1.The clinical significance of total serum bile acid test in evaluation of the prognosis of posthepatitis cirrhosis

Objective To explore the clinical significance of serum total bile acid (TBA) test in evaluation of the prognosis of posthepatitis cirrhosis.Methods 92 patients with posthepatitis cirrhosis were selected as the observation group,and divided into A,B,C groups according to the Child-Pugh grades.32 healthy people were selected as the control group.The level of serum TBA was detected and compared.Results The level of TBA in the observation group was (84.26 ±49.03) μmol/L,which was higher than that in the control group[(3.52 ± 2.67) μmol/L](t =17.53,P < 0.01).The level of TBA was gradually increased in Child-Pugh grade A,B,C grade,and the differences were statistically significant (t =3.84,6.21,5.49,all P < 0.05).Conclusion TBA test is benefit for the diagnosis and prognosis of posthepatitis cirrhosis.
2.Study on the expression of prolactin mRNA in thyroid glands of patients with Graves'disease

Abstract Objective:To investigate whether mRNA of prolactin(PRL) ,an important immunomodulatory hormone,is expressed in thyroidglands of patients with Graves'disease(GD) .Methods:The expression of PRL mRNA was examined by nonisotopic in situ hybridization in GDthyroid glands and the control ones( including multinodular goiters and samples of normal thyroid tissue adjacent to adenomas) as well as intra-thyroidal mononuclear cells isolated from GD thyroid glands and primary thyroid follicular cell(TFC) cultures derived from them.Results:PRL mRNA was expressed in GD thyroid glands, but not in the control ones. It was localized in infiltrating mononuclear cells and vascular endothelial cells adjacent to mononuclear cell infiltrates, but not in TFC .(9.8?.3)% (x ?s) of intrathyroidal mononuclear cells isolated from GD thy-roid were found containing PRL mRNA.It was absent in the primary TFC cultures. Conclusion:PRL mRNA is expressed in GD thyroid tissue. This indicates that PRL-like substance can be locally produced in GD thyroid tissue, which may play an important role in GD pathogenesis by autocrine and paracrine mechanisms.
3.Effects of iptakalim on the functions of endothelin system in human pulmonary artery endothelial cells

Aim To investigate the effects of iptakalim(IPT),a novel K_(ATP) opener,on the functions of endothelin system in human pulmonary artery endothelial cells.Methods Primary cultured human pulmonary artery endothelial cells were incubated with different concentrations iptakalim for 24 h.The levels of ET-1 in medium were observed by radioimmunoassay.Reverse transcription polymerase chain reaction(RT-PCR)was performed to analyze the expression of ET-1 and ECE.Results When endothelial cells were incubated with IPT at concentrations above 10 μmol·L~(-1),the levels of ET-1 release in medium and the levels of ET-1 mRNA were significantly inhibited.When endothelial cells were incubated with IPT at concentrations above 1 μmol·L~(-1),the levels of ECE mRNA were significantly inhibited.Conclusions IPT can inhibit the expression of ET-1 and ECE mRNA from human pulmonary artery endothelial cells, thus it inhibits the secretion of ET-1 from endothelial cells. Iptakalim may serve as a promising candidate drug to treat pulmonary hypertension.
4.Feasibility of renal hypothermia achieved by retrograde ice-cold saline infusion with perfusion pump

Objective To study the feasibility of renal hypothermia achieved by retrograde icecold saline infusion with perfusion pump. Methods Twenty-one patients who received the open radical nephreetomy were divided into three groups,A group with ice-slush renal hypothermia,B group with gravitational retrograde ice-cold infusion,C group with retrograde ice-cold infusion by perfusion pump.After the kidney was dissected and the ureter was divided,the renal vasculars were clamped.The kidney was cooled by three methods respectively.The temperature of renal parenchyma was monitored for 15 min. Results Fifteen minutes later,the temperature of renal parenchyma of A group declined from 34.4℃ to 5.4℃,B group from 34.8℃ tO 23.8℃,C group from 35.1℃ to 22.3℃.Coneiusions Renal hypothermia can be achieved by retrograde cold saline infusion and the perfusion pump may accelerate the speed of cooling.
5.Effect of CM on Interventional Therapy on Middle and Late Stage Malignant Tumors

Objective To determine the clinical use of CM on interventional therapy for middle and late stage cancer.Methods We observed the effect of clinical administration on the patients with interventional therapy or inteventional therapy plus CM,control with themselvies.Results 38(76%) out of 50 patients got better than that of preadministration,cancer mass decreased.Conclusion CM can increase the sensitivity of carcinoma cells to chemicals and decrease its sideeffect.
6.Purification and enzyme activity assay of filamentous temperature-sensitive protein Z in Streptococcus suis serotype 2

We conducted purification of filamentous temperature-sensitive protein Z of Streptococcus suis serotype 2 (S.suis 2) and measured its GTPase activity.The ftsz gene in the genome of the Chinese 05ZYH33 strain of S.suis 2 was successfully amplified using PCR,and then the ftsz gene was cloned into prokaryotic expression plasmid pET28a,and the recombinant plasmid pET28a-ftsz was transformed into E.coli BL21.After induction by IPTG,the isolated FtsZ protein was analyzed with SDS-PAGE.Then the recombinant protein was purified by Ni2+-NTA affinity chromatography.The rabbit serum was harvested after immunization with recombinant FtsZ protein,and was analyzed by indirect ELISA and Western blotting.The GTPase activity of FtsZ was measured with the malachite green method.Results showed that successfully constructed recombinant plasmid pET28a-ftsz and the recombinant protein with high purity was obtained;Western blot result indicated that FtsZ could react with the His-tag antibody and the rabbit serum;the polyclonal antibody titer of the rabbit serum reached 1 ∶ 13 107 200;FtsZ have GTPase activity.We successfully prepared S.suis 2 recombinant protein FtsZ having GTPase activity and high titer antiserum would be useful for the further study of S.suis 2 cell division mechanism.

In view of the challenges and opportunities presented in the new century and after rational deliberations on five occasions, the completely new model of "hi tech+humanistic solicitude=the green hospital" was put forward and the overall framework of "one line of thought", "two cornerstones", and "three goals" was carefully formulated. In the meantime, all staff members of the hospital were called on to be involved in the "six major activities". As a result, great changes have taken place in the appearance and development of the hospital. It has been proved through practice that "the green hospital" is a successful model conforming to the trends of the times.

Objective To investigate the effect of sleeve gastrotomy (SG) and gastric plication (GP) on the short-term metabolism in type 2 diabetes mellitus (T2DM) Zucker fatty rats.Methods Thirty healthy male rats were randomly allocated into three groups:Sham,SG and GP groups,and devices like metabolic cage were applied to detect the metabolic changes in the third week after operation.Results The body weight of the SG and GP group decreased,but no statistical significance was found when compared with that of the Sham group.Less food consumption were found in SG (P<0.05) and GP (P<0.05) groups compared with that of the Sham group.In the daytime,the energy expenditure of SG group was higher than that of the sham group(P<0.01),while in the night,the energy expenditure of both the SG (P<0.001) and GP (P<0.001) groups were higher than that of the Sham group.In the night,the activity level of SG (P<0.05) and GP (P<0.05) groups were lower than that of the Sham group,and activity level in the night was lower than that in the daytime for SG group(P<0.05).In the daytime,the respiratory exchange rate (RER) of the SG (P<0.001) and GP (P<0.001) group were lower than that of the Sham group,and the night RER of GP group was higher than that in the daytime (P<0.05).No statistical significance was found in terms of the average speed among the three groups.The average speed of the Sham (P<0.05) and SG (P<0.05) group in the night were faster that in the daytime.The fasting plasma glucose (FPG) of SG and GP group were improved compared with that of the Sham group.Conclusion SG and GP procedures can improve the metabolism of diabetic rats in the short term.
9.Analysis of preventive measures and pathogenic related factors of peri-odontitis

Objective To investigate preventive measures and pathogenic related factors of periodontitis. Methods A total of 138 patients with periodontitis from August 2012 to March 2014 in our department were selected as the experi-mental group. Meanwhile 138 healthy cases matched with the patients within 2 years age difference in the same regions were selected as the control group. Questionnaire survey was used to inquire the basic information, educational status, economic income, acquisition of knowledge in periodontal disease, brushing habits, negative life events and so on. The differences in results between two groups were compared. Results Among 138 patients with periodontitis, there was 1087 disease parts on maxillaries, 872 parts on mandibles, 1243 parts on molars, and 876 parts on anterior teeth and bicuspids. According to Amaitage standard, 138 patients with periodontitis, the rate of mild to moderate periodontitis patients was 81.16% (112 cases), and the rate of severe periodontitis was 18.84% (26 cases). Acquisition of knowledge in periodontal disease, smoking, the frequency and sustained time of brushing per day, negative life events and mouth care between two groups showed statistical significance(P<0.05). There was no significant difference between two groups in age, gender and educational status(P>0.05). Non conditional Logistic regression results showed that: independent pathogenic risk factors of periodontitis included acquisition of knowledge in periodontal disease, the frequency and sus-tained time of brushing per day, negative life events and regular mouth care, and regular mouth care ranked first among these factors(the odds ratio=3.102, P<0.05). Conclusion Periodontitis is the result of interaction of multiple risk factors, therefore, comprehensive measures especially regular mouth care should be adopted to prevent the occurrence of peri-odontitis.
10. Clinical and prognostic values of TP53 mutation in patients with acute myeloid leukemia

Objective:
To explore the clinical and prognostic values of TP53 gene mutation in patients with acute myeloid leukemia (AML) .
Methods:
A retrospective analysis of 265 newly diagnosed AML patients with next-generation sequencing (NGS) data in the Hematology Department of Changhai Hospital from January 2010 to January 2019 was performed. Mutation analysis was carried out by targeted sequencing technology including 200 hematological malignancy related genes. The association of TP53 mutation with clinical features was analyzed.
Results:
Alterations in TP53 were found in 20 (7.5%) patients, including 17 case (6.4%) of missense mutations, 2 cases (0.7%) of frame-shift deletion mutations and 1 case (0.4%) of splicing sites mutation. A total of 23 kinds of TP53 mutations were detected, most of them (16, 69.6%) were located in the DNA binding domain of exon 5-8, 4 in the DNA binding domain of exon 3-4, 2 in exon 10 and 1 in splice site, respectively. The median age of patients with TP53 alterations was higher than those without [52 (26-72) years old
