1.Theory of selectivity of RP-LC C18 column and its application

In recent years, more and more researchers focus on the selectivity of RP-HPLC columns. Among these columns, C18 column is the most commonly used the column. Yet even the same filler can not give us the same separation effect with the columns of different brands and different types, therefore, it is important to choose the suitable HPLC column for carrying out an experiment. After study in recent years, some column classification, selection methods (e.g.: column parameter method, F value method) have been achieved and contributed to the finding of suitable columns and reducing the blindness of column selection. This paper summarizes the advantages of these methods and their limitations in the application process, and gives some advices and expectations to pharmaceutical analysis works based on the works of other researchers.
2.Determination of Glycyrrhizic Acid and Chlorogenic Acid by HPLC

A method for the determination of glycyrrhizic acid and chlorogenic acid in Chinese medicinal preparation by RP-HPLC-UV was described. Mobile phase: methanol (0.5%HAc):H2O(0.5%HAc); C18 column and externat standard was used.
3.The association of occupational stress, social support, type D personality with physical health of nurses

Objective To investigate the association of occupational stress,social support,type D personality and physical health of nurses.Methods Cross-sectional study design was adopted.We used cluster sampling to investigate 1 287 nurses serving in one tertiary hospital and two second-class hospitals in Guangzhou.The questionnaire was anonymous answered.The content of questionnaire included the demographic data,occupational stress of nurses,social support,type D personality and physical health,etc.SPSS 13.0 was used for statistical analysis,including descriptive analysis,Pearson correlation analysis and path analysis.Results 1 192 of questionnaires were effective.The occupational stress was negatively related with the physical health status and social supports,and social support was positively related with the physical health status.The type D personality was negatively related with social supports and the physical health status,while positively related with occupational stress.Conclusions Occupational stress can make directly negative effect on the physical health.Social support has a directly protective effect on physical health.Type D personality makes direct and indirect negative effect on the physical health of nurses.
4.Molecular pathophysiological basis of the ocular albinism type 1

Ocular albinism type 1 (OA1),the most form of the ocular albinism,is an X-linked disorder mainly characterized by a severe reduction of visual acuity,hypopigmentation of the retina,photophobia,strabismus and nystagmus. The OA1 gene is located on chromosome Xp22.32 and the coding sequence is divided into nine exons. The OA1 gene codes for a 404 amino acid protein thought to be a melanosomal transmembrane glycoprotein. The OA1 protein is similar to the G protein-coupled receptors,but it's exact function is not clear. There are many mutations and deletions of the OA1 gene have been found.
5.Study on association between work-family conflict and turnover intentions

Objective To explore the relationship between work-family conflict and turnover inten-tions in nurses and find out the key points to reducing turnover intentions by releasing individuals' work-family conflict. Methods Cross-sectional study method was used in this investigation. By using cluster sampling method, 1287 nurses from some top-level and third-class general hospital were investigated. The questionnaire of work-family conflict and turnover intentions scale were used in this research and the re-suits underwent analysis. Results The score of turnover intentions in nurses was positively correlated with the level of their work-family conflict (r=0.233,P<0.01). Stress,behavior and time dimension were related factors of turnover intentions(r=0.243,0.191,0.176,P<0.01 ).After control of demographic factors, the turnover intentions deviation that can be explained increased by 7%. Conclusions The score of work-family conflict is one of the predicators of turnover intentions. A developing measure should be taken to reduce turnover intentions by releasing work-family conflict in nurses.
6.High cell-density fermentation of recombinant human insulin-like growth factor-1 engineering bacteria

BACKGROUND:Insulin-like growth factor-1(IGF-1)is an important cell factor which plays a special role in many disease treatments such as diabetes.The research of engineering bacteria fermentation production technology is great to IGF-1 industrialization and to clinical application.OBJECTIVE:To investigate the high cell-density fermentation and expression condition of recombinant human insulin-like growth factor-1(IGF-1) engineering bacteria.DESIGN,TIME AND SETTING:The enzyme,gene engineering,study was performed at the Biotechnological Laboratory of Zhejiang Shuren University from May 2007 to May 2008.MATERIALS:Recombinant E coli strain for IGF-1 expression as BL21 (DE3)/pET22a-IGF-1 was reserved in the Biotechnological Laboratory of Zhejiang Shuren University.Nutrient feed was composed of:glucose(300 g/L),peptone(40 g/L),yeast powder(10 g/L),Na2HPO4(280 mmol/L),Na2HPO4-2H2O2(120 mmol/L),MgSO4(10 mmol/L),and ampicillin(100 mg/L).METHODS:The strains were activated and then cultured in orbitaI shakers.Parameters such as types of media,isopropyl-β-D-thiogalactopyranoside(IPTG)concentration and induction time have been analyzed to explore optimaI fermentation conditions for expressing the recombinant protein.According to the optimal fermentation condition of orbitaI shakers.batch fermentation was carried on with 5 L-autocontroI fermentor.The process contained two stages:batch culture and fed-bafch through pH-stat solution.JPTG was added to jnduce the expression of protein in the middle and latter of the logarithmic growth phase for 4-βhours.MAIN OUTCOME MEASURES:Following described parameters were measured:fermentation and protein expression of the recombinant human IGF-1 engineering bacteria,cell concentration,cell dry weigh,objective protein,glucose concentration.RESULTS:Cells were cultured in 2×YT+5 g,L glucose medium,induced by 0.8 mmol/L IPTG for 5 hours.By controlling dissolved oxygen and by pH-stat feeding solution,high cell-density and high protein expression were achieved.Under the establishedconditions.50.1 g dry cell weight(DCW)/L could be obtained,and 5.25 g/L IGF-1 was achieved.CONCLUSION:The established high cell-density fermentative procedure of recombinant human IGF-1 engineering bacteda is the useful bases for further pudfication and large scale production of IGF-1.
7.Development and application of reference materials containing mixed degradation products of amoxicillin and ampicillin.

Reference materials containing mixed degradation products of amoxicillin and ampicillin were developed after optimization of preparation processes. The target impurities were obtained by controlled stress testing, and each major component was identified with HPLC-MS and compared with single traceable reference standard each. The developed reference materials were applied to system suitability test for verifying HPLC system performed in accordance with set forth in China Pharmacopeia and identification of major impurities in samples based on retention and spectra information, which have advantages over the methods put forth in foreign pharmacopoeias. The development and application of the reference materials offer an effective way for rapid identification of impurities in chromatograms, and provide references for analyzing source of impurities and evaluation of drug quality.
8.Influence of Shenfu injection(参附注射液)on tumor necrosis factor-?, interleukin-2 and lung function in patients with chronic obstructive pulmonary disease at acute exacerbation stage

Objective To evaluate the clinical therapeutic effect of Shenfu injection (参附注射液) on patients with chronic obstructive pulmonary disease at acute exacerbation stage (AECOPD) and approach its therapeutic mechanism. Methods Fifty-eight patients with AECOPD were divided into a Shenfu group (30 cases) and a control group(28 cases); the Shenfu group received 50 ml Shenfu injection and 5% glucose 250 ml intravenous drip once a day at the base of conventional therapy,while the control group received conventional therapy only. The therapeutic course of both groups was 14 days. The clinical therapeutic effects of both groups were observed, the serum levels of tumor necrosis factor-? (TNF-?), interleukin-2 (IL-2) were measured by enzyme-labeled immunosorbent assay (ELISA), and the lung function of both groups was measured before and after the treatment. Results The total effective rates was 93.3% (28/30) and 85.7% (24/28) in the treatment group and the control group, respectively. In the comparison before and after treatment, the levels of TNF-? in both groups were decreased obviously (both P0.05). Conclusion Shenfu injection has a definite clinical curative effect in the treatment of AECOPD, it may decrease obviously the serum level of TNF-?, increase the level of IL-2 and improve the lung function in the patients with AECOPD.
9.Genotyping and Polymorphism Analysis of Plasmodium falciparum Isolates by Nest-PCR and PCR-RFLP

Objective To analyze the efficiency and specificity of MSP2 alleles genotyping for Plasmodium falci-parum isolates by Nest-PCR and PCR-RFLP. Methods MSP2 alleles from Plasmodium falciparum isolates of Yunnan and Hainan were genotyped by Nest-PCR and PCR-RFLP, respectively, and the efficiency and specificity of the two me-thods were analyzed. Results The conventional Nest-PCR method could detect 79.8% (166/208) alleles of MSP2,and 65.7% (65/99) for 3D7 family, but could not identify the type of any allele. While PCR-RFLP showed 25.3% higher genotyping efficiency than Nest-PCR. Moreover, this method could identify the allele types. Conclusion PCR-RFLP genotyping technique is more efficient and specific than conventional Nest-PCR, and it is a convenient tool in the study on molecular epidemiology of malaria.
10.UV-Visible Spectrophotometric Assay for Saikosaponins in Bupleurum Chinense DC

Objective To set up a simple and accurate method for the determination of saikosaponins in Bupleurum chinense DC. by UV-Visible Spectrophotometry. Methods The content of saikosaponins was determined by UV-Visible Spectrophotometry. The absorbency was measured at 536nm. Results The calibration curves was linear within 194~1940mg/L(r=0.9996). The average recovery was 97.35%,and the RSD was 1.02%(n=9). Conclusion The method was proved to be simple, precise and reproduciable. It can be used for the quantitative determination of B upleurum chinense DC.
