1.Research of the correlation with pathology and kinetics of TH1/TH2 cells and expression of iNOS in animals of tuberculosis

Objective To investigate the correlation of pathology to immunology in animals with tuberculosis by molecular pathology. Methods BALB/c mice were randomly divided into two groups: tuberculosis model (A) and control mice (N). Immunohistochemistry technology was used to investigate the correlation between the expression of IFN-γ, IL-4 and iNOS and the type as well as magnitude of tissue injury. Results Two phases were observed during the 2nd week characterized by inflammatory infiltration in the alveolar-capillary interstitium and the tissues surrounding blood vessels. A predominance of TH1 cells was shown by a higher percentage of IFN-γ positive cells and a lower percentage of IL-4 positive cells in the inflammatory infiltrate identified by immunohistology. From the 4th week after infection to the end of experiment, we observed a chronic advanced phase characterized by pneumonia and local necrosis with a TH0 balance due to an equivalent proportion of IFN-γ and IL-4 positive cells in the lung lesions. The iNOS expression was increased in acute phase and decreased in chronic phase. Conclusion There is a closely correlation on the kinetics of TH1/TH2 cells and iNOS expression with the type and magnitude of tissue injury during the infection of tuberculosis.
2.Effect of ketamine anesthesia in early pregnancy on expression of hippyragranin mRNA in hippocampus in offsprings of rats

Objective To investigate the effect of ketamine anesthesia in early pregnancy on expression of hippyragranin (HGN) mRNA in hippocampus in offsprings of rats.Methods One hundred and twenty pregnant Sprague-Dawley rats at 5-7 days of gestation,aged 7 weeks,weighing 270-300 g,were randomly divided into 4 groups (n =30 each):2 h anesthesia with ketamine group (group K1),4 h anesthesia with ketamine group (group K2),8 h anesthesia with ketamine group (group K3) and control group (group C).Ketamine 20 mg/kg was injected via the caudal vein,followed by continuous intravenous infusion at 130 mg· kg-1 · h-1 after loss of righting reflex,and the infusion lasted for 2,4 and 8 h in groups K1-K3,respectively.The equal volume of normal saline was given instead of ketamine in group C.The learning ability and memory of the offsprings were tested by Morris water maze test on postnatal day 30.The offsprings were sacrificed on 2nd day after the end of Morris water maze test,their brains were removed and hippocampi were isolated for determination of the expression of HGN mRNA by RT-PCR.Results Compared with group C,the escape latency was significantly prolonged,the frequency of crossing the original platform was decreased,the time of staying at the original platform quadrant was shortened,and the expression of HGN mRNA was up-regulated in groups K1-K3 (P < 0.05).There was no significant difference in the escape latency,frequency of crossing the original platform and time of staying at the original platform quadrant among groups K1-K3 (P > 0.05).The expression of HGN mRNA was significantly higher in group K3 that in group K1 (P < 0.05).There was no significant difference in the expression of HGN mRNA between group K2 and group K3 (P > 0.05).Conclusion The mechanism by which ketamine anesthesia in the early pregnancy induces impairment of learning ability and memory of the offsprings is related to up-regulation of HGN mRNA expression in hippocampus in offsprings of rats.
3.Effect of prolonged anesthesia with propofol during early pregnancy on cognitive function of offspring rats

Objective To evaluate the effects of prolonged anesthesia with propofol during the early pregnancy on cognitive function of offspring rats.Methods One hundred and twenty female Sprague-Dawley rats at 5-7 days of gestation,weighing 280-320 g,were randomly divided into 4 groups (n =30 each) using a random number table:control group (group C),and propofol 2,4 and 8 h groups (P2,P4,P8 groups).In P2,P4,P8 groups,after propofol 20 mg/kg was injected intravenously,propofol was infused at 20 mg· kg-1 · h-1 for 2,4 and 8 h,respectively.In group C,normal saline 2 ml/kg was infused intravenously.At 30,31,32,33,34,35 and 36 days after birth,Morris Water maze was performed to evaluate the learning and memory abilities of offspring rats.At the end of Morris water maze test,the hippocampus of offspring rats was removed for microscopic examination of pathological changes with light and electron microscope.Results Compared with group C,the escape latencywas significantly prolonged,the frequency of crossing the original platform was decreased,and the time of staying at the original platform quadrant was shortened in p4 and P8 groups,and no significant changes were found in the indices mentioned above in group P2.The pathological changes of hippocampi were not found in C and P2 groups,while the pathological changes were obvious in P4 and P8 groups.Conclusion Prolonged anesthesia with propofol during the early pregnancy can induce cognitive dysfunction of offspring rats and the mechanism is related to the damage to hippocampal tissues.
4.The effect of NPY on the activation of microglia and IL-1βproduction

Objective To explore the effect of NPY on activation of primary microglia and the production of in?terleukin-1β. Methods Rat primary cortical microglia was cultured and divided into control group, LPS group, NPY+LPS group, NPY group and BIBP3226+NPY+LPS group. Microglia in control group were incubated with serum-free me?dium for 6 h;microglia in LPS group were incubated with serum-free medium plus LPS for 6 h;microglia in NPY+LPS group were incubated with serum-free medium plus NPY and LPS for 6 h; microglia cells in NPY group were incubat?ed in serum-free medium plus NPY for 6 h; microglia cells in BIBP3226+NPY+LPS group were incubated in se?rum-free medium including BIBP3226 、NPY and LPS for 6 h. After 6 h , Primary cultured microglia were stained us?ing IBA-1 antibody and examined under the fluorescence microscope. The protein levels of IL-1βin the culture media and the mRNA expression levels of IL-1βin the microglia of different groups were detected using the methods of Elisa and RT-PCR. Results After 6 h, the contents of IL-1 βin the culture media and the mRNA expression levels of IL-1βin the cells of LPS group increased remarkably compared with control group (P<0.05) and the microglia were activat? ed. Compared with LPS group, the contents of IL-1 βin the culture media. the mRNA expression levels of IL-1β and the activity of microglia in LPS+NPY group were significantly decreased .Compared with LPS+NPY group, the contents of IL-1βin the culture media. the mRNA expression levels of IL-1β and the activity of microglia in BIBP3226+NPY+LPS group were increased (P<0.05). There were no significant differences in the contents of IL-1βin the culture media. the mRNA expression levels of IL-1βand the activity of microglia between BIBP3226+NPY+LPS group and LPS group or between NPY group and the control group. Conclusion NPY can inhibit the biological activity of microglia and IL-1βproduction through NPY Y1 receptorin the microglia.
5.Meta analysis on HBsAg-positive rate among general populations aged 1-59 years, 2007-2016,China

Objective To perform a Meta-analysis on hepatitis B surface antigen (HBsAg)-positive rates among general Chinese population aged 1-59 years.Methods We systemically reviewed the related data (January 2007 to August 2016) published from Chinese National Knowledge Infrastructure (CNKI),VIP,and PubMed.We also assessed the HBsAg-positive rates among general Chinese populations aged 1-59 years,using a random effects regression model with the comprehensive Meta-analysis software 2.2.Results A total of 46 papers were finally included,with a total sample size of 625 053 individuals.Results from the Meta-analysis showed that the overall combined HBsAg-positive rate was 5.7% (95%CI:4.8%-6.6%) among general Chinese populations aged 1-59 years.When comparing the HBsAg-positive rates in different regions,data showed that the HBsAg-positive rate of was higher in the mid-western areas (6.3%,95%CI:4.9%-8.0%)than in the eastern areas (5.5%,95%CI:4.4%-6.8%).Results showed that HBsAg-positive rates was higher in males (6.1%,95%CI:5.3%-7.0%) than in females (4.8%,95%CI:4.2%-5.5%).As for the HBsAg-positive rates in different time periods,data showed positive rate of 6.3% (95%CI:5.5%-7.2%)in 2007-2009,5.9% (95%CI:4.4%-8.0%) in 2010-2012 and 3.5% (95%CI:2.0%-6.1%) in 2013-2016,respectively.Conclusion The prevalence of hepatitis B virus infection was decreasing between 2007 and 2016 in China,making the country an intermediate endemic area on HBV.
6.Brief Analysis of Clinical Evaluation Concerns of Personalized Abutments and Abutment Crown Bridge Products

Objective The applications of personalized abutments and abutment crown bridge products have increased year by year,but there is no clear requirement for clinical evaluation of the same variety of such products.This study mainly introduces the clinical evaluation concerns of personalized abutments and abutment crown bridge products,in order to provide reference for the declaration and registration of such products.Methods The clinical evaluation of personalized abutments and crown bridge products are summarized,and the research content of clinical evaluation is clarified.Results The clinical evaluation requirements that need to be considered by enterprises are introduced.Conclusion Personalized abutment and abutment crown bridge products can refer to this study when they are launched in China,mainly using in vitro performance comparison tests for equivalence verification.
7.Characteristics of natural foci and spatial distribution of mosquitoes and ticks in Yadong County, Tibet Autonomous Region

Objective:To learn about the characteristics of natural foci and the spatial distribution of mosquitoes and ticks in Yadong County, Tibet Autonomous Region (Tibet for short).Methods:The eco-geographical characteristics, demographic information, agricultural and animal husbandry data of the natural foci in Yadong County, Tibet were collected from the Yadong County 2020 statistical yearbook, vector data were collected from the Yadong County Center for Disease Control and Prevention, and the data of the meteorological station in Yadong County from 2010 to 2021 were collected from the World Weather Network. In July 2021, a total of 20 mosquito and tick sampling points were selected for field investigation according to the distribution characteristics of vegetation and animal husbandry of Yadong County. The light trap method was used to trap mosquitoes, and the manual inspection and flag dragging method were used to catch ticks. The spatial distribution characteristics of mosquitoes and ticks were analyzed.Results:The natural epidemic foci in Yadong County, Tibet, were high in the north and low in the south, with an elevation difference of about 5 000 m. It was rich in water resources, and the average runoff of Yadong River was 20.1 m 3/s. The climate in the north was cold and dry, and the south was mild and humid. From 2010 to 2021, the annual average station air pressure in Yadong County was 452.8 mmHg (1 mmHg = 0.133 kPa), the maximum monthly average temperature, precipitation and relative humidity of air were 8.7 ℃, 134.5 mm and 81.3%, respectively, all in July. A total of 6 897 diptera insects were attracted by the light trap method, all of which were non-blood-sucking mosquitoes. The total density was 163.77 pieces/(lamp·h), the dominant population was Muscaridae, accounting for 89.69% (6 186/6 897). In different sampling areas, Xiayadong Township collected the most mosquitoes, accounting for 67.17% (4 633/6 897); the others were Yadong County and Shangyadong Township, accounting for 27.36% (1 887/6 897) and 5.47% (377/6 897), respectively. A total 2 014 host animals were examined, and 23 parasitic ticks were caught, of which 20 were of the genus Ixodes and 3 were of the genus Haemophilus. No free ticks were caught in all ticks sampling points. Conclusions:The climate and ecological environment of the natural foci in Yadong County, Tibet have obvious vertical gradient. Mosquitoes and ticks are active, but the density is not high, the density of mosquitoes is generally high in the south and low in the north.
8.Application and regulatory challenges of organoid technology in medical field

BACKGROUND:3D organoids have characteristics that resemble physiological tissues and to some extent mimic organ function,making them excellent models for applications ranging from basic development/stem cell research to personalized medicine. OBJECTIVE:To review and discuss the types of diseases and application areas such as tumor modeling that organoids can be applied to,as well as their regulatory status and challenges. METHODS:With"organoid,stem cell,disease model,3D printing technology,medical field"as Chinese and English search terms,we searched PubMed,Elsevier,WanFang,and CNKI databases to summarize and analyze organoid products at home and abroad,summarize the application of organoid technology in the medical field,and prospect the future development of organoid products in the medical field. RESULTS AND CONCLUSION:Organoids can break the limitations of traditional cell and animal models,avoid the ethical problems existing in clinical research,and have a high similarity to the source organ,a more similar performance to the physiology and pathology of human systems,and genetic stability,which has great advantages in current research.Organoids have been applied in the following fields:efficacy evaluation studies(preclinical models),including intestinal organoids,kidney organoids,liver organoids,gallbladder organoids,lung organoids,brain organoids,heart organoids,skin organoids,and reproductive system organoids;research on infectious diseases;cancer research and precision therapy;regenerative medicine;immune organoids.Although the United States,the European Union and China do not have perfect regulatory provisions,they are trying to promote the formulation of organoid regulatory laws and regulations.In China,although no organoid medical device products have been listed for the time being,its related regenerative medicine products have made breakthroughs.
9.Analysis of the key points of clinical application and clinical evaluation of capsule endoscope

By summarizing the clinical use of different capsule endoscopes in small intestine,colon,stomach and esophagus,the relevant evaluation indicators of the clinical application of capsule endoscopes for patients with different diseases were sorted out.Combined with the review process of capsule endoscopes that have been marketed at home and abroad,the key points of clinical evaluation of capsule endoscopes products in China that need to be marketed through clinical trials were considered.The clinical evaluation focuses included main evaluation index,secondary evaluation index and safety evaluation index,and the main evaluation index could consider the consistency rate of lesion diagnosis results and the excellent rate of image quality.The inclusion criteria for clinical trials of capsule endoscopy products were the population who intended to use the product,and the exclusion criteria need to consider the circumstances that were not applicable to such products and the circumstances that were not applicable to enrollment.
10.Nomogram model based on multimodal ultrasound for predicting sentinel lymph node metastasis in patients with breast cancer

Objective:To analyze the multimodal ultrasound characteristics of primary breast cancer and sentinel lymph node (SLN) and to establish a nomogram model for predicting SLN metastasis in invasive breast cancer, thereby providing reference for precise clinical diagnosis and treatment.Methods:A total of 329 patients diagnosed with invasive breast cancer and admitted to the Affiliated Hospital of Nanjing University of Chinese Medicine from June 2018 to October 2023 were retrospectively enrolled. They were randomly divided into a training cohort ( n=230) and a validation cohort ( n=99) in a ratio of 7 to 3. In the training cohort, ultrasound findings and clinical parameters were analyzed, univariate and multivariate Logistic regression analyses were used to identify independent predictive factors for SLN metastasis, and a nomogram model was constructed based on these factors. The ROC curve, calibration curve, and decision curve analysis (DCA) were plotted between the training and validation cohorts to assess the discrimination, calibration, and clinical applicability of the nomogram model. Results:Regression analysis identified 3 independent risk factors for establishing the nomogram prediction model: ratio of the long diameter to the short diameter of SLN ( P=0.020), lymphatic contrast-enhanced ultrasound (LCEUS) enhancement pattern ( P<0.001) and intravenous contrast-enhanced ultrasound (ICEUS) enhancement mode ( P=0.002). The area under the curve (AUC) of the training cohort was 0.888, the accuracy was 0.865; the AUC of the validation cohort was 0.870, the accuracy was 0.859, demonstrating good predictive performance of the model in both cohorts. The calibration curve demonstrated that the nomogram has a strong concordance between predicted and actual probability. DCA demonstrated that the nomogram could increase net benefit within a certain probability threshold range. Conclusions:The nomogram based on ratio of the long diameter to the short diameter of SLN, LCEUS enhancement pattern and ICEUS enhancement mode can effectively predict SLN status in patients with invasive breast cancer, facilitating precise diagnosis and treatment.
