Purpose: To study the mechanism of chemoprevention of colorectal cancer by aspirin. Methods: Different concentrations of aspirin have been used in the culture of colonic cell-Caco-2, its proliferation and apoptosis was observed during the culture and the expression of Fas/FasL mRNA was assayed by RT-PCR. Results: Major portion of cells died in the culture medium that was added high concentration of aspirin, but the lower concentration promoted apoptosis. The Fas mRNA was expressed in cells added with 25 mmol/L aspirin, but not in that of 1640. The FasL mRNA expressed in 25mmol/L aspirin was lower than in 1640. Conclusions: The lower concentration of aspirin can promote apoptosis of coloniccell, the mechanism has correlation with induction of the expression of Fas, and down-regulation of the expression of FasL.

Purpose: To study the immunophenotype and the expression of cytotoxic granule proteins of nasal NK/T cell lymphoma and its significance. Methods: 44 cases of nasal NK/T cell lymphoma were studied by the two-step method of DAKO EnVisionTM using a series of antibodies including CD3, CD20, CD43, CD45, CEI45RO, CD56, CD57, CD79?, TIA-1, granzyme-B and perform. Results: All the cases of nasal NK/T cell lymphoma were CD45 positive. 43% of the cases expressed CD3 with positive signal located in the cytoplasm, which was different from peripheral T cell lymphoma. 45% and 52% of the cases were CD43 and CD45RO positive respectively. Cases that reacted to CD56 accounted for 52% of the cases, 43% of which were also positive to CD3. Concerning the reactions to both CD3 and CD56, 10 cases showed CD3 + CD56 + , 13 showed CD3-CD56 + , 9 were CD3 + CD56- and 12 were CD3-CD56-. None of the 44 cases showed positive reaction to CD20, CD79? and CD57. All cases were reactive to TIA-1. 93% and 95% of the cases showed the reactions to granzyme-B and perform. All the controls were negative to TIA-1, granzyme-B and perform. Conclusions: The immunophenotypes of nasal NK/T cell lymphoma showed less consistency. CD56 was not always positive in the cases of this tumor. The different locations of the positive signal to CD3 showed that the cell lineage of this tumor was different from T lymphocytes. The high frequency of the staining by cytotoxic granule proteins, TIA-1, granzyme-B and perform, showed that these cells may have originated from NK cells. The distinctive differences in immunohistochemical staining of cytotoxic granule proteins in nasal NK/T cell lymphoma make their detection very useful and important in diagnosis and differential diagnosis.

Objective To study the dose rate distribution on cultured cell plane and establish a reference dose rate table of in vitro cell line ~(125)I seed irradiator. Methods Thermoluminescence dosimetry(TLD) was used to measure the irradiation dose rate of a single 6711 model(~(125) I) seed with apparent activity of 10.323 MBq in water at point P.Meanwhile, the theoretic value of the irradiation dose rate at point P was calculated with theoretic formula. The difference between the calculated and observed values within 10% was set as standard to analyze the accuracy of the measurement. The irradiation dose rate of a single 6711 model(~(125) I) seed was measured in 1mm-thick polystyrene + water medium at point P.The value was applied to differential or non-differential proof along with the value from water medium to study the effect of 1 mm thick polystyrene on distribution of irradiation dose in water. Finally,by simulating the(~(125) I) seed plane irradiator with nine(~(125) I) seeds, the distribution table of irradiation dose rate on the cultured cell plane was calculated with theoretical formula. Results The observed value(n= 10) of irradiation dose rate with one(~(125) I) seed in water at point P was(0.359?0.023)cGy/h and the calculated value was 0.347cGy/h,the difference was within 10%.The observed value(n=10) of irradiation dose of one(~(125) I) seed in 1mm-thick polystyrene + water medium at point P was (0.350?0.027)cGy/h,which showed no statistical difference from the observed value in water under differential and non-differential proof. The reference table on dose rate distribution for cells exposed to(~(125) I) seed irradiation in vitro was developed. Conclusions 1mm-thick polystyrene gives no significant effect on irradiation dose rate distribution from(~(125) I) seeds in water.A reference table on the dose rate distribution for cells exposed to(~(125) I) seed irradiation in vitro has been developed, which can be used to determine an optimal irradiating strategy for future work.

China Oncology 2001;0(05):-
Background and purpose: About 20 cases of BML have been reported in the literature, they occurred in females with history of uterus leiomyoma.BML appears as a histopathologic benign smooth muscle tumor located far from the uterus, often in the lungs.Firstly,it should be confirmed as originating from smooth muscle by immunohistochemistry,Secondly, its primary tumor should be confirmed as benign, with mucoid degenenation,it also should be distinguished from myxoid leiomyosacroma. Methods:We report a case of benign metastasizing leiomyoma, study its clinical, histopathologic and immunohistologic features, discuss the diagnosis and differential diagnosis and give a review of the related references. Results:The case occurred in a forty-two year old female who was found to have bilateral lung nodules after hysterectomy of her uterus leiomyoma six years ago.An open lung biopsy revealed the nodules to consist of proliferating smooth muscle cells with a histopathologic feature similar to her uterus leimyoma, consistent with the diagnosis of BML.Positive staining of estrogen and progesterone receptors was detected in both the uterus leiomyoma and the metastasizing lesions. No significant increase or enlargement of the lung nodules were found seven months after the operation with no therapy except taking Chinese traditional medicine. Conclusions:BML is a rare entity usually occurring in females with history of uterus leiomyoma. It appears as a histopathologic benign smooth muscle tumors distant from the uterus. It is hormone dependent and progresses slowly.

Objective To calibrate and assess the dose distribution of 125 I seed brachytherapy. Methods Twenty 125 I seeds, each with activity of 12.2 MBq, were implanted on the circumference of a circle 15mm across in a phantom. Into a designed prostate model, 70 125 I seeds were implanted in four planes. The absorbed dose rate of the target volume was monitored by Farmar 2570 dosimeter and thermoluminescence dosimeters (TLD) with the isodose curves drawn on Kodak films. Results The central dose rates of the circular target volume assessed by Farmar 2570 and TLD were 8.4, 7.9 cGy/h in the phantom and 12.0, 11.1 cGy/h in the prostate model. For the target volume of the prostate model, the total absorbed dose was 24?219 cGy. The dose rate 4 cm from the prostate cancer as shown by the isodose curves was only 10% of the central dose rate. Conclusion The central dose rate of target volume measured by the two methods are similar.

Journal of Practical Stomatology 2010;26(1):33-37
Objective: To observe the biocompatibility of acellular nerve scaffold (ANS) via three sterilization methods, to provide experimental data for tissue engineering industrialization. Methods: Pig sciatic nerves were cut and treated using the NaOH maceration method. ANSs were sterilized by ethylene oxide, ~(60)Co-irradiation and peracetic acid. Evaluated the biocompatibility by MTT, cellular compatibility test, collagenase susceptibility test in vitro and local implantation test. Results: ANS retained the integrity of structure and major components of the basement membrane. The result of MTT test showed that the ANSs via different sterilization methods had statistical differences. There were no overall significant differences in Collagenase susceptibility test. Scanning electron microscope results showed the skin fibroblasts could attach, proliferate and grow well on the surface and holes of ANS with sterilization of PAA and Co~(60),a small quantity of cells adhered on ANS with sterilization of ETO. Tests for local effects after implantation show that different sterilization methods don't effect the ability of ANS to resist the enzyme degradation. In ETO group, rats showed an acute inflammatory response followed by chronic inflammation. In PAA and ~(60)Co group rats showed an acute inflammatory response that diminished such that the graft ultimately became indistinguishable from native tissue, observations that were consistent with graft acceptance. Conclusion: Peracetic acid sterilization offers a convenient alternative protocol for ANS processing. ANS sterilized with PAA shows good compatibility and biologic safety. It is an ideal sterilization method for ANS.
