1.Combined Lethal Toxicity of TNF and LPS in Tumor-Bearing Mice

Both TNF and LPS are toxic to host, especially to tumor - bearing host Intravenous administration at a dose of 5?g of TNF or 25?g of LPS could not result in death of mice transplanted with sarcoma 180 (S180) 10 d later, but 80% of S180-bearing mice died after iv injection of 5?g of LPS combined with 0.5?g of TNF. In contrast to TNF, combined toxicity was seriously LPS-dose dependent Combined mortality dropped from 80% to 14% when LPS dose reduced to 1?g, however, 50% of mice died of combined toxicity when only TNF dose dropped to 0.1?g. It suggested that TNF sensitized host response to toxic effect of LPS. Indomethacin could partially block the combined toxicity, demonstrating that combined toxicity of LPS and TNF was partially mediated by prostaglan-dins.

3.Surgical treatment of gastrointestinal stromal tumors: a report of 486 cases

Objective To investigate the surgical treatment of gastrointestinal stromal tumor (GIST) based on the clinical characteristics of GIST in different locations.Methods The clinical data of 486 GIST patients who received surgical treatment at the Peking Union Hospital from January 2003 to December 2012 were retrospectively analyzed.There were 461 patients with primary GIST and 25 with secondary GIST.The clinical characteristics and surgical treatment methods were analyzed.All data were analyzed using the chi-square test.Results Of the 461 patients with esophageal stromal tumors,6 received partial esophagectomy.Of the 234 patients with gastric stromal tumors,183 received partial gastrectomy,23 received proximal gastrectomy + cardiectomy,23 received distal gastrectomy,2 received total gastrectomy and 3 received exploratory laparotomy.Of the 51 patients with duodenal stromal tumors,34 received partial duodenectomy,9 received pancreatico-duodenectomy,5 received pylorus-preserving pancreaticoduodenectomy,3 received palliative surgery.Of the 116 patients with small intestinal stromal tumors,110 received partial small intestinal resection and 6 received palliative surgery.Of the 29 patients with rectal stromal tumors,13 received posterial transsphincteric surgery of the rectum,12 received transanal local resection of rectal tumors and 4 received abdominoperineal resection; laparotomy was performed on 25 patients with GIST in other positions.Of the 25 patients with secondary GIST,10 patients with liver metastasis of GIST received hepatic segmentectomy,1 received hepatobiopsy; 6 received abdominopelvic tumor resection; 5 received portial resection of the small intestine or colon; 1 received sigmoid colostomy; 1 received splenectomy and 1 received intracranial tumor resection.Of the 461 patients with primary GIST,patients who received combined devisceration accounted for 12.58% (58/461),and the ratio of combined cholecystectomy was the highest,which was 34.5% (20/58).Combined devisceration was considered for patients with duodenal stromal tumors,gastric stromal tumors and small intestinal tumors.Laparoscopic surgery accounted for 20.39% (94/461) of all the surgery,and the ratio of laparoscopic surgery which carried out in recent 5 years was 28.52% (77/270),which was significantly higher than 8.90% (17/191) of the earlier 5 years (x2=36.67,P < 0.05).Conclusion Different surgical treatment methods including minimally invasive surgery could be adopted according to different clinical characteristics of GIST in different locations,and radical resection of GIST is the main objective.

Percutaneous endoscopic gastrostomy (PEG) is an convenient, effective, and safe technique,and is especially useful for the nutritional support of patients who are unable to befedorally. PEG should be carefully arranged based on indications, ethical criteria, and contraindications. Early PEG can remarkably improve the quality of life.
5.Mechanism of the radioresistant effect of LyGDI on NSCLC A549 cells

Objective To elucidate the mechanism of radiation resistant effect of LyGDI on NSCLC A549 cells.Methods A549 and H460 cells were irradiated with X-rays of 0,2,4 and 6 Gy.The clone-forming assay was used to detect cell survival and radiosensitivity.The expressions of LyGDI and COX-2 (Cyclooxygenase-2),a key radiosensitivity-related protein,were detected using Western blot.The miR-34 families were analyzed with RT-PCR.50 nmol/L mature miR-34c was transfected into A549 cells.Results The expression levels of LyGDI and COX-2 were much higher in radioresistive A549 cells than that in H460 cells.While the expression of miR-34a was quite low and miR-34b/c was hardly found in both NSCLC cells.Transfection of miR-34c into A549 cells strongly enhanced X-ray induced apoptosis by inhibiting the activations of LyGDI,COX-2,Bcl-2 and p21.Conclusions Up-regulation of LyGDI could induce COX-2 expression.The low expression of miR-34 family might be responsible for the radiation resistance of NSCLC cells.

Iodine is a micronutrient that is essential for the production of thyroid hormones.Either iodine deficiency or excess will not only affect the synthesis and secretion of thyroid hormone but also be related to the occurrence and development of thyroid diseases.Iodine deficiency can cause endemic goiter,cretinism,hypothyroidism and thyroid cancer;high incidence of hyperthyroidism,hypothyroidism,autoimmune thyroid disease,goiter,and thyroid cancer are associated with excessive iodine intake.It is essential to assess iodine nutritional status for the populations,particularly those with thyroid diseases.
7.R-fraction as a Biological Principle in the Natural S. abortus equi Endotoxin Preparation

A typical smooth-form lipopolysaccharide (LPS) isolated from Salmonella abortus equi was fractionated into a S (smooth)-and a R (rough)-fraction and their serological and biological properties were investigated. It was shown that S- fraction expressed an O-antigenicity while R-fraction predominately a Rb-antigenicity. The R-fraction was endowed with higher bioactivities than the S-fraction in lethal toxicity, local Shwartzman reaction and pyrogenicity. Both S-and R-fractions were active in inducing mitogenicity to murine spleen cells. A reconstituted LPS preparation with fractionated S-fraction and a trace amount of R-fraction (1% of the original preparation) exhibited a same extent of lethal toxicity as that of the original one. Antr-Ra and -Rb antiserum with a liter of 1?4096 showed a .highly effective protection against the lethal challenge of LPS, indicating that the R-fraction in the natural LPS preparations plays a critical role to the lethality of LPS.
8.Effect of chronic hypoxia on TGF-?1 expression and proliferation in MDCK Cells

Objective To investigate the effect of hypoxia on renal tubular cells and the mechanism of renal interstitial fibrosis. Methods Effect of chronic hypoxia on the proliferation, cell cycle and expression of TGF-?1 mRNA in cultured MDCK cells was examined. Quiescent cells were exposed to hypoxia (3%O2) or normoxia (18%O2) for 24, 48, 72 and 96 hours. At the end of each incubation, cellular proliferation and cell cycle were assessed by MTT method and flow cytometry, respectively. TGF-?1 mRNA level of cells were analyzed by semi-quantity RT-PCR. Results Exposure of MDCK cells to hypoxia induced a significant increase in proliferation and a significant decrease in the percentage of G0-G1 stage and a significant increase in the percentage of G2-M stage at 24, 48 and 72 hours. Meanwhile, TGF-?1 mRNA expression increased in a time-dependent manner as compared with normoxia group when MDCK cells were stimulated with 24, 48, 72 and 96 hours in hypoxia condition. Conclusion Chronic hypoxia-induced proliferation of MDCK cells is associated with increased expression of TGF-?1. Chronic hypoxia of renal tubular cells may be one of the causes of renal interstitial fibrosis.

10.Correlation between serum zinc,selenium,chromium,copper and blood glucose levels in type 2 diabetes patients

Objective To investigate the correlation between levels serum zinc,selenium,chromium,copper and blood glucose in diabetic patients.Methods The 136 cases of type 2 diabetes were enrolled in our hospital during September 2013 to December 2015 in the observation group,and 136 cases of physical health in our hospital during the same period were recruited in the control group.Then the levels of serum zinc,selenium,chromium,copper contents and glycemic index,oxidative stress indicators were detected.Results The serum zinc,selenium content of the observation group were significantly lower than those in the control group(P<0.05),no significant difference were observed in the serum chromium,copper contents between two groups(P>0.05);the levels of FBG,2 h PBG,glycated hemoglobin,MDA,and HOMA-IR of observation group were higher than those of control group,the HOMA-β,I30/G30,SOD,GSH-Px levels were lower than those in the control group(P<0.05);the serum zinc,selenium contents were negatively correlated with FBG,2 h PBG,glycated hemoglobin,MDA levels,and HOMA-IR(P<0.05),while positively correlated with HOMA-β,I30/G30,SOD,GSH-Px(P<0.05).Conclusion Serum zinc,selenium contents in type 2 diabetes significantly decrease and closely be related with blood glucose levels,they can reduce blood sugar,promote insulin secretion,increase insulin sensitivity,and adjust the oxidative stress reaction.
