The clinical features of the hospital first patients were further examined with the Positive and Negative Syndrome Scale (PANSS) before trcatment with antipsychotics.ResultsTwo SNPs were genotyped in subjects.A significant association was found between rs3737597 and schizophrenia in genotype (387:133:8,350:164:14) and allele frequencies (907:149,864:192,P<0.05 ).Moreover,the haplotypes,A-A constructed from two SNPs showed significant differences between cases and controls ( P< 0.05).For rs821616,there were significant difference in positive syndrome score,delusions and poor rapport between patients with genotypes AA or not.ConclusionThis study describes a positive association between DISC(1) and schizophrenia in the Han Chinese population and DISC1 is the susceptible gene in schiz ophrenia.
4.Research progress of additives for improving therapeutic peptides and proteins stability in PLGA microspheres

Microspheres made of poly-lactic-co-glycolic acid (PLGA) have been frequently proposed as drug delivery systems.A very significant challenge in the development of controlled PLGA releasing systems is the instability of drugs especially therapeutic peptides and proteins.Additional approaches,particularly the use of additives,are needed to optimize PLGA delivery of drugs.This article reviews the effects of additives,especially the effects of stabilizing protein during the preparation of PLGA microsphere and the sustained drug releasing processes.
5.Expression of let-7a2 in lung adenocarcinoma

Objective To explore the expression and significance of let-7a2 in lung adenocarcinoma.Methods To adapt technology of realtime PCR with reference of U6, the expression of let-7a2 in tumor tissues and normal tissues was detected in 15 cases of resected lung adenocarcinoma. The laboratorial data were analysed by measure of 2~(-△△CT) method. The analysed data were managed by statistics. Results The expression of let-7a2 in lung adenocarcinoma tissue was significantly lower than that in lung normal tissue (0.8733±0.4821 vs 2.4527±1.0111). This difference possess markedness significance(t=6.816, P<0.001).Conclusion The expression of let-7a2 in lung adenocarcinoma is obviously lower and is concerned with the course of lung cancer evolution.
6.The employment of acute gastrointestinal injury grading system in the strategy of diagnosis and treatment of severe acute pancreatitis in patients for guiding nutritional support

Objective To study the utility of acute gastrointestinal injury (AGI) grading system in the strategy of diagnosis and treatment of severe acute pancreatitis (SAP) in patients for the guidance of nutritional support.Methods Forty-five patients with SAP admitted to the ICU from January 2012 to December 2013 were enrolled for study.The diagnosis of SAP was made as per the 2013 Chinese guideline of dliagnosis and treatment of acute pancreatitis.The patients with aged < 18 years,length of hospital stay <72 h,and those included in other experimental study were excluded.The patients were divided into two groups:AGI group (n =24) and control group (n =21).There were no statistically significant differences in age,gender,APACHE Ⅱ between two groups (P > 0.05).The length of hospital stay,in-hospital mortality,the incidence of complications such as infection,the time of starting enteral nutrition (EN) and reaching caloric goal were compared between the two groups.The t test or Mann-Whitney U test was used for continuous variables and x2 test or Fisher exact test was used for categorical variables,and P < 0.05 was considered statistically significant.Results (1) Between two groups,there were statistically significant differences in the time of starting EN (2.0 d vs.5.7 d,Z =-4.149,P <0.01) and reaching caloric goal (5.5 d vs.10.4 d,Z =-3.392,P =0.001).(2) Between two groups,there were statistically significant differences in the incidence of pneumonia (25% vs.57.14%,P =0.028 1) and catheterrelated infection (0% vs.23.81%,P =0.039 4).(3) Between two groups,there was no statistically significant difference in in-hospital mortality (16.67% vs.33.33%,P =0.194 3).In addition,there was significant difference in length of hospital stay between two groups (23.13 ± 10.58) d vs.(31.10 ± 15.33) d,P =0.046.Conclusions The diagnosis and treatment strategy based on AGI grading system is associated with earlier initiation of EN and reaching caloric goal,and partly reduces the incidence of infections and shortens the length of hospital stay,in the meantime,provides guidance for the rationale of nutritional support in the patients with SAP.
7.Antioxidant activity of an exopolysaccharide from cultivated Cordyceps sisnensis fungus

Aim: To preliminarily investigate the antioxidant activity of the acidic exopolysaccharide(EPS-A) from cultivated Cordyceps sisnensis in vitro and in vivo. Methods: PC12 cells were injured by H_2O_2 after the cells were treated with different doses of EPS-A for 6 h, and then the levels of malondialdelyde( MDA) and superoxide dis-mutase (SOD) in PC12 cells were determined. H22 cells were inoculated into the right oxter of each mouse after ICR mice were treated with EPS-A for 7 days, and tumor-bearing mice were treated with EPS-A for 7 days. MDA level and SOD activity in liver, brain and serum were assayed, and tumor weight of each mouse was measured as well. Results: EPS-A significantly decreased the production of MDA and increased SOD activity in H_2O_2-injured PC12 cells. Moreover, EPS-A inhibited H22 tumor growth in mice, in which the level of MDA in liver and brain was reduced, and SOD activity in liver, brain and serum was enhanced. Conclusion: EPS-A exhibited antioxidant activity in cell cultures and mice.

Object To extract and purify a novel fibrinolytic enzyme from Perinereis aibuhitensis Grube Methods The enzyme was precipitated from the extract by ammonium sulfate, dialyzed, chromatographed on Sephadex G 100 column, and then purified by polyacrylamide gel electrophoresis Its fibrinolytic activity was assessed with fibrin plate method Results The purified enzyme showed an isoelectric point (PI) around 4 5 as tested by gel isoelectric focusing It consisted of two polypeptide chains with molecular weights around 33 000 u and 14 400 u, respectively Conclusion This was a novel fibrinolytic enzyme discovered from P aibuhitensis for the first time
9.Clinical Studies on Effect of Allergic Rhinitis Oral Liquid on Perennial Allergic Rhinitis

[Objective] To observe the effect of Allergic Rhinitis Oral Liquid (AROL) on perennial allergic rhinitis (PAR) and to explore its therapeutic mechanism. AROL, composed of modified Guizhi Tang and Yupingfeng San, has the actions of strengthening spleen and lung, replenishing Qi and consolidating superficies and dispelling pathogens and dredging orifices. [Methods] One hundred and twenty-nine cases of confirmed PAR were randomized into Group A (n=43), Group B (n=43) and Group C (n=43). Group A was treated with AROL, Group B with Biyankang Tablets and Group C with normal saline. Three groups were treated for 4 weeks as one treatment course, and another treatment course was carried out one week after interval. 10.Research on the antitumor actions of extracts from the fruiting body of coriolus versicolor

Objective: To investigate the antitumor actions of polysaccharide extracts from the fruiting body of coriolus versicolor (CVE). Methods:Hepatoma HepA cells were injected into mice subcutaneously. Different doses of CVE were given by gavage. On the 7 th and 14 th day, tumor inhibitive rates were calculated. ELISA was performed to measure the serum IgG level; MTT was used to examine CVE′s effects on the proliferation of T lymphocytes of thymus. Immunohistochemistry was used to determine CVE′s influence on the expression of tumor related genes P53 and VEGF in liver. Results: CVE may evidently inhibit the growth of the transplanted HepA tumors. Its effects on the serum IgG level and on the proliferation of T lymphocytes of thymus were also significantly. Also, CVE markedly decreased the expression of P53, VEGF genes in liver. Conclustion: CVE had significant antitumor effects in vivo . The mechanisms may involve immune modulation effects and antimetastasis actions.
