AIM: To explore the different inhibitory effect of arsenic trioxide (As_2O_3) on hepatocarcinoma cell growth in SMMC-7721 and BEL-7402 cell lines and its mechanism. METHODS: The cell culture and trypan blue staining were used to study the inhibitory effect of arsenic trioxide on cell growth, and the glutathione (GSH) contents in hepatocarcinoma cells treated with arsenic trioxide were detected. RESULTS: Arsenic trioxide inhibited the growth of BEL-7402 cells in a time and dose-dependent manner. The inhibitory effect was significant at a lower dose of 0.50 ?mol/L for 24 h, however, to SMMC-7721 cells, a higher dose of 2.00 ?mol/L for 96 h was needed. The inhibitory rate of arsenic trioxide (0.25-2.00 ?mol/L) on BEL-7402 cell growth was higher than that on SMMC-7721 cells. The content of GSH in SMMC-7721 cells was much higher than that in BEL-7402 cells [(50.8?5.2) (?mol/g) protein and (18.7?1.4) ?mol/g protein, respectively]. CONCLUSION: There was a significant difference in inhibition of hepatocarcinoma cell growth by arsenic trioxide between BEL-7402 and SMMC-7721 cell lines, the cause of which may be due to the difference in GSH content in BEL-7402 and SMMC-7721 cells. Objec0tive To investigate the molecular mechanism of the mal-production of IgA and IgA1 by tonsillar mononuclear cells (TMCs) in IgA nephropathy (IgAN) patients by measuring the mRNA expression of Iα-Cα germline transcript and the mRNA and protein expression of activated induced cytidine deaminase (AID) in cultured TMCs stimulated with lipopolysaccharide (LPS) or hemolytic streptococcus (HS) in IgAN patients as well as the chronic tonsilitis patients. Methods Twent-seven IgAN patients admitted into our hospital from Jan.2009 to Feb.2010 were enrolled.Twent-seven patients with chronic tonsillitis without renal disease were selected as control.Tonsillar mononuclear cells were isolated by density gradient centrifugation in lymphocyte separation medium. The amount of IgA or IgA1 secreted in the culture supernatants was determined by specific enzyme-linked immunosorbent assay (ELISA).Expressions of Iα-Cα germline transcript and AID mRNA were examined by real-time PCR.The AID protein was determined by Western blot. Results The production of IgA and IgA1 protein, especially the ratio of IgA1/IgA and the expression of AID protein in TMCs were significantly increased in IgAN group compared with chronic tonsillitis group (all P<0.05).The IgA and IgA1 level of stimulated TMCs were obviously increased in patients with IgAN compared with control group (P<0.05).And the expressions of Iα-Cα mRNA,AID mRNA and AID protein were up-regulated significantly in stimulated TMCs (all P<0.05). Conclusions Both LPS and HS can induce the production of IgA and IgA1 and up-regulate the expressions of AID and Iα-Cα in TMCs of IgAN patients. Our results indicate that the TMCs are capable of producing high level of IgA and IgA1 stimulated by LPS or HS,whuch may be due to the incression of AID and Iα-Cα.

Purpose To analyze the image quality of brain CT with 256-slice wide detector axial scanning mode, routine axial scanning mode and spiral scanning mode, and to provide a more effective brain CT examination method for patients.Materials and Methods The prospective study was conducted on 90 patients accepting routine brain CT examination, and they were randomly divided into three groups.CT examination with 160 mm axial scanning mode,40 mm axial scanning mode and 40 mm spiral scanning mode were respectively conducted using GE Revolution CT.The scanning condition was adjusted to remain constant radiation dose, and then the image quality was analyzed.CT attenuation of gray and white matter,contrast-to-noise ratio (CNR) of white-gray matter and image noise of the three scanning modes were compared.Subjective scoring on image quality of the three scanning modes was also performed. Results On body lateral cerebral ventricle level, there were no significant difference in CT attenuation of gray and white matter and CNR (P>0.05).On centrum semiovale level, the CT attenuation of gray matter [(31.71 ± 1.82) HU],white matter [(22.97± 1.50) HU] and CNR 2.05±0.42 of 160 mm axial scanning mode was significantly different from the other two scanning modes (F=26.74,47.16 and 3.85,P<0.05).On centmm semiovale level, image noise of 160 mm axial scanning mode was lower than the other two kinds of scanning methods (F=6.31,P<0.05),in the basal ganglia and posterior fossa there were no statistically significant differences in the image noise between the three scanning modes (P>0.05).The subjective score of the three scanning modes all met the diagnostic requirements, and there was no significant difference (P>0.05).The effective dose and scanning time of 160 mm axial scanning mode was lower than those of the other two scanning modes, and the X-ray utilization was higher. Conclusion 160 mm wide detector axial scanning mode is more suitable for brain CT scan, and it can be used as the preferred scanning mode in the emergency and among non-cooperative patients.

Chinese Journal of Ultrasonography 2017;26(7):569-575
Objective To evaluate the application value of cardiopulmonary ultrasound(CPUS) in patients with interstitial lung diseases(ILD).Methods Ninety patients with clinically diagnosed interstitial lung diseases and 30 healthy subjects were included in this study. The conventional echocardiographic parameters and right ventricular free wall longitudinal strain (RVLS_FW) were measured in all patients and healthy subjects. The degree of pulmonary fibrosis or the disease severity of each ILD patient was scored using a semi-quantitative scoring method.Patients’ pulmonary function parameters and clinical outcome score (GAP score) were recorded.Fourteen patients belong to other types of ILD and 19 patients with incomplete clinical data were unable to perform GAP scoring, finally 57 patients' clinical and ultrasonographic parameters were measured. On the basis of the pulmonary arterial systolic pressure (PASP) obtained by Doppler echocardiography,90 patients were divided into two groups:ILD patients with pulmonary hypertension(ILDpH,PASP ≥36 mmHg,1 mmHg =0.133 kPa) and ILD patients without PH (ILDNpH,PASP< 36 mmHg).Comparsion between ILDpH,ILDNpH and healthy subjects were then evaluated.According to the GAP score, all cases were divided into three groups:low risk, moderate risk and high risk.Statistical analysis was performed to examine the relationship between CPUS parameters and clinical parameters. Results ①Compared with ILDNpH patients,ILDpH patients had significantly lower right ventricular function, wider basal right ventricular diameter (RVD),proximal right ventricular outflow diameter (DRvor1) and right pulmonary artery diameter (DRPA).Compared with healthy subjects,ILDNpH patients had mildly lower right ventricular function and wider left pulmonary artery diameter (DLPA) and right ventricular outflow tract dimensions at the distal or pulmonic valve (DRvoT2).②A good correlation was found between lung ultrasound (LUS) scores and pulmonary function parameters, especially diffusion capacity for carbon monoxide of the lung(DLco)(r =-0.68,P <0.001).③With the increased risk of patients, the right ventricular function was gradually decreased,DRVOT2,DRPA,DLPA were gradually widened, and the lung ultrasound score was gradually increased.A weak correlation was found between GAP scores with RVLS_FW,LUS scores,DRpA(r =0.44,0.34,0.31,P <0.05).Conclusions CPUS parameters are useful in the assessment of interstitial lung disease and could reflect the prognosis of ILD patients to some extent. Therefore,on the basis of GAP scores, addition of RVLS_FW and LUS score will be more helpful to evaluate the prognosis ofILD patients.
