1.Analysis on pathogenic bacterium distribution and drug resistance of the patients with lower respiratory tract infection

ObjectiveTo analyze pathogenic bacterium distribution and drug resistance to the inpatients suffered from lower respira.tory tract infection in our hospital,and to guide adminisrration in clinical. MethodsThe inpatients' sputum specimens were collected,and automatic microorganism identification and drug resistance test were conducted,and the records were taken down and statisticly analyzed. Results Among 321 sputum examples,194strains pathogenic bacterium were isolated.And pathogenic bacterium distributed were as following: there were 126 strains gram-negative bacilli,accounting for 64.95%,there were 32 strains of gram-postive cocci,accounting for 15.98%,and there were 37 strains of fungi,accounting for 19.08%.The drug resistance of gram-negative bacilli to ampicillin,trimethoprim-sulfamethoxazol were relatively high,accounting for 87.45%~97.74% and 75.37%~91.75%,respectively.Pseudomonas aeruginosa and baumanii were resistant to selected antibiotics at high degree,and showed multiple drug resistance. ConclusionThe species of pathogenic bacterium were various.Gram-negative bacteria were the rnain pathogenic bacteria involved in respiratory tract infection.The drug resistance was high in Pseudomonas aeruginosa and baumanii.
2.Effects of transcription factor HOXB7 silencing on viability, invasion and migration of SGC-7901 cells

[ ABSTRACT] AIM:To explore the effects of transcription factor HOXB7 silencing on the viability , invasion and migration of SGC-7901 cells.METHODS:The siRNA was designed to specifically interfere the expression of HOXB7.To assess the silence efficiency of the siRNA , the expression of HOXB7 at mRNA and protein levels was detected by real-time PCR and Western blot in the SGC-7901 cells transfected with siRNA .The cell viability was measured by MTT assay .Cell cycle-related proteins such as cyclin D 1 and CDK4 were determined by Western blot .Transwell assay was performed to an-alyze the migration ability of the SGC-7901 cells.The mRNA and protein levels of the tumor invasion-and metastasis-relat-ed molecules, such as VEGF, PTEN and p-AKT, were also detected by real-time PCR and Western blot .RESULTS:The expression of HOXB7 at mRNA and protein levels was higher in the gastric carcinoma tissues than that in the normal gastric tissues.The expression of HOXB7 at mRNA and protein levels was knockdown by siRNA .After silencing of HOXB7 gene expression, the viability of the SGC-7901 cells decreased, the expression of cyclin D1, CDK4 and VEGF was down-regula-ted, and the phosphorylation level of AKT was also obviously decreased .CONCLUSION:HOXB7 effectively promotes the viability, invasion and migration of gastric carcinoma cells by up-regulating the expression of cyclinD 1, CDK4 and VEGF, and increasing the phosphorylation level of AKT to inhibit PTEN function .

The estimation of a-tocopherol in human blood was carried out by gas chromatography. The cholesterol in plasma must he removed in advance. The a-tocopherol is chromatographed as tocopherol acetate and is eluted with a retention time of about 8 min. Tetratriacontane was used as internal standard. The average recovery rate was 76.4 per cent and coefficient of variation was 4.4 per cent (N = 10). The mean level in 25 cord blood was 2.1 ?0.5 ?g/ml. Strengthening the construction of supervisor team has great effects on the improvement of the quality of postgraduate education.Some problems exist in China's current situation of the construction of supervisor team.For example,the quality of supervisors needs to be improved,the quantitative evaluation system is not scientific,the quantity of professional degree supervisors is not enough,and the dynamic evaluation mechanism is imperfect and so on.Strengthening the construction of supervisor team from the development of scientific and rational quantitative evaluation index system,the selection of professional degree supervisors,the enhancement of supervisor training and some other aspects,can effectively improve the quality of postgraduate education.
5.Effects of Tianma Gouteng Decoction on mRNA expression of CaL-?1C and PMCA1 in vascular smooth muscle cells of spontaneously hypertensive rat

AIM:To observe the effects of Tianma Gouteng Decoction on mRNA expression of L-Type Calcium Channel ?1 C subunit (CaL-?1C) and plasma membrane calcium-ATPase (PMCA1) in vascular smooth muscle cells of spontaneously hypertensive rat (SHR). METHODS:Twelve-week-old SHRs were assigned randomly to 5 groups:group A (treated with Tianma Gouteng Decoction),group B (treated with Tianma Gouteng Decoction excluded concha haliotidis),group C (treated with Nifedipine),group D (treated with concha haliotidis),group E (treated with normal sodium,NS),each group consisted of 9 rats was adminstrated,once a day for 4 weeks and mRNA expression of CaL-?1C and PMCA1 was measured with RT-PCR half-fix-quantify method. RESULTS:After 4-week-treatment,both Tianma Gouteng Decoction,Tianma Gouteng Decoction excluded concha haliotidis and Nifedipine could regulate downward mRNA expression of CaL-?1C in vascular smooth muscle cells and regulate upward mRNA expression of PMCA1 in vascular smooth muscle cells,concha haliotidis has no effect on mRNA expression of CaL-?1C and PMCA1. CONCLUSION:Tianma Gouteng Decoction can regulate downward expression of CaL-?1C,and regulate upward expression of PMCA1 at the same time,and meliorate the Ca 2+ overload in vascular smooth muscle cells with hypertension.
6.The clinical effects of valsartan on the insulin resistance of type 2 diabetes patients with Ⅰ hypertension

Objective To investigate the effects of valsartan on insulin resistance of type 2 diabetes in patients with Ⅰ hypertension.Methods Newly diagnosed 152 patients of type 2 diabetes with Ⅰ hypertension were randomly divided into treatment group and control group.The treatment group and control group undergone lifestyle changes( diet and adequate exercise) for two weeks.After that,the control group were given Diltiazem,while the treatment group were given valsartan,an angiotensin receptor blocker(ARB) drugs.Both groups were treated for 8 weeks.Fasting plasma glucose (FPG) and fasting plasma insulin (FINS) before and after treatment in both groups were determined.Homeostasis model assessment evaluation of insulin resistance index(HOMA-IR) and insulin sensitivity index (IAI) were carried out and compared between the two groups.Results After 8 weeks of treatment,HOMA-IR in the treatment group was significantly decreased compared with the control group ( P < 0.01 ),while IAI was significantly increased( P < 0.01 ).Conclusion Application of valsartan in type 2 diabetes mellitus with Ⅰ hypertension can significantly improve the level of insulin resistance.
7.Variation of serum ghrelin and leptin in patients with functional dyspepsia

Objective To investigate the alteration of serum ghrelin and leptin in patients with functional dyspepsia (FD) and their clinical significance. Methods Sixty patients with FD [-30 patients with postprandial distress syndrome(PDS) and 30 patinets with epigastric pain syndrome(EPS)] were enrolled. Thirty healthy subjects were severed as controls. The serum ghrelin and leptin were measured by enzyme linked immunosorbent assay and radioimmunosorbent assay, respectively. Results The concentrations of serum ghrelin and leptin were both significantly lower in patients with FD than in controls (P<0.01). Meanwhile, the concentrations of serum ghrelin and leptin in PDS patients were significantly lower compared with controls (P<0.01) and EPS patients (P<0.01). There was no significant difference in levels of serum ghrelin and leptin between controls and EPS patients(P>0.05). Conclusions The alterations of serum ghrelin and leptin in PDS patients are responsible for their reduction in FD patients. The PDS may he associated with abnormalities in gastrointestinal motility. There is an interaction between serum ghrelin and leptin in development of FD. The detection of serum ghrelin or leptin is helpful in classification and treatment of FD.
8.Mechanism of effect of Ginkgolide B on lipopolysaccharide induced microvascular endothelial cells

Objective To investigate the effect of Ginkgolide B (BN52021) on lipopolysaccharide (LPS) induced pancreas microvascular endothelialv (MS1) cells, and to explore its molecular mechanism.Methods The optimal concentration and best time point of LPS inhibing MS1 cell survival and the optimal concentration of BN52021 increasing survival of LPS induced MS1 cells were determined by MTT.The mRNA and protein expression of adenylate cyclase (AC), phospholipase A2 (PLA2), phospholipase Cβ (PLCβ),protein tyrosine kinase (PTK) and G protein coupled receptor kinase (GRK) in platelet activating factor receptor(PAFR) signal pathway in MS1 cells were determined by real-time PCR and Western blot.Results It was showed that 10 μg/ml LPS for 24 h was the optimal concentration and best time point to induce the decrease of MS1 cells.50 mmol/L of BN52021 was the optimal concentration of increacing survival of LPS induced MS1 cells.After LPS induction, AC, GRK, PLA2, PLCβ, PTK mRNA expressions of MS1 cells were 4.02 ±0.14, 2.63 ± 0.03, 3.31 ± 0.12, 2.09 ± 0.08, 1.85 ± 0.07, which were significantly higher than those in control group (P < 0.01).After BN52021 treatment, AC, GRK, PLA2, PLCβ mRNA expressions of LPS induced MS1 cells were 2.35 ±0.13, 1.17 ±0.14, 1.87 ±0.11, 1.65 ±0.10, which were significantly lower than those in LPS induction group (P < 0.01).The expression of PTK mRNA was 1.83 ± 0.13, which was not significantly different from that in LPS induction group.Western blot showed that the levels of protein expression were consistent with those of mRNA expression.Conclusions BN52021 can down-regulate the up-regulated genes expression of AC, GRK, PLA2 and PLCβ in the PAFR signal pathway in LPS induced MS1 cells.
9.ANALYSIS OF FREE FATTY ACID IN HUMAN BLOOD BY GAS CHROMATOGRAPHY

Serum free fatty acids were isolated with trichloromethane and esterified with boratotrifluoricether/methanol. Heptadecanoic acid (17:0) was used as internal standard. The recovery rate of this method was 86.4% and coefficient of variation was 6.9%. The mean level of 25 normal adult serum samples was coincident with the data reported.
10.The application effects of PDCA circulation on behavior management in discharged patients with first cerebral infarction

Objective To explore the application effects of PDCA circulation on behavior management in discharged patients with first cerebral infarction. Methods Patients with first attack of cerebral infarction were enrolled from the department of neurology of the First Hospital of Anhui Medical University from April 2013 to March 2015. According to their admission numbers, patients were divided into the observation group (94 cases) and the control group (80 cases). Besides of the regular personalized secondary prevention, the observation group was also applied for the PDCA cycle for nine months. Stroke Behavior Change Inventory was used to evaluate the changes of their lifestyle and medication compliance one year after discharge for two groups. Results After the observed patients were discharged one year, the changed extent of these five behaviors including quitting smoking, regular monitoring blood pressure and sugar, appropriate exercise, pressure control in the observation group were 0.00 (0.00, 3.00), 1.68 (1.00, 3.00), 1.00 (0.00, 2.00), 1.20 (0.75, 2.00), 1.00 (0.00, 1.00) points, and these in the control group were 0.00 (0.00, 1.0)0, 1.00 (0.00, 2.00), 0.00 (0.00, 1.00), 1.00 (1.00, 2.00), 0.00(0.00, 1.00) points. The observation group in these parameters had significantly higher than the control group (Z=-3.166--2.419, P < 0.05). The changed extent of these behaviors in the medication compliance such as reducing blood glucose, blood pressure, atrial fibrillation, cardiovascular disease, and stroke prevention in the observed group were 0.00 (0.00, 0.00), 0.00 (0.00, 2.00), 0.00 (0.00, 0.00), 0.00 (0.00, 0.00), 3.00 (3.00, 3.00) points, and these in the control group were 0.00 (0.00, 0.00), 0.00 (0.00, 0.79), 0.00 (0.00, 0.00), 0.00 (0.00, 0.80), 0.00 (0.00, 1.50) points. The observation group in these parameters had significantly higher the above parameters than the control group (Z=-8.081--2.595, P<0.05). Conclusions Application of the PDCA circulation in the patients with first attack of cerebral infarction after their discharge can help them correct lifestyle and increase their medical treatment compliance, which is beneficial for secondary prevention of cerebral infarction.
