1.Anti-oxidation Effects of Compound Chinese Medicine of Yiqiling on Hypertensive Rats

Objective To investigate the anti-oxidation effects of compound Chinese medicine of Yiqiling,on the spon- taneously hypertensive rats(SHR) and to explore the new target for improving the prognosis of hypertension.Method SHR were randomly divided into 2 groups:Yiqiling group and the control group.The serum content of malondiadehyde (MDA) and superoxide dismutase(SOD),and the urine content of 8-iso prostaglandin F2?(8-isoPGF2?) in two groups were measured.Examination of the gene expression profile was analyzed by using cDNA microarray.Three of the anti-oxidation genes were selected to undergo RT-PCR and quantitative real-time PCR.Results The 8-isoPGF2?urine level and MDA serum level were lower,and SOD serum level was higher in Yiqiling group than those in the control group,the differences of the above indexes had statistical significance.In Yiqiling group the expression of 354 genes was up-regulated and that of 121 genes was down-regulated in comparison with the control group.The results of the Hsphl,Hspca and Pam genes detected by RT-PCR and quantitative real-time PCR were consistent with those detected by the microarray examination.Conclusion Yiqiling has anti-oxidation effects and improvement on the prognosis of SHR,and its possible mechanism is related with the regulation of Hsphl,Hspca and Pam genes expression.
2.Clinical analysis of psychological problems in patients with rheumatic diseases

Objective To study the mental status and its influencing factors of patients with rheumatic disease. Methods Three hundreds and eighty-one patients with rheumatic diseases were assessed with the self-rating anxiety scale, self-rating depressive scale, Eysenck Personality Questionnaire symptom checklist-90, and demographic characteristics questionnaire. The comparison among groups were tested by t tests, Spearman's correlation analysis. Results Patients with anxiety symptoms accounted for 35.96%, and pre-valence rate of depression was 48.03%. The gender (r=0.87, OR=2.34) and duration (r=0.006, OR=1.006) were the risk factors and the C4 (r=-2.63, OR=0.07) was the protective factor. In patients with systemic lupus erythematosus, depression was related to education (Z=2.228, P=0.026), disease duration (Z=-2.015,P=0.044), IgG level (t=-2.266, P=0.027), T cell subsets CD8 (Z=1.981, P=0.047) and CD3 (Z=1.967, P=0.049). Anxiety was related to education (Z=2.452, P=0.014), duration (Z=-2.054, P=0.04), complement C3 (t=2.035, P=0.045), C4 (Z=-2.204, P=0.028) and IgG level (t=-2.126, P=0.036). The value of CRP(Z=-1.984, P=0.047) and IgM (t=2.243, P=0.028) of rheumatoid arthritis patients were higher in the depression group. The depression group was different in disease duration (Z=-2.631, P=0.009), gender ( χ2=4.415, P=0.042), ESR (Z=2.17, P=0.023) and CD3 cell (Z=2.023, P=0.043) in Sj?gren's syndrome patients. Correlation analysis showed positive correlation between the score of SDS, SAS and the P value (r=0.21, P=0.01) (r=0.14, P=0.007) N dimension (r=0.52, P=0.01) (r=0.55, P=0.007) of EPQ. There were negative correlation between the score of SDS, SAS and the E dimension (r=-0.23, P=0.01) (r=-0.14, P=0.006) of EPQ. Conclusion The prevalence of depres-sion and anxiety is higher in patients with depressive and anxious symptoms. They are in older age, female gender, poor education, longer duration, lower complement and lower T cell subsets when compared with those patients without depression and anxiety. Patients who have an introverted, instable, nervous temperament are more likely to be depressed and anxious. Psychological assessment intervention and therapy should be carried out as early as possible.
3.Biomechanics of posterior lumbar interbody fusion with unilateral fixation

Objective To study the biomechanics of posterior lumbar interbody fusion with unilateral fixation.Methods 15 bovine lumbers were divided into three groups(intact group,spondylolisthesis group,posterior lumbar interbody fusion with unilateral fixation group as experimental group).Nondestructive tests were performed in pure compression,flexion,extension,and lateral bending on every group.Results Compared with intact group and spondylolisthesis group,after posterior lumbar interbody fusion with unilateral fixation,the mean straining of vertebral body in experimental group increased.The value of experimental group had significant difference from spondylolisthesis group(P < 0.01,P <0.05).And the value of experimental group was proximal to intact group (P > 0.05).Conclusion After operation with posterior lumbar interbody fusion with unilateral fixation,the stiffness of vertebral body was approached the intact group during axis compression,flexion,extension and lateral bending.And the initial stability was enough after operation.

5.Influence of interbody fusion intervertebral fixation on adjacent joint degeneration in patients with lumbar diseases

Objective To explore the influence of interbody fusion intervertebral fixation on adjacent joint degeneration in patients with lumbar diseases .Methods 56 patients with lumbar diseases who were taken interbody fusion intervertebral fixation were selected as the research subjects ,and the adjacent intervertebral joint degeneration in patients were followed up .Results 56 patients were followed up for 24-72 months,the average time was (41.2 ± 8.3)months.The new clinical symptoms were occurred in 12 cases(21.4%),including 8 cases of low back pain, 3 patients with leg pain,1 case of lumbocrural pain.Radiographic changes occurred in 9 cases(16.1%),9 cases of patients with new clinical symptoms ,including 5 cases of osteophyte formation or exacerbation ,2 cases for adjacent segment instability ( I degree of vertebral spondylolisthesis ) ,1 case of small joint hyperplasia stage spinal stenosis , 1 case of protrusion of intervertebral disc .In 9 patients with imaging performance ,internal fixation of upper adjacent segment degeneration in 7 cases,internal fixation of the adjacent segment degeneration in 2 cases.Conclusion Inter-body fusion intervertebral fixation in the treatment of lumbar diseases ,adjacent joint is prone to degeneration ,so the destruction of adjacent joint operation should be reduced .
6.Clinical efficacy of sequential therapy for butylphthalide combined with alteplase in the treatment of acute cerebral infarction

Objective To observe the effect of butylphthalide sequential therapy combined with injection of alteplase(injection with recombinant human collagenous tissue plasminogen activator) in the treatment of acute cerebral infarction (ACI).Methods 61 patients with ACI and within 4.5 hours of onset were selected,and they were divided into treatment group(butylphthalide combined with alteplase group,31 cases) and the control group (alteplase only group,30 cases) by random number table.The two groups were given glucose-lowering,statins for maintaining the blood pressure,as well as aspirin enteric tablets or clopidogrel for those without bleeding on the 24 hour review head CT in the acute phase.The treatment group was given alteplase for injection with sequential butylphthalide therapy (butylphthalide injection for 2 weeks followed by an oral administration of butylphthalide soft capsules for 3 months),and the control group was given only alteplase for injection.The National Institute of Health stroke scale (NIHSS) and Barthel index (BI) were used for evaluating the final clinical efficacy.Results The NIHSS scores of the treatment group compared with the control group at 24 hour,14 days and 3.5 months after the treatment had statistically significant differences (t =-0.102,-1.964,-2.444,P =0.037,0.018 and 0.012).The NIHSS scores of the treatment group had decreased 2%,27% and 41% compared with the control group at 24 hour,14 days and 3.5 months after the treatment,respectively.The BI score of the treatment group and control group at 3.5 months after treatment had statistically significant difference (t =-1.956,P =0.029).Conclusion The sequential butylphthalide therapy combined with injection of alteplase has good synergistic effect for the patients with onset of ACI less than 4.5 hours,and it is a safe and effective treatment method for ACI within 4.5 h and it is worthy of clinical promotion.
7.Application of Propofol in Painless Fibrobronchoscopy and Its Effects

OBJECTIVE:To evaluate the effects of propofol in relieving pain during fibrobronchoscopy when it is used in general intravenous anesthesia.METHODS:160patients undergoing fibrobronchoscopy were randomly divided into propofol group and control group.90patients in the propofol group were anesthetized intravenously by injection of propofol at the dosage of1.5mg/kg and speed of30mg/10s and then underwent fibrobronchoscopy;While70patients in the control group underwent regular fibrobronchoscopy.RESULTS:The lash reflex disappeared within(40.73?7.91)seconds after propofol injection,and patients became conscious within(5.39?1.85)minutes after stopping injection,full consciousness occurred at(10.82?2.73)minutes.Electrocardiogram did not show any signs of change in blood pressure,myocardial ischemia and cardiac dysrhythmia,the post-operative satisfaction rate was96%as compared with81%in the control group.The patients in the propofol group showed extensive willingness for second fibrobronchoscopy,while the patients in the control group presented cough,struggle,and20%of them refused the second fibrobronchoscopy.CONCLUSION:It is safe and effective to apply propofol in painless fibrobronchoscopy.
8.Expression of Topo-Ⅱ, GST-π in gastric cancer tissues and their significance

Objective To explore the expressions of Topo-Ⅱ,GST-π in gastric cancer tissues and normal gastric mucosa tissues,to reveal its relationship with clinical pathological features and significance.Methods Immunohistochemical method was used to detect Topo-Ⅱ,GST-π expressions in 100 cases of gastric carcinoma and 20 cases of normal gastric mucosa,make a comprehensive analysis combined with clinical pathology data.Results There were significant difference of expression of Topo-Ⅱ,GST-π between the normal gastric mucosa tissues and gastric cancer of different degree of differentiation.Topo-Ⅱ positive expression rate of 5.0 % (1/20),100 % (30/30),96.7 % (29/30) and 87.5 % (35/40) respectively; GST-π positive expression rate were 60.0 % (12/20),83.3 % (24/30),96.7 % (29/30) and 100.0 % (40/40) respectively (P < 0.05).The expressions of Topo-Ⅱ,GST-π in gastric cancer tissue were not relevant to patient' s sex,age,tumor location,infiltration depth (P > 0.05).Topo-Ⅱ associated with the differentiation degree and lymph node metastasis of tumors,with the decreasing degree of tumor differentiation,lymph node metastasis,Topo-Ⅱ expression also decreased.GST-π was associated with tumor diameter,degree of differentiation and lymph node metastasis,the lower the degree of differentiation,lymph node metastasis,tumor diameter the more,GST-π expression increased (P < 0.05).GST-π and Topo-Ⅱ were negative correlation and both expressed in gastric cancer tissue (P < 0.01).Conclusions The expressions of Topo-Ⅱ in gastric cancer tissue is associated with the differentiation degree and lymph node metastasis of the tumor.GST-π is associated with tumor diameter,the degree of differentiation and lymph node metastasis.GST-π and Topo-Ⅱ in gastric cancer tissues are negatively correlated.
9.DADS induces G2/M arrest through Chk1/Cdc25C/CyclinB1/CDK1 pathway in human leukemia HL-60 cells

Aim To study the effects of cycle arrest and molecular mechanism in human leukemia HL-60 cells induced by diallyl disulfide ( DADS ) . Methods Cell count, colony formation in soft agar experiments and flow cytometry analysis were employed to observe the DADS-induced cell growth inhibition and the effect of cycle arrest in HL-60 cells. The expressions of Chk1/2 and its downstream element in HL-60 cells were detected by Western blot. Results Cell count revealed that population doubling time increased to 35. 03 h and 71. 82 h, respectively, from 19. 14 h in HL-60 cells treated with 60 and 120 μmol·L-1 DADS ( P<0. 05 ) . Colony formation in soft agar experiments showed that colony formation inhibition rate of HL-60 cells exposed to 30, 60, 90 and 120μmol·L-1 DADS increased to 35. 06%, 62. 10%, 93. 79% and 99. 35%, respectively ( P<0. 05 ) . Flow cytometry a-nalysis exhibited that HL-60 cells treated with 60 and 120 μmol · L-1 DADS for 24 h and 48 h arrested in G2/M phase in a concentration-and time-dependent manner ( P <0. 05 ) . Western blot disclosed that the expression of p-Chk1 increased in a time-dependent manner ( P <0. 05 ); however, Chk1, Chk2 and p-Chk2 were not changed in HL-60 cells treated with 60μmol·L-1 DADS (P >0. 05). The expression of Cdc25C, CyclinB1 and CDK1 decreased after treated with 60 μmol·L-1 DADS in a time-dependent manner ( P<0. 05 ) , but the expression of 14-3-3 protein did not change ( P>0. 05 ) . Conclusion DADS can in-hibit the proliferation of HL-60 cells, and induce G2/M arrest through Chk1/Cdc25 C/CyclinB1/CDK1 path-way.
10.The imbalance of CD4~+ CD25~+ Foxp3~(high) regulatory T cells/Thl7 cells in infant with sepsis

Objective To further explore the pathogenesis of disturbed adaptive immune response in infants with sepsis. Methods Forty-eight infants with sepsis and 26 age-matched healthy infants were enrolled in this study. The HLA-DR expression of CD14~+ monocyte, the proportion of CD4~+ CD25~+ Foxp3~(high) Tr cells and the proportion of Thl7 cells were measured by flow cytometry. Cytokines (IL-1β, IL-6, TNF-α, IL-10, TGF-β and IL-17A) were measured by ELISA. Real-time PCR were used to evaluate the mRNA levels of Foxp3, ROR-γt in CD4-positive cells and IL-17A. Forty-eight infants with sepsis were divided into two groups according to HLA-DR expression of CD14~+ monocyte: DR-H group ( > 30% ) and DR-L group ( < 30% ). Results The ratio of IL-10/TNF-α in DR-L group was higher than that in healthy control or DR-H group(P <0.05). The proportion of CD4~+ CD25~+ Foxp3~(high) Tr cells and mRNA expression of transcription factor Foxp3 in DR-L group was found to be significantly higher than that in healthy control or DR-H group(P<0.05). The proportion of Thl7 cells, Serum concentration of IL-17A, the mRNA expression of IL-17A and transcription factor ROR-γt were significantly increased in DR-H group and DR-L group (P < 0.05) , while there is no significant difference between DR-H and DR-L group( P >0.05). Serum levels of Th17-inducing cytokine such as IL-1β, IL-6 were significantly elevated in DR-H group and DR-L group (P<0.05), while there is no significant difference between DR-H and DR-L group( P>0.05). Serum level of CD4~+ CD25~+ Foxp3~(high) Tr-inducing cytokine TGF-p in DR-L group was higher than that in DR-H or healthy control group(P<0. 05). Conclusion Over-activation of Th17 cells may be one of the factors causing aberrant increase of pro-inflammatory cytokine/chemotatic factor in infant with sepsis. The imbalance of CD4~+ CD25~+ Foxp3~(high) Tr cells/Th17 cells may be contributed to the pathogenic mechanism of mixed antagonist response syndrome ( MARS) in infant with sepsis. The changes of cytokine environment in infants with sepsis may be one of the factors causing the imbalance of CD4~+ CD25~+ Foxp3~(high) Tr cells/Th17 cells.
