Objective:To explore the effects and mechanisms of insulin postconditioning on myocardial protection in isolated rat hearts.Methods:32 Wistar rats were randomly divided intocontrol group,ischemia/reperfusion group(I/R group),low dose insulin postconditioning group(L-POST group)and high dose insulin postconditioning group(H-POST group),with 8 rats in each group.Langendorff and neely perfusion models were steadily established,then the rat hearts were arrested by cardioplegia(St.Thomas No.2 solution,STH-2)after perfusion for 30 min.These arrested hearts were remained for 30 min at 4℃and then underwent reperfusion for 60 min.In L-POST group and H-POST group,the arrested hearts were reperfused by Krebs-Henseleit buffer solution(K-H)with insulin.The content changes of myocardial eNOS and PKC were detected by immunohistochemistry and Western blo(tWB),the AI(Apoptosis index,AI)of cardiomyocyte was detected by TUNEL.Results:Compared with control group,higher expression of eNOS and PKC and higher AI were detected in I/R group、L-POST group and H-POST group (P

The effects of ranitidine (Ran) on cultured myocardial cells of neonatal rats were observed. Ran 100 ?mol?L-1 took no effect on spontaneous beating rates of cultured myocardial cells, but it decreased the beating rates increased by histamine 10 ?mol?L-1 and did not inhibit the beating rates increased by isoprenaline 2?mol?L-1. Histamine 10?mol?L-1 increased the amplitude, maximal rate of rise and pro-longed action potential duration at 50% (APD50) and 90% (APD90) repolarization. The effect was abolished by Ran 100?mol?L-1. The results suggested that Ran may possess antiarrhythmic action induced by histamine's increase.

Objective To discuss the applied value of multislice spiral CT(MSCT) in diagnosing and localizing the embedded supernumerary teeth. Methods A total of 43 cases with the embedded supernumerary teeth were undergone MSCT using shaded surface display(SSD), minimum intensity projection(MIP) and multiplanar reformation(MPR) to present the form and location of the embedded supernumerary teeth and to determine the surgical style.The result of image was retrospectively analysed after operation.Results The reconstruction images,could clearly show the dental surface image including crown, root, neck and root bifurcation, labial or palatal location, eruption orientation and relation with dentition in anterior maxilla. The position of 52 embedded supernumerary teeth in 43 patients were completely confirmed by surgery with accuracy of 100%. All patients were well healed with no infection and injury to nearby teeth. Conclusion The Embedded supernumerary teeth in bone can be detected by MSCT and MSCT can be used as an important examination technique before surgical and orthodontic treatment.

AIM To study the relation between the antiarrthymic effect and the inhibition of slow reaction cardiomyocyte of ethacizine(EC). METHODS The beating rate and action potentials of cultured slow response cardiomyocyte from neonatal rats was recorded. RESULTS EC 0 1～5 0 ?mol?L -1 produced a concentration dependent suppressing effect on spontaneous beating rate of cultured cardiac cells with IC 50 of 0 17 ?mol?L -1 and the maximal inhibitory rate was 52%.The steady state of inhibition to beating rate was achieved after adding EC for 15～30 min. The effective inhibition of beating rate by EC lasted for over 1 h.As EC 2 5 ?mol?L -1 depressed beating rate to the steady state ,effects of CaCl 2 2 mmol?L -1 and Iso 1 ?mol?L -1 on beating rate were partially reduced,and the effect of aconitine 0 2 mg?L -1 was abolished. EC 0 3～2 5 ?mol?L -1 pruduced the concentration dependent depression on APA,V max ,OS,SP 4 and MDP,and prolongation of SCL of the slow response action potentials.APD 90 was also lengthened at the concentration of EC 0 9～2 5 ?mol?L -1 . CONCLUSIONS The effective depression of EC on abnormal automaticity and conductance of the slow response cardiomyocyte in heart ischemic zone may be an important mechanism for its effective termination on some arrhythimas.EC may depress Ca 2+ ,especially Na + currents on slow response cardiomyocyte.

Objective To investigate the clinical value of radioactive seed 125Ⅰ implantation combined with gemcitabine(GEM)in treatment for peripheral non-small cell lung cancer(NSCLC).Methods 60 patients(male 35,female 27,mean age 73.3)with peripheral lung carcinoma were confirmed under CT-guided biopsy including 8 bronchoalveolar caicinomas and 17 squamous carcinomas,6 in the stage I,14 in the stage II,34 in the stage III and 6 in the stage IV were designated under clinical staging.All patients were devided into two groups as GEM group and GEM-125Ⅰ group.GEM group underwent chemotherapy with GEM only and GEM-125Ⅰ group was treated under CT guided radioactive seed 125Ⅰ implantation combined with gemcitabine.Results GEM group showed tumors under controlling as PR in 5,SD in 14,PD in 11 cases;the total effective rate was 16% and the total control rate was 63%.The median survival time was 7 months and one years survival rate was 26%.GEM-125Ⅰ group revealed CR in 5,PR in 11,SD in 8,PD in 6 cases and the total effective rate reached 36% with total control rate as 80%.The median survival time was 12.3 months and one year survival rate was 50%.There was a significant difference(P

Objective To explore the expression and clinical significance of constitutive androstane receptor(CAR)in placenta syntrophoblast from patients with intrahepatic cholestasis of pregnancy(ICP).Methods Placenta were collected from women with ICP who delivered from April 2009 to March 2010 in First Affiliated Hospital of Chongqing Medical University.According to the severity of ICP,patients were classified into mild ICP group(n=10)and severe ICP group(n=10).Ten healthy pregnant women who delivered in the same period were chosen as control group.The location of CAR protein in placenta was studied by immunohistochemical streptavidin-biotin complex(SABC)method.CAR mRNA level was determined by reverse transcription(RT)-PCR technique and CAR protein expression level was determined by western blot.Results(1)CAR was located in the placenta syncytiotrophoblastic cells in control group and mild ICP group,showed light tan when stained,and was mainly in the cytoplasm.In severe ICP group,CAR was also located in placenta syncytiotrophoblastic cells but mainly in the nucleolus,showed dark tan when stained.(2)The mRNA expressions of CAR in control group,mild ICP group,severe ICP group were 0.06 ±0.03,0.07 ±0.03 and 0.56±0.03.respectively.CAR in severe ICP group was significantly higher than those in control group and mild ICP group(P＜0.05).The difference of mRNA between control group and mild ICP group wag not statistically significant(P＞0.05).(3)The CAR protein levels in control group,mild ICP group,severe ICP group were 0.74±0.03,0.79±01 03 and 1.02±0.04,respectively.CAR protein expression in the severe ICP group was significantly higher than the other two groups(P＜0.05).And there was no statistical significance between mild group and control group(P＞0.05).Conclusion In ICP women.especially severe ICP patients,the CAR expression in placenta syncytiotrophoblastic cells increased appreciably,which may be involved in the maintenance of placenta barrier function and protection in ICP pathogenesis.

Objective: To investigate the application value of diffusion kurtosis imaging (DKI) for evaluating brain parenchyma microstructures changes in hypertensive patients with normal blood pressure control. Methods: Totally 22 patients with primary hypertension with blood pressure controlled in normal range were enrolled in observation group, while 19 healthy volunteers were selected as control group. DKI was performed in all subjects, and the mean kurtosis (MK), kurtosis anisotropy (KA), axial kurtosis (AK) and radial kurtosis (RK) of each brain area were measured and compared between the two groups. Results: MK values in right frontal lobe, right inner capsule knee and right cerebellum of observation group were lower than that of control group, whereas in right parietal lobe was higher than in control group(all P<0.05). The pontine KA value in observation group was lower than that in control group (P<0.05). AK values of left parietal lobe and right pallidum of observation group were lower than that in control group, of left red nucleus, right cerebellum and pontine were higher than that in control group (all P<0.05). The values of RK in right inner capsule knee, right cerebellum and left occipital lobe in observation group were lower than that in control group, and of right putamen was higher than that in control group (all P<0.05). Conclusion: DKI can be used to monitor changes of brain parenchyma microstructures in hypertensive patients with normal blood pressure control.

The scrota and testes of the rabbits were exposed to 2450 mHz microwave field and the temperature of scrotal skin was raised to 41-42℃ for 20 minutes. Ultrastructural observations on the peritubular tissue of ductus epididymides were studied at different intervals from 30 minutes to 4 months after the treatment, and alkaline phosphatase cytochemical changes were observed at intervals from 12 hours to 30 days. Pronounced morphological alterations of the smooth muscle cells and endothelial cells of capillaries were detected within 30 minutes to I hour following exposure. After 24 hours to 3 days, the thickening of the peritubular tissue were noted. The thickening and infolding of the proper lamina were also prominent. The degenerative smooth muscle cells generally returned to normal appearance after 3-4 months, but the thickening of the peritubular tissue and infolding of the proper lamina still appeared. Alkaline phosphatase was localized in the basal lamina, proper lamina, the innermost smooth muscle cells cytoplasm and surface vesicles. There were no obvious alterations of alkaline phosphatase activity after 12 hours. After 15 days, the alkaline phosphatase prominently decreased, while after 30 days the alkaline phosphatase increased nearly to normal.
