2.Status and influencing factors of transitional nursing needs of patients with transjugular intrahepatic portosystemic shunt

Objective To focus on the transitional nursing needs and its influencing factors of patients accepted transjugular intrahepatic portosystemic shunt (TIPS). Methods The study was conducted with a self-designed questionnaire involving 54 patients accepted TIPS. Also, in-depth semi-structured interviews were conducted among 10 patients. Results The total score of transitional nursing needs among patients accepted TIPS was (4.53 ± 0.48) points, which was in the relatively high lever. The multiple linear regression analysis showed that age, hepatic function and levels of venous ammonia were influence factors of transitional nursing needs among patients accepted TIPS (P<0.05). Conclusions The transitional nursing needs of patients accepted TIPS is urgent. Postoperative transitional nursing should be focused on younger, poor hepatic function and venous ammonia big change among patients accepted TIPS. Low-protein diet and the prevention of complications should be taken priority over all others.
3.Bonemorphogeneticproteins-2/Osterix signaling pathway regulates the differentiation of preosteoblasts

BACKGROUND:Bone formation and development are reported to be regulated by bone morphogenetic protein2(BMP2)-induced Osterix expression. OBJECTIVE:To investigate the regulatory effect of BMP2/Osterix signaling pathway on differentiation of preosteoblasts in mice. METHODS:mRNA and protein expression of Osterix wasdetermined by real-time RT-PCR and western blot assay, respectively at various time points after mouse preosteoblasts were treated with BMP2. pcDNA3.1/myc-Osterix eukaryotic expression vector was constructed and transducted into preosteoblasts, and then mRNA expression of alkaline phosphatase, bone sialoprotein, and matrix extracelular phosphoglycoprotein wasdetected by real-time RT-PCR after transduction and BMP2 treatment. RESULTSANDCONCLUSION:Osterix mRNA expression was up-regulated when treated with BMP2 in mouse preosteoblasts, and reached the peak at 24 hours. In addition, the protein expression of Osterix was increased after BMP2 treatment. Alkaline phosphatase, bone sialoprotein, and matrix extracelular phosphoglycoprotein mRNA expression wasup-regulated after transfection of mouse preosteoblasts with pcDNA3.1/myc-Osterix eukaryotic expression vector and BMP2 treatment. Our results indicate that BMP2 regulates the synthesis of genetic markers of osteogenesis,such asalkaline phosphatase,matrix extracelular phosphoglycoproteinviaup-regulating Osterix expression in mouse preosteoblasts, suggesting BMP2/Osterix signaling pathway plays a critical role in bone development.
4.Different inhibition of hepatocarcinoma cell growth by As_2O_3 in SMMC-7721 and BEL-7402 cell lines

AIM: To explore the different inhibitory effect of arsenic trioxide (As_2O_3) on hepatocarcinoma cell growth in SMMC-7721 and BEL-7402 cell lines and its mechanism. METHODS: The cell culture and trypan blue staining were used to study the inhibitory effect of arsenic trioxide on cell growth, and the glutathione (GSH) contents in hepatocarcinoma cells treated with arsenic trioxide were detected. RESULTS: Arsenic trioxide inhibited the growth of BEL-7402 cells in a time and dose-dependent manner. The inhibitory effect was significant at a lower dose of 0.50 ?mol/L for 24 h, however, to SMMC-7721 cells, a higher dose of 2.00 ?mol/L for 96 h was needed. The inhibitory rate of arsenic trioxide (0.25-2.00 ?mol/L) on BEL-7402 cell growth was higher than that on SMMC-7721 cells. The content of GSH in SMMC-7721 cells was much higher than that in BEL-7402 cells [(50.8?5.2) (?mol/g) protein and (18.7?1.4) ?mol/g protein, respectively]. CONCLUSION: There was a significant difference in inhibition of hepatocarcinoma cell growth by arsenic trioxide between BEL-7402 and SMMC-7721 cell lines, the cause of which may be due to the difference in GSH content in BEL-7402 and SMMC-7721 cells. 5.Health status and needs for health promotion of a company staff

Objective To understand the affecting factors of health and health promotion needs of one company staff so as to provide evidence for effective health intervention.Methods A convenience sampling method was used to enroll subjects in this cross-sectional study.All the participants were required to fill in a questionnaire to learn their demographics,health related factors and health promotion needs in the 2 work days of June 2012 had been studied.Chi-square test and unconditional logistic regression were used for data analysis.Results More males showed higher risk of disease than females (x2 =5.394,P < 0.05).In conditional logistic regression,age was the main factor affecting health (odds ratio (OR) =3.127,95% (confidence interval) CI 1.020-9.591,P < 0.05) when gender,marital status,educational level and occupation were adjusted.Dietary nutritional knowledge (69.6%) and physical check-up (64.6%) were urgent needs among those surveyed.Conclusion Staff with varied demographic characteristics have different health related factors and physical check-up may provide a tool to meet their urgent needs of health promotion.
6.Concept and significance of a subjective and multiple index system of clinical evaluation for traditional Chinese medicine.

Traditional Chinese medicine (TCM) has its own system of diagnosis and treatment theory, and its methods for evaluating clinical efficacy are different from those of Western medicine. Applying evaluation techniques and methods that are used in Western medicine mechanically to TCM will not work. So building evaluation techniques, which adhere to regulations and characteristics of TCM, is necessary and imperative. As the quality of life and patient-reported outcome instruments were brought into practice and developed, clinical evaluation ideas and methods for TCM are provided with an opportunity for development. This article puts forward the concept of subjective complex outcomes (SCOs), which constitutes subjective feelings gained from the patient, doctor and caregiver, different from laboratory parameters. SCOs provide a multidimensional and complex health-related quality of life (HRQL) assessment and focus on the source of assessment information of diseases. This article also introduces a case study building SCO methods of TCM treatment for chronic obstructive pulmonary diseases, in order to promote discussion and provide a platform for future research.
7.Comparative?analysis?of?two?different?DNA?purification?methods?for?bones?and?teeth

Objective To compare the effect of silica-extraction method and Silico membrane based method in DNA purification from bones and teeth.Methods DNA samples were purified respectively with the silica-extraction method and MinElute PCR Purification kit from 6 bones and 8 teeth,then tested STR types by GlobalFiler? kits. And evaluated the two methods with the success rate and the peak height. Results Both of the two purification methods can successfully obtain the STR markers of the 14 samples. And there was no statistical difference between the two methods in the average peak height from bones and teeth. Conclusion The Silico membrane based method which have more advantages in operation is an efficient method to purify DNA from bones and teeth, and there is no significant difference compared with the silica-extraction method. But the cost is higher. It can be selectively used in forensic practice.
8.High glucose induces a metabolic memory in human periodontal ligament cells

BACKGROUND:Studies on high glucose exposure in human periodontal ligament cel s usual y focus on the biological behaviors, pathways and secretory factors, but whether the metabolic memory is involved is little known. OBJECTIVE:To investigate the metabolic memory of high glucose exposure in human periodontal ligament cel s. METHODS:Human periodontal ligament cel s were primarily cultured and identified. Cel s at 5-8 passages were selected and randomized into four groups. Group A (controls):DMEM containing 5.5 mmol/L glucose for 8 days;group B (5-day memory group):DMEM containing 35 mmol/L glucose for 3 days and DMEM containing 5.5 mmol/L glucose for 5 days;group C (3-day memory group):DMEM containing 35 mmol/L glucose for 5 days and DMEM containing 5.5 mmol/L glucose for 3 days;group D (8-day high glucose group):DMEM containing 35 mmol/L glucose for 8 days. The cel proliferation was detected by cel counting kit-8, the cel apoptosis was determined by flow cytometry, and the levels of total proteins and alkaline phosphatase were investigated using ELISA. RESULTS AND CONCLUSION:Compared with the control group, the cel proliferation in the other three groups was significantly reduced (P<0.05), the number of apoptotic cel s was significantly increased, while the levels of total proteins and alkaline phosphatase were significantly decreased (P<0.05). These results suggest that high glucose causes persistent changes in human periodontal ligament cel s by inhibiting cel viability, increasing the apoptosis and downregulating the levels of the total proteins and alkaline phosphatase

Abstact:Aim To identify whether the petroleum e-ther fraction of Yueju pill (YJ-PE)could exhibit a po-tent rapid antidepressant effect and to investigate the underlying biological mechanism involved.Methods Fourty-four male KM mice were randomly assigned to five groups:the control group (saline group),low dosage of YJ-PE,middle dosage of YJ-PE,high dos-age of YJ-PE and ketamine.After a single drug admin-istration,we used tail suspension test (TST)to test the rapid antidepressant effect of YJ-PE in KMmice at 30 min.Another twenty-four male KM mice were ass-inged to three groups:control group,screened effective low dosage of YJ-PE group and ketamine group.We usd TST to assess these mice at 24 h post a single ad-
ministration.Western blot was performed to examine the expression of BDNF and TrkB in hippocampus of KMmice at 30 min and 24 h post a single administra-tion.Results An acute administration of YJ-PE de-creased the immobility time of KMmice in TST and in-creased the expression of BDNF and TrkB as soon as 30min post a single administration and lasted at least for 24 hours.Conclusion YJ-PE (low dosage)ex-hibits a rapid antidepressant-like potent,and the po-tent rapid antidepressant effects of YJ-PE are associat-ed with the up-regulation of BDNF and TrkB.
10.Evaluation of the effects of intervention on employees' health risk factors in a security corporation in Beijing

Objective To evaluate whether an intervention with a health promotion program can reduce prevalence of the health risk factors of non-communicable chronic diseases (NCD) and improve the knowledge awareness of NCD prevention of employees from a security corporation in Beijing selected by the study team. Methods A cross-sectional study was conducted by selecting 157 employees of a security corporation in Beijing, and a quasi-experiment method and a self-developed Questionnaire of Intervention for Health Risk Factors were used. Based on the results of the distribution of risk factors for NCD and the focus-group discussion, one year health promotion program intervention was performed. According to the comparison of prevalence of risk factors between baselines and after intervention of the studied employees, the intervention effectiveness was evaluated. Results After the health intervention, rate of reported unhealthy diet reduced from 75.0%to 56.8%, which showed statistically significant (?2=8.53, P<0.05);rate of reported less physical activity reduced from 35.5%to 17.4%, with statistical significance (?2=19.60, P<0.05);rate of reported cigarette smoking reduced from 16.8%to 11.6%with statistically significance. There was no statistically significant reduction in the rate of the other health risk factors (P>0.05). The median value of health knowledge awareness of the studied employees was significantly improved from 10.20 to 11.33(S=-2 545, P<0.05) and awareness of recommended daily intake of dietary salt increased significantly from 46.20%to 67.09%after intervention (χ2=17.85, P<0.05). Conclusion The non-communicable chronic diseases intervention program can reduce the health risk factors and improve health knowledge awareness of the studied population.
