1.Expression and function of CD226 with CD16 on NK subsets

Objective:To investigate the expression and function of CD226 on NK s ubsets and its coexpression with other activation receptor and inhibition recept or on NK cells. Methods:The expression of CD226 on CD56 bright and CD56 dim NK subsets and coe xpress ion with CD16 and NKG2A in PBMC and MLC in the presence or absence of IL-2 or IL-15 were detected by double fluorescent staining and flow cytometry analysis. The level of IFN-? in the supernatants of PBMC culture and MLC treated with o r without IL-2 or IL-15 were evaluated by ELISA. 51Cr release assay w as employed to measure the specific lysis of NK cells killing target K562 cells. Results:CD226 was mainly expressed on CD56 dim NK subsets in PBMC. When s imulated by IL-2, CD226 expression was shifted to CD56 bright NK subsets, while IL-15 increased the subpopulation of NKG2A+CD226+double positive cell s. In MLC-generated NK cells, CD226 was mainly expressed on CD56 dim NK su bsets, and also shifted to CD56 bright NK subsets in the addition of IL-15 . Furt hermore, the percentage of CD16+CD226+和NKG2A+CD226+ subsets were increa sed when stimulated by IL-2 or IL-15. There was great increase in IFN-? lev el in the supernatants of PBMC culture in the presence of IL-2 or IL-15, but no difference in the supernatants of MLC treated with or without two cytokine s. Moreover, the cytotoxicity of NK cells in PBMC and MLC were greatly enhanced by IL-2 or IL-15. Conclusion:CD226 is mainly expressed on CD56 bright NK subsets in IL-2 or IL-1 5 activated NK cells, and is coexpressed with CD16 and NKG2A preferentiatly, whi ch maybe involve in the modulation of cytotoxicity of NK cells based on the balance of coexpressed activation and inhibition receptors. 2.Comparison of effects of propylthiouracil and methimazole on pregnancy outcomes and glucose and lipid metabolism in patients with hyperthyroidism in pregnancy

Objective To compare the effects of propylthiouracil and methimazole on pregnancy outcomes and glucose and lipid metabolism in patients with hyperthyroidism in pregnancy, and to analyze the security.Methods Totally 92patients with hyperthyroidism in pregnancy were divided into the observation group (46cases) and control group (46cases).Patients in both groups were given routine adjuvant therapy, such asβ-blocker, liver protection, increasing white blood cells.The patients in observation group were treated with propylthiouracil while the patients in control group were treated with methimazole.The thyroxine levels (TSH, T3, T4, FT3, FT4), pregnancy outcomes, glucose and lipid metabolism (FPG, FINS, TC, TG), liver function indicators (ALT, ALP, AST) and maternal complications of two groups were compared after treatment for 3months.Results The TSH level of the observation group was significantly higher than that of the control group, and the T3, T4, FT3, FT4levels of the observation group were significantly lower than those of the control group (P<0.05).The incidence of premature birth, miscarriage, intrauterine fetal death, fetal distress and gestational hypertension of observation group were significantly lower than those of the control group, but the incidence of full-term production of observation group was significantly higher than that of the control group (P<0.05).The FPG, FINS, TC and TG levels of the observation group were significantly lower than those of the control group (P<0.05).The increased amplitude of ALT, ALP, AST of the observation group were significantly lower than those of the control group (P<0.05).There were puerperal infection, hyperthyroid heart disease, diabetes, oligohydramnios, placental abruption, pregnancy-induced hypertension and other complications occurred in both two groups, but the incidence of the observation group were significantly lower than those of the control group (P<0.05).Conclusion Compared with methimazole, propylthiouracil is more beneficial to improve thyroid function, pregnancy outcome and glucose and lipid metabolism in patients with hyperthyroidism in pregnancy, and has higher security.
3.The preliminary study of the origin characters of snore in simple snorers.

OBJECTIVE:
to investigate the origin characters of snore in simple snorers and provide the basis for its treatment.
METHOD:
Thirty-two simple snorers diagnosed by polysomnography were induced to sleep by propofol and dexmedetomidine, then we observed the vibration sites, pattern and concomitant collapse of soft tissue in pharyngeal cavity by nasendoscopy.
RESULT:
Thirteen cases showed palatal fluttering only, and 1 case showed vibration of epiglottis only. Six cases showed palatal fluttering with vibration of epiglottis, and 2 cases showed palatal fluttering with vibration of epiglottis and tongue base. Five cases showed palatal fluttering with vibration of pharyngeal lateral wall, and 5 cases showed palatal fluttering with vibration of lateral wall, epiglottis and tongue base together. Palate and pharyngeal lateral wall vibrated strongly and always collapsed with vibrating, but epiglottis and tongue base usually vibrated slightly and seldom collapsed.
CONCLUSION
The palatal fluttering is the main source of snoring sounds for most simple snorers, then followed by vibration of palatal and pharyngeal lateral wall together. The site of collapse in pharyngeal cavity is consistent with the main site of vibration.

4.Effects of Transcutaneous Electrical Acupoint Stimulation on Hand Dysfunction after Stroke

Objective To observe the effects of transcutaneous electrical acupoint stimulation (TEAS) on hand dysfunction after stroke. Methods From March, 2013 to June, 2015, 56 cases of stroke with hand dysfunction were divided into group A (n=28) and group B (n=28). Both groups received basic rehabilitation, while group B received TEAS in addition, for six weeks. They were evaluated with Brunnstrom Grades, Manunl Muscle Test (MMT), Fugl- Meyer Assessment (FMA) of fingers, Motor Status Scale (MSS), modified Ashworth Scale (MAS), National Institutes of Health Stroke Scale (NIHSS), Motor Hand Functional Status Score and Barthel Index (BI). Results The scores of FMA of fingers, MMT of wrist flexion, MSS, MAS and BI were more in group B than in group A (t>2.2527, P<0.05), and the score of NI-HSS was less in group B (t=3.556, P<0.001). There was no significant difference between two groups in the score of Motor Hand Functional Status Score and MMT of wrist extension (t<0.310, P>0.05). Conclusion TEAS can promote the recovery of hand function and the activi-ties of daily living in patients after stroke.
5.The effect of prunella vulgaris extract on inflammatory response and peritoneal macrophages in septic mice

Objective To investigate the effect of prunella vulgaris extract on inflammation,macrophage phenotype,and phagocytic ability in septic mice,and analyze whether Toll like receptor 4(TLR4)/nuclear factor-κB(NF-κB)signaling pathway involved in its mechanism.Methods C57BL/6 mice were divided into the control group,the model group and the prunella vulgaris extract low(25 mg/kg),medium(50 mg/kg)and high(100 mg/kg)dose groups.Except for the control group,all other groups of mice were injected intraperitoneally with lipopolysaccharide(LPS)to prepare sepsis model.Each group was given corresponding medication by gavage.After 24 hours of administration,serum tumor necrosis factor-α(TNF-α),interleukin(IL)-1β,high mobility group protein B1(HMGB1),IL-10 levels,the proportion of M1 type(CD11b+F4/80+)and M2 type(CD206+F4/80+)macrophages in peritoneal macrophages,the phagocytotic capacity of macrophages,the expression of inducible nitric oxide synthase(iNOS)messenger RNA(mRNA)and arginase 1(Arg1)mRNA in peritoneal macrophages and expression levels of TLR4,NF-κB p65 and their phosphorylated proteins in macrophages were detected.Results Compared with the control group,serum TNF-α,IL-1β,HMGB1,proportion of M1 type macrophages in abdominal cavity,mean fluorescence intensity and phagocytotic capacity of macrophages,iNOS mRNA,TLR4,phosphorylated NF-κB p65(p-NF-κB p65)/NF-κB p65 protein expression were increased in the model group(P<0.05).IL-10,proportion of M2 type macrophages and Arg1 mRNA expression were decreased(P<0.05).Compared with the model group,serum TNF-α,IL-1β,HMGB1,proportion of M1 type macrophages in abdominal cavity,iNOS mRNA,TLR4,p-NF-κB p65/NF-κB p65 protein expression were decreased successively in the prunella vulgaris extract low,medium and high dose groups(P<0.05).IL-10,proportion of M2 macrophages,mean fluorescence intensity and phagocytotic capacity of macrophages and Arg1 mRNA expression were increased successively(P<0.05).Conclusion By inhibiting TLR4/NF-κB pathway,prunella vulgaris extract may inhibit the polarization of peritoneal macrophages into M1 type and promote their polarization to M2 type,enhance macrophage phagocytic ability and alleviate LPS induced inflammatory response in septic mice.
6.Generation of GABAergic interneuron-specific PGC-1α knockout mice.

OBJECTIVETo generate mice which are specific for peroxisomproliferator-activated receptor-γ coactivator-1(PGC-1α) knockout in the GABAergic interneuron.
METHODSConditional mice specific for PGC-1αwere introduced from the Jackson Laboratory, USA and initially inbred to obtain homozygote PGC-1αmice. The PGC-1αconditional mice were further crossed with Dlx5/6-Cre-IRES-EGFP transgenic mice to achieve specific knockout of PGC-1α in the GABAergic interneuron.
RESULTSThe offspring with specific knockout PGC-1α gene were successful for the generation of GABAergic interneuron, with the resulting genotype being PGC-1α.
CONCLUSIONThe PGC-1αmice were obtained through a proper crossing strategy, which has provided a suitable platform for studying the function of PGC-1α in neuropsychiatric diseases.

7.Fear memory and heart rate and blood pressure during conditioned fear in rats with hippocampal lesion or fimbria-fornix transection

Objective To investigate the effects of dorsal hippocampal lesions (DH) or fimbria-fornix transection (FF) on the learning and memory of conditioned fear and the heart rate and blood pressure in rats.Methods Nineteen male adult Wistar rats were used in this experiment.They were implanted telemetry sensors in their abdominal aortas.Two week later,six of the rats were subjected to permanent NMDA-induced neurotoxic lesions to the dorsal hippocampus (DH) and seven for the fimbria-fornix transection (FF)through stereotactic brain surgery,the left six were treated with saline as the control (Sham).All rats were subjected to a conditioned fear experiment.Meanwhile,changes in heart rate and blood pressure were measured.Results There was no significant difference in heart rate and blood pressure among the rats with the hippocampal operation or fimbria-fornix transection.In the acquisition of conditioned fear,there were significant difference in freezing time among the three group in both inter-trial-interval (ITI) and conditioned stimulus (CS) process (all P<0.05).The freezing time of the FF group showed significantly lower than that of the Sham group (P<0.05).There was no significant difference in heart rate and blood pressure among the three group(P>0.05).In the test of conditioned contextual fear memory,the freezing time percentage in the FF group ((0.31±0.16) %) significantly lower than that in the Sham group ((2.78± 1.23) %) (P<0.05)at the first 3 min of the test.There was a significant difference in heart rate among the three group.The heart rate of FF group ((436.42± 10.16) times/min) was significantly lower than that of the Sham group ((472.48±4.43) times/min,P<0.01) and the DH group ((469.94 ±7.36)times/min,P<0.01).In the test of conditioned tone fear memory.The freezing time percentage in FF group ((18.78±6.29) %) was significantly lower than that in the Sham ((51.77±9.33)%,P<0.01) and DH group ((59.19±8.13)%,P<0.01),but the freezing time percentage between the later two groups had no difference (P=0.52).The synchronous telemetry measurement showed there was no significant difference both in the heart rate and the blood pressure among the groups (all P>0.05) during the conditioned tone test.Conclusion The dorsal hippocampal lesions and fimbria-fornix transection in rats can significantly reduce the learning and memory ability in conditioned fear and scene fear in rats,and the effect of fimbria-fornix transection is more obvious.The decrease in,fear memory is not synchronously reflected in heart rate and blood pressure in rats.
8.Effects of resveratrol on cognitive dysfunction and the expression of parvalbumin in depression model mice

Objective To examine the antidepressant effects of resveratrol (RSV),and its possible mechanism specialized on improving cognitive function.Methods Thirty-two C57BL/6J mice were randomly divided into four groups:Control,Model,Model+RSV and Model+NA+RSV groups.The mice were subjected corticosterone (20 mg/(kg · d)) intraperitoneal injection for 21 consecutive days except the control mice.From the 22nd to 42nd day,the mice in different groups received further treatment with vehicle/ RSV (400 mg/(kg · d),op)/NA (100 mg/(kg · d),ip)+RSV (400 mg/(kg · d),op).The sugar preference test,novel object recognition test,novel location recognition test and water maze test were applied to evaluate the cognitive effects of RSV on mice.Subsequently,the silence information regulation factor 1 (SIRT1),peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α),parvalbumin (PV) transcription and translation level were evaluated by RT-PCR and Western blotting.Results The sugar preference test showed that the depression model mediated by CORT injection was successfully established(F(1,30) =6,P=0.038).In in the novel object learning test,resveratrol significantly increased the proration on the frequency ((-0.20±0.37) vs (0.16±0.29))and duration((0.10±0.45) vs (0.62±0.29)) and decreased the proration on the distance((0.09±0.36) vs (0.55±0.27)).In the water maze test,resveratrol reduced the time((41±9)s;(26± 8) s) and distance ((295± 70) cm;(224±43) cm) to find the platform.All the results were accompanied with the increased expression of protein SIRT1 (F(3,29) =15.60,P<0.01),PGC-1α(F(3,29)=7.51,P=0.0006) and PV (F(3,29) =17.87,P=0.0004).While pretreatment with nicotinamide,resveratrol could not rescue the cognitive impairment and could not reverse the iecreased expression level of protein SIRT1,PGC-1 α and PV.Conelusion Resveratrol can reverse the cognitive dysfunction of depressant mice,which may be achieved by activating the SIRT1/PGC-1α signaling pathway and increasing the transcription and protein expression of PV.
9.Application values of multiple detection methods of bone marrow in newly diagnosed multiple myeloma

Objective:To investigate the application values of bone marrow morphology, bone marrow immunohistochemistry, flow cytometry, fluorescence in situ hybridization (FISH) and cytogenetic testing in newly diagnosed multiple myeloma.Methods:A total of 280 patients with multiple myeloma who were newly diagnosed in Tianjin KingMed Diagnosis Center from September 2018 to August 2019 were collected. The bone marrow biopsy was carried out according to the routine method, and bone marrow morphology, bone marrow immunohistochemistry, flow cytometry immunophenotyping, FISH and cytogenetic testing were performed. The detection results of each method were compared.Results:In 280 patients, the bone marrow immunohistochemistry results showed that the median ratio of plasma cells was higher than those of bone marrow morphology (20 cases, 0.675 vs. 0.300) and flow cytometry (47 cases, 0.650 vs. 0.147), and the differences were statistically significant ( Z = -3.883, P < 0.01; Z = -5.947, P < 0.01). Flow cytometry results showed that the positive rates of CD38, CD138, κ, λ, CD56 and CD19 were 100.0% (280/280), 100.0% (280/280), 57.5% (161/280), 42.5% (119/280), 62.1% (174/280) and 19.3% (54/280); bone marrow immunohistochemistry results showed that the positive rates of CD38, CD138, κ, λ and CD56 were 98.9% (277/280), 98.2% (275/280), 57.5% (161/280), 42.5% (119/280) and 62.1% (174/280); there was no statistical difference between the two detection methods in the detection coincidence rate of the same detection index (all P > 0.05). Among patients who underwent FISH detection, the detection rate of gene abnormalities was 69.9% (93/133); the detection rate of abnormalities by direct fluorescence in situ hybridization (D-FISH) was 42.9% (57/133); the detection rate of abnormalities by CD138 immunomagnetic sorting myeloma cells (MACS)-FISH was 82.7% (110/133). Among patients who underwent G-band karyotyping, the detection rate of abnormal karyotype was 38.5% (85/221). FSIH, especially MACS-FISH, had a higher detection rate of cytogenetic abnormalities than G-band karyotyping, and the difference was statistically significant ( χ2 = 65.697, P < 0.05). Conclusion:The comprehensive application of bone marrow morphology, bone marrow immunohistochemistry, flow cytometry, FISH (especially MACS-FISH), cytogenetic testing and other detection methods is more helpful for the diagnosis of multiple myeloma, and may be useful for prognostic judgment.
