AIM: To study protective effects as myocardial ischemic preconditioning of sasanquasaponin (SQS) and its relationship with K ATP channel. METHODS: The study adopted the model of myocardial ischemic injury induced by subcutaneous injection of isoproterenol (ISO) in rats, administering specific K ATP channel blocker gliberclamide (GLI 5 mg?kg -1 ). Four groups were set as NS group, I/R group, SQS group ( 0.2 mg?kg -1 ), and GLI group (5 mg?kg -1 ). Prior to injection of ISO, all agents were intraveneously injected into rats for 3 days, one time per day. Subsequently, ISO was subcutaneuously injected into rats by the ways of many different sites, and some indices were measured including ECG, serum creatine kinase (CK) activity, free fatty acid (FFA), and adenosine contents in rats. RESULTS: Preconditioningly intravenous injection of SQS could effectively protect myocardium from ischemic injury induced by ISO. With GLI injected prior to SQS, the cardioprotective effects of SQS were significantly attenuated. CONCLUSION: SQS can protect myocardium from ischemic injury induced by ISO, and the protection may be mediated by K ATP channel.

Chinese Pharmacological Bulletin 2003;0(09):-
AIM In this study, we observe the effects of SQS on contents of myocardial malondialdehyde(MDA)and activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH Px) through adopting the model of myocardial ischemic injury induced by subcutaneous injection of isoprenaline(ISO 4 mg?kg -1 ?d -1 ?2 d) into rats. METHODS Four groups were divided, namely NS, I/R, SQS1 and SQS2 group. The contents of MDA and activities of SOD and GSH Px were examined after administering SQS for 3 days(1 time per day). RESULTS The results show the elevation of S T in ECG, increase of MDA contents and decrease of SOD and GSH Px activities in I/R group. SQS may antagonize the changes of MDA, SOD and GSH Px induced by ISO, revealing the relationship to dose dependence. CONCLUSION SQS is most likely to possess the capabilities of anti oxygen free radicals and anti lipoperoxidation to myocardial ischemic injury induced by ISO.

BACKGROUND:For pediatric patients with aplastic anemia in China, it is difficult to find human leucocyte antigen-matched sibling donors that are mostly replaced by parental donors.
OBJECTIVE:To retrospectively analyze the clinical efficacy and safety of parental haploidentical peripheral blood hematopoietic stem cel transplantation in children with relapsed and refractory severe aplastic anemia.
METHODS:Seventeen children with relapsed and refractory severe aplastic anemia who had no matched sibling or unrelated donor and failed to respond to immunosuppressive therapy were subjected to parental haploidentical peripheral blood hematopoietic stem cel transplantation. A conditioning regimen of fludarabine+cyclophosphamide+rabbit anti-human thymocyte immunoglobulin antibody and the triple therapy of methotrexate, cyclosporine A and mycophenolate mofetil were applied to prevent graft-versus-host disease.
RESULTS AND CONCLUSION: (1) Of the 17 children, 16 cases (94%) reached hematopoietic reconstitution, and the median time of neutrophils≥ 0.5×109/L and platelets≥ 20×109/L was 13 (11-15) days and 17 (12-28) days, respectively. (2) Incidence of acute graft-versus-host disease was 47% (8 of 17 cases), including 29% (5/17) of grades I-II and 18% (3/17) of grades III-IV. Incidence of chronic graft-versus-host disease was 41% (7/17). (3) With a median folow-up duration of 268 (43-753) days, the overal survival rate was 70.6% (12/17). Five dead cases (29%) belonged to transplantation-related death, including one case of fungal skin infections, one case of graft-versus-host disease, three cases of severe lung infection. No relapse case was reported. These findings indicate that if there are no matched sibling or unrelated donors and the immunosuppression effect is poor, parental haploidentical peripheral blood hematopoietic stem cel transplantation is a safe and effective salvage treatment for children with relapsed and refractory severe aplastic anemia.

BACKGROUND:Cytokine induced kil er cells therapy as an effective means of adoptive immunotherapy, becomes a new way to treat acute myeloid leukemia. But, the researches about sequential cytokine induced kil er cells therapy after autologous peripheral blood stem celltransplantation in acute myeloid leukemia patients are stil less, which deserve further research.
OBJECTIVE:To observe the clinical efficiency and safety of sequential cytokine induced kil er cells therapy after autologous peripheral blood stem celltransplantation in acute myeloid leukemia M2 patients.
METHODS:Total y 45 patients with low-or intermediate-risk acute myeloid leukemia M2 were recruited in this study. Among them, 19 patients received sequential cytokine induced kil er cells therapy after autologous peripheral blood stem celltransplantation and 26 patients only received autologous peripheral blood stem celltransplantation. The relapse rate, disease-free survival, and overal survival were compared between two groups, and safety of cytokine induced kil er cells therapy was observed.
RESULTS AND CONCLUSION:(1) Compared with the patients only receiving autologous peripheral blood stem celltransplantation, the relapse rate was lower (21.05%vs. 38.46%;P<0.05), and elevated percentages of the disease-free survival and overal survival were observed in the patients receiving sequential cytokine induced kil er cells therapy after autologous peripheral blood stem celltransplantation (P<0.05). (2) The 19 patients who received sequential cytokine induced kil er cells therapy after autologous peripheral blood stem celltransplantation al completed the treatment scheme successful y. Only four patients appeared to have chil s and fever, and no more side effects were observed. These findings suggested that the sequential cytokine induced kil er cells therapy after autologous peripheral blood stem celltransplantation can improve the disease-free survival and overal survival of low-or intermediate-risk acute myeloid leukemia M2 patients without remarkable side effects, which is a safe, effective and feasible way for the treatment of acute myeloid leukemia M2.

Objective To explore the effect of group cognitive-behavior therapy intervention on dys-thymic disorders in children and adolescents. Methods From July 2015 to December 2017,68 cases of chil-dren and adolescents treated in Lishui Second People's Hospital were divided into two groups randomly. The study group ( 34 cases) were treated with group cognitive-behavior therapy, and the control group ( 34 cases) were treated with selective 5-hydroxytryptamine reuptake blocker ( SSRI ) . The achenbach child behavior check list(CBCL),self-rating depression scale(SDS) and self-rating anxiety scale(SAS) scores of the two groups before and after the intervention were compared. Results Before the intervention, there was no differ-ence in the score of CBCL,SDS and SAS between the study group and the control group (P>0.05). After drug treatment or cognitive-behavior therapy, the social, activity, thinking and aggressive behavior of both groups were obviously improved. The SDS and SAS scores were 43.64±4.86 and 42.27±3.74 in cognitive-be-havior therapy group, which had significant difference (P<0.05) compared with pre-intervention score of SDS (82.91±3.95) and SAS (78.61±6.28). The scores of SDS (45.61±8.03) and SAS (44.09±7.04) in treat-ment group were significantly (P<0.05) decreased compared with those before treatment (84.03±5.11 and 77.30±9.55,respectively). The satisfaction of the study group and control group was 94.1% and 67.6%,re-spectively,and the difference was significant between the two groups(P<0.05).Conclusion The effect of cognitive-behavior therapy in children and adolescents on reducing the negative emotion is remarkable, which is equivalent to the drug treatment. The cognitive-behavior therapy in children and adolescents can be used as a auxiliary means to treat children's dysthymic disorders in clinical practice.

Vesicouterine fistula (VUF) represents a rare urogenital complication. It is considered to be the least common type of urogenital fistulae. Surgical repair remains the mainstay of treatment. Transabdominal repair is the most common surgical method and transvaginal route is rarely reported. Herein, we presented the successful transvaginal repair of 3 cases of VUF in our center, including 2 cases of vesicocervical fistula and 1 case of high vesico-uterine body fistula. All the 3 VUF cases were repaired through vagina and followed up for 6 months, 1 year and 2 years, respectively. There was no vaginal leakage or hematuria postoperatively. VUF was cured. Transvaginal repair of VUF was shown to be a feasible, safe and effective procedure with small trauma.

Objective To explore the effect of group cognitive behavior intervention on self-rated health of middle school students with emotional disorders. Methods From January 2018 to June 2018,79 middle school students with emotional disorders were randomly divided into intervention group ( 41 cases) and control group (38 cases) according to the single or double number of medical records. The control group only received drug treatment, while the intervention group received group cognitive behavior intervention on the basis of drug treatment. All the students in the two groups completed the self-rated health measurement scale before intervention (T0),after intervention (T1) and 8 weeks after intervention (T2). Results (1) There were no significant differences in total health score and dimension score between the two groups before intervention (both P<0. 05). (2)The repeated measurement variance analysis showed that there was a signif-icant group × time interaction effect on total health score and dimensions(P>0. 05). (3) The group effect of physical health was not significant (P>0. 05). The group effect of total mental health, social health and health score at T1 and T2 time points were significant (all P>0. 05). (4)Compared with before intervention, mental health ((123. 34±9. 33),( 122. 63± 9. 11)),social health ((102. 89 ± 7. 28),( 101. 89± 7. 73)) and total health score ((370. 34±17. 99),(367. 63±17. 89)) of intervention group at T1 and T2 increased ( all P<0. 05),while that of control group increased only at T1 (all P<0. 05). Conclusion Group cognitive behavioral intervention has no obvious effect on physical health of middle school students with emotional dis-orders. And group cognitive behavioral intervention can effectively improve their mental health, social health and overall health level, and the long-term effect is better.
