1.Effects of acute hypervolemic hemodilution with HES130/0.4 and HES200/0.5 on blood coagulation

Objective To evaluate the effects of 6% hydroxyethyl starch (HES)130/0.4 and 6% HES 200/0.5 on blood coagulation in patients undergoing major operation under general anesthesia.Methods Sixty ASA Ⅰ or Ⅱ patients aged 18-64 yr undergoing major operation under general anesthesia during which the blood loss was expected to be>20%of blood volume were allocated randomly into 3 groups(n=20 each):Ⅰ control group(C);Ⅱ6% HES200/0.5 group (H) and Ⅲ HES130/0.4 group (V).In group H and V the patients Arterial blood samples were taken before(T1),immediately(T2)and 1 h after the end of fluid infusion(T3)for determination of hematocrit(Hct),platelet count(PLT)and prothrombin time(PT),platelet maximum aggregation test induced by 5 μmol/L ADP,and Sonoclot coagulation and platelet function analysis(gbACT,CR,PF,TP).Activated partial thromboplastin time(Am),coagulation factor Ⅷ (FⅧ)activity and von Willebrand factor(vWF)were measured only in group H and V.Results (1) PT and APTT were significantly prolonged after fluid infusion at T2 and T3 as compared with the baseline values at T1 in group H and V.(2)FⅧ activity and platelet maximum aggregation induced by ADP were significantly decreased at the end of fluid infusion at T2 as compared with the baseline at T1 but increased again at T3 in group H and V.(3)There was no significant change in Sonoclot coagulation parameters after fluid infusion as compared with the baseline except clot formation rate (CR)in group H and V.Conclusion Acute hypervolemic hemodilution with 6% HES130/0.4 or 200/0.5 has minimal effect on blood coagulation and there is no significant difference in blood coagulation changes induced by 6%HES130/0.4 and 200/0.5.
2.The comparative analysis of seven tumor markers in the two detection systems between roche E411 and E601

Objective To investigate the comparability of test seven tumor marks results of two electrochemiluminescence immu-noassay analyzer between Roche Cobas E411 and Cobas E601 .Methods According to United States Clinical and Laboratory Stand-ards Institute (CLSI) EP9-A2 document ,E411 was acted as comparison method (X) and E601 was as the experimental method (Y ) .Alpha fetoprotein (AFP ) ,carcinoembryonic antigen (CEA ) ,carbohydrate antigen125 (CA125 ) ,carbohydrate antigen199 (CA199) ,carbohydrate antigen153(CA153) ,total prostate specific antigen(TPSA) and free prostate specific antigen(FPSA) levels of patient samples were tested and the results were analyzed .Results The correlation between the two systems was good and the bias was acceptable in the medical decision level .Conclusion Two systems have good concordance in detecting the seven tumor markers ,w hich can meet the clinical need .
3.Combined detection of anti CCP antibodies,AKA and APF in the diagnosis of rheumatoid arthritis

Objective To investigate the diagnostic value for rheumatoid arthritis(RA) by using combined detection of anti‐cy‐clic citrullinated peptide antibody (anti CCP ) ,anti keratin antibody (AKA ) and the antiperinuclear factor (APF ) in serum . Methods A total of 110 RA patients(RA group) ,50 patients with other autoimmune diseases(non RA group) and 110 healthy subjects (control group) were enrolled in the study .The concentrations of AKA ,APF were detected by using indirect immunofluo‐rescence assay and anti CCP by using ELISA .Results The positive rates of anti‐CCP ,AKA ,APF in RA group were higher than those in non RA group and control group(P<0 .05) .In the series detection of the three indicators ,the sensibility and specificity were 44 .55% and 99 .38% respectively ;in the parallel detection of the three indicators the sensibility and specificity were 93 .64%and 85 .63% respectively .Conclusion Anti CCP detection exhibits relatively higher sensitivity and specificity in the diagnosis of RA .Series detection of the 3 indicators can improve the specificity ,reduce the rate of misdiagnosis;parallel detection of the 3 indica‐tors can improve the sensitivity ,reduce the rate of misdiagnosis .The Combined detection of anti CCP ,AKA and APF has better di‐agnostic efficiency than single detection .
4.THE VALUE OF CELL DNA QUANTITATIVE ANALYSIS IN THE EARLY SCREENING OF CERVICAL CANCER AND PRE-CANCERATION

Objective To evaluate the application of DNA image cytometry in screening cervical intraepithe-lial neoplasia and cervical cancer.Methods 80 patients with cervical intraepithelial neoplasia admitted during No-vember 2012 and November 2014 were involved in this study.The samples from all the patients were taken by cervix brushes and prepared for 2 slides:one for liquid-based cytology staining, read by cytologist, and the other for Thi-onin-Feulgen staining, tested by DNA image cytometry.The biopsy was performed with direction of colposcopy if one test was positive to verify the sensitivity and specificity of the two methods of staining.Results 70 cases were positive by DNA image cytometry and 27 ones were positive by liquid-based cytology.There was statistically signifi-cant difference in the sensitivity between the DNA image cytometry and the liquid -based cytology ( 94.8% vs. 38.8%;p<0.05), but no difference in the specificity between them (99.6%vs.99.8%).Conclusion Besides its good specificity, DNA image cytometry is more sensitive in screening cervical intraepithelial neoplasia and cervical cancer.
5.Urodynamic analysis of changed length of functional urethra in male dogs

Objective To evaluate the variation of functional urethra length on urine control,explore a new method to prevent urinary incontinence and seek urodynamic evidence after radical prostatectomy. Methods Sixteen male dogs,matched in body weight,were randomly divided into two groups. Group A were used to test the different rest urethral pressure profile after resection of prostate,resection of distant prostate plus posterior urethra in length of 1.5 cm and 2.0 cm respectively; Group B to test the different rest urethral pressure profile on pedicle myotube in length of 2.0 cm and 1.0 cm using anterior bladder flap respectively after resection of prostate and posterior urethra in length of 2.0 cm. Results The urodynamic indexes fell off while the functional urethra length decurtated gradually in group A,but they went up in group B. The two groups showed significant difference before and after the resection of prostate and distant urethra in length of 2. 0 cm. Conclusion There was a direct ratio between the functional urethra length and the capability of urethral urine control. Reconstruction of functional urethra using anterior bladder flap pedicle myotube is a good choice to treat urinary incontinence after radical prostatectomy.
6.Comparison of ELISA and colloidal gold immunochromatographic assay for detection of serum anti-CCP antibody in the diagnosis of RA

Objective To evaluate the diagnostic values of enzyme‐linked immunosorbent assay (ELISA ) and colloidal gold im‐munochromatographic assay in detecting serum anti‐cyclic citrullinated peptide(anti‐CCP) antibody .Methods Colloidal gold immu‐nochromatographic assay and ELISA were used to detect serum anti‐CCP antibody in 110 RA patients and 110 healthy cases . Results The data were analysed with the paired Chi‐Square Test (χ2 = 174 .354 ,P < 0 .01) ,the difference was significant .The K ap pa value was 0 .890(P< 0 .01) ,indicating that the coincidence between the two kinds of methods was good .The sensitivity , speciality of ELISA were 83 .64% and 92 .73% respectively .Conclusion In detecting serum anti‐CCP Antibody ,there was dife‐rence between colloidal gold immunochromatographic assay and ELISA according to statistics analysis .The sensitivity and speciality of ELISA were higher than those of colloidal gold immunochromatographic assay .
7.Expression of centrosome-associated kinase Aurora-A, mt-P53, and c-myc in colorectal cancer and its significance

Objective:This study aimed to investigate the expression levels of centrosome-associated kinase Aurora-A, mutant type P53 (mt-P53), and c-myc in colorectal cancer. This study was also conducted to investigate the mutual relationship and functions of these factors in tumorigenesis and tumor progression. Methods:We examined the pathological specimens obtained from colorectal cancer, pericancerous tissues, and normal colorectal tissues by tissue microarray technique and immunohistochemistry (SP method) to determine the expression levels of Aurora-A, mt-P53, and c-myc proteins. The clinicopathological parameters were then analyzed. Re-sults: The positive rates of Aurora-A expression in normal colorectal tissues, pericancerous tissues, and colorectal cancer were 0%, 35%, and 69%respectively;by comparison, the positive rates of mt-P53 were 0%, 20%, and 57%, respectively. For c-myc, the positive rates were 0, 37%, and 76%, respectively. The expression levels of Aurora-A, mt-P53, and c-myc were significantly higher in tumor tis-sues than in normal colorectal tissues and pericancerous tissues (P<0.01). Aurora-A overexpression was related to the depth of invasion (P<0.05). Mt-P53 and c-myc overexpression was related to the depth of invasion, lymph node metastasis, and Dukes' classification (P<0.05). A strong positive correlation was observed between the expressions of Aurora-A, mt-P53, and c-myc in colorectal cancer (r=0.362, P<0.01; r=0.487, P<0.01). A strong positive correlation was also observed between the expressions of mt-P53 and c-myc in colorectal cancer (r=0.242, P<0.01). Conclusion:The overexpression of Aurora-A and c-myc and the mutation of P53 were important in tumorigenesis, tumor invasion, and metastasis of colorectal cancer. Thus, the co-detection of Aurora-A, mt-P53, and c-myc may be useful for the early diagnosis and prognosis of colorectal cancer.
8.Chondrogenic differentiation of mouse bone marrow mesenchymal stem cells induced by cartilage-derived morphogenetic protein-2 in vitro.

To study the cartilage differentiation of mouse mesenchymal stem cells (MSCs) induced by cartilage-derived morphogenetic proteins-2 in vitro, the MSCs were isolated from mouse bone marrow and cultured in vitro. The cells in passage 3 were induced into chondrogenic differentiation with different concentrations of recombinant human cartilage-derived morphogenetic proteins-2 (0, 10, 20, 50 and 100 ng/mL). After 14 days of induction, morphology of cells was observed under phase-contrast microscope. Collagen II mRNA and protein were examined with RT-PCR, Western blotting and immunocytochemistry respectively and the sulfate glycosaminoglycan was measured by Alcian blue staining. RT-PCR showed that CDMP-2 could promote expression of collagen II mRNA in an dose-dependant manner, especially at the concentration of 50 ng/mL and 100 ng/mL. Immunocytochemistry and Western blotting revealed a similar change. Alcian blue staining exhibited deposition of typical cartilage extracellular matrix. Our results suggest that mouse bone marrow mesenchymal stem cells can differentiate into chondrogenic phonotype with the induction of CDMP-2 in vitro, which provides a basis for further research on the role of CDMP-2 in chondrogenesis.

9.The correlation analysis of indoor carbon monoxide concentration and chronic diseases

Objective To explore the correlation of the indoor carbon monoxide concentration and chronic diseases by monitoring the indoor carbon monoxide concentration timely.Methods Collected the data of non-traumatic patients(n=80)from July 201 1 to January 2014,and assigned them into the experiment group with carbon monoxide concentration measured and the control group without measured.All the subjects were further divided into one group with complaint of carbon monoxide poisoning and the other one without.For patients exposed to high concentration of carbon monoxide,carboxyhemoglobin was also measured and APACHEⅡ was scaled,and the relationship between them was analyzed.The confirmation and misdiagnosis rate of carbon monoxide poison-ing were calculated.For the patients with acute exacerbation of chronic diseases like chronic cardiac failure,chronic obstructive pul-monary disease,cerebral infarction induced by carbon monoxide poisoning,BNP,CAT and NIHSS were documented on admission and during follow-up with removal of carbon monoxide exposure and compared respectively.Results The relationship between blood carboxyhemoglobin,APACHEⅡ scores on admission and indoor carbon monoxide concentration was linear,and obviously positive.Between the experiment group(n=40)and the control group(n=40),there was significant difference(P <0.05)in confirma-tion rate and misdiagnosis rate with 80.00% vs.55.00% and 6.25% vs.81.80% respectively.For the 54 patients with carbon monoxide poisoning diagnosed,the changes between before and after removal of carbon monoxide exposure of the CAT scores of chronic obstructive pulmonary disease and NIHSS score of CI disease and myoglobin,troponin I,creatine kinase isoenzyme,BNP for chronic cardiac failure were significantly different(P <0.01).Conclusion The indoor carbon monoxide concentration may indicate the severity of carbon monoxide poi-soning,which could increase the confirmation rate of carbon monoxide poisoning,and reduce the misdiagnosis rate.It is helpful to identify carbon monoxide exposure,the common inducing factor,so as to improve the patients′clinical symptoms.
10.Control observation between mild moxibustion and TDP for obsolete collateral ligament injury of interphalangeal joints.

OBJECTIVETo compare the effects between mild moxibustion and specific electromagnetic spectrum therapy apparatus (TDP) for obsolete collateral ligament injury of interphalangeal joints.
METHODSSixty patients were randomly divided into a mild moxibustion group and a TDP group, 30 cases in each one. In the mild moxibustion group, pure moxa sticks were used at the affected digital joints locally for 20-30 min a time. In the TDP group, TDP was applied at the affected digital joints locally for 20-30 min a time. The treatment was given once a day for two courses, and 10-day treatment was made into a course. Visual analogue scale (VAS) for pain, swelling degree of the affected digital joints before and after treatment were observed and the clinical efficacy and safety were evaluated in the two groups.
RESULTSThe excellent rate was 56.7% (17/30) and the excellent and, good rate was 83.4% (25/30) in the mild moxibustion group,which were better than 36.7% (11/30) and 76.7% (23/30) in the TDP group respectively (both P < 0.01). After treatment the score of VAS and digital joints swelling degree were improved than those before treatment in the two groups (P < 0.01, P < 0.05), and the improvements of the mild moxibustion group were better than those of the TDP group (P < 0.01, P < 0.05).
CONCLUSIONMild moxibustion can apparently relieve the painful and swelling degree of obsolete collateral ligament injury of interphalangeal joints, which is superior to TDP.
