1.The role of nicotinamide adenine dinucleotide phosphate oxidase in vascular cognitive impairment and its mechanism: a research progress

Oxidative stress(OS)is one of the factors leading to vascular cognitive impairment(VCI). Reactive oxygen species(ROS)produced by Nicotinamide adenine dinucleotide phosphate oxidase(NOX)is an important pathogenic molecule of VCI.Risk factors for VCI such as hyperglycemia and decomposition/re-implantation always increases an upstream NOX activation.Other risk factors such as advanced age, hypertension and stroke are the downstream damage caused by NOX-dependent ROS.The causes of VCI include both risk factors related to NOX and the cell damage caused by NOX-dependent ROS.Studies have shown that the inhibiting of NOX activity can reduce cognitive impairment and the risk factors in animal models of VCI.In this article, we review the research progress of the role and mechanism of NOX in VCI.
2.Tensile mechanical properties of lateral collateral ligaments in animal models of osteoarthritis

BACKGROUND:It is very nscessary for the prevention and treatment of ostaoarthritis(OA)to understand the tensile mechanical properties of lateral collateral ligaments(LCLs)of knee joints with osteoarthritis,while reports on it are still few.OBJECTIVE:To compare the LCL tensile mechanical property indexes of knee joints between normal animals and OA medels established for this purpose,and to provide qualitative and quantitative referential data about effects of OA on the LCL tensile mechanical properties of animal knee joints.DESlGN,TIME AND SETTING:A randomized controlled trial was finished at the Mechanical Expedmental Center of Jilin University from December 20th to 30th in 2008.MATERIALS:A total of 20 male SD rats of 6 months old were divided into a normal control group and a model group,with 10ones in each group.METHOOS:Rats in the model group were copied into OA models.after which test on tensile was performed to each of the ten rats in the two groups at the speed of 5mm/min on Universal Testing Machine,Shimadzu.Data obtained from the test were processed in polynomial though least square method.MAIN OUTCOME MEASURES:Tensile maximum load.maximum displacement,maximum stress,maximum strain and stress-strain curves.RESULTS:The tensile maximum load,maximum stress,maximum displacement and maximum strain in the normal control group and the model group were(12.754±2.795)N,(27.681±5.832)MPa,(2.754±0.707)mm,(11.679±2.373)%and (7.183±1.817)N,(16.162±3.403)MPa,(1.827±0.768)mm,(8.019±2.811)% respectively,from which we could see that each index in the normal control group was greater than that in the model group obviously (P<0.05).CONCLUSION:The tensile maximum load,maximum stress,maximum displacement and maximum strain all decreased after OA,which indicates that osteoarthritis has certain effects on tensile mechanical properties of LCLs of knee joints as well as stress relaxation.
3.Establishment of Hyperkalemia Model in Rabbits

Objective According to the special changes of ECG in hyperkalemia animals, explore a method to reproduce a rabbits model for hyperkalemia. Methods Three programs were applied by intravenous drip of 3%KCl to reproduce the hyperkalemia model. In the first program,once the wide and deformed QRS complex occurred, the potassium drip should be stopped immediately. In the second program,the wide and deformed QRS complex was kept for 30 min by adjusting the speed of potassium infusion. In the third program, once low and flat P wave and peaked T wave appeared, immediately adjusted the speed of potassium infusion in order to keep for 30 min. And HR, BP, urine output and serum potassium were monitored simultaneously. Hyperkalemia is defined as serum potassium≥5.1 mol/L. The successful hyperkalemia model should keep serum potassium≥5.1 mol/L after ceasing potassium given 30 min. Results The second and third programs could reproduce a hyperkalemia model successfully. The serum potassium returned to normal within 30 min after stopping potassium given in the first program. Conclusion The method which keep low and flat P wave and peaked T wave for 30 min and keep the wide and deformed QRS complex 30 min could reproduce the hyperkalemia model successfully.
4.Effect of pulse electro-magnetic fields on the repair of osteo chondral fracture in rabbits

Objective:To study the effects of pulse electro-magnetic fields (PEMFs) on the healing of tibial condylar osteochondral in rabbits.Methods:Sixteen rabbits were divided into two groups,and osteochondral fractures of left tibia condyle were created,internal fixation was performed with steel wire.Group 1 was constantly treated with PEMFs six hours per day;group 2 was not treated.The results were studied by gross morphology,light microscopy and elect ro-microscopy.Results:75% and 13% fractures were repaired by hyalin-like cartilage in group 1 and in group 2 respectively.Conclusion:PEMFs are essential to facilitation of the cartilage repair.
5.Simultaneous Determination of 6 Organic Solvents in Tosufloxacin Tosylate by Headspace GC

OBJECTIVE:To establish a method for the residual simultaneous determination of methanol,alcohol,dichlorometh-ane,n-hexane,tetrahydrofuran and benzene in tosufloxacin tosylat. METHODS:Headspace GC was performed on the capillary col-umn with 6%cyanopropylphenyl-94%dimethylpolysiloxane(DB-624)as fixative lipid,temperature programmed,the inlet temper-ature was 180 ℃,detector was flame ionization detector with temperature of 300 ℃,carrier gas was high purity N2 at a flow rate of 1.5 mL/min,split ratio was 10:1,headspace equilibrium temperature was 100℃,equilibrium time was 40 min,headspace sam-ple volume was 10 mL,and the headspace sample volume was 1 mL. RESULTS:The linear range was 178.3-1782.7 μg/mL for methanol (r=0.9991),301.2-3012.1 μg/mL for alcohol(r=0.9997),33.81-338.10 μg/mL for dichloromethane(r=0.9993), 18.02-180.22 μg/mL for n-hexane(r=0.9991),43.26-432.58 μg/mL for tetrahydrofuran(r=0.9991)and 0.1268-1.2681 μg/mL for benzene(r=0.9991);limits of quantification were 0.31,3.00,0.67,0.02,0.005,0.10 μg/mL,limits of detection were 0.15, 1.51,0.22,0.01,0.001,0.05 μg/mL;RSD of precision test was no higher than 3.1%;RSDs of methanol and n-hexane in stabili-ty and reproducibility tests were no higher than 5%;recoveries were 93.72%-102.20%(RSD=3.1%,n=9),90.10%-101.79%(RSD=4.0%,n=9),97.07%-103.11%(RSD=2.0%,n=9),92.38%-103.83%(RSD=3.9%,n=9),95.44%-103.62%(RSD=2.8%,n=9),and 94.00%-104.73%(RSD=4.1%,n=9). CONCLUSIONS:The method is rapid,sensitive,accurate,and suitable for the residual determination of methanol,alcohol,dichloromethane,n-hexane,tetrahydrofuran and benzene in tosufloxacin tosylat.

Patient satisfaction is an important index of hospital quality evaluation and performance evaluation.This study established multi-attribute patient satisfaction index system based on the theory of customer satisfaction index,determined the index weight by combination weighting approach,and graded the hospitals and adjusted the weights by the RSR method.A set of comprehensive evaluation scheme is initially formed,which is suitable for patient satisfaction evaluation and performance evaluation.

Objective To investigate the distribution of high pathogenicity island(HPI)in multiple-drug-resistance gram-negative bacilli and analyze the protein sequence.Methods To amplify thefyuA-irp2 gene cluster of the 84 isolates by multiple polymerase chain reaction(PCR),the product was subsequently sequenced.Results The positive rate ofirpl,irp2,irp3,irp4 and fyuA was 40.48%,41,67%,5.95%,O%and 16.67%,respectively.Theamino sequence offyuA comefromEC06748,Kp7151 and PAE7 was usedto compare with AL590842,there are 100%identities.Amino sequence ofirp2 come from Kp49 and Kp51 have 99%identities with AAA27636.1,but amino sequence of irp2 come from EC04 and EC07 only have 90%identities with 1176840.The GenBank accession number is FJ211852 and FJ211851.Amino sequence ofirpl come fromKp 10,Kp49 and Kp51 have 99%identities with AL590842。and amino sequence ofirp3 come from EC03,Kp51,Kp10 and Kp49 have 97%identities with CAA73128.There are the same mutation among the same species,and different mutation among different species.Conclusion There was different extant mutant lost in thefy~t-i,v2 gene cluster in multiple-drug-resistanee gram-negative bacilli.

Objective Examination and evaluation of employee satisfaction index in the hospital performance evaluation.Methods Stratified sampling,field survey and telephone survey were used in customizing a questionnaire for two surveys in July and December 201 5 respectively.Results The standardized score of employee satisfaction was 86.252±1 5.1 53,and the lowest score was found in the canteen environment and food quality.Conclusion Employee satisfaction is found as good overall,and targeted measures are recommended to improve insufficiencies for better employee satisfaction.
9.The protective effect of paeoniflorin in retina ischemia animal model through regulation of NLRP3 inflammasomes

Objective To investigate the mechanisms of paeoniflorin in protection of retinal ischemia injury. Methods Fifty.four male specefic pathogen free ( SPF) degree Wistar rats were randomly divided into normal control group,model control group and paeoniflorin group. Retinal ischemia injury was induced by raising the intraocular pressure of right eyes of rats to 110 mmHg for 30 minutes. The rats of paeoniflorin group were administrated through intraperitoneal injection of 5 mg/kg paeoniflorin each day for 14 days. OCT and electroretinogram ( ERG ) were performed to detect the thickness of retinal nerve fiber layer+retinal ganglion cell layer+inner plexiform layer ( NGI) and electrophysiological changes of retina, respectively. Retrograde labelling of retinal ganglion cells ( RGCs ) was used to evaluate the survival number of RGCs. Western blot analysis was used to detect NLRP3,apoptosis.associated speck.like protein containing a caspase activation and recruitment domain (ASC),cleaved caspase 1 (c.caspase 1), IL.18,and IL.1β expression. The use and care of animals complied with the statement of the Association for Research in Vision and Ophthalmology ( ARVO ) and Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results The thickness of retinal NGI in model control group was ( 58. 2 ± 1. 7)μm, which was significantly lower than ( 84. 8 ± 1. 9)μm in normal control group and(71. 1±2. 4)μm in paeoniflorin group (both at P<0. 05). The amplitudes of A and B waves in paeoniflorin group and normal control group were significantly higher than those in model control group ( both at P<0. 05 ) . The number of RGC in model control group was significantly lower than that in paeoniflorin group and normal control group ( both at P<0. 05).
