1.Effect of medroxyprogesterone acetate on proliferation and apoptosis of human colorectal cancer cells

Objective:To investigate the effects of medroxyprogesterone acetate(MPA) on the proliferation and apoptosis of human colorectal cancer cells.Methods: Human colorectal cancer cell line LS174T was treated with different concentrations of MPA(12.5,25,50 and 100 ?mol/L) for 120 h.The inhibitory effect of MPA was measured with MTT method;the cell cycle,cell apoptosis,and cell surface Fas expression were analyzed with flow cytometry.Results: MPA had significant inhibitory effect on the growth of LS174T human colon cancer cells.Cell growth was found to be arrested in G_(1) phase and cells in S phase and G_(2)/M phase were decreased.The natural apoptosis rate and the apoptosis rates at MPA 12.5 and 25 ?mol/L were 4.8%,15.1%and 25.6%,respectively.Cell surface Fas expression in cancer cells increased after treated with MPA.Conclusion: MPA has significant inhibitory effect on the growth of human colon cancer cells,which may be related to Fas expression upregulation on cancer cell surface and G_(1) phase arrest.
2.Molecular cloning and expression of a stress-inducible gene highly homologous to human structural maintenance of chromosome 6

BACKGROUND: Previous studies have demonstrated that the responses of myocardium to short periods of ischemia/reperfusion are associated with changes in the expressions of a variety of genes. However, the characteristics of these changes are unknown.OBJECTIVE: To clone and analyze a stress-inducible gene characteristics in order to study the molecular mechanism of myocardium to short periods of ischemia/reperfusion damage.DESIGN: A controlled animal study.SETTING: Department of Hematology, Xiangya Hospital of Central South University.MATERIALS: Sixteen castrated male German Landrace-type domestic porcines, weighing between 21 and 39 kg, were provided by the laboratory of Department of Experimental of Cardiology at Bad Nauheim in Germany. The porcines were randomly divided into two groups ischemia/reperfusion group (n =14) and control group (n =2). The porcines in the ischemia/reperfusion group were observed at 0, 30 and 90 minutes of reperfusion after reocclusion, and ischemia and non-ischemia myocardial tissues were harvested from each pocrine.METHODS: The experiments were performed in the laboratory of Department of Experimental Cardiology of Max-Planck Institute for Physiological and Clinical Research at Bad Nauheim in Germany and the Department of Hematology, Xiangya Hospital of Central South University from July in 1998 to May in 2007. All the animals were anesthetized and thoracotomized. Following 30 minutes of stabilization, the left anterior descending coronary artery (LAD) was occluded for 10 minutes followed by 30-minute reperfusion and then another 10-minute reocclusion. The porcines were killed immediately at corresponding time points. The sham-operated animals were killed without occlusion. Experimental tissue was removed from the LAD area and control tissue from the region of the left circumflex coronary artery area. Firstly, a porcine heart cDNA library was screened, DNA and deduced amino acid sequences were then analyzed. Meanwhile, short periods of myocardium ischemia/reperfusion was performed by occluding porcine LAD followed by reperfusion as mentioned above, and total RNAs isolated from myocardium or a variety of other tissues were used for Northern blotting. Quantitation of mRNA levels was accomplished by using a PhosphorImager and Image Quant software, the ration of mRNA/18S rRNA was suggested as the level of gene expression.MAIN OUTCOME MEASURES: ① DNA and amino acid sequences analysis of the cloned gene; ② Analysis of the cloned gene expression.RESULTS: All the 16 porcines were involved in the final analysis of results. After screening, a cDNA fragment with 3 461 base pairs was obtained. DNA sequencing and searching revealed that this cDNA shared 86% identity to human structural maintenance of chromosome 6 gene (hSMC6), and 84% identity to Mus musculus SMC6 (mSMC6). Furthermore, a largest polypeptide deduced from this cDNA contained 1007 amino acid residues, and protein homology searching indicated that the predicted polypeptide carried 92% and 89% identity to hSMC6 or mSMC6 protein respectively. Thus, the cloned cDNA was referred to as a porcine homology gene to hSMC6 (pSMC6). The results of Northern blotting showed that the expression of the pSMC6 mRNA in ischemia/reperfusion myocardium was increased, and its mRNA existed in all organs tested. CONCLUSION: A stress-inducible gene of pSMC6 from porcine myocardium has been cloned, which is high homology to hSMC6 gene, and the pSMC6 or along with other molecules is possible involved in the early repair of myocardium to DNA damage caused by ischemia/reperfusion in the porcine heart.
3.The impact of the Jiawei Xiaochaihu particle and it's prescription on immune function of ITP mice

Objective:To observe the impact of the Jiawei Xiaochaihu particle and it's prescription on immune organ and immune function of ITP mice.Methods:The ITP mice were divided into five groups at random:(1)Nomal group;(2)ITP model group;(3)Jiawei Xiaochaihu particle group;(4)Xiaochaihu particle group;(5)Danshen and Sanqi particle group.All ITP mice were administrated 7days later by 30ml per kilogram the rat's weight once a day for 14 days.21 days after the model mice were made,peripheral blood T lymphocyte subpopulation was detected with flow cytometry.Antibody against platelets was measured by ELISA.Peripheral blood IL-2 and IL-10 were measured by radio immuno assay(RIA).Thoracic gland and spleen were weighted to compute the immune organ index.Results:Spleens of ITP mice had intumesce of spleens and a rise of the index;Peripheral blood platelet antibody was also increased;A disproportion was found in T lymphocyte subpopulation,CD8 upgrade,CD3 and CD4 descent,CD4/CD8 also descent.As one of the Th1 cytokine,IL-2 expression accentuated while IL-10,the kind of Th2 cytokine,was inhibited.There were significances when compared with the nomal group(P0.05).Conclusion:There are indeed dysfunction in immune organ and its function of ITP mice.Jiawei Xiaochaihu particle and Xiaochaihu particle can exert a regulation,but the Danshen and Sanqi particle not.
4.The application of high-quality nursing care principle in nursing students′clinical tutoring at oral and maxillofacial-head and neck oncology ward

Objective To apply high-quality nursing principles in the nursing education, allow nursing students to adapt to the modern nursing pattern more effectively in the oral and maxillofacial-head and neck oncology ward, and then to evaluate the effect of this application. Methods From June 2013 to March 2014, 60 nursing students had practiced in our department were randomly divided into the control group (30 cases, traditional teaching method) and the experimental group (30 cases, high-quality nursing teaching method). Nursing expertise, nursing procedures, teachers′ evaluation and patients′ satisfaction degree were evaluated. Results The nursing expertise, nursing procedures, teachers′ evaluation and patients′ satisfaction scores was (92.97 ± 5.65), (92.93 ± 4.38), (94.20 ± 2.95), (93.60 ± 3.97) in the experimental group respectively, while in the control group was(87.93 ± 4.62),(87.55 ± 7.54),(89.67 ± 6.21),(89.50 ± 5.80)respectively. All the four evaluation indexes of the experimental group were better than that of the control group(t=-3.78,-3.38,-3.61,-3.20), P﹤0.05. Conclusions Applying high quality nursing care principle in the nursing education allows students to adapt to the modern nursing pattern more quickly, and to establish foundations leading to their future professions.
5."Analysis of ""one-card""mode for division-level cadres hospitalized at military hospitals"

Objective To analyze the feasibility of theone-cardmode for division-level cadres hospitalized at military hospitals, and to provide reference for application of this mode to cadres under the regimental level who are hospitalized at military hospitals.Methods A 4-month pilot project was carried out among division-level cadres in Lanzhou, Jinan, Nanjing and Chengdu.Data collection and statistical analysis were conducted through the monitoring platform established in the administrative section.Results Division-level cadres outside the system mostly chose division-level hospitals.Patients thended to be hospitalized at nearby or high-quality hospitals mainly due to cardiovascular and cerebrovascular diseases and malignant tumor.Accommodation was not difficult.Conclusion The one-cardmode is feasible at military hospitals for division-level cadres, but the current system of initial diagnosis at basic medical institutions should be maintained.An information sharing platform of electronic medical records should be built to implement inter-hospital linking and sharing of diagnosis and treatment information.A budget subsidy mechanism should be established to normalize hospitalization.
6.Facial nerve and the surrounding vessels: an anatomic study

Objective:To study the relationship between facial nerve and surrounding vessels. Methods:Dissect skull of cadaver 40 sides of 20 cases: open the calvaria and clean the cerebrum, then open the tentorium of cerebellum, object and measure distance between the facial nerve and the nearest vessel, follow the course of it. Results:60%(24/40) of the arteries have the comprission with the facial nerve; among the nearest vessels, 87.5%(35/40) of them are anterior inferior cerebellar artery, and 12.5% (5/40) are posterior inferior cerebellar artery; the distances between facial nerve and the nearest vessels are from zero to 5.78mm, with an average of 1.14 mm; 94.3%(33/35) of the anterior inferior cerebellar artery emitted from the lower 2/3 of the basic artery. Conclusion:The most familiar vessel next to the facill nerve is the anterior inferior cerebellar artery, and the second is the posterior inferior cerebellar artery; most of the anterior inferior cerebellar artery emitted from the basic artery, exactly, the lower 2/3 of the artery.
7.Analysis of screw placement accuracy following mini-open anterior correction for adolescent idiopathic scoliosis

1 mm); A, the screw tip was adjacent to the aorta (≤1 mm); C, the screw tip was felt to be against the aorta and creating contour deformity. The angle composed of the central line of the screw and the central line of vertebral body on posterior X-ray film was also measured. Results The average correction of the main thoracic curve was 77.7% postoperatively. 155 screws were inserted, of which 134 screws(86.5%) had a bicortical purchase. 123 screws(79.4%) were distant from the aorta. Two screws encroached into the spinal canal. There were no significant difference between the proximal screws, the periapical screws and the distal screws. There were no vascular or neurologic complications or instrumentation failure during operation or follow-up. Conclusion Mini-open anterior correction for type Lenke 1 scoliosis enable a satisfy bicortical screw placement and curve correction. Sequential CT scan before surgery may improve the accuracy of screw placement, thus avoid the aortic or neurologic complication.
8.Video-assisted Thoracoscopic Cardiac Surgery for Congenital Heart Diseases

Objective To probe the feasibility of video-assisted thoracoscopic operation for congenital heart diseases.Methods From October 2004 to August 2008,156 patients with congenital heart disease,including 79 cases of atrial septal defect(ASD),65 cases of ventricular septal defect(VSD),1 case of aortic aneurysm rupture combined with VSD,3 cases of double chamber right ventricles(DCRV) combined with VSD,and 8 cases of partial anomalous pulmonary venous drainage,were treated totally under thoracoscope.Surgical procedures were performed through 3 trocars inserted at the right chest wall,and catheters were placed in the right femoral artery and vein to set up extracorporeal circulation.The ascending aorta was cross-clamped with long tailor-made forceps and the myocardium was protected by coronary perfusion with cold crystalloid cardioplegia. Results All the thoracoscopic operations were completed successfully.The mean extracorporeal circulation and cross-clamping time were(74?28) min and(29?13) min respectively.Postoperative ventilation was withdrawn in(3.2?1.1) h,and the patients were discharged from the hospital in(6.1?1.2) d.Thirteen of the patients had postoperative complications,including 7 cases of right pneumothorax(healed by thoracentesis),and 6 cases of fat liquefaction of the incision at the right axillary(delayed healing).No severe complications occurred in this series.UCG performed 5-7 days after the operation revealed no residual shunt of ASD or VSD;in the patients who had aortic aneurysm rupture combined with VSD,slight aortic valve regurgitation was detected after the neoplasty;and in the 3 patients with DCRV combined with VSD,the right ventricular outflow tract was patent.ECG showed sinus rhythm without AVB in the patients.Follow-up up to 3 months to 3 years were available in 132 cases(over 12 months in 95 cases).During the period,no patient had complaints;and the heart function was confirmed as level Ⅰ.Conclusions Video-assisted thoracoscopic cardiac surgery is feasible,safe,and minimal invasive for patients with VSD or ASD,resulting in quick recovery and good cosmetic outcomes.
9.Progress on tissue engineered trachea research

10.An application of template in intensity modulated radiotherapy planning

Objeetive To study the application of template in intensity modulated radiotherapy (IMRT)planning.Methods With Philips Pinnacle3(R) version 7.6c treatment planning system(TPS) platform.a template for left breast cancer's IMRT planning was customized by using the function of scripting.Ten patients received IMRT alone after breast-conserving surgery were selected,two IMRT plans were designed for each patient:individual plan and the plan based on the template.Dose parameters of target,including conformal index(CI)and homogeneity index(HI),dose-volume histogram parameters,including V30,V20 of lung and cardiac,and the planning time were compared respectively.Results Of the two IMRT plans based on the template method and individual method.The average time of design an IMRT plan were 7±0.82 min and 36.5±6.84 min(t=13.55,P=0.000),the PTV average CI were 0.851± 0.015 and 0.845±0.032(t=0.59,P=0.560),the PTV average HI were(13.2±1.3)%and(14.4±1.6)%(t=1.804,P=0.08),the lung average V30 were(8.80±0.53)%and(8.31±0.74)%(t= cardiac average V30 were(9.89±1.14)%and(9.66±1.43)%(t=0.41,P=0.688).Conclusion For the tumor with the same location and stage,the application of template can simplify planning process and reduce working time,which was worthy of expansion.
