1.The application of MiniFiler~(TM) kit in LCN-STR genotyping

Objective To evaluate the application value of MiniFilerTM kit in LCN-STR genotyping in forensic science.Methods Compared the testing results of MiniFilerTM and IdentifilerTM kits in 49 general quantity biological samples and 39 LCN-biological samples,including blood stains,sperm stains,bones,and so on.Meanwhile the sensitivity of these two kits were also compared.Results Full concordance between IdentifilerTM and MiniFilerTM kits was observed in all of 49 general quantity biological samples.For 39 LCN-biological samples,only 22 samples could be genotyped in partial loci and 17 samples negative with the IdentifilerTM kit,while 30 samples could be genotyped in all loci,5 samples in partial loci and 4 samples negative with the MiniFilerTM kit.Therefore,there was a higher success rate for LCN-biological samples typing with MiniFilerTM kit than with IdentifilerTM kit.In addition,the sensitivity of the MiniFilerTMkit was also a little higer than the IdentifilerTMkit.Conclusion The results demonstrated that the MiniFilerTM kit can markedly improve the typing success rate of LCN-biological samples,and is suitable for analyzing difficult biological samples in forensic practice.
2.Determination of two kinds of alkaloids in Zhitong Capsule by HPLC

AIM:To establish a HPLC method for determining contents of tetrahydropalmatine and pepper alkali in Zhitong Capsule(Rhizoma Corydalis,Fructus Piperis,Radix Paeoniae alba,etc.)to control its quality.METHODS:The samples were extracted in water by ultrasonic wave,and then extracted by aether to refine after being acidized by 1 mol/mL HCl.Following that the pH of water solution was adjusted to 9.0-10.0 by NH_3?H_2O,and then extracted by aether again.After that,the aether solution was collected to evaporation to dryness and metered by methanol.The sample solution was determined by high performance liquid chromatography on a Hypersil ODS 2 C_ 18 column(4.6 mm?250 mm,5 ?m)with mobile phase composed methanol-0.1% phosphoric acid water solution(the pH was adjusted to 6.0 by triethylamine)(55∶45).The flow rate was 1.0 mL/min,the column temperature was at 30 ℃ and the signal from 0 to 14.5 minutes were acquired at 280 nm,and that from 14.5 to 22 minutes were detected at 328 nm.RESULTS:The resolution of tetrahydropalmatine and pepper alkali was good,with no miscellaneous peak.The linear range was at 0.196-1.96 ?g(r=0.999 6)for tetrahydropalmatine,0.03-0.3 ?g(r=0.999 7)for pepper alkali.The average rate of recovery of tetrahydropalmatine was 97.74%(RSD= 0.42%,n=6),and that of pepper alkali was 104.51%(RSD=2.01%,n=6).CONCLUSION:The method is simple,reliable,accurate and can be applied to the quality control of the preparation.
3.Expression of Toll-like receptors in lesions of condyloma acuminatum

Objective To measure the expressions of Toll like receptors (TLRs) in lesions of condyloma acuminatum (CA). Methods Tissue samples were resected from lesions of 25 patients with CA and normal skin of 16 human controls. Frozen sections were prepared and the expressions of TLRs 1 - 10 were measured by immunohistochemical staining. Results In normal skin samples, there was a strong expression of TLR 6 and 9, medium expression of TLR 1, 3, 10 and weak expression of TLR 2, 4, 5, 7, 8 in the stratum corneum and stratum granulosum, but a weak or negative expression of TLRs in the stratum spinosum and stratum basale. In contrast, a strong expression of TLR 1, 3, 6, 9, medium expression of TLR 4, 8, 10 and weak expression of TLR 2, 5, 7 were detected in keratinocytes of almost all epidermal layers except for stratum basale in CA lesions. Conclusions The expressions of TLRs, especially TLR 1, 3, 6, 9, are markedly enhanced in proliferated keratinocytes in CA lesions compared with normal skin, which is likely to be associated with anti-viral immunity triggered by HPV infection.
4.Comparative?analysis?of?two?different?DNA?purification?methods?for?bones?and?teeth

Objective To compare the effect of silica-extraction method and Silico membrane based method in DNA purification from bones and teeth.Methods DNA samples were purified respectively with the silica-extraction method and MinElute PCR Purification kit from 6 bones and 8 teeth,then tested STR types by GlobalFiler? kits. And evaluated the two methods with the success rate and the peak height. Results Both of the two purification methods can successfully obtain the STR markers of the 14 samples. And there was no statistical difference between the two methods in the average peak height from bones and teeth. Conclusion The Silico membrane based method which have more advantages in operation is an efficient method to purify DNA from bones and teeth, and there is no significant difference compared with the silica-extraction method. But the cost is higher. It can be selectively used in forensic practice.
5.Lorrelation of gene mutation of EGFR,the expressions of HER-2,VEGF in lung adenocarcinoma and clinical pathological factors

Objective To investigate the gene mutation of epithelial growth factor receptor(EGFR) and the expressions of hu‐man epiderma1 growth factor receptor 2(HER‐2) and vascular endothelial growth factor(VEGF) in lung adenocarcinoma and their relationships with the clinical pathological factors .Methods ARMS‐PCR method was used to detect the gene mutation of EGFR in 49 lung adenocarcinoma specimens and 10 normal tissue specimens .Immunohistochemical method was also used to detect the ex‐pressions of the HER‐2 and VEGF in them .Results (1) The mutation rates of EGFR and the positive rates of HER‐2 and VEGF in lung adenocarcinoma were significantly higher than those in the normal tissues(P< 0 .01) .(2) The occurs of EGFR mutation were correlated with sex and the smoking habit(P<0 .05);but not correlated with age ,the size of tumor ,differential ,lymph nodes metastasis ,TNM stages and pleural invasion(P>0 .05) .The HER‐2 and VEGF protein expressions in lung adenocarcinoma were correlated with differential ,the size of tumor ,TNM stages ,lymph nodes metastasis and pleural invasion(P<0 .05);but not correla‐ted with sex ,age ,and the smoking habit(P>0 .05) .(3) The expressions of HER‐2 and VEGF protein in the lung adenocarcinoma were positively correlated with each other(P<0 .01) .Conclusion EGFR mutation is closely related to the occurrence of lung ade‐nocarcinoma ,its high expressions in the women ,non smoking people have important clinical significance .HER‐2 and VEGF could promote the lung adenocarcinoma′s occurrence ,development and transfer .They could be used to evaluate the patients′prognosis ,and provide new molecular targeted therapy .
6.Psychological rehabilitation of paraplegic patients with spinal fracture

Objective To study the effects of psychological rehabilitation in the treatment of paraplegic patients with spinal cord injury (SCI).Methods A hundred paraplegic spinal fracture survivors were divided into a research group and a control group with 50 cases in each.Systematic rehabilitation was given to the patients in the control group,while this was combined with individualized psychological rehabilitation protocols for the patients in the research group.The daily treatment lasted 10 weeks,6 days a week.All of the patients were assessed with the Barthel index (BI),a functional independence measure (FIM),the Hamilton depression scale (HAMD) and the Hamilton anxiety scale (HAMA) pre-treatment and post-treatment.Results Before the intervention there was no significant difference between the 2 groups in any of the assessments.After ten weeks the average BI,FIM,HAMD and HAMA results in the research group were significantly better than those pre-treatment and also significantly better than those in the control group.Conclusions Psychological rehabilitation can distinctly improve the functioning and psychological state of paraplegic patients after SCI.
7.Analysis of clinical factors in achalasia complicated by esophageal cancer

Objective To analyze the clinical pathological characteristics of achalasia patients complicated by esophageal cancer.Methods From January 2005 to May 2013,among 658 patients with achalasia,the clinical data,pathological characteristics of tumor and the treatment of those complicated by esophageal cancer were collected and analyzed.At the same time,receiver operating characteristic (ROC) curve analysis was performed to analyze the relationship between the course of achalasia and esophageal cancer.Results Among the 658 patients with achalasia,297(45.1%) cases were male and 361(54.9%) cases were female.A total of 62 cases (9.4%) were lost to follow-up and of 596 cases who completed the follow-up,26 cases (4.4 %) were complicated with esophageal cancer.Among the 26 patients complicated with esophageal cancer,69.2% (18/26) were male;the course from achalasia diagnosed to esophageal cancer was (16.5±8.2) years.The patients with a disease duration more than 10 years accounted for 88.5%(23/26),and 80.8% (21/26) underwent previous balloon dilatation or Heller surgery.The predilection site of esophageal cancer was in the middle esophagus of patients complicated by esophageal cancer (69.2%,18/26),the main type was squamous carcinoma (88.5%,23/26),65.4%(17/26) of the lesions invaded to muscular layer (stage T3 to T4),61.5%(16/26) had the late stage tumor (stage Ⅲ to Ⅳ),and 23.1%(6/26) had distant metastasis.The results of ROC curve analysis showed that when the duration was 10.5 years,the Youden index reached the maximum value (0.71).And 38.5% (10/26) of patients with esophageal cancer could not be treated with esophageal cancer radical surgery,and their average survival time was (11.4±6.6) months.Conclusion The patients with the duration of achalasia over 10.5 years are required for regular endoscopic examination to improve the prognosis.
8.Antibacterial Activities of Cefoperazone-sulbactam Against Gram-negative Bacilli

OBJECTIVE To evaluate the antibacterial activities of cefoperazone combined with sulbactam against Gram negative bacteria,and compare with other antimicrobial agents.METHODS The antibacterial activities of 10 frequently used antibiotics against 1 670 strains of clinical isolated Gram negative bacteria were studied by using agar dilution methods according to the NCCLS 2002.RESULTS The results showed imipenem was the most active tested against Enterobacteriaceae such as Escherichia coli and Klebsiella pneumoniae,followed by cefoperazone-(sulbactam),which had similar activities as imipenem against the non-fermentative strains such as Pseudononas aeruginosa,and Acinetobacter spp,but cefoperazone-(sulbactam) had higher susceptibility to imipenem-(resistant) Stenotrophomonas maltophilia.CONCLUSIONS Cefoperazone-sulbactam has good and broad spectrum(antibacterial) activities especially against Gram negative(bacteria,) and is expected to have a bright prospect in the treatment of severe hospital infections induced by Gram negative bacteria.
9.Matrine sustained release tablet, capsule and injection: comparison of pharmacokinetics and relative bioavailability

Objective:To compare the pharmacokinetics and relative bioavailability between matrine sustained release tablet,matrine conventional capsule and matrine injection. Methods: Dogs were given single oral dose and multiple doses of matrine sustained release tablet, capsule or injection in a randomized crossover way. Matrine concentrations in dog plasma were determined by RP-HPLC method. Results: The results showed that the t max and c max were 300 min and (5.088?0.490) ?g/ml for matrine sustained release tablet, (85?12) min and (6.360?0.215) ?g/ml for the conventional capsule, and 10 min and (6.500?0.404) ?g/ml for the injection. The relative bioavailability of the sustained release tablet was (153.7?9.4)% compared with that of the conventional capsule; the absolute bioavailability of the sustained release tablet was (73.5?14.2)% compared with that of the injecetion. 10.Effect of Proprioceptive Training on Lumbar Disc Herniation

Objective To investigate the effects of proprioceptive training on lumbar disc herniation (LDH). Methods 50 patients with LDH were divided into experimental group (n=25) and control group (n=25). Both groups received physiotherapy and core stability exercise, and the experimental group received proprioceptive training with BIODEX Balanced System in addition. They were assessed with the Visual Analogous Scale (VAS) of pain, Oswestry Disability Index (ODI), and posture stability and limits of stability of BIODEX Balanced System. The incidence of relapse was followed up in a year. Results The scores of VAS, ODI, posture stability and limits of stability improved in both groups after treatment (P<0.05), and the ODI, posture stability and limits of stability improved more in the experimental group than in the control group (P<0.05). There were 5 cases relapsed in the control group, and 2 cases in the experimental group. Conclusion The proprioceptive training may further improve the function of lower back, and motor control in patients with LDH, and prevent the relapse.
