1.Influence of Anshen Bunao syrup combined with escitalopram oxalate on nervous function defect in patients with post stroke depression

Objective To observe the effect of Anshen Bunao syrup combined with escitalopram oxalate on nervous function defect in patients with post stroke depression,and to provide effective treatment for the disease with integrated traditional Chinese and western medicine.Methods 257 patients with post stroke depression were selected,and they were divided into observation group(130 cases) and control group(127 cases) according to the digital table.The two groups were given the same basic treatment,the control group was given escitalopram oxalate,and the observation group was given Anshen Bunao syrup combined with escitalopram oxalate.The NIHSS and HAMD scores of the two groups were evaluated before and 2,4,8 weeks after treatment.The adverse drug reaction was observed during the treatment period.The treatment effect was determined according to the HAMD reduction rate after 8 weeks.Results 2,4 and 8 weeks after treatment,the NIHSS scores of the observation group were (18.9 ±6.2)points,(15.6 ± 6.4) points,(12.2 ± 4.3) points,respectively,which were lower than (22.5 ± 7.1) points before treatment (t =4.355,8.230,14.148,all P < 0.05).4 and 8 weeks after treatment,the NIHSS scores of the control group were (17.2 ± 7.5) points,(14.4 6.6) points,respectively,which were lower than (22.1 ± 8.4) points before treatment (t =4.904,8.123,all P < 0.05).4 and 8 weeks after treatment,the NIHSS scores of the observation group were lower than those of the control group(t =3.640,1.855,all P < 0.05).2,4 and 8 weeks after treatment,the HAMD scores of the two groups[(15.4 ± 5.1) points,(14.0 ± 5.2) points,(10.5 ± 4.7) points and (14.8 ± 4.6) points,(12.4 ± 5.8) points,(7.9 ± 3.8) points] were lower than before treatment [(23.9 ± 4.7) points,(23.2 ± 5.4) points] (t =13.501,15.539,26.419,13.812,4.748,4.748,all P < 0.05).8 weeks after treatment,the HAMD score of the observation group was lower than that of the control group (t =3.640,P < 0.05).8 weeks after treatment,the treatment effect was determined according to the HAMD reduction rate,the total effective rates of the observation group and the control group were 95.4% (124/130),91.3 % (116/127),respectively.The curative effect of the observation group was better than the control group(Z =3.104,P < 0.05).In the course of treatment,the incidence rate of adverse drug reactions of the control group was 6.30% (8/127),which of the observation group was 5.38% (7/130),there was no statistically significant difference between the two groups(x2 =0.098,P > 0.05).Conclusion Anshen Bunao syrup combined with escitalopram oxalate can improve the nervous function defect in patients with post stroke depression,and the incidence rate of adverse drug reactions is low,which has good clinical value.
2.Bladder Cancer J82 Cells Apoptosis Induced by Brucea Javanica Oil Emulsion and the Mechanism

Objective To study the effect of Brucea javanica oil emulsion on apoptosis of bladder cancer J82 cells and the possible mechanism.Methods Brucea javanica oil emulsion of different concentrations (0.5~4.0 ml/L) was added to the bladder cancer J82 cell cultured in vitro.After 24 h, the proliferation of J82 cells was measured by methyl thiazole tetrazolium (MTT) assay, cell apoptosis was detected by morphological observation and flow cytometer analysis, and caspase-3 activity was assessed using a caspase-3 kit. Results 0.5~4.0 ml/L Brucea javanica oil emulsion inhibited J82 cell proliferation in a dose-dependant manner.2.0 ml/L Brucea javanica oil emulsion induced typical apoptosis 24 h after incubation. The caspase-3 activity of 2.0 ml/L Brucea javanica oil emulsion group was higher than those of control group or Brucea javanica oil emulsion plus Ac-DEVD-CHO group. Conclusion Brucea javanica oil emulsion induces J82 cell apoptosis, and this effect includes activation of caspase-3.
3.Effect of Radix Salviae Miltiorrhizae on Invasion and Metastasis of Melanoma Cells

Objective To study the effect of Radix Salviae Miltiorrhizae (RSM), a kind of Chinese Traditional Medicine which can dissolve stasis by activating blood circulation, on proliferation, invasion, adhesion, migration and metastasis of B16-BL6 metastatic mouse melanoma cells and discuss its functional mechanism. Methods The proliferation, adhesion, invasion and migration capacity of B16-BL6 metastatic cells was evaluated by MTT assay, adhesion assay and reconstituted basement membrane invasion and migration assay in vitro respectively. Mouse spontaneous melanoma model was used to study the effect of RSM on metastasis in vivo. Results The extract of RSM bidirectionally adjusted the multiplication of B16-BL6 cells, promoted prominently the adhesion of B16-BL6 to Laminin, inhibited significantly B16-BL6 invading reconstituted basement membrane and the migration of B16-BL6. In the mouse spontaneous melanoma model, it suppressed significantly the volume of lung metastatic nodes but had little effect on the number of lung metastatic nodes. Conclusion The extract of RSM can alleviate the degree of the metastasis of B16-BL6 metastatic mouse melanoma cells, which may be related with inhibiting the B16-BL6 cells invading the extracellular matrix and reducing the migration of B16-BL6 cells.
4.Evaluation of recurrent gastric malignancy with computed tomography

Purpose:To evaluate the role of CT in diagnosing local recurrence after surgery of gastric carcinoma. Methods:Two groups of 51 patients after surgery of gastric carcinoma were prospectively studied by means of CT performed with the water as oral contrast,hypotonic agent and dynamic or spiral CT technique. 15 patients proved to be without local recurrence measured the thickness and observed the CT findings of anastomosis stoma ,36 patients proved to have local recurrence ,those patients included male 21 and female 15.Results:90.19%(46/51)anastomosis stoma was clearly demonstrated ,and 32 lesions was detected and confirmed the diagnosis in 29 of 32 cases . The CT findings of local recurrence after gastric resection for gastric cancer were irregular gastric wall thickening ( n =14),local mass ( n =6) and mixed lesions ( n =9).Conclusions:Using the optimal technique can improve the detectability and accuracy of CT for diagnosing the local recurrence after surgery for gastric carcinoma.
5.Preventing superficial bladder cancer from recurrence by intravesical instillation with Brucea Javanica oil emulsion after TUR-Bt operation

ObjectiveTo observe the effect of intravesical instillation of Brucea Javanica oil emulsion on recurrence of bladder cancer after transurethral resection of bladder tumor (TUR-Bt) operation.Methods187 patients with superficial bladder carcinoma after TUR-Bt operation were randomly divided into the group A (85 cases) and group B (102 cases). Patients of the group A were treated with instillation of Brucea Javanica oil emulsion; those of the group B were treated with mitomycin. A three-years following up was performed to observe the recurrence and side effects.ResultsAfter a 3-years following up, the recurrence rate of group A was 12.94%, lower than that of group B (34.31%). The side effects were seldom seen in the group A.ConclusionThe effect of intravesical instillation of Brucea Javanica oil emulsion to prevent the recurrence of bladder cancer after TUR-Bt operation is favorable.
6.An in vitro skin sensitization test based on THP-1 cell line

Objective To establish an in vitro skin sensitization test,human cell line activation test (h-CLAT),based on THP-1 cell line (a human acute monocytic leukemia cell line),and to assess the sensitizing potency of plant raw materials of chemical and cosmetic products by this in vitro skin sensitization test.Method THP-1 cells were cultured in vitro and exposed to 11 reference skin sensitization chemicals and 9 samples,by monitoring the cell viability,cell surface marker CD54 /CD86 and relative fluorescence intensity of cells surface after the cells was exposures to the substances,and to discover whether there is a positive reaction.At the same time,Buehler test was used to validate the results of samples tested by h-CLAT.Results 11 reference chemicals were distinguished correctly by h-CLAT.Among the 9 samples tested,7 samples were recognized as negative sensitizer and 2 plant extracted substances were identified as suspicious skin sensitizer.The qualitative classification of the 9 samples by h-CLAT test was consistent with the results obtained by animal test.Conclusions The h-CLAT-in vitro test can be used to replace some animal tests for the prediction of soluble skin sensitizing substances.
7.Research progress of in vivo animal models of prostate cancer

Prostate cancer is one of the most common malignant tumors in men and related studies have achieved great breakthrough in recent years.But because of the lack of effective in vivo animal models, the process to translate basic research into clinical application has been severely hampered.Patient derived prostate tumor xenograft ( PDPTX) model is an ideal animal model in which freshly isolated tumor tissues from patients were inoculated into immunodeficient mice.This model can duplicate the heterogeneity of primary tumor in a better way and keep the tumor complexity at molecular, genetic and pathological levels.Particularly, the PDPTX model, in which the isolated tumor tissue is inoculated under the renal capsule, is even better, because it solves the clrawbacks of traditional subcutaneous inoculation model.In traditional mod-els, the success rate is low, it’s not easy for lower grade tumor to form xenograft, and it’s not easy to reconstruct metasta-sis, etc.PDPTX provides a more ideal in vivo model for prostate cancer studies.It has irreplaceable advantages, especially in target therapy, new drug screening and individualized tumor treatment.
8.Association between serum levels of SP-A, SP-C and the impairment of lung function in coal workers with pneumoconiosis

Objective To investigate the relationship between serum levels of surfactant protein( SP)-A,SP-C and lung function impairment in coal workers with pneumoconiosis(CWP) in order to provide evidence for the biomarker study of pneumoconiosis.Methods Thirty-two coal workers with pneumoconiosis snd 41 healthy controls were included in this study.Serum levels of SP-A and SP-C were measured.Grading assessment of dyspnea and pulmonary function including predicted percentages of FVC,FEV1,FEV/FVC,MVV,and DLCO were conducted.Results Among the 32 participants with CWP,the severity of dyspnea was rated as level Ⅲ for 14 and level Ⅳ for 18 individuals.The pulmonary function was significantly impaired in CWP patients with level Ⅳ dyspnea compared with the healthy controls (FEV1% predicted:[69.38 ± 15.17 ]% vs.[96.35 ±10.24 ] % ; MVV% predicted:[ 65.89± 8.14 ] % vs.[ 94.13 ± 10.38 ] % ; DLCO% predicted:[ 96.51 ±11.37 ] %.The serum levels of SP-A and SP-C were significantly higher in CWP patients than that in the healthy controls (SP-A:[4.02 ± 1.22] μg/L vs.[2.41 ±0.68 ] μg/L,t =6.480,P =0.001 ;SP-C:[3.58 ±0.67 ] ng/L vs.[ 2.31 ± 0.29] ng/L,t =9.290,P < 0.001 ).Serum SP-A and SP-C levels in CWP patients were found to be significantly correlated with exposure to dust,dyspnea severity,FEV1% predicted and DLCO% predicted.Conclusion Serum SP-A and SP-C levels in CWP patients are closely associated with lung function,suggesting their role as candidate biomarkers for CWP.
9.A comparative study of the speech respiratory characteristics of 7-15 years old ordinary children and children with spastic cerebral palsy

Objective To explore the speech respiratory characteristics of 7-15 years old children with spastic cerebral palsy and the differences between ordinary children and children with cerebral palsy.Methods The maximum phonation time (MPT) and maximum counting ability (MCA) of 90 ordinary children and 58 children with cerebral palsy aged 7-15 years were measured and a comparison between the 2 groups were conducted.Results (1) Age has significant effects on MPT and MCA of the ordinary children (P <0.05).The MPT and MCA of the ordinary children at the age of 7 ~9 years and 10 ~ 12 years are lower than the ones at the age of 13 ~ 15 years.Age has almost no effects on MPT and MCA of children with cerebral palsy(P >0.05) ; (2)Sex has no significant effects on MPT and MCA of ordinary children and children with cerebral palsy (P >0.05) ; (3) The MPT and MCA of children with cerebral palsy are significantly lower than those of ordinary children (P <0.05).Conclusions The MPT and MCA of ordinary children are increased with the growth of age,especially after the age of 13 years,but the children with spastic cerebral palsy do not show that kind of trend and demonstrate a state of retardation of speech respiratory function.Compared with ordinary children,the children with spastic cerebral palsy are easier to suffer from insufficient respiratory support and poor respiratory-phonatory coordination.
10.The different responses of diaphragm and peripheral muscles to cisatracurium in rabbits

Objective To investigate the different responses of diaphragm and peripheral muscles to cisatracurium in rabbits.Methods 8 male New zealand rabbits were anaesthetized with pentobarbital,and then the diaphragm,tibialis anterior,soleus muscles,phrenic nerves,tibial nerve,and peroneal nerve were gently freeded.The muscles were secured to force displacement transducers,and the nerves were directly stimulated by electrodes with supramaximal square waves.The isometric force of twitch tention of each muscle was recorded.The cumulative dose-response technique was separately used for obtaining the ED50and ED95 values of the cisatracurium in each muscle.Results The muscle-relaxing of cisatracurium on the three muscles in were observed in a dose-dependent manner.The ED50 values and ED95 values were: diaphragm(39.3 ± 2.5)μg/kg and(75.7 ± 4.2)μg/kg,tibialis anterior(80.6 ± 7.5)μg/kg and(123.3 ±9.3)μg/kg,soleus(80.0 ± 7.1)μg/kg and(126.9 ± 9.4)μg/kg,respectively.It had significant difference between diaphragm vs tibialis or soleus,P < 0.05.Conclusions The muscle relaxant effects of cisatracurium on diaphragm and peripheral muscles were different,and diaphragm was more sensitive than peripheral muscles.
