1.CALCIUM IONOPHORE STRONGLY INHIBITS THE GENERATION OF EFFECTOR KILLERS FROM PROLIFERATIVE CYTOTOXIC T CELLS

The Ly2~+ Cortisone-Resistant Thymocytes(CRT)were cultured in mediumcontaining IL2,CAS and various mitogens for 7 days.The cytolytic activityof effector CTL was assayed by ~(111)In-release cytotoxicity.At the K/T ratio32/1,the cytotoxicity of effector CTL generated from the stimulus of ConA,PMA.Calcium ionophore(CaI)and PMA+CaI was 46%,24%,13% and 5%respectively.This was the first time that the Cal showed a stronger effecton the inhibition of the generation of effector CTL than that of PMA.The inhibition of PMA and CaI on the differentiation of CTL could bepartially reversed by the removal of PMA and CaI-containing medium andreplaced with cytokine-containing fresh medium.The cytolytic activity wasincreased by three fold,but still 2.4 fold lower than that stimulated byConA at the same K/T ratio. The critical cytokine in the replaced medium was IL2 for the differentia-tion of CTL.Supplementation of IFNr,IL4 or CAS could not further increasethe cytotoxicity.However,the cytolytic activity was slightly increased whenIFNr was added at the initiation of culture. Early T cells could be induced to generate antigen-specific cytotoxic Tcells when cultured in the system with our designed protocol,
2.Clinical observation on irinotecan as second-line treatment of patients with advanced colorectal carcinoma

Background and purpose:Colorectal carcinoma(CRC) is a major cause of cancer-related mortality. In the 1990s, several fi rst-line phase Ⅲ trails showed a signifi cant improvement in result with the addition of CPT-11(irinotecan, which is a specifi c inhibitor of topoisomerase Ⅰ) to FU-LV combination therapy(FOLFIRI).We observed the survival rate, effi cacy and adverse reaction of the combination chemotherapy of irinotecan plus folinic acid/continuous 5-? uorouracil bimonthly FOLFIRI regimen as second chemotherapy in treating advanced colorectal adenocarcinoma. Methods:Sixty patients with histologically proved advanced colorectal adenocarcinoma whose disease had progressed after treatment with first-line oxaliplatin or other chemotherapeutics agents were included to receive at least 2 cycles of chemotherapy with irinotecan 180 mg/m2 day 1 and folinic acid 200 mg/m2 iv,5-FU 400 mg/m2 iv bolus,days 1 and 2, 5-FU 600 mg/m2 iv infusion over 22 hours, days 1 and 2.Treatment was repeated every two weeks. Results:All patients were assessable for toxicity and 58 patients were evaluable for treatment response. The non-hematological toxicity was mild. Most was grade Ⅰ or Ⅱ. Only two patients experienced grade Ⅲ diarrhea and one patient experienced grade Ⅲ nausea and vomiting. There were no cases with grade Ⅳ toxicity. The most common hematological toxicity was neutropenia. Grade Ⅲ neutropenia were observed in fi ve patients. There was no case of febrile neutropenia. Based on intention to treat analysis, there were no complete responses (CR), 14(24.14%) partial response (PR), and 30 (51.72%) stable disease. the median time to disease progression was 6.09 months. The median overall survival was 9.65 months. Conclusions:Bimonthly irinotecan in combination with folinic acid and 5-fluorouracil was active with acceptable toxicities and a prolonged survival time in retreated colorectal cancer.

Objective To investigate the Correlation between Plasma Total Homocysteine and Copper in Patients with coronary artery disease complicated with diabetes mellitus. Methods 31 pateints with coronary artery disease complicated with diabetes mellitus were studied. 36 age-matched and gender-matched patients with coronary artery disease not complicated with diabetes mellitus and 30 normal subjects were served as control groups. The concentrations of copper and total homocysteine was determined and compared. Objective To investigate the association of Lipoprotein(a) concentrations and patients with aortic dissection.Methods 27 pateints with aortic dissection were studied. Sixty age-matched and gender-matched patients with acute myocardial infarction and fifty normal subjects were served as control groups. The levels of Serum lipoprotein(a), plasm C-reactive protein, white blood cell count were compared between the three groups. Observed the changes of the levels of lipoprotein(a), C-reactive protein, white blood cell count 24 hours, 72 hours and 5 days after the initial of disease in eight of the patients with aortic dissection. 6.Study of Quality Standard for Tianqi Granules

Objective To establish the quality standard for Tianqi Granule.Methods Radix Notoginseng in Tianqi Granule was identified by TLC.The ginsenoside Rg1 and notoginsenoide R1 in Tianqi were determined by HPLC.Result Radix Notoginseng could be identified by TLC.The linear ranges of ginsenoside Rg1 and notoginsenoide R1 were at 0.628~5.652 ?g(r =0.999 8)and 0.155~1.395 ?g(r =0.999 8),respectively.The average recoveries were 99.24%(RSD=0.84%)and 99.14%(RSD=1.13%).Conclusion The method is simple,accurate and with good reproducibility and high precision,and can be used for quality control of Tianqi Granule.
7.Quality standard for Capsicum Rheumatism Gel

AIM:To develop the quality standard for Capsicum Rheumatism Gel(Fructus Capsici extract,menthol,borneol). METHODS:HPLC was used to determine capsaicin in Capsaicum Rheumatism Gel.The se-(paration) was performed on zorbax C_(18) column with methanol-water-phosphoric acid (70∶30∶0.085) as a mobile phase.The flow rate was 0.8 mL/min,and the detection wavelength was at 280 nm.Menthol and borneol contents in Capsaicum Rheumatiosm Gel were determined by GC and naphthalene was adopted as internal standard substance. RESULTS:There was a good linear relationship for capsaicin at a range of 0.082 65-0.578 55 ?g and the average revovery was 97.86% with RSD=1.28%.There were good liner relationships for menthol and borneol at a range of 0.081 0-1.053 ?g,0.080 8-1.050 4 ?g and the average revovery was 98.668%,97.536%. CONCLUSION:The method proves to be simple,precise and reproducible and is suitable for quantitative control of Capsicum Rheumatism Gel.
8.The meta-analysis of Laurance-Moon-Bardet-Biedl syndrome.

Objective To deepen the clinicians' impression on the Laurance-Moon-Bardet-Biedl syndrome(LMBBS),we made a summary in the incidence and clinical manifestations of the disease in China comparing with foreigners.Methods We made evidence-based meta-analysis about the 2 cases reports in 1987 and 2003 in our hospital and literature review of 462 cases in foreign countries and 94 cases in China.Results LMBBS patients,with more frequency of consanguinity and family history,retinal dystrophy,mental retardation,obesity,polydactyly,hypogenitalism,often had many complicated and variable clinical manifestations.Conclusion To avoid intrafamiliy marriage would reduce the incidence rate.The diagnostic criteria and ascertainment methods introduced recently do benefit the early diagnosis in their childhood period.
9.Interleukin-6 is involved in the enhanced proliferation of HEK293 cells by the tumor specific antigen of HCA520

Objective: To investigate the effect of tumor specific antigen of HCA520 on the proliferation of HEK293 cells and to obtain some functional implications for HCA520. Methods: MTT assay was performed with HEK293 stable cell lines transfected with pcDNA3-HCA520-flag construct. Cytokines probably involved in HCA520 enhanced proliferation were screened by RT-PCR, and effect of these cytokines on the HEK293 cell proliferation was further confirmed by MTT assay. Results: HCA520 significantly promoted the proliferation of HEK293 cells, which was at least partially attributed to the up-regulation of IL-6 in HEK293 cells by HCA520. Conclusion: HCA520 might accelerate tumorigenesis by promoting proliferation of cancerous cells.
10.Studies on the characterization of a molecule expressed on murine thymic stromalcells and activated thymocytes

To identify the expression of the molecule recognized by Pf18-3 mAb (Pf18-3 molecule) on various cells. Meth-ods: The expression of pr18-3 molecule was assayed by flow cytometry and confocal laser scanning microscope . Result: The molecule recog-nized by Pf18-3 mAb expressed on TSC and other stromal cells. Whereas, fresh thymocytes were Pf18-3 negative. Interestingly, the expressionof Pf18-3 molecule was gradually up-regulated on thymocytes after activation by ConA. This molecule mainly expressed on CD4+ CD8+ andCD4+ CD8- cells. Under confocal laser scanning microscope, the staining of fluorescence showed as ring around the cell, it changed grsduallystronger and thicker with activation. Conclusion: This study indicated that the Pf18-3 molecule was co-expressive molecule of MTSC and acti-vated tlymocytes,it was concemed closely about the activation of CD4+ C D8+ and CD4+ CD8- cells.
