1.Effect of two kinds of injected biomaterial combined with BMP on tendon-to-bone healing after anterior cruciate ligament reconstruction in rabbits

Objective To investigate the effect of two kinds of injected biomaterial combined with bone morphological protein (BMP) on tendon-to-bone healing after anterior cruciate ligament reconstruction (ACL) in rabbits. Methods ACL reconstruction was performed bilaterally in 50 skeletally mature rabbits using long digital extensor tendon grafts. Injected fibrin glue (FG) or injected calcium phosphate cements (CPC) combined with BMP were implanted into the treated knee of each rabbit,with the contralateral knees as controls. Every three rabbits were killed at 2,6 and 12 weeks postoperatively for routine histological studies. The samples were processed through Micro CT and subsequent HE and toludine blue staining. The remaining 16 rabbits were sacrificed at 6 and 12 weeks. Their femur-tendon graft-tibia complexes were harvested for subsequent mechanical testing. Results Imageological results showed that 6 weeks after operation,bone mineral density (BMD) values in FG-bBMP group were higher than those in CPC-bBMP group. However,12 weeks after operation,BMD values in CPC-bBMP group were the highest in the three groups. Histological findings of the interface between the tendon and bone differed in the three groups. Two weeks after operation,more chondrocyte-like cells that were fairly disorganized were noted between the tendon and bone in FG-bBMP group. Six weeks after operation,mature bone formation around the tendon was observed in FG-bBMP group while more immature bone ingrew toward implanted tendon in CPC-bBMP group; at 12 weeks,more chondrocyte cells and new bone formation were seen in CPC-bBMP group while a small quantity of mature bone was around tendon in FG-bBMP group. 2.Effect of temperature on crosslinking of collagen/chitosan scaffolds with genipin

[Objective]To investigate the changes of pore sizes,crosslinking index,swelling ration,degradation rate,cytotoxicity and biocompatibility of genipin crosslinked collagen/chitosan scaffolds affected by the different crosslinking temperature points.[Method]The freeze-dried collgaen/chitosan scaffolds crosslinked with 0.5% genipin within 24 h were divided into 3 groups by crosslinking temperature:4℃ group,20℃ group and 36℃ group.The characteristics of pore sizes,crosslinking index,swelling ratio,degradation rate,cytotoxicity and biocompatibility were evaluated.Collagen/chitosan scaffolds without crosslinking were chosen as control group.[Result]With the increase of temperature,crosslinking index was increased,but swelling ratio and degradation rate were decreased.In 4℃ group,the crosslinking index was 47.88%?6.4%,the swelling ratio was 721%?46%,and the degradation rate was decreased by 3.95%?6.4% at 4 weeks.In 20℃ group,the crosslinking index was 67.69%?3.6%,the swelling ration was 662%?72%,and the degradation rate was 0.91%?5.9% in 4 weeks.In 36℃ group,the crosslinking index was 70.32%?5.7%,the swelling ration was 635%?27%,and the degradation rate was 0.66%?7.3% at 4 weeks.The above indexes of the three groups were much better than those of control group(P

5.Study on the Outcome of Acetabular Fractures Operative Treatment

Objective To explore the outcome of acetabular fractures operative treatment. Methods 62 patients with acetabular fractures were enrolled in this study. According to classification of Letournel, fracture of posterior wall 13 cases, fracture of posterior column 9 cases, fracture of anterior column 5 cases, transverse fracture 4 cases, transverse fracture with fracture of posterior wall 6 cases, double column fracture 10 cases, fracture of posterior wall and posterior column 8 cases, fracture of T type 7 cases. Results 62 patients were followed-up for more than one year after operation. All cases had bone union without complications of the infection and sciatic nerve injury. Traumatic arthritis occurred in 2 cases, and the function of the hip-joint was limited in 3 cases. Conclusion Operative reduction and internal fixation can notably increase forward outcome of acetabular fractures.
6.Nevus of Ota resistant to multiple sessions of Q-switched laser treatment: two case reports

Two cases of nevus of Ota resistant to multiple sessions of Q-switched laser treatment are reported.Both of the patients were females,who developed beige or blue patches on the left side of the face (zygomatic region,cheek)several months after birth.No conjunctiva or sclera was involved.The lesions spread to the left retroauricular region and posterior neck during pregnancy in one patient.Histopathological examination of the lesions showed scattered melanocytes and melanophages in lower dermis and subcutaneous adipose tissue as well as apparent proliferation of collagen.Immunostaining for CD34 showed no abnormality in the distribution of vascular endothelial cells in these tissues.No obvious improvement was observed in either of the patient after more than 10 sessions(12 sessions in 1 case,and 13 sessions in the other case)of treatments with Q-switched alexandrite(755 nm)or Q-switched Nd:YAG laser(1064 nm).The immediate response of lesions to laser irradiation was unusual.Purpura or errhysis was absent in the lesions after repetitive irradiation with high-energy laser,but both were visible in the adjacent normal skin even after irradiation with low-energy laser.
7.Manual reduction for external fixation of distal radius fractures

BACKGROUND:Manual reduction for external fixation of distal radius fractures can achieve good outcomes.
OBJECTIVE:To carry out the bibliometric analysis on manual reduction for external fixation of distal radius fractures.
METHODS:Literatures concerning reduction for external fixation of distal radius fractures were retrieved in CNKI database from 2003 to 2012. The keywords were“external fixation;distal radius fractures”. Duplicate and irrelevant articles were eliminated, and a total of 408 articles were retrieved. A bibliometric analysis of these 408 articles was performed in terms of publishing time and number, subject categories, source journals, research institutions, times cited and download frequency.
RESULTS AND CONCLUSION:Literatures concerning manual reduction for external fixation of distal radius fractures exhibit an upward trend in number. In 2012, there were 85 relevant articles. In the past 10 years, 18 relevant articles have been published in Chinese Journal of Traditional Medical Traumatology&Orthopedics, which is the most. There are not many relevant studies in various research units, only 1-7 articles published per unit, and five articles are funded.
8.Role of cancer stem cells in EMT-induced acquired resistance to EGFR-TKIs in NSCLC cells

Objective To explore the role of cancer stem cells in EMT-induced acquired resistance to EGFR-TKIs in NSCLC. Methods The EGFR del E746-A750 mutated human lung adenocarcinoma PC-9 cell line and gefitinib acquired resistance cell line PC-9/AB were used in this study. EMT was assessed by western blotting assay. The sensitivity to gefitinib was tested with CCK8. Flow cytometry for antibody analysis was used to quantify CSCs within the cell lines. Results Compared with PC-9, PC-9/AB underwent EMT and showed no-table resistance to gefitinib (P < 0.01). Compared with PC-9, the proportions of CSCs were much higher in PC-9/AB. Conclusion EMT plays an important role in the acquired resistance to EGFR-TKIs in NSCLC , possibly through the up-regulation of CSCs.
9.Diagnostic Value of 64-MSCT and 1.5T MRI in Nurseling With Congenital Vascular Rings

Objective: To investigate the diagnostic value of 64 multislice spiral CT (64-MSCT) and 1.5T MRI in nurseling with congenital vascular rings.
Methods:We retrospectively studied the diagnostic records of 64-MSCT and 1.5T MRI in 48 nurseling with congenital vascular rings. There were 42 cases received CTA, 12 received MRA and 6 received both CTA and MRA. We classiifed the types of vascular rings by Edwards sketch map of aortic arch embryonic development and trachea wrapping conditions. The diagnostic values of 64-MSCT and 1.5T MRI on vascular rings were compared.
Results: There were 25 patients with complete vascular rings including 3 of double aortic arch, 12 right aortic arch with left subclavian artery and patent ductus arteriosus (PDA), 9 combining with arterial ligament, 1 neck right aortic arch with left subclavian artery combining left PDA;and 23 patients with in-complete vascular rings including 2 pulmonary artery sling, 9 left aortic arch with right subclavian artery combining left PDA, 10 combining with arterial ligament, 1 right aortic arch with left subclavian artery combining right PDA, 1 with innominate artery compressed trachea. There were 32/48 patients combined with cardiac malformations, both 64-MSCT and 1.5T MRI may well reveal the composition and special relationship of vessel rings. The minimum intensity projection (MinIP) and air volume rendering (VR) of 64-MSCT may clearly display the morphology and development of trachea, image fusion technique could directly show the special relationships among vascular rings, trachea and esophagus. While 1.5T MRI was relatively poor to present tracheal image.
Conclusion: 64-MSCT could accurately and comprehensively display the special relationships among the vessels rings, trachea and esophagus;which was the best method for diagnosing the nurseling with vessel rings in clinical practice.
10.Study on relationship between conserved motif of sphingosine 1-phosphate receptor type 1 and FTY720-induced internalization

Objective:To study the relation between conserved motif of sphingosine 1-phosphate receptor type 1(S1P1) and FTY720-induced internalization.Methods:With HA-S1P1(WT)-Myc-EGFP-N1 fusion vector as template,HA-S1P1(R142N)-Myc-EGFP-N1 fusion vector was constructed by overlap PCR.The conserved ERY motif of wild type S1P1 was mutated into ENY.The recombinant vectors were confirmed by sequencing,then they were transfected into HEK293 cells by Polyfect.The transfected HEK293 cells were selected with G418.Cells were incubated for 3,6,12 hours in the absence or presence of 100 nmol/L FTY720,then S1P1 gene expression was analyzed by fluorescence microscopy.Results:Sequencing confirmed HA-S1P1(R142N)-Myc-EGFP-N1 vectors was successfully constructed.S1P1(WT) protein and S1P1(R142N) protein were expressed on the stably transfected-HEK293 cell surface.FTY720 induced S1P1(WT) internalization,but not S1P1(R142N).Conclusion:FTY720-induced S1P1 internalization is related with conserved ERY motif.
