1.A Review of Environmental Estrogenic Effects of Alkylphenols

Alkylphenols(APs) , the second most widely used commercial non-ionic surfactants in the world, are the main degradated productions of alkylphenol ethoxylates (APEs). The structure, biodegradation, transportation and distribution in environment, environmental monitoring and management are reviewed in this paper. Many meaurement methods of APs show that APs possess estrogenoid effects, that mainly disturb endocrinal and reproductive system, especially, estrogen-dependent organs. APs are associated with decreased fertility, reproductive disfunction, inferbility and cancer formation. APs are able to bind estrogen receptors, to imitate or antagonize estrogen effects, to disturb formation and metabolism of endogenous hormones and receptors. APs are also able to change the structure, quantity and function of cells and to disturb the kinetics of cellcycle at cytological and molecular levels. The more work on epidemiological study and mechanism of APs are needed. Management program for controlling environmental pollution caused by APs is being speedily investigated and put into practice progressively. It is necessary to restrict the use of APEs.
2.Measurement of ceftriaxone concentrations in aqueous humor of human eye after intravenous administration of the drug

Objective To measure ceftriaxone concentrations in aqueous humor of human eye after one gram of the drug was administered intravenously. Methods 75 subjects (75 eyes) were studied, 60 of whom were divided randomly into 4 test groups according to different time intervals between drug administration and extraction of aqueous humor (30min, 60min, 120min, 180min), and the rest 15 were controls. One-gram of ceferiaxone was administered intravenously before cataract operation. About 200-300?l aqueous humor was withdrawn from every patient during the operation at respective time point mentioned above and reversed phase high performance liquid chromatography (HPLC) was used to measure the concentration of the drug. Results The concentrations of ceftriaxone in aqueous humor were 0.235?0.121mg/L, 0.474?0.224mg/L, 0.453?0.162mg/L, and 0.534?0.202mg/L, respectively, after a time interval of 30min, 60min, 120min, 180min. The drug concentration in the 30 min group was lower compared with the other 3 test groups (P0.05), and the concentrations of ceftrixone in aqueous humor were all above the minimum inhibitory concentration for 50%(MIC 50 ) of ceftriaxone against most pathogenic bacteria. Conclusion Ceftriaxone may be an effective antibiotic for prophylactic use in ophthalmic surgery and also for the treatment of intraocular infection due to organisms susceptible to the drug.
3.Determination of related substances of vitamin B6 for injection by HPLC

Objective To establish a HPLC method to determine the related substances of vitamin B6 for injection.Methods HPLC method was adopted with DiamonsilTM C18,5μm 250mm×4.6mm;the mobile phrase was length was 291nm.Results The calibration curve was linear in the concentration range of 600~1400μg/ml(r=0.9998).Conclusion The method is simple,rapid,accurate and reliable.As the supplement method of active pharmacopoeia,it can effectively control the quality of the product.
4.Long-time follow-up study of the changes of morphology of central corneal after LASIK using an in vivo confocal microscopy

Objective To morphologically observe the changes of cornea tissue a long period after myopic LASIK with confocal microscopy. Methods 34 patients (68 eyes) from April 1999 to July 2004, who had undertook LASIK in our Hospital, were involved in present study. The confocal microscopy was employed to examine the changes of cornea tissue. The average spherical equivalence was -6.81?2.88D (ranged from -3.0 to -16D). The average age of the patients was 24.17?5.93 (ranged from 18 to 42). The average follow-up time was 39?3 months (ranged from 6 to 73 months). Results The Micro-folds at the Bowman's layer were found in 68 eyes (100%), as well as variously reflectivity particles located at the interface in all eyes examined after operation. The density of high reflective particles did not decrease significantly with time. In the middle stroma, the mean depth of acellular area was 44.01?10.67?m in all 68 eyes (100%). The appearance of morphology of subbasel corneal nerve fiber bundles after operation recovered. Conclusions Morphologic changes were found in central cornea. Acellular area, micro-folds as well as variously reflectivity particles located at the interface in all eyes existed in the central cornea for long time.
5.Effects of Estrogen Octylphenol on the Expression of Apoptosis Regulator Gene bcl-2 in Human Breast Cancer Cell Line MCF-7

Objective To explore the effects of octylphenol, an environmental contaminant with estrogenic activity, on the expression of apoptosis regulator gene bcl-2 mRNA in human breast cancer cell line MCF-7 and to compare its effects with 17?-estradiol (E 2). Methods MCF-7 cells were cultured in vitro and exposed to various concentrations of octylphenol and E 2 from 12 h to 120 h respectively. Total RNA was abstracted and the expression of bcl-2 mRNA was measured using reverse transcription polymerase chain reaction (RT-PCR) approach. Results The expression of bcl-2 mRNA in MCF-7 cells increased significantly after exposure to octylphenol and E 2 24 h and the effects continued to 120 h. The stimulation effect on the expression of bcl-2 mRNA was induced by octylphenol with a wide range of concentrations, and the strongest effect was found at the concentration of 10-6 mol/L of octylphenol. Conclusion The results suggested that octylphenol had similar estrogenic activities as E 2 on regulating the expression of bcl-2 mRNA in MCF-7 cells, but the effect was weaker than that of E 2.
6.Organization and management in hospital's humanistic services

7.Observation of the estrogenic activity of Octylphenol in vitro

Objective To observe the estrogenic activity of octylphenol(OP) in vitro and to conduct a preliminary study of its mechanism. Methods The estrogenic activity of OP was detected by cell proliferation test of MCF 7 cells in vitro and the mechanism was preliminarily studied by growth curve analysis, cell cycle analysis, tamoxifen(Tam) antagonistic test and apoptosis detection. Results OP was found to have estrogenic activity to stimulate the proliferation of MCF 7 cells. Cell cycle analysis revealed that the cell proliferation indexes of OP and 17? estradiol(E 2) were higher than those of alcohol. The estrogenic activities of OP and E 2 to stimulate the proliferation of MCR 7 could be antagonized by Tam. Both OP and E 2 could inhibit the cell apoptosis of MCF 7 cells. Conclusion OP possesses estrogenic activity to stimulate the proliferation of MCF 7 cells. The mechanism may be due to binding to the estrogen receptor, which may have effect on cell proliferation and apoptosis.
8.The adhesive mechanical properties of renal tubular epithelial cells on matrigel

purpose: to investigate the adhesive properties of renal tubular epithelial cells on matrigel and compared with the following three cases: ischemia、hypoxia and ischemia & hypoxia(I/H).materials and methods: A micropipette aspiration technique was adopted to determine the adhesive mechanics of renal tubular epithelial cells on matrige. results: it showed that the adhesion of renal tubular epithelial cells on matrigel was higher than that of those three model, further more, a different factor was followed by different adhesive mechanic. conclusion: the adhesion of I/H is lower, the ischemia is higher, but all were lower compared with control. It suggested that effect of hypoxia on adhesive properties of renal tubular epithelial cells on matrigel is bigger than that of ichemia.
9.Expression of apoptosis-related protein in epithelial dysplasia and squamous cell carcinoma.

OBJECTIVETo study the expression and significance of apoptosis-related protein p53, Bcl-2, and Bax during the development of oral squamous cell carcinoma (SCC).
METHODSThe expression was observed in 10 normal oral epithelia, 48 dysplasia epithelia and 42 SCC by immunohistochemical evaluation.
RESULTSIn normal mucosa, the positive rate of p53, Bcl-2 and Bax were 0%, 20% and 60%. In dysplasia epithelia, the positive rate of p53 is increased (P < 0.05), the positive rate of Bcl-2 and Bax remained no significant change (P > 0.05), but the positive intensity in severe dysplasia was higher than in mild group. In SCC, the positive rate of Bcl-2 increased significantly (compared with dysplasia, P < 0.05), while the expression of Bax was decreased with the increase of SCC histological grade. Further analysis showed the correlation was evident in p53 and Bax in dysplasia, and in p53 and Bcl-2 in SCC.
CONCLUSIONSIn dysplasia, p53 gene mutation results in accumulation of dysplasia cells. In SCC, the cooperation of p53, Bcl-2 and Bax results in the progression of SCC. Apoptosis genes could work either independently or cooperatively.

Inthis study, the DNA fragment encoding the regions Ⅰ to Ⅱ of CSP gene from Plasmodium falciparm isolate FCC1/HN was cloned into an. expression vector pcDNA3 contained cytomegalovius (CMV) and transformed into human Hela cell line. The expressed protein PfCSP (condidate vaccine) was used to immunize BALB/c mice by subcutaneous、 intravenous or intraperitoneal administration respectively. The splenocyte of BALB/c mice immumized with the condidate vaccine released significantly IL-2 and IFN-γ following stimulation with this vaccine. It is associated with increase of the splenic T lymphocyte proliferation stimulated by this vaccine and enhancement of NK cell killing activity in the former studies. These results suggested that the vaccine could stimulate T cell response and enhance the cell-mediated immunity.
