1.Effects of epidural capsaicin with different contrention on pain threshold and nerve tissue structure in rats

Objective To evaluate the effects of epidural capsaicin on pain threshold and nerve tissue structure in adult rats. Methods Twenty-four adult male Wistar rats weighing 250-300 g were used in this study. A catheter was inserted into epidural space at L5,6 according to the method of Philippe. 0.4 ml of capsaicin 0.1% (group A), 0.25% (group B) or 0.5% (group C) or 10% Tween 80 (control group) was injected via the epidural catheter. Pain threshold was measured by thermal stimulation of the tail before (baseline) and on the 3rd, 7th and 14th day after epidural capsaicin. The animals were then killed. The lumbar segment (L4-6) of the spinal cord and spinal nerve roots were removed immediately for light and electron microscopic examination. 2.Role of mitochondrial KATP channel in reduction of renal ischemia- reperfusion injury by ischemic postconditioning in rats

Objective To investigate the role of mitochondrial KATP (mito-KATP) channel in reduction of renal ischemia-repeerfusion (I/R) injury by ischemic postconditioning (IPo) in rats. Methods Thirty-five adult male SD rats weighing 250-280 g were randomly divided into 5 groups ( n = 7 each): group Ⅰ sham operation (group S); group Ⅱ I/R; group Ⅲ IPo; group Ⅳ 5-HD + I/R and group V 5-HD + IPo. The rats were anesthetized with intraperitoneal (IP) chloral hydrate 300 mg/kg. Bilateral kidneys were exposed and their pedicles were occluded for 45 min with atraumatic mini-clamp followed by 6 h reperfusion in group Ⅱ - Ⅴ . In group Ⅲ and Ⅴ 3 cycles of 10 s reperfusion followed by 10 s ischemia were applied immediately after 45 min kidney ischemia. In group Ⅳ and Ⅴ 5-HD (a specific blocker of the mito-KATP channel) 10 mg/kg was given IP at 30 min before ischemia. Blood samples were obtained at 6 h of reperfusion for determination of serum creatinine (Cr) and urea nitrogen (BUN) concentrations. The animals were then killed. Bilateral kidneys were removed for determination of mitochondrial membrane potential in the renal tubular epidural cell and intracellular reactive oxygen species (ROS)content and free Ca2+ concentrations. Results Renal I/R significantly increased serum Cr and BUN concentrations and intracellular free Ca2+ concentration and ROC content and decreased mitochondrial membrane potential as compared with sham operation group. IPo significantly attenuated the I/R-induced changes mentioned above. The protective effects of IPo against renal I/R injury was reversed by 5-HD. Conclusion Mito-KATP channel is involved in reduction of I/R-induced renal injury by ischemic postconditioning.
3.Clinical study on changes of serum IL-17 and IL-35 levels in patients with heart failure

Objective To study changes of serum IL-17 and IL-35 levels in patients with heart failure. Methods 60 patients with heart failure (observation group)were selected as research subjects.60 patients accord-ing to different severity were divided into acute period heart failure (34 cases)and stable stage heart failure (26 ca-ses);60 patients graded according to the NYHA standards were divided into 24 cases of heart failure with grade Ⅱ, 20 cases of grade Ⅲ ,16 cases of grade Ⅳ.According to the different primary diseases :expansion cardiomyopathy group (20 cases in group A),the coronary heart disease group (group B,24 cases),hypertensive heart disease group (group C,16 cases).During the same period,42 healthy elderly people in our hospital were selected as control group. The serum IL-17,IL-35 levels were tested,and the serum IL-17,IL-35 levels in patients with heart failure were ana-lyzed.Results Serum level of IL-17 in the observation group was higher than the control group,and the difference was significant [(15.61 ±4.02)pg/mL vs (9.49 ±3.96)pg/mL,t =9.018,P <0.01].Serum level of IL-35 in the observation group was significantly lower than that of the control group,and the difference was significant[(52.78 ± 4.29)pg/mL vs (61.49 ±4.81)pg/mL,t =11.963,P <0.01].The level of serum IL-17 in acute stage of patients with heart failure was higher than that of stable heart failure,and the difference was significant (t =6.278,P <0.01);IL-35 level in serum of patients with heart failure in acute phase was lower than that of stable heart failure,the difference was significant (t =9.529,P <0.01).With the increase in heart failure grade,serum IL-17 level showed a rising trend,and the differences among three groups had statistical differences (F =6.098,P <0.01);serum IL-35 level decreased,and the differences among three groups had statistical differences(F =8.978,P <0.01).The serum IL-17 level of A group was higher than that in B group and C group,there were significant differences (F =6.096, P <0.01),the serum IL-17 level between B group and C group had no statistical difference (t =0.172,P >0.05). The serum IL-35 level of A group was lower than that of B group and C group,there were significant differences (F =8.978,P <0.01),the serum IL-35 level between B group and C group had no statistical difference (t =0.208,P >0.05).Serum IL-17 and serum IL-35 level was negatively correlated (r =-0.429,P =0.009).Conclusion High expression of IL-17 in elderly patients with heart failure,while IL-35 decreased in elderly patients with heart failure, IL-17,IL-35 are closely related to the senile congestive heart failure and the severity of illness.Serum IL-17 is nega-tively correlated with the level of serum IL-35.
4.Applications of Reference Extract from Ginkgo Leaf Total Lactones for Quantitative Assay of Ginkgo Folium

Objective To explore the feasibility of using the quantitative reference extract of ginkgo leaf total lactones instead of single component reference for the quantitative assay of Ginkgo Folium.Methods HPLC-ELSD method was performed by using a Diamonsil C18 column (250 mm×4.6 mm,5 μm) with methanol-water as the mobile phase at the gradient elution mode.Flow rate was 1.0 mL·min-1.The parameters of ELSD detector were as follows,the drifit tube temperature was 105 ℃,and the flow rate of nitrogen(N2) was 3 L·min-1.Results The linear ranges of ginkgolide A,ginkgolide B,ginkgolide C,and bilobalide were 0.735-5.879 μg (r=0.999 6),0.404-6.060 μg (r=0.999 6),0.296-4.439 μg (r=0.999 6),and 1.001-6.006 μg (r=0.999 7),respectively.The recoveries and RSD of the four components were 95.6% (4.0%),97.3% (4.5%),99.3% (5.0%),and 100.4% (2.1%),respectively.Conclusion The quantitative reference extract of ginkgo leaf total lactones can be used as the substitute for the determination of terpene lactones.
5.Effect of early enteral nutrition on mucosal structure and absorption of small bowel allograft

Objectives:To investigate the effect of early EN on mucosal structure and absorption function of allograft after living-related small bowel transplantation. Methods:Early enteral nutrition was used.The image analysis,D-xylose absorbtion test and blood culture were used to measure the villus height and area,D-xylose absorption and bacterial translocation in blood. Results:The morphometric examination showed that villus height and villus area were decreased on the 7th postoperative day,began to increase on the 12th day after transplantation and had been the 80% of normal level.The percentage of D-xylose absorption was gradually increased and reached the normal level on the postoperative 38th day.Postoperative blood cultures were always negative. Conclusions:Early EN support can improve the mucosal structure and absorption function of small bowel allograft after living-related small bowel transplantation.
6.Liver histological changes and influencing factors in patients with chronic hepatitis C

ObjectiveTo investigate the histological features as well as the factors influencing liver disease progression in Chinese patients with chronic hepatitis C (CHC). MethodsA total of 102 CHC patients who underwent percutaneous liver biopsy between August 2007 and May 2010 were recruited. Age, gender, body mass index (BMI) and transmission route of recruited patients were recorded. Serum levels of alanine transaminase (ALT) and aspartate transaminase (AST), HCV genotypes, HCV viral load and liver histological changes were detected. Statistical analysis was done by t test and Logistic regression. ResultsThe serum levels of ALT and AST in CHC patients with histological activity index (HAI) ≥4 were much higher, while platelet (PLT) counts were lower than those with HAI <4(t=2.209, 2. 298 and 2. 565, respectively; all P<0.05). Likewise, in patients with F≥3, the serum levels of ALT and AST as well as the mean age and the duration of infection were significantly elevated compared with F < 3 group ( t = 3.497, 2. 758, 2. 340 and 2. 570,respectively; all P<0. 05), while PLT counts were much lower (t = 2. 761, P=0. 007). The unvariate predictors for HAI≥4 were female, ALT>1 × upper limits of normal (ULN), AST level,F≥3, HCV RNA≥6 lgIU/mL and PLT counts. By mutivariate analysis, the Ishak stage score was the only independent predictor for HAI≥4 (OR 3.098, 95%CI 1.884-5. 092; P<0.01). Finally,the univariate predictors for F≥3 were age, BMI≥24 kg/m2 , ALT>1 × ULN, AST level, HAI≥4,PLT counts and duration of infection≥ 15 years. Multivariate analysis revealed that age (OR 1. 074,95%CI 1.006-1. 146; P=0.033), ALT level (OR 1. 035, 95%CI 1.015-1.055; P<0.01), ASTlevel (OR 0. 969, 95%CI 0. 948-0. 990; P=0. 005), the duration of infection ≥15 years (OR 37. 215, 95%CI 5. 816-238. 127; P<0.01) and HAI≥4 (OR 1. 939, 95%CI 1. 426-2. 636; P<0.01) were independent predictors for F≥ 3. ConclusionAge, ALT level, AST level, duration of infection≥15 years, HAI≥4 are independent predictors for liver fibrosis.
7.Effects of family function, resilience and focal lesion characteristics on acute stress disorder in patients with cerebral injury

Objective To investigate the status quo of the cerebral injury in patients with acute stress disorder (ASD) and the impact of family function,mental toughness,and the characteristics of brain lesions on it.Methods A total of 349 patients were enrolled from North China University of Science and Technology affiliated hospital neurology department and neurosurgery department from May 2016 to November 2016 and they were tested with Stanford Acute Stress Reaction Questionnaire (SASRQ),the family function assessment scale (APGAR),Chinese version of mental toughness scale (CD-RISC).Results 349 cases of patients with SASRQ score (57.21±44.97),8 to 39 160 people (45.8%),40 to 56 85 people (24.4%),57 to 150 104 people (29.8%).The results showed that whether the hemiplegia (β=-0.030),family function (β=-0.032),mental toughness (β=0.886),disturbing degree (β=0.052),bad days (β=0.060)were picked in the regression equation (P<0.05).Conclusion There were obvious acute stress symptoms in patients with brain injury.The individuals who have family dysfunction,the worse the psychological resilience and the higher the disturbance degree of the patients with hemiplegia,may be likely to get acute stress disorder.
8.Effect of mindfulness music training on sleep-wakefulness behavior in patients with acute stress disorder sleep disorder with cerebral hemorrhage

Objective To explore the effect of mindfulness-based music therapy (MBMT) on sleep-wakefulness behavior in patients with acute stress disorder (ASD) coexisting sleep disorder with cerebral hemorrhage. Methods A total of 70 patients with cerebral hemorrhage and ASD coexisting sleep disorders were randomly divided into two groups (n=35 each group). The study group was treated with mindfulness and soothing music training on the basis of routine treatment for 28 consecutive days. The control group only received routine treatment. Polysomnography (PSG) was used to compare sleep-wake behavior before and after intervention. Results After the intervention, the numbers of awakenings, awakenings greater than 5 min, micro awakenings, NREM micro awakenings, micro awakenings in phase 1, micro awakenings in phase 2, and REM awakenings were decreased in both groups (P<0.05). The degree of reduction of those parameters were greater and the proportion of total sleep time to recording time was longer in the study group than in the control group (P<0.05). Conclusion Music therapy based on mindfulness can effectively improve the sleep-wakeful behavior of ASD patients with cerebral hemorrhage, change the sleep process, and improve their sleep quality.
9.Effects of sevoflurane post treatment on cerebral ischemia-reperfusion injury in diabetic rats and non-diabetic rats

Objective To investigate the effects of sevoflurane post treatment on cerebral ischemia-reperfusion injury in diabetic rats and non-diabetic rats.Methods Thirty-two male SD rats,aged 3.0-3.5 months,weighing 280-320 g,were divided into four groups by random number method (n =8):non diabetic ischemia reperfusion group (group NDC);diabetic ischemia reperfusion group (group DC2);non diabetic ischemia reperfusion group with the sevoflurane post-treatment (group NDS);diabetic ischemia reperfusion group with the sevoflurane post-treatment (group DS).The middle cerebral artery occlusion method and streptozotocin were used to establish the ischemiareperfusion injury model and diabetic model.2 h after ischemia and 24 h after reperfusion,the rats nerve defect scale was detected,the cerebral infarction volume was evaluated by TTC method,and Western blot method was used to detect angiogenin-1 (Ang 1) expression.Results Between the four groups of rats,nerve deficit score and infarct volume were significantly higher,Ang-1 protein relative expression was significantly lower in group DC than those in group NDC (P<0.05);neural deficit score and infarct volume were significantly lower,Ang-1 protein relative expression was significantly higher in group NDS than those in group NDC (P<0.05);nerve deficit score and infarct volume were significantly lower,Ang-1 protein relative expression was significantly higher in group DS than those in group DC (P<0.05).Conclusion In the rats after cerebral ischemia reperfusion,diabetes could aggravate the nerve defect,increase the volume of cerebral infarction,reduce the expression of Ang-1;sevoflurane could reduce nerve defects,reduce infarct volume,increase the expression of Ang-1.The expression of Ang-1 and degree of injury after cerebral ischemia reperfusion in rats have relationship.
10.Mechanism of Yifei Sanjie Prescription in Treatment of Lung Adenocarcinoma via JAK2/STAT3/VEGF Signaling Pathway Based on Network Pharmacology and Experimental Verification

Objective To predict the potential mechanism of Yifei Sanjie prescription in the treatment of lung adenocarcinoma based on network pharmacology,and to verify one of the key signal pathways,Janus protein tyrosine kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3),by cell experiments in vitro.Methods To screen the main active components and potential action targets of Yifei Sanjie prescription,with traditional Chinese medicine system pharmacological database(TCMSP).To search and retrieve the main targets of lung adenocarcinoma,with human genetic database(GeneCards)and online human Mendelian genetic database(OMIM).To obtain the intersection targets by screening and apply Wayne diagram,then analysis the topology and establish the traditional Chinese medicine-active compound-target network diagram by using of Cytoscape 3.7.2 software.To construct the protein-protein interaction(PPI)network,with the protein-protein interaction platform(STRING)and Cytoscape3.7.2 software.To analyze the functional enrichment of gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG),with the Metascape database.To carry out the molecular docking verification by using of Vina1.2.3 software.Using CCK-8 method to detect the effect of Yifei Sanjie prescription on cell activity.Using the cell scratch test to observe the effect on cell migration.And using Western blot method to test the expression of p-STAT3,STAT3,p-JAK2,JAK2 and VEGF-A.Results 94 active components,329 related drug targets and 1358 lung adenocarcinoma targets were obtained from Yifei Sanjie prescription,among which,150 of them intersected.PPI network visualization analysis shows that the potential key targets of Yifei Sanjie prescription in the treatment of lung adenocarcinoma are protein kinase B1(AKT1),β-actin(ACTB),tumor suppressor gene p53(TP53),serum albumin(ALB),caspase-3(CASP3)and vascular endothelial growth factor A(VEGFA).KEGG enrichment analysis screened 138 related signal pathways,indicating that JAK/STAT signaling pathway may play a key role in the treatment of lung adenocarcinoma with Yifei Sanjie prescription.Molecular docking results showed that quercetin,luteolin,and ursolic acid had good binding activities with JAK2 and STAT3.The cell experiment showed that compared with the blank group,Yifei Sanjie prescription could significantly inhibit the activity of A549 cells,inhibit the migration of A549 cells,and decrease the expression of p-JAK2/JAK2,p-STAT3/STAT3 and VEGF-A protein.In addition,Colivelin,an activator of JAK2/STAT3 pathway,could reverse the effect of Yifei Sanjie prescription on the expression of A549 related proteins.Conclusion Yifei Sanjie prescription has the characteristics of multi-component,multi target and multi pathway in the treatment of lung adenocarcinoma,and its mechanism may be related to the down-regulation of p-JAK2,p-STAT3 and VEGF-A protein expression,thereby inhibiting cell proliferation and migration.
