1.Experimental Studies on Attenuating and Synergic effects of Modified Yigong Powder

Objective To observe the effects of modified Yigong Powder (MYP) combined with chemotherapy on transplanted hepatocarcinoma of mice and to study its mechanisms. Methods Hepatocarcinoma 22 mouse model was established and then was used to observe the attenuating and synergic effects of MYP when applied together with 5-fluorouracil (5-FU). After 8 days of treatment,the tumor-inhibiting rate,activity of natural killer (NK)cells and interleukin-2 (IL-2) and small intestinal malondialdehyde (MDA) content were examined. Results MYP combined with 5-FU could increased the tumor-inhibiting rate to some extent and improve the immune function by increasing immune organ weight and increasing the activity of NK cells and IL-2. MYP combined with 5-FU could also reduce the 5-FU-induced intestinal injuries by relieving the damage of free radicals and inhibiting the lipid peroxidation and a good prognosis was expected in tumor-bearing animals treated with chemotherapy. Conclusion MYP exerts an attenuating and synergic effect when used together with 5-FU in treating tumor-bearing mice and its mechamism may be related to the improvement of immune function and reduction of intestinal injuries.
2.Renal insufficiency and its associated factors in type 2 diabetic patients with normoalbuminuria

Objective To investigate the prevalence of renal insufficiency and its associated factors in type 2 diabetes mellitus with normoalbuminuria using estimated glomerular filtration rate ( eGFR) .Methods We retrospectively analyzed 10-year data of chronic complications in type 2 diabetics in-patient from the Chinese Diabetes Society.eGFR was estimated using the equation from Modification of Diet in Renal Disease(MDRD) study.The clinical characteristics as well as associated factors for low eGFR were analyzed among the normoalbuminuric type 2 diabetic patients.Results A total of 1351 type 2 diabetic patients were included, 755 patients with normoalbuminuria, 466 patients with microalbuminuria and 130 patients with macroalbuminuria respectively.Among the patients, 310 (22.9% ) had low eGFR (GFR <60 ml · min~(-1) · 1.73 m~(-2) ) , 19.7% (149/755) in the patients with normoalbuminuria, 21.9% (102/466) in microalbuminuria and 45.4% ( 59/130 ) in macroalbuminuria Patients with normoalbuminuria and low eGFR suffered more chronic complications than those with normoalbuminuria and normal eGFR, mainly retinopathy, cerebrovascular diseases and sensory neuropathy.Stepwise logistic regression analysis revealed that age ( OR = 1.042, P < 0.001), diabetic duration ( OR = 1.038, P = 0.045), systolic blood pressure (OR = 1.017, P < 0.001) were independently associated with renal impairment among the patients with normoalbuminuria.Body mass index ( OR = 0.868, P < 0.001) and HbAlc (OR =0.898, P =0.021) were also related with renal insufficiency.Conclusion A considerable proportion in type 2 diabetic patients without albuminuria may exist renal impairment, and eGFR estimation could benefit the evaluation of renal function in such patients.
3.Clinical effect of creatine phosphate on infantile with rotaviral enteritis and myocardial damage

Objective To explore the clinical effect of creatine phosphate on infantile with rotaviral enteritis and myocardial damage.Methods One hundred and twenty children with rotaviral enteritis and myocardial damage were randomly divided into the control group (n =60) and observation group (n =60).Two groups were given antiviral,rehydration,nutritional support,redressing acid-base imbalance and electrolyte disorders and other conventional treatment.The patients of the control group were given 160 mg/(kg · d) 1,6Fructose Diphosphate(1 times/d) on the basis of routine treatment,while those in the observation group were given 1.0 g creatine phosphate + 150 ml 5% glucose injection(1 times/d) on the basis of routine treatment,both courses of two groups were 7 days.The levels of myocardial enzymes were measured and the clinical effect and adverse reactions were recorded.Results The total effective rate in observation group was 95.0% (57/60),obviously higher than that of control group (80.0% (48/60)) and the differences were statistically significant (x2 =6.268,P < 0.05).There were no statistical significant difference in terms of myocardial enzyme between the two groups before treatment(P >0.05).The level of lactate dehydmgenase(LDH),creatine kinase (CK),creatine kinase isoenzyme (CK-MB) and aspartate aminotransferase (AST) of observation group were (163.8 ±18.5) U/L,(152.8±26.4) U/L,(19.4±7.8) U/Land (24.4 ±6.8) U/Lafter treatment,and (193.4 ± 22.8) U/L,(168.6 ± 32.6) U/L,(27.4 ± 9.2) U/L and (35.4 ± 9.8) U/L of control group,and the differences between the two groups were significant (t =3.484,3.228,4.278 and 4.729; P < 0.05).There was no obvious adverse reaction during the treatment course.Conclusion The clinical effect of creatine phosphate on infantile with rotaviral enteritis and myocardial damage is remarkable and safe,and it is worth of clinical application.
4.A control study on the development of "cool" and "hot" executive functions between cerebral palsy children and normal children

Objective To explore the differences and development of executive function(EF) between cerebral palsy(CP) children and normal children. Methods Forty-eight 4 ~6 years old CP children and fiftyeight normal children were tested by "cool" and "hot" EFs. Results There were significant differences between "cool" EF of CP children(18.34±14.31) and normal children(6.94 ±3. 18) ( t = 3. 83, P<0.01 ) ;and there were significant differences between "hot" EF of CP children(279.67 ±330. 18) and normal children(709.31 ± 304. 13)( t = -4.93, P< 0.01). There were significant age differences on the "cool" EF ( F=8.689, P< 0.01) and "hot" EF ( F=3. 833, P<0.05) of CP children. There were significant age differences on the "cool" EF ( F= 15.469, P<0.01) and on the "hot" EF ( F=8.470, P<0.01) of normal children. There was negative correlation between "cool" EFs and "hot" EFs( r= -0.440, P<0.01). Conclusion "Cool" and "hot" EFs of CP children are lower than those of normal children. "Cool" and "hot" EFs can develop from 4 to 6 years old,but the development of EFs are not absolutely consistent between CP children and normal children. There is correlation between "cool" and "hot" EFs in patients with CP,but not dissociation.
5.Combination of Cervical Perivascular Sympathectomy and Selective Posterior Rhizotomy in Spasticity of Cerebral Palsy: 11 Cases Report

Objective To investigate the effect of combined application of cervical perivascular sympathectomy (CPVS) and selective posterior rhizotomy (SPR) on spasticity of cerebral palsy. Methods The data on 11 children with cerebral palsy following spasm who were treated by CPVS and SPR were retrospectively analyzed. Results Spasm of most children relieved significantly 1 month after surgery and limb movement function got notable improvement. The effect was confirmed stable and durable 4 months after surgery. Conclusion CPVS and SPR are effective surgical methods for treating spasticity of cerebral palsy.
6.Screening and characterization of human phage antibody to permethrin

Objective To screen permethrin human single-chain variable region (scFv) antibody for aims of developing rapid detection kit. Methods Phage display technology was used in this study. Permethrin was solid phase coated on Nunc plate as antigen. Semi-synthetic single-chain variable region of human antibody library technology was applied, and single chain variable region was screened from phage antibody library after 3 rounds "adsorption - elution - amplification" of the selection process. 100 clones were random selected as resistance to permethrin clones , enzyme-linked immunosorbent assay (ELISA), crossreactivity and competitive inhibition experiments were used to validate permethrin binding activity with strong scFv clones from the selected phage antibody clones plasmid. The plasmid was digested with restriction enzyme Sfi Ⅰ / Not Ⅰ and subcloned into pCANTAB5E vector. After transformed into E. coli XL1BIue, the plasmid was identified by restriction enzyme analysis. Results After screening in 100 clones, 18 clones had high ELISA absorbance values ( A value) at 490nm wavelength ( A490nm), then bovine serum albumin (BSA) cross-reactions identified five weak cross-reaction. Combined with the triplicate ELISA and competitive inhibition experiment results, one positive clone was acquired at last. And this clone was subcloned into pCANTAB5E vector and transformed into competent cells XL1-Blue. Conclusion Plasmid fragment was consistent with the purpose, which provided the foundation for further study of its specific affinity.
7.Preparation of SiO2/Ni core-shell type nanoparticles and their magnetic properties

BACKGROUND: Inhibitory coating can prevent nanoparticle oxidation, grain growth, corrosion and agglomeration, and endow nanoparticle with special properties. ABJECTIVE: To prepare SiO2/Ni core-shell type nanoparticles and assess their magnetic properties. DESIGN, TIME AND SETTING: The observation experiment was performed between November 2005 and March 2006 at Nanometer Compound Material Research Laboratory of Dalian University of Technology, Dalian, Liaoning Province, China. MATERIALS: Nanometer nickel powder prepared by DC arc plasma jet method, Na2SiO3 produced by Bazhou Chemical Industry Branch Factory of Tianjin Quartz Clock Factory (China).METHODS: SiO2/Ni core-shell type nanoparticles were synthesized by coating a layer of SiO2 on the surface of manometer nickel powder via liquid deposition method using Na2SiO3 as the main source material. MAIN OUTCOME MEASURES: Their microstructures and material properties were investigated by X-ray diffraction, Fourier transform infrared spectrometer, transmission electron microscopy, thermo-gravimetric analysis, differential scanning calorimetry and vibrating sample magnetometer. RESULTS: The experimental results showed that SiO2 shell was in an amorphous state around Ni cores and it avoided agglomeration of the Ni nanoparticles. The oxidation temperature of nanometer nickel powder coated by SiO2 elevated from 287 ℃ to 385 ℃. The analysis result of magnetic properties indicated that the hysteresis loop of Ni had an excursion for the existence of anti-ferromagnetic NiO, the silica coating reduced the saturation magnetization and improved the coercivity. CONCLUSION: Preparation of SiO2/Ni core-shell type nanopartieles was successful; silica coating improved the oxidation resistance of nanometer nickel powder, endowed nanometer nickel powder better ferromagnetism and improved the coercivity.

Objective To do screening acute lymphoblastic leukemia patients scFv antibody single chain variable region to cre-ate conditions for the expression and obtain further specificity of antibody fragments.Methods In this study,patients with newly diagnosed acute lymphoblastic leukemia serum as coating antigen using phage display technology,screening phage an-tibody specificity from the semi-synthetic human phage antibody libraries,the first to target the immune antigen-coated tab-let,phage library was added,so that with the target antigen-specific binding phage antibody was immobilized on plates immu-nization,could not be specifically bound phages were rinsed.The eluted specific binding phage,E.coli infection.Could get the specific antibody gene containing phagemid.Results After three “adsorption-elution-amplification”screening process,got stronger leukemia patient antigen-specific phage antibody variable region fragment and identification.Conclusion Got better strain affinity antibody fragments,to create the conditions for the next fragment expression,identification and clinical appli-cation.
9.Expression of OTUB1 in colon cancer and its clinical significance

Purpose To investigate the expression of OTUB1 in colon cancer and the relationship between its expression and some clinicopathologic parameters. Methods Quantitative real-time PCR and immunohistochemical SP method were carried out in selected colon cancer and normal mucosa tissues. Results OTUB1 mRNA in colon cancer was 3. 5-fold higher than the normal mucosa. The expression of OTUB1 protein in the colon cancer was significantly higher than normal mucosa (P<0. 05). Moreover, its expression in normal tissues, adenoma and colon cancer showed a gradually increasing trend (P<0. 05). The higher expression of OTUB1 in colon cancer was related with tumor size, differentiation and lymph node metastasis. Conclusions OTUB1 may play an important role in co-lon cancer development.
10.Clinical Study on Deep Insertion at Tianshu(ST 25)for Colonic Slow Transit Constipation

Objective:To observe the clinical effect of deep insertion at Tianshu(ST 25)for colonic slow transit constipation(STC).Methods:120 cases of STC patients were randomly divided,60 cases in a deep insertion group,30 cases in an electroacupuncture group and 30 cases in a medication group by 2:1:1 ratio.The deep insertion group was treated with deep insertion at Tianshu(ST 25).The electroacupuncture group was treated with routine insertion at Tianshu(ST 25).The medication group was treated with oral administration of Lactulose oral liquid.The first voluntary defecation time,and constipation scores before the treatment,four weeks after the treatment and relevant scores of clinical symptoms were assessed in the three groups of the patients.Results:The scores of the clinical symptoms in improvement of constipation were better in the deep insertion group than in the electroacupuncture group and medication group,with differences in statistical significance(P<0.01).The unsuccessful numbers in the improvement of defecation and abdominal pain were also better in the deep insertion group than in the other two groups,and better in instant effect in the deep insertion.Conclusion:The improvement of STC clinical symptoms was better by deep insertion at Tianshu(ST 25)than by medication and routine acupuncture method at Tianshu(ST 25).
