1.The expression and clinical signiifcance of microRNA-192 and ZEB2 in colorectal cancer

Background and purpose: MicroRNA(miRNA) is a class of small non-coding RNA playing an important regulatory role in many diseases. In this study, we explore the levels of miR-192 and zinc ifnger E-box binding homeobox 2 (ZEB2) in CRC, the clinical signiifcance of miR-192 and the correlation between the expression of miR-192 and ZEB2 protein. Methods: The expression levels of miR-192 and ZEB2 mRNA in 30 colorectal carcinoma samples, 30 corresponding cancer-adjacent tissue samples (from the edge of tumor≥5 cm), 25 colorectal adenoma samples, and 15 normal tissue samples were quantiifed using quantitative real-time polymerase chain reaction (qRT-PCR). ZEB2 protein expression was determined using Western blot. The relationship between the miR-192 and clinicopathological factors, miR-192 and ZEB2 protein expression was analyzed. Results:Signiifcant upregulation of miR-192 expression and reduction of ZEB2 mRNA and protein expression were identiifed in CRC tissues, compared to cancer-adjacent tissues, colorectal adenoma samples, and normal tissues (P<0.05). Low miR-192 levels were signiifcantly associated with lymph node (P=0.021) and distant metastasis (P=0.023). An inverse relationship between miR-192 and ZEB2 protein expression was identified in CRC group (r=-0.365, P<0.05). Conclusion: MiR-192 downexpression was correlated with CRC metastasis. MiR-192 may play a role in the development and progression of CRC through ZEB2.
2.Effect of dexamethasone combined with oridonin on proliferation and apoptosis of multiple myeloma cell line U266

Objective To investigate the effect of dexamethasone combined with oridonin on proliferation and apoptosis in multiple myeloma cells U266 and the related molecular mechanism. Methods Exponential phase of growth U266 cells were treated with different concentrations of oridonin combined with dexamethasone or alone. U266 cells treated by DMSO were taken as control group. The proliferation inhibitory ratios were measured by CCK-8 assay followed by 24 h, 48 h and 72 h. Apoptosis induction was assessed by using Annexin V-FITC kit. Real time PCR was used to examine the mRNA changes of Notch1, NF-κB/p65 and bcl-2. Western blot assay was applied to detect the protein expression of Notch1, cleaved Notch1, NF-κB/p65 and bcl-2. Results Compared with that in control group, proliferation in all the experimental groups was inhibited (P<0.05), and the apoptosis was promoted (P<0.05); especially the combination of dexamethasone and oridonin had a synergistic effect on the proliferation and apoptosis of U266 cells (P<0.05). The results of PCR and Western blot showed that after treatment of U266 cells with dexamethasone, the mRNA as well as their protein levels of NF-κB/p65 and bcl-2 were decreased compared with those in the control group (P<0.05). Moreover, the mRNA and protein expression of Notch1, cleaved Notch1, NF-κB/p65 and bcl-2 was obviously down-regulated in oridonin group and the combination group (P<0.05). Conclusion Combination of dexamethasone and oridonin can significantly increase the anti-tumor effect by inhibiting proliferation and inducing apoptosis of U266 cells, which may be related to the inhibition of the Notch1 pathway.
3.Imaging Characteristics and Diagnostic Value of Primary Ureter Carcinoma

Objective To explore the imaging characteristics and the diagnostic value of primary ureter cancer.Methods 24 cases of primary ureter carcinoma underwent ultrasound(US), intravenous pyelography(IVP),retrograde pyelography and CT scan.Those imaging features were analyzed retrospectively.Results The postive rate of US,IVP,retrograde pyelography and CT was 45.8%(11/24),37.5%(9/24),88.8%(16/18)and 62.5%(10/16).Conclusion Ultrasound(US) is a more value examination for making the choice of cases; IVP is an essential method for understood function of the kidneys, showing carcinoma; retrograde pyelography is most value in the diagnosis of primary ureter carcinoma;CT scan is more important diagnostic values in area,properties and extent of tumor.

6.Predictors of chronic post-thoracotomy pain in rats

Objective A rat model of chronic post-thoracotomy pain is used to study whether acute pain and pre-operative diffused noxious inhibitory controls(DNIC) can predict chronic pain and how DNIC changes when pain maintains.Methods Rats were randomly divided into three groups:naive group,sham group and model group.DNIC was constantly assessed in individual rats,along with each animal's mechanical hyperalgesia and cold allodynia after thoracotomy.Results In model group,the incidence of chronic post-thoracotomy pain was 55%(11 of 20),which was named CPTP group,and the other 9 rats without chronic pain was defined as non-CPTP group.The pre-operative DNIC was significantly weaker in CPTP group with lower mechanical threshold on 6 days after surgery and higher cold sensitivity on 6 days after surgery comparing with non-CPTP group.In the acute pain phase (day 3),DNIC was decreased in both CPTP group and non-CPTP group as compared with pre-operative period.Besides,DNIC was recovered in non-CPTP group while kept impaired in CPTP group on 21 days after surgery.Conclusions Pre-operatively assessed DNIC efficiency and acute post-operative pain intensity were two independent predictors for CPTP.DNIC was decreased both in acute pain and chronic state,while returned to normal when pain sense was normal.
7.Study on UPLC Fingerprint of Glycyrrhiza uralensis

OBJECTIVE:To establish UPLC fingerprint of Glycyrrhiza uralensis. METHODS:UPLC method was adopted. The determination was performed on Waters CORTECS UPLC C18column with mobile phase consisted of acetonitrile-0.1% formic acid(gradient elution)at the flow rate of 0.3 mL/min. The detection wavelength was set at 254 nm,and column temperature was 30 ℃. The sample size was 1 μ L. Using glycyrrhizic acid as control,UPLC chromatograms of 27 batches of sample were determined. Similarity evaluation was conducted by using TCM Chromatogram Fingerprint Similarity Evaluation System(2004 A edition)to determine common peak and conduct cluster analysis of 27 batches of samples. RESULTS:There were 20 common peaks in UPLC chromatograms of 27 batches of samples. The similarity degree of S2,S4,S19,S21,S22,S24 were less than 0.90, the others samples were more than 0.90.After validation,UPLC chromatograms of 21 batches of batches of samples were in good agreement with control fingerprint. 27 batches of samples were clustered into 3 categories,in which S24 was categoryⅠ;S2,S4, S12,S19,S21,S22 were categoryⅡ;other were categoryⅢ. CONCLUSIONS:Established fingerprint can provide reference for quality evaluation of G.uralensis.
8.Scoliosis and associated factors among middle school students in Guangzhou City

Objective:
To investigate the general situation of scoliosis and influencing factors among middle school students in Guangzhou, so as to provide evidence for behavioral intervention measures.
Methods:
By stratified cluster random sampling method, 2 121 students from 8 middle schools of Guangzhou were selected to conduct questionnaire survey and physical examination.
Results:
The detection rate of scoliosis among middle school students in Guangzhou was 8.20%, girls (9.50%)>boys (7.00%), urban area (10.45%)>suburb area (4.77%), senior high school (10.08%)>junior school (6.39%) ( P <0.05). Among the most of the 174 positive students were found to have large thoracic curve, right lateral bending, being girls with medium scoliosis. Multivariate Logistic regression analysis showed that students in urban area ( OR=2.56, 95%CI =1.71-3.82), with mobile electronic devices usage time≥3 h/d( OR=1.59, 95%CI =1.12-2.27), prolonged near vision work ≥1 h ( OR=1.40, 95%CI =1.00-1.95), outdoor activity time<2 h/d( OR=1.82, 95%CI =1.24-2.67) had a higher detection rate of scoliosis ( P <0.05).
Conclusion
The detection rate of scoliosis among middle school students in Guangzhou is much higher, which might be related to district, mobile electronic devices usage, prolonged near vision work and insufficient outdoor activity. Health education regarding spinal knowledge should be strengthened.
9.Differences in the detection rates of depression and anxiety among middle school students in different time periods

Objective:
To investigates the prevalence of depression and anxiety among middle school students at different times, and to provide a reference for mental health promotion among adolescents.
Methods:
A total of 1 505 middle school students, selected from Chongqing in 2021 by using multi stage stratified random sampling method, were surveyed by using the Center for Epidemiologic Studies Depression Scale(CSE-D) and Zung Self Rating Anxiety Scale (SAS).
Results:
At the beginning of the semester, 434(27.6%) and 601 students( 38.2 %) reported depressive and anxiety symptoms,respectively. While at the end of the semester showed that 463 cases (30.8%) were prone to depression, and 653 cases ( 43.4 %) reported depressive and anxiety symptoms,respectively. The detection rate of depressive symptoms at the end of the semester was significantly higher than that at the beginning of semester. (43.4%, 38.2 %, χ 2=8.55, P <0.05). Depressive of female students at the end of the semester( 52.8 %) was higher than that at the beginning of the semester(46.3%)( χ 2=6.19, P <0.05) The detection rate of anxiety symptoms in senior high school students at the end of the semester(44.9%) was significantly higher than that of at the beginning of the semester(28.6%) ( χ 2= 43.33 , P <0.01). No significant difference in anxiety symptoms boys and junior high school students between the beginning and the end of the semester ( χ 2=2.34, 0.71, P >0.05).
Conclusion
Substantial changes are observed in anxiety symptoms among female middle school students and depressive and anxiety symptoms among high school students in different periods of a semester. When investigating depression and anxiety of middle school students, time effect should be considered.
10. Advances in Research on Regulatory Mechanism of Abnormal MiRNA Expression in Digestive System Diseases

MicroRNA (miRNA) is a non-coding single strand endogenous RNA, with a length of 20-25 bp. It regulates the transcription and translation of target mRNA by binding to its 3' untranslated regions (UTR). Abnormal expression of miRNA plays an important role in the occurrence and development of digestive system diseases. Clarifying the regulatory mechanism of miRNA will be of great help for early diagnosis and treatment of diseases. This article reviewed the advances in research on regulatory mechanism of abnormal miRNA expression in digestive system diseases.
