AIM:To investigate the role of nerve growth factor (NGF) and leukemia inhibitory factor (LIF) in the expression of IL-4 and IL-5 in the splenic lymphocytes of asthmatic rats. METHODS:Sixteen Sprague-Dawley rats were randomly divided into two groups:control group (n = 8) and asthma group (n = 8). The asthmatic model was set up by intraperitoneal injection of 100 mg ovalbumin and 100 mg aluminum hydroxide,and receiving nebulization aspiration of 1% ovalbumin solution 2 weeks later. The splenic lymphocytes from the control rats and asthmatic rats were isolated and cultured with exogenous NGF or LIF at different doses. The mRNA expression of IL-4 and IL-5 in the splenic lymphocytes was respectively detected by reverse transcription-polymerase chain reaction (RT-PCR). Objective:To compare the effects of different polishing techniques on the surface roughness of ceramic surfaces,provi-ding reference to restoration polishing in clinic.Methods:70 disc-shaped ceramic specimens were randomly divided into 7 groups(n=10).The glazing treated samples were served as the controls(group 1),the rest samples were treated by white aluminum oxide stone only(group 2)or followed by the polishing head of Ceramiste(group 3),Ceramiste and Ceramaster(group 4),Komer porcelain (group 5),Komer zirconium oxide(group 6)and EVE zirconium oxide,respectively.Roughness of the ceramic samples(including Ra,Rz and Rp)was measured with surface profilometry after different process and the data were analyzed by one way ANOVA.The surface characteristics of the specimens were observed by scanning electron microscopy.Results:The Ra and Rp of group 2 and 3 was higher than those of group 1(P <0.05).The Rz of group 2,3,4 and 5 was higher than that of group 1(P <0.05).Conclusion:Polishers interspersed with diamond grit may achieve a finish equal in smoothness to the glazed surface.

Objective To investigate the Correlation between Plasma Total Homocysteine and Copper in Patients with coronary artery disease complicated with diabetes mellitus. Methods 31 pateints with coronary artery disease complicated with diabetes mellitus were studied. 36 age-matched and gender-matched patients with coronary artery disease not complicated with diabetes mellitus and 30 normal subjects were served as control groups. The concentrations of copper and total homocysteine was determined and compared. Objective To investigate the association of Lipoprotein(a) concentrations and patients with aortic dissection.Methods 27 pateints with aortic dissection were studied. Sixty age-matched and gender-matched patients with acute myocardial infarction and fifty normal subjects were served as control groups. Objective To compare the advantages and disadvantages of the amplification refractory mutation system -polymerase chain reaction (ARMS-PCR) with restriction fragment length polymorphism polymerase chain reaction(RFLP-PCR) technology, then explore its application in detection and analysis of single nucleotide polymorphism (SNP). Methods By using ARMS-PCR and RFLP-PCR the polymorphisms of the MMP-2 in cervical cancer and the normal-control group were analyzed. The research sites were-1306 C/T.The final result was verified by DNA sequencing. Results The results of amplified used ARMS-PCR showed that the genotypes of the SNP site in MMP-2 gene promoter were CC, CT, TT, and the DNA fragments were corresponding with the results of DNA sequencing. Conclusion ARMS-PCR technology has the advantages of lower cost time-saving simple operation and reliable results, while has a high value for research of SNP.

7.Color and opacity variations of veneer after bonding by resin cements and accelerated aging

Objective:To assess the effect of accelerated aging on the color and opacity of veneers bonded resin.Methods:28 discs were made with IPS e.max Press A1 shade with the thickness of 0.4 mm.Ceramic discs were divided into 4 groups(n=7). BEAUTIFIL Flow A1 ,DEO-Link SE Transparent,Panavia F Light color and Viriolink N transparent resin cements were applied on the porcelain discs with a thickness of 0.1 mm.Curing was performed with a calibrated LED curing-light for 40 seconds.Color and opac-ity differences of the porcelain substructures after 65 h(150 kJ/m2 )of UV ageing test were examined with a colorimeter.The results were analysed statistically with One-way ANOVA,LSD(L)test.Results:All samples showed significant changes in color and opaci-ty.The ΔE of all materials ranged from 0.6 to 2.29.DEO-Link SE showed the highest color change,while BEAUTIFIL Flow the lowest.The opacity changes ranged from 0.46 to 2.55 and its variation was not significant among groups except for Panavia F.Con-clusion:The accelerated ageing may lead to color changes of ceramic veneer bonded by resins in a clinically acceptable range(ΔE<3).
8.The expression of hypoxia-inducible factor-1α and HK-Ⅱ in esophageal squamous cell carcinoma and its effect in glycolysis

Objective To investigate the changes of hypoxia-inducible factor(HIF)-1α and hexokinase-Ⅱ(HK-Ⅱ)expression in human esophageal squamous cell carcinoma and its effect in glycolysis.Methods TE13 cells and Eca109 cells were cultured under hypoxic condition(1 %O2)for different hypoxic time(6,12,24 and 48 hours).Cells cultured under normal oxygen condition(20%O2)were set as control.The changes of HIF-1α and HK-Ⅱ expressions at protein level were detected by Western blot.HIF-1α genes were specifically silenced with RNA interference technology(RNAi),and then the changes of HIF-1α and HK-Ⅱ expression were determined by realtime PCR and Western blot.Under normal oxygen and hypoxic condition,the changes of lactic acid concentration in cell culture medium were detected by spectrophotometric method.Results Under hypoxic condition,the expression of HIF-1α and HK-Ⅱ gradually increased as hypoxic time extended(P<0.05),reached a peak at 12h and then gradually decreased as time extended.Compared with that under normal oxygen condition,the expression of HK-Ⅱ in TE13 cells and Eca109 cells significantly increased under hypoxic condition(P<0.05),which was more significant after 12 hours hypoxia.The result of realtime PCR indicated that under normal oxygen condition the expression of HIF-1α at RNA level in TE13/shRNA cells and Eca109/shRNA cells significantly decreased compared with TE13 cells and Eca109 cells without interference(P<0.05).The expression of HK-Ⅱ at RNA level was consistent with the result of HIF-1α.Under normal and hypoxia condition,the expression of HK-Ⅱ at protein level in TE13/shRNA cells and Eca109/shRNA cells significantly decreased compared with TE13 cells and Eca109 cells without interference,and the difference was statistic significant(P<0.05).The lactic acid secretion of TE13 cells and Eca109 cells under hypoxia condition(14.707 ± 3.594 and 15.062 ±3.901)was higher than that under normal oxygen condition(6.070±1.839 and 6.891±1.592,P<0.05).The lactic acid secretion of TE13/shRNA cells and Eca109/shRNA cells significantly decreased compared with TE13 cells and Eca109 cells without interference,and the difference was statistic significant(P<0.05).Conclusion The expressions of HIF-1α and HK-Ⅱ in human esophageal squamous cell carcinoma significantly increased under hypoxia conditions.The expression of HK-Ⅱ is closely correlated with lactic acid concentration and HIF-1α expression.HIF-1α may affect cell glycolysis through HK-Ⅱ.

Objective To investigate the feasibility and efficiency of unilateral pedicle screw combined with contralateral translaminar facet screw fixation by percutaneous and interbody fusion to treat low lumbar vertebra diseases. Methods Thirty patients with low lumbar vertebra diseases were entered into the study, including 8 males and 22 females with an average age of 53.7 years. All patients underwent discectomy, spinal canal decompression, cage implantation and lumbar fixation by unilateral pedicle screw combined with contralateral translaminar facet screw under gunsight guiding by percutaneous. Clinical outcomes were assed by JOA questionnaires before and after operation. Operative time, blood loss, and postoperative draiming were recorded. Radiological examination was obtained to assess position of translaminar facet screw.Results Mean operation time was 89 min with a blood loss of 285 ml. Position of translaminar facet screw grade Ⅰ were 24 cases, and grade 11 were 6. Mean follow-up was 22.5 months. 29 cases got bony fusion, and the fusion rate was 96.7%. There were no instability and evidence instrument failure during follow-up. The JOA grades improved from 13.0 preoperation to 25.2 at final follow-up, with the excellent and good rate of 72.5 %. Conclusion Unilateral pedicle screw combined with contralateral translaminar facet screw fixation by percutaneous and interbody fusion provide simple procedure, little trauma, forceful fixation, high fusion rate, and less complication, etc. Therefore, the surgical maneuver is a good choice for partial low lumbar vertebra diseases.
