1.Study on the effect of triplex forming oligonucleotide on tumor growth and angiogenesis in rats with glioma

Objective To observe the inhibit effect of triplex forming oligonucleotide (TFO)(platelet-derived growth factor-B chain, PDGF) on tumor growth and angiogenesis in rats with glioma.Methods 1?10~6 C_6 glioma cells with high-flow microinfusion were seed into right caudate putamens of 18 rats by stereotaxic technique. TFO was injected in situ 1 week after glioma cells inoculation. Treat group Ⅰ and treat group Ⅱ received TFO at dose of 1.5 mg/20 ?l and 3.0 mg/20 ?l, respectively. The same doses were given again at 8, 11 and 14th day after glioma cells inoculation. The control group was treated with 20 ?l normal saline at same time like treat groups. Three weeks after glioma cells inoculation, all the rats were killed. The expressions of, PDGF-B, vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen (PCNA) were detected with microscopic histology.Results The inhibition rate of tumor growth was 66.0% in treat groupⅠand 92.2% in treat group Ⅱ. AIM: To analyze the role of complement system(CS)and its classical pathway in the pathogenesis of lacrimal gland benign lymphoepithelial lesions(LGBLEL).METHODS: The tissues of patients with LGBLEL or orbital cavernous hemangioma(CH)were collected. Proteomics analysis was used for the identification of different proteins. Reverse transcription-polymerase chain reaction(RT-PCR), immunohistochemical staining(IHC)and Western Blotting were employed to verify the changes of the differential proteins in CS signal pathway, in order to identify its role in the pathogenesis of LGBLEL.RESULTS: The results of proteomic analysis showed that the expression levels of proteins C3, C5, C9 and C1q in CS signal pathway in the lacrimal gland tissues of LGBLEL patients were all changed compared with those of orbital CH patients.The results of RT-PCR showed that the mRNA expression levels of C1qA, C5 and C9 in patients with LGBLEL were significantly higher than those patients with orbital CH. The results of IHC showed that the expression levels of C1qA, C3, C5 and C9 were significantly increased in patients with LGBLEL compared with those patients with orbital CH. The results of Western Blotting showed that the protein expression levels of C1qA, C3, and C9 were significantly increased in patients with LGBLEL compared with those patients with orbital CH.CONCLUSION: The CS has been shown to participate in the pathogenesis of LGBLEL and its classical pathway may be one of the pathways which plays a role.
3. Clinical Effect of Modified Huangqi Guizhi Wuwu Tang on Patient with Lower-extremity Arterial Disease

Objective:To observe the clinical effect of modified Huangqi Guizhi Wuwu Tang on patient with lower-extremity arterial disease (LEAD) with deficiency of Qi and Yin and blood stasis syndrome, and study the antioxidant mechanism. Method:One hundred and twenty-eight patients were randomly divided into control group (64 cases) and observation group (64 cases) by random number table. Patients in control group was treated for controlling blood sugar, blood pressure and blood fat, and got probucol in the morning and evening, 0.5 g/time, 2 times/days, aspirin enteric-coated tablets for 3 months, 100 mg·d-1, and alprostadil injection diluted with 10 mL normal saline for 15 days, 10 μg·d-1, 1 time/day. A course of the treatment in control group was 15 days, and there were 4 courses. In addition to the therapy of control group, patients in observation group were additionally given modified Huangqi Guizhi Wuwu Tang, 1 dose/day, for 3 months. Before and after treatment, ankle brachial index (ABI) and toe branch index (TBI) were detected, and internal diameter of dorsal artery of foot, peak velocity and blood flow were detected by color Doppler ultrasound. Main symptoms and sign were scored. And levels of interleukin-1 (IL-1), Homocysteine (Hcy), tumor necrosis factor-α (TNF-α), high sensitive C reactive protein (hs-CRP), cystatin C (CysC), malondialdehyde (MDA), superoxide dismutase (SOD) and oxidized low density lipoprotein (OX-LDL) were detected. Result:According to the rank test, clinical effect in observation group was better than that in control group (P<0.05). And levels of ABI, TBI and SOD were higher than those in control group (P<0.01). The ameliorate of internal diameter of dorsal artery of foot, peak velocity and blood flow were better than those in control group, and scores of intermittent postscript, lower extremity pain, dorsalis pedis artery pulsation, skin color, skin temperature, numbness of limbs, swelling of extremities and the sign and levels of IL-1, Hcy TNF-α, hs-CRP, Cys-C and Ox-LDL were lower than those in control group (P<0.01). Conclusion:Modified Huangqi Guizhi Wuwu Tang can relieve symptoms and signs, lower limb vascular function and hemodynamic, with certain anti-inflammatory effect and oxidative stress. It can also reduce vascular endothelial cell injury, and relieve and postpone the progress of LEAD
6.Three cases of occupational acute trimethyltin chloride poisoning.

Trimethyltin chloride is a highly toxic substance, which is absorbed through respiratory tract, skin and digestive tract, with central nervous system injury as the main clinical manifestations, and can be accompanied by damage to various organs. In this paper, the treatment process of 3 patients with acute trimethyltin chloride poisoning was reviewed, and their clinical manifestations, auxiliary examination, diagnosis and treatment were analyzed. Three patients were misdiagnosed as mental abnormality, encephalitis, and hepatic encephalopathy in different hospitals in the early stage of medical treatment, suggesting that clinicians should pay attention to the occupational contact history of poisoned patients and conduct toxicant detection in time to avoid misdiagnosis and mistreatment.
7.Clinical observation of minimally invasive scleral buckling combined with 25G in the treatment of rhegmatogenous retinal detachment

AIM: To observe the clinical effects of minimally invasive scleral buckling combined with 25G cannula intra-optical fiber lighting in the treatment of rhegmatogenous retinal detachment with the help of non-contact wide angle lens.
METHODS: We retrospectively analyzed 43 patients with rhegmatogenous retinal detachment from May 2011 to March 2015 in our hospital. The retinal tear locations of these patients analyzed preoperatively by pre-set lens and three-mirror contact-lens were uncertain. We detected the retinal tears intraoperatively by non-contact wide angle lens with the help of 25G cannula intra-optical fiber lighting. The tears were sealed by minimally invasive scleral buckling. The patients were followed up at 1wk, 1mo, 3mo and 6mo postoperatively. The vison and intraocular pressure were recorded with the same equipment and methods as preoperatively did. The retina reattachment and tear sealing status were also observed.
RESULTS: The retina were reattached by one operation in 41 patients and the reattachment rate was 95.3%. One patient suffered from incomplete retina reattachment, effusion under the retina, poor position of compressed retinal area, and succeeded by minimally invasive scleral buckling once again. One patient developed new retina tear and completely reattached by vitrectomy.
CONCLUSION:For those patients with uncertain retinal detachment preoperatively, minimally invasive scleral buckling combined with 25G cannula intra-optical fiber lighting could increase the success rate. The statistical analysis in large samples and the long-term complications should be further investigated.
8.Effects of endothelial progenitor cells on proliferation and biological function of hepatic stellate cells under shear stress.

OBJECTIVE:
To investigate the effects of endothelial progenitor cells (EPCs) under shear stress on the biological function such as proliferation, adhesion, migration, apoptosis and expression of α-smooth muscle actin (α-SMA), collagen-I and collagen-Ⅲ of hepatic stellate cells (HSCs).
METHODS:
HSCs and EPCs were inoculated into the upper and lower layers of the co-culture chamber respectively and co-incubated for 24 hours. Then, 12 dyne/cm shear stress was applied to EPCs cells for another 24 hours. After that, proliferation, adhesion, migration and apoptosis of HSCs were detected by cell counting kit-8 (CCK-8) kit, cell adherent assay, Boyden cell migration assay and flow cytometry respectively. Fluorescence quantitative PCR and Western blot were used to detect the mRNA and protein expression of alpha -SMA, collagen I and collagen-Ⅲ in HSCs.
RESULTS:
Under shear stress, EPCs ecological niche could obviously inhibit the proliferation, adhesion and migration of HSCs, promote the apoptosis of HSCs, and down-regulate the mRNA and protein expression of collagen-I, collagen-Ⅲ in HSC cells.
CONCLUSIONS
Under shear stress, EPCs ecological niche could inhibit the fibrosis development of HSCs to a certain extent.
