1.Pharmacological Target of Spleen-Strengthening and Qi-Invigorating Herbal Medicine on Small Intestinal Crypt-like Cells

Achievements of pharmacological studies on IEC-6 small intestinal crypt-like cells (IEC-6 cells) were reviewed. Based on the physiological research of IEC-6 cells, the pharmacological experiment model of IEC-6 cells was established and the pharmacological actions of spleen-strengthening and Qi-invigorating herbal medicine (SQHM) on intestinal epithelial stem cells were observed. Thiazolyl blue colorimetry (MTF), reverse transcription polymerase chain reaction (RT-PCR), enzyme-linked immunosorbent assay (ELISA), high performance liquid chromatography (HPEC) and immune fluorescence labeling technology were applied in the experiments to observe the effect of SQHM on the proliferation, migration and differentiation of IEC-6. Its influences on the expression of villus protein, divalent mental transferrin, interleukin-6 (IL-6) and ornithine decarboxylase (ODC) and on ODC activity and putrescine content were also observed. The results showed that the extracted chemical components of Radix Codonopsis, Rhizoma Atractylodis Macrocephalae, Radix Astragali seu Hedysari and Radix Glycyrrhizae promoted or inhibited the proliferation, migration, differentiation and absorption function of IEC-6 cells by regulating ODC activity and putrescine production. It is indicated that IEC-6 cells are the pharmacological target of spleen-strengthening and Qi-invigorating herbal medicine.
2.ANTAGONISTIC EFFECT OF CR1409 ON CHOLECYSTOKININ-INDUCED INCREASE IN CYTOSOLIC CALCIUM CONCENTRATION

The glutaramic acid derivative cholecystokinin ( CCK ) receptor antagonist, CR1409 was tested for its ability to inhibit CCK induced increase in cytosolic calcium concentration in isolated rat pancreatic acinar cells using fura-2 as calcium probe. Preaddition of 10?mol/L CR1409 completely inhibited the increase of cytosolic calcium concertration induced with 1 nmol/L CCK, but had no antagonistic effect on carbacholine bombesin-induced increase in cytosolic calcium concentration. The half maximal inhibition was obtained for 0.37?mol/L, corresponding to a potency a hundred fold higher than that of db cGMP. In the presence of increasing concentrations of CR1409, the response curves of CCK concentrations were shifted to the right but the same maximum was achieved. Schild representation gave a straight line with a slope close to one ( n = 0.92 ) and pA2 value of 6.93. These findings therefore support that CR1409 is a powerful competitive and specific antagonist bf CCK-induced increase in cytosolic calcium Concentration in isolated rat pancreatic acinar cells and provide further evidence for the role of this ion as an intracellular mediator of pancreatic CCK receptor.
3.Rapid Method for Determination of Coliform Bacteria in Water

ve To explore the feasibility of the application of rapid method of LTSE in determination of coliform bacteria in water. Methods The 205 water samples were determined for coliform bacteria by rapid method of LTSE, and at the same time by routine method ruled by National Standard Regulation as control. Results The re-sults obtained from rapid LTSE accorded with those from routine method on the whole, and the detectable rates of these two methods showed a consistent rate of 96.6% . Conclusion Rapid method of LTSE could be applied to de-termination of coliform bacteria.
4.Effects of Astragali Injection and extracts from Atractylodes macrocephala on IEC-6 cell migration

Methods IEC 6 cells were inoculated in 6 well microplates and injured in 72 h, and added with Hi, B1, B4, B9, B13. Then D PBS to negative control in the same amount and EGF to positived control, the cell migration was observed in 24, 48 and 72 h after treatment. Methods:492 community peopl e were sampled from community and tested by Generic Quality of L if e Inventory-74.Results:(1) Generic quality of life of comm unity people was well, and their body and mind were in good health; (2) Effects of mental health、physic health and social function on quality life were over ef fect of material life conditions;(3) Qualities of life of elders、workers、non-p rofession people were worse than qualities of young、profession people and cadre (P<0.01).Conclusion:Quality of life of community peop le of Hangzhou was in good condition, but attentions should be paid to qualities of life of elders and workers.
6.Influence of different modes of teaching presentations on the education of medical neurobiology for the college students of clinical medicine

The expression mode in presentations may affect the efficiency during the teaching. In this study, we used a random-controlled trial to compare the educational efficiency in three different expression modes: script, pictures, animation. Test scores and questionnaire survey were statistically analyzed. It showed that the presentations with animation led to the highest average test score of the students. Visual objects could attract students' interest, enhance memory, and academic achievement. A proper insert of visual objects in teaching presentation is encouraged in the education of medical neurobiology for those medical students.
7.Applicotion of intraoperative Irrigation of Colon in Traumatic Rupture of Left Hemicolon

Objective To study the effect of intraoperative irrigation in management of traumatic rupture of left hemicolon.Methods 36 traumatic rupture of left hemicolon patients in our hospital were randomly divided into treatment group and control group.The treatment group received primary reparation and intraoperative irrigation of colon,and the control group received colostomy and repair.The therapeutic efficacy were compared between the two groups.Results There were no significant differences in complication rates and perioperative mortality between two groups( P > 0.05 ),and the control group's the average hospitalization day and average hospitalization charge are more than the treatment group( P < 0.01 ).Conclusion Intraoperative irrigation of colon and primary repair is a safe and feasible method for the traumatic rupture of left hemicolon.
8.Effect on growth of human hepatoma cells by antisense gene of type Ⅱ insulin-like growth factor receptor transfection

Objective To study the growth inhibition of human hepatoma cells tranfected with type Ⅱ insulin like growth factor receptor (IGF ⅡR) antisense gene. Methods We constructed the recombinant eucaryotic expression plasmids of IGF ⅡR sense and antisense genes, which were transfected into SMMC 7721 human hepatoma cell line using calcium phosphate coprecipitation, and selected in DMEM supplemented with 500 ?g/ml G418(Geneticin) for 2~3 weeks. Then the total number of colonies formed was counted. The effect of IGF ⅡR antisense gene transfection on endogenous IGF ⅡR mRNA levels was examined by Northern blot, and the growth rate and the ability of the transfected cells to form colonies in soft agar medium were also examined. Results The hepatoma cells transfected with IGF ⅡR antisense gene exhibited significant reduction in endogenous IGF ⅡR messenger RNA(mRNA) levels and loss of their ability for anchorage independent growth in soft agar. The IGF ⅡR antisense gene supressed the formation of colonies of SMMC 7721 cells resistant to G418 without alteration on cell growth curve. Conclusion The IGF ⅡR antisense gene expression effectively blocks IGF Ⅱ to IGF ⅡR autocrine and/or paracrine signal transduction pathway and reverses certain aspects of the cell malignant phenotype.

【Objective】To analyze the structure of N-linked glycans in salivary glycoproteins and amylase of chronic gastritis(CG)patients with spleen deficiency after stimulation of acid,and to explore the relationship of N-linked glycans structure with salivary amylase activity and splenic hypofunction.【Methods】Iodine-spectrophotometry was used to detect the amylase activity in 47 CG patients with spleen deficiency,15 CG patients with dampness-heat in the spleen and stomach,and 12 healthy volunteers before and after acid stimulation.Method of concanacalin A(ConA) affinity chromatography was used to separate total salivary glycoproteins and amylase and to analyze N-linked glycans structure in 27 CG patients with spleen deficiency,14 CG patients with dampnessheat in the spleen and stomach,and 5 healthy volunteers after acid stimulation.ConA affinity reversed-phase two-dimension chromatography was applied to separate total salivary glycoproteins and to analyze the constitute and distribution of total salivary glycoproteins with different N-linked glycans in 13 CG patients with spleen deficiency,6 CG patients with dampness-heat in the spleen and stomach,and 5 healthy volunteers after acid stimulation.【Results】After acid stimulation,salivary amylase activity in CG with spleen deficiency decreased as compared with that before acid stimulation(P
10.Extraction and Purification of Total Flavonoids in Radix Glycyrrhizae

Objective To study the method of extraction,purification,and analysis of total flavonoids of Radix Glycyrrhizae(TFG).Methods The extracting effects of three different ways(ultrasonic wave,heating recirculation,and maceration)for TFG were compared.TFG was refined with resin absorption and the effects of different parameters on yield and purity were observed.Naringin scanning method was used to analyze TFG.Results TFG extracting percentage is 5.46 %by ultrasonic wave method,6.12 %by heating recirculation,and 5.04 %by maceration,the effect of heating recirculation being the best.In the test of resin absorption of TFG,the heating recirculation composition is mainly located in 40 %~60 %ethanol elution parts.Conclusion The best method for extracting TFG is heating recirculation in above three methods,Elution with 80%ethanol continuously after water elution is the the optimal resin purification method for TFG.
