1.Effect of high-dose vitamin C on MMP2 expression and invasive ability in human pancreatic cancer cell line PANC-1

Objective To observe the effect of high-dose vitamin C on MMP2 expression and invasive ability in PANC-1. Methods Transwell invasion assay was used to compare the invasive ability of PANC-1 cells in different concentrations of vitamin C treated groups. RT-PCR and Western blot were used to detect and compare the levels of MMP2 mRNA and protein expression in each group. Results Compared with the 0mM vitamin C treated group, the mRNA expression of MMP2 was significantly decreased in 1.0,5.0,10. 0mM group(0. 510 ±0. 004 vs 0. 792 ±0. 006, 0. 391 ±0. 007 vs 0. 792 ±0. 006, 0. 282 ±0. 008vs 0. 792 ± 0. 006, P < 0. 05 ). Compared with the 0mM vitamin C treated group, the protein expression of MMP2 was significantly decreased in 1.0,5.0,10. 0mM group(0. 519 ±0. 004 vs 0. 761 ±0. 014,0. 310 ±0. 007 vs 0. 761 ±0. 014,0. 297 ±0. 008 vs 0. 761 ±0. 014, P <0. 05). Compared with the 0mM vitamin C treated group, the invaded cell number was significantly decreased in 1.0,5.0,10. 0mM groups ( 452 ± 22 vs653 ± 28,340 ± 32 vs 653 ± 28,409 ± 33 vs 653 ± 28, P < 0. 05 ). Conclusion High-dose vitamin C can decrease the expression of MMP2 in PANC-1 cells, and weaken its invasive ability.
2.Expression of transcription factor Sp3 in hepatocellular carcinoma

Objective To research the expression of Sp3 andβ‐Catenin in HCC and study the assessable factors of them for prognosis in patients with hepatocellular carcinoma .Methods Western blot and RT‐PCR methods were used to detect the expres‐sion of Sp3 andβ‐Catenin in HCC and the liver tissue beside tumor among 49 cases .We analyzed the difference of these two indexes expressed in HCC and the liver tissue beside tumor .Then we detected the correlation between these two indexes and the character of clinic pathology ,and researched the correlation between Sp3 and the prognosis of HCC .Results The high expression rate of Sp3 in HCC was higher than that of liver tissue beside tumor(P<0 .05) according to Western blot and RT‐PCR ,the same toβ‐Catenin (P<0 .05) .Expression of Sp3 andβ‐Catenin were both related with size of tumor and degree of differentiation .Positive correlation existed between these two indexes according to Western blot method(r=0 .681 ,P=0 .000) and RT‐PCR method(r=0 .641 ,P=0 .000) .The prognosis of cases with high expression of Sp3 was poorer than the low expression cases(P<0 .05) .Conclusion Sp3 plays a promoter role in occurrence of HCC ,which is correlated with the grade malignancy of HCC .Sp3 might participated in occur‐rence and development of HCC via the Wnt pathway .
3.Effect of BuShen Prescription on metabolism of collagen Ⅰ in ovariectomized rats

Objective:To study the effect of BuShen Prescription on metabolism of collagen Ⅰ in ovariectomized rats,which will provide experimental evidence for treating osteoporosis. Methods:50 Ovariectomized SD rats were randomLy divided into 5 groups,including normal control group,model group,positive control (OVX) group,Zuogui Wan group and Yougui Wan group,each group with 10 rats. After 12 weeks' routine feed,normal control group and model group were intragastric administrated with pure water,positive control group was intragastric administrated with conjugated estrogens water-solution,Zuogui Wan group was intragastric administrated with Zuogui Wan decoction,and Yougui Wan group was intragastric administrated with Yougui Wan decoction,then the PINP,DPYD/Cr,NTX/Cr were detected after ovariectomized rats were intragastric administrated with the schedule for 12 weeks. 4.Analysis of reason and countermeasures of 11 024 unqualified blood specimens

Objective To analyze reason and countermeasures of unqualified blood specimens ,improve the qualified rate of sam-ples ,to strengthen the quality control before analysis .Methods A retrospective statistical analysis were conducted to analyze the characteristics of the unqualified specimen and reasons from January 2013 to June 2014 .Results A total of 11 024 unqualified spec-imens accepted in the inspection center from January 2013 to June 2014 ,accounting for 0 .331% of reasons of unqualified specimens including hemolysis(26 .7% ) ,blood coagulation(25 .8% ) .The unqualified specimen in surgical system was higher than that in med-ical system .Conclusion Control specimen qualified rate system should be established in clinical laboratory ,the fraction defective samples should be reduced through continuous analysis and communication with clinical medical personnel to ensure the quality be-fore analyzing .
5.The optimal cryopreservation liquid of the stable transfected cell line named hepatic carcinoma HepG 2 Transfected by Lintivirus

Objective To investigate the optimal cryopreservation liquid of the stable transfected cell line named hepatic carcino-ma HepG2 transfected by lentivirus .Methods Glycerol and DMSO as cryopreservation liguid ,The HepG2 transfected by lentivirus and HepG2 cells were both cryopreserved with four various proportion of cryopreservation liquid .After thawing ,the survival rate of the two cells were observed by inverted contrast microscope and the viability and proliferation were detected with MTT assay .Re-sults Using glycerol as cryoprotectant liquid ,the survival rate of the HepG2 cells transfected by lentivirus was obviously higher than other 3 groups when the proportion of the cryopreservation liquid(DMEM ∶FBS∶Glycerol)was 0∶9∶1(P=0 .001) ,where-as there was no significant difference among the groups of HepG2 cells with various proportion of cryopreservation liquid (P=0 .293) .Using DMSO as cryoprotectant liquid ,the survival rate of the cells transfected by lentivirus was 0% ,whereas the survival rate of the HepG2 cells was higher than 60% ,and there was no significant difference between groups of various proportion of cryo-preservation liquid(P=0 .487) .The MTT assay demonstrated that the higher the serum levels was ,the better proliferation capacity those cells had(P<0 .05) .Conclusion The optimal cryopreservation liquid for the hepatic carcinoma cell line HepG2 transfected by lentivirus is used glycerol as cryoprotectant solution ,and the proportion of cryopreservation solution was 90% FBS + 10%Glycerol .

Objective: To study the effect of Chinese medicinal herb Hongqu on induction differentiation of bone marrow stromal cells to osteoblasts.Methods:60 SD rats were divided randomly into 6 groups: blank control group,Low,medium and high dose of Hongqu groups,Provastain group,Premarin group.Each group was given distilled water,Hongqu extraction,Provastain solution,and Premarin solution through intragastric administration.10 days later,killed the rats and took the rats’ blood to prepared medicated serum.Meanwhile,the bone marrow stromal cells which comes from femur and tibia of the 4-5 week’s rats were cultured in medicated serum culture-medium,and MTT,para-nitro-pheneye phosphate,and the tinctorial method of alizarin Bordeaux were used to observe the effect of Hongqu on such aspects as the proliferation,ALP expression,and mineralization tuberculation of marrow stromal cells induction differentiation to osteoblasts.Results: Mmedicated serum of Hongqucould obviously increace the quantity of marrow stromal cells induction differentiation to osteoblasts,the level of the ALP expression and mineralization tuberculation(P
7.Genetics analysis of monochorionic diamniotic twins discordant for karyotypes

Objective:To explore the genetic background in monochorionic diamniotic (MCDA) twins discordant for karyotypes.Methods:This study retrospectively analyzed four pregnant women who were diagnosed as having MCDA twins with prenatal diagnostic indications by ultrasound at Maternity and Child Health Care of Guangxi Zhuang Autonomous Region from January 2016 to December 2019. Dual amniocentesis was performed for all cases guided by ultrasound and the twin amniotic fluids were taken for karyotyping and single nucleotide polymorphism (SNP) array testing. Zygosity was determined by SNP genotype analysis.Results:The karyotypes of four MCDA twins were 47,XX,+18 and 46,XX, 45,X and 46,XX, 47,XXY[17]/46,XY[33] and 47,XXY, and 47,XX,+21 and 46,XX, respectively. The results of SNP were arr(18)×3 and arr(1-22,X)×2, arr(X)×1 and arr(1-22,X)×2, arr(X)×1~2,(Y)×1 and arr(X)×2,(Y)×1, arr(21)×3 and arr(21)×2~3. All of them were monozygotic twins according to the SNP genotypes. Three out of the four cases chose to terminate the pregnancy due to fetal chromosomal abnormalities and one was lost to follow-up. One gave birth to a healthy child in the next pregnancy.Conclusions:Clinicians should be alert to the discordant karyotypes in MCDA twins, of which the mechanism is yet to be explored.
8.A noninvasive and continuous method for blood pressure measurement using pulse wave.

This paper presents a method of noninvasive and continuous measurement of blood pressure by pulse wave. The method is realized by extracting the characteristic parameters of human brachial artery pulse wave and developing the stepwise regression equation. The experiment results showed that blood pressure measured by this method was well correlated with which measured by mercury sphygmomanometer. The mean difference of blood pressure was smaller than 3 mmHg, and the standard deviation was smaller than 5 mmHg.

9.Chemotherapy resistance induced by interleukin-6 in ovarian cancer cells and its signal transduction pathways

Objective To study the mechanism of chemotherapy resistance caused by interleukin-6 (IL-6) in ovarian cancer cells and its related signal pathways. Methods Ovarian cancer cell lines A2780(IL-6 receptor positive, while non-IL-6-expressing and cisplatin/paclitaxel-responsive) and SKOV3 cell lines( overexpressing of IL-6 receptor and IL-6 and cisplatin/paclitaxel-resistant) were suitable models for this study. The effect of exogenous (a short period of treatment with recombination IL-6) and endogenous IL-6(by transfecting with plasmid encoding for sense IL-6 ) in A2780 cells or deleting of endogenous IL-6expression in SKOV3 cells (by transfecting with plasmid encoding for antisense IL-6) on the sensitivity to cisplatin and paclitaxel was investigated. Meanwhile, the mechanism of chemotherapy resistance caused by IL-6 in ovarian cancer cells and its related signal pathways were also analyzed. Results We found that both exogenous and endogenous IL-6 induce cisplatin and paclitaxel resistance in non-IL-6-expressing A2780 cells (the resistance multiple to cisplatin and paclitaxel was: exogenous, 6. 25 and 7.31; endogenous, 7. 13 -8. 34 and 7. 61 - 10. 70), while deleting of endogenous IL-6 expression in IL-6-overexpressing SKOV3 cells promotes its sensitivity to anticancer drugs ( the resistance multiple to cisplatin and paclitaxel was 0. 15 and 0. 10, 0. 10 and 0. 08). IL-6 significantly up-regulated the expression levels of mRNA and protein of drug resistance-associated genes, MDR1 and GST-π, and apoptosis-inhibiting genes, bcl-2, bcl-xL and XIAP in a dose-dependent manner in A2780 cells. In accordance with this finding, the mRNA and protein levels of MDR1 and GST-π enhanced in sense IL-6-transfected A2780 cells, and reduced in antisense IL-6-transfected SKOV3 cells compared with the corresponding parental and control vector-transfected cells, which had no difference. It was found that PD98059 [ mitogen-activated protein kinase-extracellular signalregulated kinase (MEK) inhibitor ] and wortmannin [ phosphatidylinositol 3-kinase (PI3K) inhibitor ]significantly antagonized IL-6-induced phosphorylation of extracellular signal-regulated kinase ( ERK ) and protein kinase B (Akt), respectively, and both of them blocked IL-6-induced cisplatin and paclitaxel resistance and the inhibitory effects of PD98059 and wortmannin were dependent on its concentration.Conclusions These data suggest that IL-6-induced chemoresistance may be associated with increase of both drug resistance-associated genes ( MDR1 and GST-π) and apoptosis-inhibiting genes ( bcl-2, bcl-xL and XIAP), and activation of MEK/ERK and PL3K/Akt. Therefore, modulation of IL-6 expression or its related signaling pathway may be a promising strategy of treatment for drug-resistant ovarian cancer.
10.A method of real-time sampling and smooth scrolling in epicardial mapping system.

In order to control real-time sampling and achieve smooth scroll wave in the epicardial mapping system, which has 128 channels and a sampling rate of 2kHz/channel, we have designed a user interface using Direct-Draw in cooperation with multiple-thread technology and extracting method. This method has proven to be feasible in animal experiment.
