1.Inhibitive effect of specific stealth siRNAs on TGF-?_1 expression of mouse lung fibroblasts

Objective To design and produce specific stealth siRNAs and detect their inhibitive effects on TGF-?1 expression.Methods Three stealth siRNAs aimed directly at different sequences(stealth-48,stealth-166,stealth-594) in TGF-?1 mRNA were made.These siRNAs were transfected into BALB/c mouse lung fibroblast in vitro,then the TGF-?1 expression in those lung fibroblasts was detected by immunohistochemistry.Results In different time periods,the TGF-?1 expression was differently depressed by three stealth siRNAs in vitro.The inhibitory effects of stealth-166 was better than the other two stealth siRNAs.The inhibition could be detected in 48 h,reached the highest level in 72 h and began to attenuate 96 h later.Conclusion The TGF-?1 expression of mouse lung fibroblasts in vitro can be depressed by synthetic stealth siRNAs.It seems possible to provide a new way for the treatment of pulmonary interstitial fibrosis.
2.Combined use of RT-PCR and gel electrophoresis to detect expression of transforming growth factor beta1 in mouse lung fibroblasts in vitro

BACKGROUND: As a combination of reverse transcription (RT) and polymerase chain reaction (PCR), RT-PCR has been used to detect gene expression levels in cells and tissues, RNA virus contents in cells and specific gene cloned cDNA sequences.OBJECTIVE: To detect the inhibitory effcet of Stealth siRNAs on the expression of transforming growth factor β1 (TGF-β1).METHODS: There were blank control, empty vector transfection, stealth-48, stealth-166, and stealth-594 groups. Three stealth siRNAs aimed at different sequences in TGF-β1 mRNA were made, and were then transfected into BALB/c mouse lung fibroblasts in vitro. The expressions of TGF-β1 and connective tissue growth factor were detected by RT-PCR.RESULTS AND CONCLUSION: In different time periods, the TGF-β1 expression was differentially depressed by three stealth siRNAs, especially stealth-166. The inhibitory effects varied with time, which could be detective at 48 hours,reached the peak at 72 hours and then began to attenuate at 96 hours. Our findings show that the inhibitory effect of stealth siRNAs on the TGF-β1 expression in mouse lung fibroblasts can be detected by RT-PCR.
3.Development of the state-owned enterprise worker's stressor scale

Objective To develop the state-owned enterprise worker's stressor scale and assess its reliability and validity.Methods Combined qualitative and quantitive methods,the state-owned enterprise worker's stressor scale was developed based deep interviews.Data were collected from 6400 state-owned enterprise workers in China.The initial sample was split randomly into two samples (n 1 =2835 ; n 2 =2834).The first data were used to set for exploratory factor analysis (EFA) and the second data were used for confirmatory factor analysis (CFA).3 subscales (anxiety,depression and somatization) of symptom checklist 90(SCL-90) were used for testing criterion validity.Results The state-owned enterprise worker's stressor scale got 4 factors and 15 items,including job stressor,relationship stressor,development stressor and family stressor.The Cronbach α coefficients of the full scale and its four factors were 0.66 ~ 0.86.The results of CFA showed RMSEA =0.066,NFI =0.97,NNFI =0.97,CFI =0.97,IFI =0.97,GFI =0.95.In addition,stressor and its four factors had significant positive correlations with anxiety,depression and somatization (r =0.29-0.43,P < 0.01).Conclusion The stateowned enterprise worker's stressor scale meets psychometric criteria and is an effective tool.
4.The initial experience of ~(125)I seeds brachytherapy for patients with oral carcinoma

Objective To evaluate the efficacy of ~(125)I radioactive seeds implantation and ~(125)I plaque brachytherapy for oral carcinoma. Methods Eighteen patients with oral carcinoma confirmed by cytology or histopathology were included in this study, Twelve patients with tongue cancer and six patients with gingival carcinoma, there were 20 ulcerative lesions and 10 metastatic cervical lymph nodes. The mean diameter is (2.3±0.7)cm and(2.8±1.7)cm respectively. The patients were treated with both interstitial implantation of ~(125)I seeds and ~(125)I plaque brachytherapy or with ~(125)I plaque brachytherapy only according to patient's individual conditions.The metastatic cervical lymph nodes were treated with CT-guided interstitial implantation of ~(125)I seeds.The sizes of ulcerative lesions and lymph nodes were observed at 1,3,6 months following treatment,and statistical analysis of the sizes of ulcerative lesions were evaluated by paired t-test.Results After 1,3,6 months follow-up,The mean diameters of ulcerative lesions were(2.1±0.6)cm(t=3.559,P<0.01),(1.7±0.5)cm(t=7.609,P<0.01),(0.7±0.6)cm(t=11.508,P<0.01),the cervical lymph nodes showed reduced size. Furthermore, PET-CT images showed a significant decrease in the metabolic activity of treated tumor. After six months, the focus of infection were healing in 8 patients, the cervical lymph nodes of one patient relapsed after ~(125)I implantation again. Patients were followed for 7 to 28 months, all patients were still alive. Conclusion Interstitial ~(125)I radioactive seeds implantation and ~(125)I plaque brachytherapy provide an effective, safe treatment for oral cancer.
5.Regulation mechanism of Compound Yihe Tea on improving insulin resistance in obesity mice

The aim of this study was to investigate the effect of Compound Yihe Tea on improving insulin resistance in obesity mice. Thirty-two male C57BL/6J mice were randomly divided into 4 groups: the normal fat diet group(NFD group), high fat diet group(HFD group), Compound Yihe Tea low dosage group[20 mg/(kg ·d), YH-L group] and high dosage group[40 mg/(kg ·d), YH-H group]. NFD group was given standard feed, and the remaining mice were administered with high fat diet. After 6 weeks, YH-H and YH-L groups were given Compound Yihe Tea for 6 weeks. Blood glucose was measured at week 11 and serum levels of total cholesterol(TC), serum triglyceride(TG), low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)were measured at week 12. Liver tissues were prepared for oil red O and HE staining. Immunohistochemical analysis was used to test the protein expression of GLUT4 in liver. Protein expressions of PI3K, Akt and GLUT4 in epididymis white adipose tissue(WAT)were tested by Western blot. The results showed that Compound Yihe Tea could effectively reduce body weights and the serum levels of TC, TG and LDL-C. Furthermore Compound Yihe Tea could improve the histopathological changes of liver, up-regulate the protein expression of PI3K, Akt and GLUT4 in epididymis WAT and the protein expression of GLUT4 in liver. Compound Yihe Tea can reduce the fat accumulation in liver tissue, improve the indexes of blood glucose and lipid levels, and improve insulin resistance via PI3K-AKT-GLUT4 pathway.

Objective To select and evaluate the diluents for quantitative detection of HBV DNA of high loads sample,hope to find the most applicable diluents which could be used in clinical test.Methods The standard substance(2.00 × 109IU/ml)was 10 and 100 times diluted by different diluents,compare the result of test,and the bias was analysis taking negative quality control as standard diluents,negative serum,physiological saline,and distilled water as candidate diluents.Results When 10 times diluted,there was no statistically difference between the standard diluents and distilled water as diluents(t =2.04,P > 0.05),the bias were less than the TEa regulated by professional standard.When used negative serum and physiological saline as diluents,the results were higher than that of standard diluents (P < 0.05),and the ratio of the bias higher than the TEa was 16.67% and 20.00%.When 100 times diluted,the results of candidate diluents were higher than that of standard diluents.In this time,the result of distilled water diluted detection presented a good linear relationship with the result of standard diluted detection,the formula was Y =0.963X + 0.267 (Y =result of standard diluted detection,X =result of distilled water diluted detection).All the bias were less than the TEa,the sequence of bias sort by ascending counts were negative quality control,distilled water,physiological saline and negative serum.Conclusions The most applicable diluents were negative quality control and distilled water with 10 times dilution.When 100 times diluted was used,the most applicable diluents was negative quality control,then was distilled water,physiological saline and negative serum.If using the distilled water to dilute,we could corrected the result by the formula Y =0.963X + 0.267 to ensure the result to be more exactly.
7.Effects of simple vacuum sealing drainage on the prevention and treatment of the subcutaneous effusion after breast cancer radical correlation

Objective To evaluate the curative effects of vacuum sealing drainage equipment on prevention and treatment of subcutaneous effusion after breast cancer radical correction. Methods A total of 122 cases of breast cancer patients from June 2006 to June 2012 were randomly divided into control group ( n=62) and intervention group (n=60). The patients of control group underwent the conventional drainage while the patients of intervention group received the vacuum sealing drainage. The situation of subcutaneous effusion, infection, skin flap necrosis, decannulation time as well as day flow conditions in two groups were observed and compared. Results The incidence of subcutaneous effusion, infection and skin flap necrosis in control group were 12. 9%, 6. 4%, 8. 1% respectively. In intervention group, the incidence rate of subcutaneous effusion was 1. 7%, and no infection, and skin flap necrosis had been observed (χ2 =7. 90,8. 31,9. 12;P<0. 01). The extubation time and day flow conditions in control group was(6. 8 ± 2. 3) d and (70 ± 38) ml, while the interventiongroupwere(10.5±3.1)dand(105±28)mlwithstatisticalsignificance(t =2.10,3.95;P <0. 05). Conclusions Simple vacuum sealing drainage device can not only prevent the subcutaneous effusion, infection and skin flap necrosis after breast cancer radical correction, but also can realize the early extubation, therefore it is worthy of promotion in clinical.

In recent years, with the accumulated explore of epigenetics in RNA modification, many studies have reported that m 6A methylation modification contributes to development and progression of lung cancer. m 6A-related modified regulator has potential application value as a clinical target for diagnosis and treatment of lung cancer.

Objective To investigate the immediate brain effect of acupuncture at Fengchi using amplitude of low-frequency fluctuation(ALFF)and functional connectivity by the resting-state functional magnetic resonance imaging(rs-fMRI)in patients with posterior circulation ischemia vertigo(PCIV).Methods Twenty patients with PCIV were enrolled.The dizziness handicap inventory(DHI)was used to evaluate the severity of vertigo.The patients were randomly divided into acupuncture group and sham acupoint acupuncture group.Rs-fMRI scan was performed before and after acupuncture.MATLAB-based DPABI 6.1 software was used to analyze rs-fMRI data.Correlation analysis was used between the altered ALFF values and DHI scores.The regions of altered ALFF were taken as seeds to analyze functional connectivity.Results Compared with the sham acupoint acupuncture group,the increased ALFF values were mainly located on the left precuneus,left superior frontal gyrus and left caudate nucleus after acupuncture in the acupuncture group;the decreased ALFF values were mainly located on the left cerebellum and right inferior occipital gyrus.The ALFF value of the left superior frontal gyrus was negatively correlated with the DHI score(P=0.04).The increased functional connectivity was mainly found between left precuneus and the right middle frontal gyrus,the right superior frontal gyrus,the decreased functional connectivity was mainly found between left precuneus and the bilateral paracentral lobule and right cerebellum.Conclusion The ALFF value and functional connectivity are different before and after acupuncture,indicating that the vestibular network,visual and motor brain regions functional activities are changed after needling at Fengchi,which may be the brain functional basis of Fengchi for vertigo in PCIV.
10.Excessive bleeding and reexploration after cardiac surgery is a simple and worth thinking about issue

The incidence of postoperative excessive bleeding and reexploration is relatively high, and it will bring a series of serious complications, such as an additional surgery, a longer intensive care unit stay, longer time on mechanical ventilation, an increase need of allogeneic blood product transfusions and increased mortality. However, the understanding of postoperative excessive bleeding and reexploration in China is significantly different from the international level, and the understanding of postoperative excessive bleeding and reexploration after cardiac surgery in clinical work is still not enough. This review will focus on some problems, such as related factors analysis of postoperative excessive bleeding, hemostatic process optimization and the serious complications of reexploration after cardiac surgery.
