1.The modulation of met-enkephalin and naloxone on the excitatory effects of kainic acid and glutamic acid in hippocampus of rats
Journal of Third Military Medical University 1983;0(03):-
Objective To observe the modulation of met enkephalin (ME) and naloxone (NLX) on the excitatory effects of kainic acid (KA) and glutamic acid (GLU) in hippocampus of rats and to explore the possible mechanism of the effects of ME in the KA epilepsy model. Methods A total of 45 healthy Wistar rats were anesthetized with 1% sodium pentobarbital (3 ml/kg intraperitoneal injection). After endotracheal intubation, the animal was mounted onto a SN 1stereotaxic apparatus (Narishige) and routine craniotomy was performed. Then the animal was immobilized by 0.01% curare (1 mg/kg) after it woke up. The peripheral pipettes of a multiple pipette microelectrode were used for microelectrophoretic application of drugs; the central pipette was used for extracellular recording of the hippocampal unit discharges (HUDs). Results ①ME excited most of the HUDs; opioid receptor antagonist NLX could reverse the excitatory effects of met enkephalin. NLX applied alone had no effects on the firing rate of HUDs. ②KA and GLU excited the HUDs intensively but ME and NLX could modulate their excitatory effects. Conclusion ME may promote the development of epilepsy by modulating the excitatory effects of KA and GLU in hippocampus.

Objective To Analyze the distribution characteristics of Hyperthyroidism's syndrome and the relationship between symptoms and thyroid hormones. Methods The TCM syndromes of a total of 78 patients with hyperthyroidism were differentiated. The relationship between syndrome and general condition of patients (gender and age, etc.) were analyzed;correlation between symptoms and thyroid hormones were also studied. Results Of all 78 patients, patients with yin deficiency and hyperactive yang accounted for 43.6%; patients with internal disturbance of phlegm-fire accounted for 30.8 %; and patients with hypteracitivity of heart-fire and liver-fire accounted for 25.6%. The severity of all the symptoms showed positive correlation with hyroid hormones (the value of r is positive, P<0.05); and no linear relationship with TSH (P>0.05).Conclusion The syndrome of yin deficiency and hyperactive yangI was the most common syndrome type. Thyroid hormones had direct proportion with the severity of syndrome.

Objective To investigate the diagnostic value of treadmill exercise tests (TET), Hoher electro-cardiogram in coronary artery disease(CAD) patients. Methods 144 patients who clinically diagnosed as CAD,un-derwent TET and Hoher electrocardiogram. Then the patients with the positive results were examined by coronary arte-riography(CAG). According to the results of CAG the diagnostic value of TET and Hoher electrocardiogram was per-formed with comparative analysis. Results The sensitivity, specificity and evaluated accuracy rates of TET were 93.3% ,72. 2% and 73. 8% respectively. But the sensitivity, specificity and positive evaluated accuracy rates of Hol-ter electrocardiogram to examine myocardial iachemia were 76.7%, 88.4%, 57.1%. Conclusions The combined examination of TET and Holter electrocardiogram could preliminarily estimate the stenosis site of coronary artery. For the patients with unconditional GAG, those two examinations could increase the diagnostic reliability of positive rate of myocardial isehemia, and on this basis to enhance the diagnostic level of the stenosis site of coronary artery.

Chinese Journal of Clinical Oncology 2014;(16):1017-1020
Objective: To investigate the mechanism responsible for homoharringtonine (HHT), which contributes to imatinib (IM) sensitivity in the chronic myeloid leukemia (CML) cell line. Methods:We established cell lines from a patient with CML at the time of first diagnosis and relapse phase, and designated the cell lines as NPHA1 and NPHA2, respectively. Stable underexpressed EphB4 cells (NPHA2-EphB4-sh) were obtained. Leukemia cell lines were incubated with HHT. The activated signal proteins in cells were tested by Western blot. Results:EphB4 was overexpressed in IM-resistant NPHA2 compared with the NPHA1 cell line. However, the expression of EphB4 mRNA and protein were significantly decreased in knockdown NPHA2-EphB4-sh cells compared with the NPHA2 and NPHA1 (P<0.001) cell lines. NPHA2-EphB4-sh cells were sensitive to IM (IC50:0.93 mg/L), and NPHA2 showed IM re-sistance (IC50 : 5.45 mg/L) (P<0.001). However, co-stimulation with HHT+IM decreased IC50 of NPHA2 cells to 1.17 mg/L (P<0.001). Meanwhile, phospho-Rac1/cdc42 was significantly increased in NPHA2 cells compared with NPHA2-EphB4-sh (P<0.001). HHT blocked the expression of EphB4/RhoA. Conclusion: The overexpression of EphB4 contributed to IM resistance in CML line cells. EphB4/RhoA may be a new marker of IM resistance. HHT with IM yielded more treatment advantages than IM alone by blocking EphB4/RhoA pathways.
