1.Intravascular ultrasound assessment of the causes of coronary angiographic hazy lesions

Objective To identify the causes of coronary angiographic hazy lesions by intravascular ultrasound (IVUS) to avoid inappropriate stenting. Methods Twenty-five cases with hazy regions on coronary angiogram were consecutively identified from July 2009 to March 2010. Hazy regions were defined by coronary arteriongraphy as reduced contrast density without a clearly defined intimal tear, dissection,thrombus,or stenosis ( > 50% ). This cohort of patients were subsequently underwent IVUS examinations and treated according to the results of IVUS. Results The lumen CSAs were settled as > 4. 0 mm2 in all examinations. Among all 25 cases,hazy lesions were located in left anterior descending in 12 patients, right coronary artery in 6 patients, left circumflex in 5 patients, and left main artery in 2 patients. According to the IVUS findings, 2 cases showed absolutely normal or near-normal arterial wall structure image, 10 cases showed calcified plaque,5 cases showed plaque rupture,3 cases showed eccentric plaque ,2 cases showed thrombosis formation,2cases showed dissection,1 case showed subintimal hematoma. Seven patients received stent implantation, and the rest accepted medical therapy. There were no in-hospital MACEs reported among all patients. Conclusion Nearly half of the coronary arteriongraphic hazy lesions were caused by calcified plaque. IVUS can distinguish calcified plaques from intimal tears, thrombus and other underlying etiologies,and help to avoid unnecessary stenting.

OBJECTIVE:To establish a method for the bacterial endotoxin test of ~(131)Ⅰ-epidepride injection.METHO_ DS:After dilution with 10 parts of water,~(131)Ⅰ-epidepride injection was detected with a method of tachypleus amebocyte lysa_te(TAL) and subjected to verification test.RESULTS:The interference of the dilute ~(131)Ⅰ-epidepride injection on TAL can be eliminated after dilution,the bacterial endotoxin levels for 3 batches of 131Ⅰ-epidepride injection were all below 50EU/ml.CONCLUSION:The method for the bacterial endotoxin test of ~(131)Ⅰ-epidepride injection is feasible.
5.SYMPATHETIC PREGANGLIONIC NEURONS IN THE RABBIT SPINAL CORD PROJECTING TO THE SUPERIOR CERVICAL GANGLION——HRP STUDY

The purpose of this research work is to find out the longitudinal distribution of the preganglionic neurons which project to the superior cervical sympathetic ganglion (SCSG). Experiments were performed on 12 adult rabbits and a monkey. Under sodium pentobarbital anesthesia, 20 microliters of 10% HRP were injected slowly into the rabbit's SCSG. In the monkey, 20 microliters of 15% HRP was injected.After a postoperative survival time of 3~6 days, the animals were perfused through,the ascending aorta with a cold fixative mixture composed of 2% paraformaldehyde and 1.25% glutaraldyhyde in 0.1mol phosphatebuffer at pH 7.4. The spinal cord segments C_1~L_3 were cut serially in transverse plane on a cryostat at 48 micra.The HRP reaction product was demonstrated according to Mesulam's (1976) benzidine blue reaction method, and counterstained with neutral red.HRP labeled neurons in the spinal cord were located exclusively on the side ipsilateral to the injected SCSG. The total number of labeled cells were 7908 in 12 rabbits, but the number of labeled cells varied from animal to animal. The highest amount was 1690 (423~#) and the lowest amount was 71 (425~#). The longitudinal distribution of the labeled cells in 12 rabbits was 12 segaments of the spinal cord (C_6~T_9), but the largest proportion (86.18%) of them was concentrated from T_1 to T_4, especially at the level of T_2 and T_3 (56.22%), and with a peak at T_2 (29.10%).In cross section of the spinal cord. HRP-labeled cells were concentrated in four cell groups, they are: nucleus intermediolateralis pars principalis (ILp), nucleus intermediolateralis pars funicularis (ILf), nucleus intercalatus (IC) and nucleus intercalatus pars paraependymalis (ICpe). The latter is subdivided into dorsal portion and ventral portion. HRP positive cells were mainly located in the ILp, In 12 rabbits about 92.99% cells were located in it. A small portion of labeled cells(6.25%) were seen in the ILf. A few labeled neurons could be detected within,the IC (0.68%) and ICpe (0.08%). Furthermore, occasionally, very few labeled cells were found at the dorsol portion of the anterior horn.In the monkey, generally speaking, the pattern of the distribution of labeled cells was the same as the rabbit.
6.Expression and significance of P-JNK and mutlidrug resistance protein in gastric cancer

Objective To investigate the clinicopathogenical significance of phosphorylation type of c-jun N-terminal kinase(p-JNK)and muhidmg resistance protein P-glycopretein(P-gP),multidrug resistance proteinl(MRP1)and lung resistance protein(LRP)in gastric cancer. Methods The expression of p-JNK,P-gP,MRP1 and LRP was detected in a tissue microarray containing 168 spots of gastric cancer tissue and 27 spots of normal gastric tissue by immunohistochemistry.Results The positive expression rate of p-JNK,P-gp,MRP1 and LRP in gastric cancer wag 45.8%,51.8%,45.8%and 55.4%respectively,which Was significantly higher than that in normal gastric tissue(P<0.05).The p-JNK expression correlated with depth of invasion(P<0.01),histology grade(P<0.05),vessel invasion (P<0.01),lymph node metastasis and distant metastasis(P<0.01).The expression of p-JNK,P-gp and MRP1 in a positive relationship(P<0.05).Kaplan-Meier analysis revealed a significant impact on survival by p-JNK,P-gp and MRPl(P<0.05). Conclusion The p-JNK expression in gastric cancer is correlated with malignant biological behavior and may be involved in the chemotherapeutic resistance by upregulating the expression of P-g[ and MRP1.

8.Preparation of Electrochemical Microfluidic Device Based onPulse Driving and Controlling of Microfluids Technique

The fabrication system for the electrochemical microfluidic device was set up based on the pulse driving and controlling of microfluids technology.The nano silver ink and glycerol solution were jetted on the glass substrates to form the microelectrode pattern and the liquid mold pattern for the microchannel.Then the microelectrode and microchannel were obtained through a sintering process and a molding process, respectively.The electrochemical mircrofluidic device was Finally prepared through a bonding process with the microelectrode and the microchannel.The influences of the system parameters on the formation of the droplet were studied, as well as the influences of the droplets diameter and the overlap on the formation of the liquid lines.The minimal width, the thickness and the resistance of the prepared microelectrode were 45 μm, 2.2 μm and 5.2 μΩ cm, respectively.The minimal width of the microelectrode was 35 μm and the surface was smooth.The electrochemical flow detection of glucose concentration was carried out with the device, and the results showed that the glucose concentration had a high linear correlation with the response current, which could be used in the quantitative detection of glucose concentration.The fabrication of the electrochemical microfluidic device based on the pulse driving and controlling of micro fluids technology has many advantages such as simple system structure, lower cost and higher accuracy of the micro droplet and can be used in the preparation of the devices in the biochemical analysis and biosensor areas.
9.Correlative cell signaling pathway for expression of heme oxygenase-1 induced by ginkgo biloba extract in rat vascular smooth muscle cells

AIM: To explore the effects of heme oxygenase-1(HO-1) protein expression induced by ginkgo biloba extract(EGB761) in rat vascular smooth muscle cells(RVSMC) and the correlative cell signaling pathway.METHODS: The RVSMC lines were revived.Serial passage to 6 generation was carried out and divided into different groups.The cells were treated respectively with vehicle,purely EGB761,EGB761 plus zinc protoporphyrin IX or other specific inhibitors of cell signaling pathway.Western blotting method was used to detect the expression of HO-1 in RVSMC.RESULTS: EGB761 induced HO-1 protein expression in a dose dependent manner.ZnPPⅨ and genitein significantly inhibited HO-1 protein expression induced by EGB761(0.10?0.01,0.07?0.01 vs 0.61?0.07,P0.05,respectively).CONCLUSION:(1) EGB761 significantly induces HO-1 protein expression in RVSMC,and the effect can be inhibited by a specific HO inhibitor ZnPPⅨ.(2) The HO-1 protein expression induced by EGB761 in RVSMC is mediated by tyrosine protein kinase pathway.
10.Effect of polysaccharide ingredients from TCM complex prescription on releasing cytokines of mouse peritoneal macrophages

Objective: To observe the effect of polysaccharide ingredients from six TCM complex prescriptions on releasing the cytokine(CK) level of mouse celiac macrophages (M?), to explore the possible mechanism and direct the extractionand separation of active components for above compounds. Methods: The total polysaccharide ingredients of six complex prescriptions were prepared: Siwu Decoction, Sijunzi Decoction, Liuweidihuang Decoction, Guizhi Decoction, Longdanxiegan Decoction and Yupingfeng Pulveres, then the mouse celiac M? were incubated together in 96 holes board.
