1.Expression of midkine and microvessel density in salivary adenoid cystic carcinoma.

OBJECTIVEThis study aimed to investigate the expression of midkine (MK) and microvessel density (MVD) in patients with salivary adenoid cystic carcinoma (SACC) and its clinical significance, as well as detect the correlation between the expression of MK and MVD in SACC.
METHODSImmunohistochemistry analysis (SP method) for MK and MVD were performed on 60 cases of SACC and 26 cases of normal salivary gland tissue. The expression of MK and MVD, as well as the correlation between the expression of MK and MVD in SACC were detected.
RESULTSIn SACC, the MK expression rate was 70.0% (42/60), and MK was not expressed in normal tissue. Statistical significance was found between SACC and normal tissue (P<0.05). The MVD values in SACC and normal salivary gland tissues were 38.73 +/- 8.96 and 11.15 +/- 3.33, respectively. These values were statistically significant (P<0.05). The expression levels of MK and MVD were unrelated to age, gender, and type in SACC (P>0.05), but correlated with tumor size, lymph node metastasis, and tumor-node-metastasis in SACC (P<0.05). The expression of MK and MVD was positively correlated with SACC (r=0.560, P<0.05).
CONCLUSIONSACC is correlated with the expression of MK protein and the increase in MVD, which may be some of the early diagnostic markers in SACC.

2.Cloning of flavone synthase (FNSII) gene and expression in three cell lines of Saussurea medusa.

Saussurea medusa is a rare traditional Chinese medicinal herb, of which luteolin is the niain active medicinal compound for cancer prevention and treatment. A full-length FNSII gene, namely SmFNSII (GenBank Accession No. KF170286), was obtained from green cell line of Saussurea medusa by RT-PCR and RACE-PCR. Sequence analysis indicated that SmFNSII is 1 710 bp in full length, containing a 34 bp 5'-untranslated region (5'-UTR), a 125 bp 3'-UTR, and a 1 551 bp open reading frame (ORF) encoding 516 amino acid residues. Amino acid sequence analysis indicated that SmFNSII belonged to subfamily CYP93B of plant cytochrome P450. Sequence alignment and phylogenetic analysis revealed that amino acid sequences of SmFNSII shared 87% homology with the protein in Hieracium pilosella. Quantitative real-time PCR analysis indicated that SmFNSII expression is the highest in red cell line and the lowest in white cell line, corresponding to quantitative analysis of luteolin concentration. pET-SmFNSII, a prokaryotic expression recombinant plasmid, was constructed and transferred into Escherichia coli, and the expressed protein band was the same size with predicted protein. Saussurea medusa cultivars with high anti-inflammatory, anti-cancer activities and health care function would be cultivated through filtering cell lines and plants with high expression level of FNSII gene and luteolin accumulation.

3.Effects of Bushen Wenyang Huayu Recipe on TRPV1 and Sensitization Factor NGF in Experimental Endometriosis.

OBJECTIVETo study the effect of Bushen Wenyang Huayu Recipe (BSWYHYR) on nerve growth factor (NGF) and transient receptor potential vanilloid receptor I (TRPV1) in experimental endometriosis (EMT), and to explore its mechanism for treating EMT-induced pain.
METHODSTotally in-bred line BALB/c 75 female mice were divided into five groups, i.e., the sham-operation group, the model group, the high dose BSWYHYR group, the low dose BSWYHYR group, the gestrinone group, 15 in each group. Writhing response was observed in each group. Serum contents of NGF were detected using ELISA. Expression levels of NGF and TRPV1 in uterus and ectopic foci were detected using immunohistochemical staining SP and Western blot. mRNA expression levels of NGF and TRPV1 in uterus and ectopic foci were detected by Real-time PCR.
RESULTSThe serum NGF content in the model group was higher than that in the sham-operation group (P < 0.01), and there was positive correlation between NGF and the writhing frequency (r = 0.574, P < 0.01). Compared with the model group, serum levels of NGF significantly decreased in the 3 treatment groups (P < 0.05). Compared with the sham-operation group, mRNA and protein expression levels of NGF and TRPV1 increased significantly in the model group (P < 0.01). Protein expression levels of NGF and TRPV1 decreased significantly in the 3 treatment groups, when compared with the model group (P < 0.01). mRNA expression levels of NGF and TRPV1 decreased most in the high dose BSWYHYR group (P < 0.01). NGF in uterus and ectopic foci was positively correlated with protein and mRNA expression levels of TRPV1 (P < 0.01).
CONCLUSIONSNGF and TRPV1 participated in the occurrence of pain in EMS. BSWYHYR played an important role in inhibiting EMT-induced pain through reducing the up-regulation of NGF on TRPV1.

4.Affect of protein tyrosine phosphatase non-receptor type 2 and nuclear factor-kappaB on periodontal destruction with diabetes.

OBJECTIVETo investigate the expression of protein tyrosine phosphatase non-receptor type 2 (PTPN2) and nuclear factor-kappaB (NF-kappaB), as well as the relationship between their expression and periodontal destruction in mice with diabetic periodontitis.
METHODSFour weeks old healthy C57BL/6J mice were randomly divided into normal control group (Group N), periodontitis group (Group P) and diabetic periodontitis group (Group DP), and each group had six mice. Mice in Group P were inoculated Porphyromonas gingivalis orally to induce periodontitis. Mice in Group DP had high sugary and fatty food, streptozotocin intraperitoneal injection and Porphyromonas gingivalis oral inoculation to induce diabetic periodontitis. All mice were sacrificed 4 weeks after the last bacterium inoculation of Groups P and DP. Stereo microscope was chosen to detect morphological changes and bone loss areas of the alveolar bone. Hematoxylin-eosin (HE) staining was selected to observe loss heights of periodontal attachment. Immunohistochemical staining was used to detect PTPN2 and NF-kappaB expression in periodontal tissues.
RESULTSGroup P and Group DP had significantly more attachment loss heights and areas than Group N (P < 0.05), and showed less PTPN2 expression (P < 0.05) and higher NF-kappaB levels (P < 0.01).
CONCLUSIONPTPN2 may negatively regulate the development of diabetic periodontitis, while NF-kappaB may have opposite effects. PTPN2 down-regulation might contribute to NF-kappaB over-expression leading to exacerbated periodontal destruction.

5.Algorithm of brain pathology detection based on statistical atlas of attribute vectors.

To detect lesions of brain CT automatically, a statistical atlas of attribute vectors (SAAV) was designed and created to describe the multiple features of medical images. By comparing the features of study image with those of SAAV, we successfully detected various kinds of brain lesion. It was demonstrated that the algorithm is effective in detecting various kinds of lesions found on brain CT images. Further studies are needed to make the algorithm more acceptable.

6.Long-term benefit of male circumcision to the reduction of urinary tract infections and genitourinary cancers in China.

Increasingly accumulated results from randomized controlled trials and other clinical studies have demonstrated that male circumcision reduces the risks of acquisition and transmission of HIV, HPV, HSV-2, and other sexually transmitted infections, and thus has a potential role in preventing cervical cancer, penile cancer and prostate cancer. The prevalence of male circumcision in China is currently less than 5%. The clinical evaluation studies and randomized controlled trials of the Shang Ring device showed excellent safety profiles, extremely high acceptability, and satisfaction among the participants and service providers in Africa and China. Given the recent recommendations by the World Health Organization and the Joint United Nations Program on HIV/AIDS (UNAIDS), voluntary medical male circumcision should be promoted in China at the national level as an important alternative intervention to reduce reproductive tract infections and prevent both males and females from reproductive tract cancers. More emphasis is required on the studies of the long-term health benefits of male circumcision in uro-andrology.
