Objectve To find out an effective method for treatment of postoperative wounds of the patients with anal and rectal diseases.Methods Cryoprecipitate was smeared over postoperative wounds of the patients with anal and bowel diseases(the experimental group),and the mean healing time,infection rates and hemostatic effect were observed and compared to the patients who were treated by routine medicine(the control group).Results The mean healing time of patients with hemorrhoid,anal fistula and perianal abscess was shorter than the control group by 5.68 days,6.01 days and 1.87 days respectively.And the infection rates of the experimental and control group were 9.18%(9/98)and 44.68%(42/94)respectively,significant difference between them was found( P

Objective To screen and identify the human phage engineered antibodies against HBsAg, and to analyze their gene sequence. Methods The phage antibody library was constructed by phage surface display techniques, and then the antibody genes were isolated and amplified from human peripheral blood lymphocyte. The human phage engineered antibody against HBsAg was selected through bio-panning from the phage antibody library. The affinity and specificity of the phage antibody was identified by ELISA and inhibition ELISA assay. The genes of heavy chain and light chain of the phage antibody against HBsAg were analyzed by sequencing. Results 30 colonies were obtained after three rounds of bio-panning. One of them had the highest A490 (A490=1.47?0.08) with ELISA, and the inhibition rate was 76% with inhibition assay (A490=0.35?0.10), implying that the phage antibody against HBsAg was of high specificity. The findings of enzyme digestion demonstrated that the phage antibody included genes of heavy chain and light chain. Sequencing results indicated that the VH region of heavy chain belonged to VHI subgroup and the VL region of light chain belonged to V? Ⅰ and V? Ⅲ subgroup. Conclusions The specific human phage engineered antibody against HBsAg was selected successfully from the phage antibody library. These results suggest that screening and identification of human phage engineered antibody against HBsAg through the phage antibody library are technologically feasible. It lays a foundation for application of human engineered antibody against HBsAg to design new targeted therapy strategy for HBV.

Objective To study the pharmacokinetics of aristolochic acid A in Radix Aristolochiae and the compound preparation of Guanxinsuhe Capsule in mice in vivo after single-dose oral administration and observe the difference of aristolochic acid A absorption and distribution. Methods Aristolochic acid A assay was performed by RP-HPLC on a Waters apparatus with a DiamonsilTM C18 column (250 mm × 4.6mm, 5 μm), a mobil phase: a mixture of methanol-water-acetic acid (72: 27 : 1), flow rate: 1.0 mL/min, detection wavelength: 315 nm, and column temperature: 20 ℃. Results Mice were given Radix Aristolochiae and Guanxinsuhe Capsule by ig at the same level of 2. 5 mg/kg of aristolochic acid A, respectively, which were suspended in 0. 3% CMC-Na solution. Plasma concentrations were determined by RPHPLC. After single-dose ig administration of Radix Aristolochiae or Guanxinsuhe Capsule to mice, the mean plasma concentration-time courses of aristolochic acid A obtained fitted the one-compartment model.The main pharmacokinetic parameters of aristolochic acid A in Radix Aristolochiae, t1/2ka, t1/2 ke, tmax,AUC, Cmax are 5. 103 min, 43. 63 min, 17.89 min, 80. 45 (μg · min)/mL, and 0. 916 8 μg/mL; the rela tive pharmacokinetic parameters in Guanxinsuhe Capsule are 5. 294 min, 43.50 min, 18. 32 min, 33.08(μg · min)/mL, and 0. 381 8 μg/mL. Conclusion The Cmax of aristolochic acid A in Guanxinsuhe Capsule is significantly less than that in Radix Aristolochiae, which indicates that the compound compability could decrease the absorption of aristolochiae acid A.

Objective To study the clinical feature of ciguatera fish poisoning.Methods Thirty-nine patients with ciguatera fish poisoning were observed.Results The clinical symlptoms of ciguatera poisoning usually began to appear 2～10 hours after eating a poisonous fish and could be classified into three broad groups:neurological,gastrointestinal cardiovascular symptoms.Gastrointes tinal symptoms were the most common complaint.Neurological symptoms generally stayed for 1～2 weeks.The clinical picture was characterized by temperature reversal.Conclusion Ciguatera poisoning can be differentiated by unique features affecting the neurological system.

Objective To optimize the producing area and parts of Gardenia roots. Methods Oleanolic acid 3-acetate was hydrolysed into Oleanolic acid in Gardenia roots from 10 different origins, and root, stem, leaf of Gardenia from Liuyang Hunan, and the content was determined by HPLC. Results The content of oleanolic acid 3-acetate in Gardenia roots of different origins from high to low was:Shaodong, Liuyang of Hunan>Anji of Zhejiang, Guiyang of Hunan>Ningxiang, Anhua of Hunan, Zhangshu of Jiangxi>Liling, Pinjiang, Youxian of Hunan. The content in root was 2 times of that in stem and leaf. Conclusion Experimental data were provided for the optimization of producing area and part of Gardenia roots.

Objective To investigate the changes of excitatory amino acids (EAAs) of cerebrospinalfluid (CSF) in patients with delayed encephalopathy after acute carbon monoxide poisoning(DEACMP).Method The EAAs levels of CSF including glutamate (Glu) and aspartate (Asp) in 24 patients with DEACMP and 20 controls with migraine were measured by high performance liquid chromatography( HPLC ).Results Glu and Asp levels in patients with DEACMP were significantly higher than those of the controls [(3.76 ± 1.52) μmol/L vs ( 1.55 ± 1.03 ) μmol/L, (0.73 ± 0.44) μmol/L vs (0.38 ± 0.33 ) μmol/L, P all <0.01]. Glu and Asp levels in moderate and severe DEACMP patients were higher than those in mild DEACMP patients. Conclusion It suggests that EAAs participated in the pathogenesis of DEACMP. The Glu and Asp levels in CSF may be regarded as indicator of DEACMP.
