Objective To explore the role of nuclear factor-kappa B(NF-?B)in regulating tumor necrosis factor-?(TNF-?)expression in acute pancreatitis (AP)in rats.Methods Sixty-four Wistar rats were randomly divided into two groups:the control group(n=32)and the AP group(n=32).Acute pancreatitis was induced by introperitoneal injection of caerulein,and the animals were killded at 12 hours,24 hours,36 hours,and 72 hours respectively.The activity of NF-?B and expression of TNF-? mRNA in the pancreas were determined by flow cytometry (FCM) method and semi-quantitave RT-PCR analysis,respectively,and the correlation coefficients were analyzed.Results The levels of activated NF-?B protein and TNF-?mRNA were all significantly higher in the AP group than those in the control group at any time point(P
2.Occlusal stimulation regulates bone remodeling during bone defect repair using collagen substitutes:a morphological analysis

BACKGROUND:Occlusal stimulation is essential for mandible function and remodeling, but there is stil a lack of clear understanding about the effect of occlusal stimulation on the bone remodeling in the process of bone defect repair using bone grafts. OBJECTIVE:To analyze the possible regulative effect of occlusal stimulation on bone remodeling in the process of bone defect repair using colagen substitutes. METHODS:Standard models of bone defects were respectively established in left mandible and parietal bone area of adult Sprague-Dawley rats. Then the bone defects area were filed with colagen and bone meal. The differences of two bone defects areas were observed by X-ray, hematoxylin-eosin staining, Gomori staining, tartrate-resistant acid phosphatase staining and bone morphogenetic protein 2 immunohistochemical staining at the 12th week after operation. RESULTS AND CONCLUSION: New bone formation was visible in the bone defect regions of the mandible and parietal bone. The amount of lamelar bone formation and the degree of mineralization of the new bone were significantly increased in the parietal bone defect compared with the mandibular bone defect area, indicating the bone remodeling in the parietal bone defect area was better than that in the mandible bone defect area. The integral absorbance values of tartrate-resistant acid phosphatase and bone morphogenetic protein 2 in the parietal bone defect area were lower than those in the mandibular bone defect area, indicating that the viabilities of osteoblasts and osteoclasts in the parietal bone defect area were lower than those in the mandible bone defect area. These results demonstrate that occlusal stimulation may delay the bone remodeling during the repair of mandibular bone defects by regulating bone mineralization and maturation.
3.Correlation of innervation to osseointegration and osseoperception of implants

BACKGROUND: Consummate innervation plays an important role in bone formation. Nerve injury can impact normal bone metabolism, whereas bone regeneration depends on nerve regeneration in the innervation site to some degrees. Modified implant surface structure or usage of growth factors in local region can promote osseointegretion.OBJECTIVE: To retrospectively analyze effects of nervous system dominance bone metabolism and denervation on bone remodeling, innervated establishment and osseoperception during osseointegration following implantation.METHODS: The first author retrieved Pubmed database (http://www.ncbi.nlm.nih.gov/PubMed) and Wanfang database (http://www.wanfangdata.com.cn) (1990/2008) for publications of effects of nervous system dominance bone metabolism and denervation on bone remodeling, innervated establishment and osseoperception during osseointegration following implantation,with the key words of "bone metabolism, innervation, osseointegrationosseoperception" . Articles of repetitive contents were excluded. A total of 54 literatures were primarily obtained. According to inclusion criteria, 28 literatures were selected for summary.RESULTS AND CONCLUSION: Signals derived from the nervous system exert potent effects on osteoclast and osteoblast function. A ubiquitous innervation of all periosteal surfaces exists and its disruption affects bone remodeling. Several neuropeptides, neurohormones, nerotransmitters and their receptors are detectable in bone, and make it clear that nervous system is involved in bone metabolism and regeneration. Establishment of innervation of soft and hard tissue around implants may play an important role in osseointegration and osseoperception. Further studies will be required to explore the basic of anatomy,neurophysiology and neuropathology, which forms osseoperception.

5.Protective Effects of Salvia Yunnanensis Extract on Hypoxia/Reoxygenation Injury in Cultured Rat H9c2 Cardiomyocytes

Objective: To study the protective effects and the mechanism of Salvia yunnanensis extract on hypoxia/reoxygenation injury in cultured rat H9c2 cardiomyocytes.Methods: The hypoxia/reoxygen (H/R) injury model was established in H9c2 cell strain with or without the extract of Salvia yunnanensis.The cultured H9c2 cardiomyocytes were randomly divided into 6 groups: the normal control (C) group, H/R group, H/R+verapamil (H/R+V) group, H/R+Salvia yunnanensis extract at low dose (H/R+L, 0.01 mg·L-1) group, medium dose (H/R+M, 0.1 mg·L-1) group and high dose (H/R+H, 1.0 mg·L-1) group.The cell viability was measured by MTT assay and the activity of malondialdehyde (MDA) and lactate dehydrogenase (LDH) was measured by a detection kit.Fluorescence absorbance (A) value was measured by a fluoroscopy to show the intracellular reactive oxygen species (ROS) levels.Results: Compared with that in the model group, the survival rate of myocardial cells was significantly higher in Salvia yunnanensis extract at low, medium and high dose groups (P<0.05 or P<0.01), and the intracellular LDH leakage (P<0.05 or P<0.01), the content of MDA in cytoplasm (P<0.01) and the intracellular ROS levels significantly decreased in Salvia yunnanensis extract at high dose group (P<0.05).Conclusion: The extract of Salvia yunnanensis has protective effect on hypoxia/reoxygenation injury in cultured rat H9c2 cardiomyocytes, and the mechanism may be related to the reduction of lipid peroxides and removal of cell oxygen free radicals.
6.Analysis of changes of the platelet count after splenectomy in patients with cirrhosis and portal hypertension

Objective:To investigate the clinical significance of changes of platelet count after splenectomy in patients with cirrhosis and portal hypertension.Methods:In this study,from January 2002 to January 2006,the platelet count and liver function in 153 splenomegaly and hypersplenism due to cirrhosis patients who had been carried out splenectomy were selected.The change tendency of platelet count was observed,the relationship of changes of platelet count and liver function was analysed,and compared with patients of spleen trauma.Results:Postoperative platelet count was higher than preoperatively in most patients with cirrhosis,the portion did not elevate,even decreased.The change tendency of platelet count was correlated with the liver function.The change in cirrhosis patients is not more than patients of spleen trauma.Conclusions:The reason of thrombocytopenia in patients with cirrhosis may be correlated with the change of liver function besides the splenomegaly.

8. Effect of Modified Buyang Huanwu Tang on Serum PNT and IGF-1 in Patients with Diabetic Peripheral Neuropathy

Objective: To investigate the changes of serum proneurotensin (PNT) and insulin-like growth factor-1 (IGF-1) levels in patients with diabetic peripheral neuropathy (DPN) and their relationship with nerve conduction velocity (NCV) and Toronto Clinical Scoring System (TCSS), so as to observe the clinical efficacy of modified Buyang Huanwu Tang on DPN patients and its effect on the levels of PNT and IGF-1. Method: Totally 68 patients with DPN were randomly divided into treatment group and control group, with 34 cases in each group. Both groups were given mecobalamin capsules orally in addition to routine therapy of diabetes, while the treatment group was given modified Buyang Huanwu Tang orally for 4 weeks. The sensory nerve conduction velocity (SNCV) of median nerve, motor nerve conduction velocity (MNCV) of median nerve, SNCV of common peroneal nerve, MNCV of common peroneal nerve, TCSS score and serum PNT, IGF-1 levels were observed before and after treatment. Result: Before treatment, there was no significant difference in SNCV of median nerve, MNCV of median nerve, SNCV of common peroneal nerve, MNCV of common peroneal nerve, TCSS score and serum levels of PNT and IGF-1 between two groups. Serum levels of PNT and IGF-1 were positively correlated with SNCV of median nerve, MNCV of median nerve, SNCV of common peroneal nerve and MNCV of common peroneal nerve (P<0.05), and negatively correlated with TCSS score (P<0.05). Compared with those before treatment, SNCV of median nerve, MNCV of median nerve, SNCV of common peroneal nerve, MNCV of common peroneal nerve and the levels of serum PNT and IGF-1 in both groups were significantly increased (P<0.05), while TCSS scores were significantly decreased (P<0.05). Compared with control group, SNCV of median nerve, MNCV of median nerve, SNCV of common peroneal nerve, MNCV of common peroneal nerve and the levels of serum PNT and IGF-1 in treatment group were higher than those in control group (P<0.05), TCSS score was lower than that in the control group (P<0.05), and the total effective rate in treatment group was higher than that in control group (P<0.05). Conclusion: Serum PNT and IGF-1 may be involved in the occurrence and progress of DPN in patients with diabetes mellitus. Modified Buyang Huanwu Tang can increase the levels of serum PNT, IGF-1, NCV but reduce TCSS score in patients with DPN. It has a definite curative effect on DPN.
9.Denervation of the inferior alveolar nerve is associated with bone formation and bone mass maintenance in extraction sockets

BACKGROUND: There is a close relationship between innervations and bone formation and bone mass maintenance in the extraction sockets. OBJECTIVE:To study the possible effect of denervations on the regulation of new bone formation and bone mass maintenance in the extraction sockets. METHODS:The unilateral inferior nerve of dogs was sectioned to establish an animal denervation model. The normal side was used as control. After model establishment, the premolars of denervated side and normal side were extracted. Histological method was used to test new bone formation and bone mass maintenance in the extraction sockets at the 2nd, 4th, 8th and 12th weeks after tooth extraction. RESULTS AND CONCLUSION:The percentage of new bone areas in the extraction sockets was significantly lower in the experiment group than the control group at weeks 2, 4, 8 after tooth extraction (P < 0.01). The height difference between the buccal and lingual alveolar ridge was higher in the experimental group than the control group at weeks 2, 4, 8, 12 after tooth extraction (P < 0.05 or P < 0.01). These findings indicate that denervation is closely related with new bone formation and bone mass maintenance in the extraction sockets.
10.The relationship between the characteristics of type 3 secretion system and biofilm of Pseudomonas aeruginosa in diabetic foot wound with antibiotic resistance

Objective:To explore the characteristics of type 3 secretion system and biofilm of Pseudomonas aeruginosa in diabetic foot wound, and to analyze the relationship between these factors, as well as to the antibiotic sensitivity.Methods:Thirty-three strains of Pseudomonas aeruginosa were collected from the foot wounds of diabetic foot inpatients in Tianjin Medical University Chu Hsien-I Memorial Hospital from February 1, 2018 to December 31, 2018. Thirteen strains of Pseudomonas aeruginosa were collected from non-diabetic wounds. All strains were tested for antibiotic sensitivity. The virulence genes exoS or exoU of Pseudomonas aeruginosa and the ability of biofilm formation were tested. The characteristics of exoS or exoU and biofilm of Pseudomonas aeruginosa were analyzed. Patients′ clinical outcomes were also analyzed.Results:Pseudomonas aeruginosa with exoS gene was the major pathogen, 90.9% found in diabetic foot group and 84.6% in control group, with no significant difference( χ2=0.54, P=0.46). The drug-resistant strains of Pseudomonas aeruginosa with exoS accounted for 16.7% in diabetic foot group and 18.2% in control group, also with no significant difference( χ2=0.18, P=0.83). There were 5 strains of Pseudomonas aeruginosa carrying exoU, 3 strains in diabetic foot group, of which 1 was resistant, 2 in control group, no resistant strain. Pseudomonas aeruginosa increased the ability of biofilm formation in diabetic foot group, accounting for 57.6%, and for resistant strains, 83.3% of them increased the biofilm formation ability. Two kinds of Pseudomonas aeruginosa produced different biofilms, but they were effectiveless for carbapenem antibiotics. The times of debridement ( P<0.01), time of antibiotic use ( P<0.01) were more in biofilm wound, but the healing rate reached 75%-90%. Conclusion:Pseudomonas aeruginosa secreting ExoS is the main one in the diabetic foot wound. The ability of Pseudomonas aeruginosa to produce biofilm in DF wound is increased. Biofilm is one reason for its antibiotic resistance. Multiple debridement combined with sensitive antibiotics is an effective method to remove biofilm.
