1.Application of three-dimensional cell culture model in cancer research

Compared to the conventional two-dimensional (2D) culture,in vitro three-dimensional cell culture (3DCC) can mimic the microenvironment more exactly.Compared to in vivo animal models,3DCC is less time-consuming and can be real-time recorded.Thus,3DCC fills the gap between 2D culture and animal models,which is used for oncologic research quickly.In 3DCC,the characteristics of cancer cells,such as morphology,the capability of invasion and migration and the resistance to chemo-radiotherapy,are quite different from those in 2D culture.The development of 3DCC technology provides a platform for oncology,organization engineering and gene or drug screening.
2.Reasons for Clinical Shortage of Certain Drugs

OBJECTIVE: To draw the attention of our country and the concerned party on the shortage of certain drugs. METHODS: Based on the drug shortage condition in our hospital in recent years, we analyzed the reasons for drug shortage, put forward the solution and summarized the clinically indispensable categories. RESULTS & CONCLUSIONS: We should draw experiences from US by solving shortage problem of certain drugs through the co-efforts of government, manufactures and medical institutions to satisfy patients' needs of disease prevention and treatment.
3.Progression of tumor dormancy models

Tumor dormancy is a state that tumor cells long-term exist in human body,without obvious growth. Dormant cell is one of the origins of tumor recurrence or metastasis. The exact mechanism of tumor dor-mancy is still unclear. Combined with cellular and molecular biological techniques,tumor cell transplantation and culture in vitro can establish experimental tumor dormancy-recurrence models. These in vivo and in vitro models can be used to investigate mechanisms of tumor dormancy and to explore novel treatments.
4.The medical equipment management base on spot tracing method risk loop in hospital accreditation

Objective:Reference to the risk loop in hospital accreditation spot tracing method, enhancing the management of medical equipment.Methods: By six link as planning, training, implementation, monitoring, feedback, improvement in the spot tracing method to implement the management of medical equipment.Results: Evaluate the risk level of medical equipment by the risk value evaluation, thereby setting clinical equipment training period and mode, and apply the result of training assessment to feedback adjusting ,use monitoring and use effect improving.Conclusion: By preventing and controlling the risk of medical equipment, the original medical management was promoted to a new level, more in line with the requirements of medical service safety and standardized level in hospital accreditation.
5.The response of GABA eliciting the rats outer hair cells during development.

OBJECTIVE:
We used electrophysiological methods to study that whether GABA could elicit OHCs outward currents provide evidence for exsitence of GABA-A receptor and investige the relationship between the effect of GABA and the development of OHCs.
METHOD:
We used whole-cell recording OHCs at current-clamp or voltage-clamp to verify the function of GABA receptor on OHCs. Then we counteds the responsive cells vs. total number cells, and according to results to study the relationships between the GABA receptor and development of OHCs.
RESULT:
OHC was elicited outward current or hyperpolarized by GABA and the responsive cells were decreased with development.
CONCLUSION
The result of GABA receptor decreasing with development suggested that the receptor may draw efferents to OHCs or facilitate the MOC-OHC synapse formation.

6.Clinical analysis of 38 systemic lupus erythematosus patients complicated with pulmonary hypertension

Objective To analyze the pathogenesis, clinical features, treatment and prognosis of pulmonary hypertension (PHT) in systemic lupus erythematosus (SLE) patients. Methods Thirty-eight patients admitted during Jan 2001 to Aug 2008 were diagnosed as SLE associated with PHT and were analyzed retrospectively. Results All cases were female with the mean age of 35.7 yds. The mean period from PHT to the diagnosis of SLE was 2.3 years. Three cases were severe PHT with remarkable heart failure. Twenty-seven eases demonstrated Raynaund's phenomenon. Antinuclear antibody (ANA) was positive in all 38 cases and the titer was 1:320. Anti-DNA antibody, anti-Sin antibody, anti-U1RNP antibody and anti-SSA, anti-SSB were positive in 21 cases, 16 cases, 15 cases, 8 cases respectively. Antiphospholipid antibody and rheumatoid factor (RF) were elevated in 4 cases and 13 cases respectively. Seven cases were involved in lung fibrosis and pulmonary function test showed mild to moderate restrictive abnormality in these 7 cases. After treated with the combination of steroid and immunosuppressive agents, one case died on the 10th day and 2 cases died in 2 to 3 years after the diagnosis was established. The other 35 cases stayed stable. Conclusion PHT is one of the severe complications of SLE and often accompanied by poor outcome. Therefore early diagnosis and early combined therapy is very important.
7.The management strategy of hospital new financial system-based medical equipment archives

Objective:To establish the medical equipment archives management system to adapt to the Finance Ministry new hospital financial system. Methods:According to the new financial system requirements, implement the new file management strategy, as adjusting the original classification, management object and process. Results: After the implementation of the new management strategy, medical equipment archives management and new financial system in our hospital realize synchronous in January 1, 2012, thethree Zhang cardsystem optimizing, and the results have been applied to the existing hospital medical equipment management software, greatly improving the efficiency of management, provides a strong basis for medical equipment economic benefit and social benefit evaluation. Conclusion:The medical equipment archives were giving full play to improve the quality of medical care.

Numb,one of the important cell fate determinants,plays a key role in regulation of nerve system development,and it is closely related to the occurrence of tumors.It is proven that Numb is implicated in the processes of occurrence,invasion and metastasis of tumors through regulating the way of cell division,cell polarity,epithelial-mesenchymal transitions and several signal pathways,e.g.,Notch,Hh,p53.The expression levels of Numb in cells are related to the malignant degree and prognosis of tumors.It is vital to explore the role of Numb in tumors for treating tumors at the standpoint of Numb.
9.In vitro evaluation of the antibacterial activities of different irrigants against Enterococcus faecalis

90 extracted single rooted human teeth were inoculated with E.faecalis.The cannals of the teeth were irrigated with MTAD, 5.25% NaClO,compound chlorhexidine gargle,tinidazole mouthwash,distilled H2 O(infection control)and autoclave followed by distilled H2 O(autoclaving control),respectively(n =1 5).Then E.faecalis infection of the samples were examined.The results showed that MTAD had significantly greater antibacterial effects than 5.25% NaClO,tinidazole mouthwashes,compound chlorhexidine gargle and tinidazole mouthwash(P <0.05).There was no significant difference between each 2 of the other 3 irrigants.
10.Comparison on rehabilitation effect and safety of laparoscopic herniorrhaphy through different approaches in the treatment of inguinal hernia

Objective:To explore the rehabilitation effect and safety of laparoscopic herniorrhaphy through different approaches in the treatment of inguinal hernia.Methods:The clinical data of 132 patients with inguinal hernia in Shushan Branch of Hefei First People′s Hospital from January 2018 to May 2020 were retrospectively analyzed. Laparoscopic herniorrhaphy was performed in all patients. The 46 patients who were treated with transabdominal preperitoneal patch (TAPP) repair were included in TAPP group, while another 86 patients who were treated with totally extraperitoneal patch implantation (TEP) were included in TEP group. The surgery-related indexes, influencing indexes of pneumoperitoneum on pathophysiology, inflammation indexes, pain score and postoperative complications were compared between 2 groups.Results:There were no significant differences in intraoperative blood loss, recovery time of gastrointestinal function, hospitalization time and non-surgery cost between 2 groups ( P>0.05). The operation time, leaving bed time and surgery cost in TEP group were significantly lower than those in TAPP group: (45.08 ± 4.92) min vs. (51.03 ± 5.62) min, (20.93 ± 2.64) h vs. (22.98 ± 2.01) h and (6 887 ± 603) yuan vs. (8 276 ± 813) yuan, and there were statistical differences ( P<0.01). Before pneumoperitoneum, at 10 min after pneumoperitoneum and after extubation, there were no significant differences in levels of total carbon dioxide (TCO 2), partial pressure of carbon dioxide (PaCO 2), partial pressure of oxygen (PaO 2) and bicarbonate radical between 2 groups ( P>0.05). Before and after surgery, there were no significant differences in interleukin (IL)-6, IL-10 and C-reactive protein (CRP) between 2 groups ( P>0.05). At 1, 3 and 7 d after surgery, there were no significant differences in visual analogue scale (VAS) between 2 groups ( P>0.05). There was no significant difference in the total incidence of complications between 2 groups ( P>0.05). Conclusions:Both TAPP and TEP are safe and effective in the treatment of inguinal hernia. The surgical cost of TEP is relatively lower, and operation time and leaving bed time are shorter. The choices of clinical surgical methods should base on surgeons′ experience and individualized features of patients.
