1.Research Progress of Mesenchymal Stem Cells and Their Exosomes on Tumors.

In China, malignant tumor is the main cause of death in both urban and rural areas. Mesenchymal stem cells (MSCs) have multidirectional differentiation potential, self-renewal ability and good immunomodulatory properties. Exosomes, as important paracrine substances of MSCs, mediate information exchange and transmission between cells in tumor microenvironment and influence the occurrence and development of tumors. Recently, conflicting findings have been reported on the effects of MSCs and their exosomes on tumors. On the one hand, MSCs and their exosomes are tumorigenic and can target specific sites to inhibit tumor growth; On the other hand, there is also evidence that MSCs could affect tumor growth and migration as part of the tumor microenvironment. In this paper, we will review the relationship between MSCs and exosomes and tumorgenesis and development, as well as how MSCs and exosomes play different roles in tumorgenesis and development, in order to provide beneficial help for tumor diagnosis, prognosis and precise treatment.

2.The study for the hemodynamics of expansive skin and the transplant time of skin flap designed by expansive skin.

OBJECTIVETo study the change rule of the hemodynamics of expanded skins and research the transplant time of random-pattern flaps that are made of the expanded skin.
METHODS72 ears of 64 rabbits were used as a subject of experiment. Four items of the contrasted skin (n = 8) and expanded skin (n = 8 x 8), Which include the microvessel diameter, the microvessel density, the blood velocity and the blood volume, were observed and measured by the microcirculatory imaging computer analysis system, the enzyme histochemical dyeing and the more functions colored pathological imaging computer analysis system. The length -width proportion and the largest area of the flaps that had made of the expansive skin and survived were measured.
RESULTS(1) The four items of expanded skin, that the microvessel diameter, the microvessel density, the blood velocity and the blood volume are 13.43 +/- 0.98 micrometer, 0.0472 +/- 0.0022, 1012.70 +/- 65.51 micrometer/s, 14.71 +/- 0.74 micrometer(3)/s respectively, are much bigger or quicker than those of contrasted skin [(7.22 +/- 0.71) micrometer, 0.0108 +/- 0.0002, (327.01 +/- 65.51) micrometer/s, (1.46 +/- 0.41) micrometer(3)/s] and there is the significant difference between both (6.49 < t < 49.49, P < 0.01). (2) The blood velocity increases in the expanding phases and always keeps on higher level. The other items all change periodically. Span of the period is four weeks. The microvessel diameter and the blood volume have a peak value at third weekend of every period and in the later period enhance more on the basis of the former period. The microvessel density changes in definite range in every period. (3) The surviving area of the random skin flaps is the largest which were made of the expanded skin in third weekend of every period, it changes more than those of other weekends in same period and later period is superior to the former period. There is a significant difference compared with contrasted random skin flaps (q = 3.7215, P < 0.05; q = 5.5026, P < 0.01).
CONCLUSIONThe hemodynamics of the expanded skin changes periodically in expansive course. The time of peak value of the blood volume is the best opportunity that random-pattern skin flaps are made of the expanded skin in every period.

3. Clinical Effect of Modified Huangqi Guizhi Wuwu Tang on Patient with Lower-extremity Arterial Disease

Objective:To observe the clinical effect of modified Huangqi Guizhi Wuwu Tang on patient with lower-extremity arterial disease (LEAD) with deficiency of Qi and Yin and blood stasis syndrome, and study the antioxidant mechanism. Method:One hundred and twenty-eight patients were randomly divided into control group (64 cases) and observation group (64 cases) by random number table. Patients in control group was treated for controlling blood sugar, blood pressure and blood fat, and got probucol in the morning and evening, 0.5 g/time, 2 times/days, aspirin enteric-coated tablets for 3 months, 100 mg·d-1, and alprostadil injection diluted with 10 mL normal saline for 15 days, 10 μg·d-1, 1 time/day. A course of the treatment in control group was 15 days, and there were 4 courses. In addition to the therapy of control group, patients in observation group were additionally given modified Huangqi Guizhi Wuwu Tang, 1 dose/day, for 3 months. Before and after treatment, ankle brachial index (ABI) and toe branch index (TBI) were detected, and internal diameter of dorsal artery of foot, peak velocity and blood flow were detected by color Doppler ultrasound. Main symptoms and sign were scored. And levels of interleukin-1 (IL-1), Homocysteine (Hcy), tumor necrosis factor-α (TNF-α), high sensitive C reactive protein (hs-CRP), cystatin C (CysC), malondialdehyde (MDA), superoxide dismutase (SOD) and oxidized low density lipoprotein (OX-LDL) were detected. Result:According to the rank test, clinical effect in observation group was better than that in control group (P<0.05). And levels of ABI, TBI and SOD were higher than those in control group (P<0.01). The ameliorate of internal diameter of dorsal artery of foot, peak velocity and blood flow were better than those in control group, and scores of intermittent postscript, lower extremity pain, dorsalis pedis artery pulsation, skin color, skin temperature, numbness of limbs, swelling of extremities and the sign and levels of IL-1, Hcy TNF-α, hs-CRP, Cys-C and Ox-LDL were lower than those in control group (P<0.01). Conclusion:Modified Huangqi Guizhi Wuwu Tang can relieve symptoms and signs, lower limb vascular function and hemodynamic, with certain anti-inflammatory effect and oxidative stress. It can also reduce vascular endothelial cell injury, and relieve and postpone the progress of LEAD
4.Fibroblast growth factor 23 expression in rat calvarial osteoblastic cells and its responses to hormones in vitro

Objective To investigate the expression patterns of fibroblast growth factor 23 (FGF23) in osteoblast and its responses to calcium, phosphate, exogenous PTH and 1,25(OH)_2D~3. Methods The primary rat calvarial osteoblasts were cultured in MEM medium which containing 10% FBS, then were harvested when cells were in half-confluence, confluence, osteoid deposition and osteoid mineralization stages respectively. The procedure was monitored under microscopy. Total RNA was extracted from cells according to the Trizol procedure. FGF23 mRNA levels were determined by Real-time PCR. Further, the confluent osteoblasts were treated with 3.2 mmol/L CaCl_2, 4.4 mmol/L β-glycerophosphate, 10~(-9) mol/L rhPTH(1-34) and 10~(-8) mol/L 1,25(OH)_2D_3 respectively for 3 days, and same volume of the medium was added as the control. The gene expressions were determined by Real-time PCR. Results FGF23 expression was transiently up-regulated at cell confluent stage and down-regulated after that. The FGF23 mRNA levels were 7.5-fold higher in confluent cells compared with that in half-confluent cells (P<0.001). The markedlly stimulating effect (about 16 times) on FGF23 expression was stimulated by exogenous 1,25(OH)_2D_3 treatment while no significant effect was found on FGF23 mRNA levels by CaCl_2,β-glycerophosphate, and rhPTH(1-34) treatments when compared with the control. Conclusions The FGF23 expression in osteoblast is developmental stage-related and its powerful stimulator is 1,25(OH)_2D.
5.Synthesis and Antihypertensive Activity of 1-Phenylethyl(or 3,4-Dimethyloxyphenylethyl)-4-substituted Aryloxypiperidines

AIM The purpose is to make a search for new antihypertensive agents with higher activity and lower side effect. METHODS As DDPH a lead compound, ten 1-Phenylethyl (or 3,4-dimethyloxyphenylethyl)-4-substituted aryloxypiperidine compounds were designed and synthesized, keeping the two structural fragments of DDPH: phenylethylamine and aryloxyalkylamine. Their structures were confirmed by Elemental Analysis, IR, 1H-NMR and Mass Spectrum. RESULTS The majority of the compounds possessed potent antihypertensive activity and the antihypertensive effect of I3 and I4 was stronger than that of DDPH. CONCLUSION The experimental results were accordant with designed thought and provided useful information for modifying the structure of DDPH deeply.
7.Effect of chromodomain helicase/ATPase DNA binding protein 1-like gene on the invasion and metastasis of tongue squamous cell carcinoma CAL27 cells.

OBJECTIVES:
A study was conducted to investigate the molecular mechanism of chromodomain helicase/ATPase DNA binding protein 1-like gene (CHD1L) influencing the invasion and metastasis of tongue squamous cell carcinoma and to provide a new target for clinical inhibition of invasion and metastasis of tongue squamous cell carcinoma.
METHODS:
Ualcan website was used to analyze the expression of CHD1L in normal epithelial tissue and primary head and neck squamous cell carcinoma and to analyze the effect of lymph node metastasis on the expression of CHD1L in tissues with head and neck squamous cell carcinoma. The relationship between CHD1L expression and the survival rate of patients with head and neck squamous cell carcinoma was tested by the GEPIA website. Western blot was used to quantify the levels of CHD1L protein in human tongue squamous cell carcinoma CAL27 and immortalized human skin keratinocyte cell HaCaT. After knocking down CAL27 in human tongue squamous cell carcinoma cells with an RNA interference plasmid, the cells were designated as SiCHD1L/CAL27 and Scr/CAL27. Western blot was utilized to detect the expression of CHD1L in each group of cells. The change in CAL27 cell proliferation ability was tested by EdU proliferation test after CHD1L knockdown. The change of cell migration ability of each group cells was tested through the wound healing assay. Western blot was used to detect epithelial-mesenchymal transition (EMT) marker E-cadherin and Vimentin protein expression levels.
RESULTS:
Ualcan database showed that the expression of CHD1L in primary head and neck squamous cell carcinoma tissues was higher than in normal epithelial tissues and in head and neck squamous cell carcinoma tissues with lymph node metastasis. GEPIA website analysis showed that the overall survival rate of patients with head and neck squamous cell carcinoma with high expression of CHD1L was significantly lower than that of patients with low expression. Western blot results showed that CHD1L expression in human tongue squamous carcinoma cells CAL27 was higher than that of human normal skin cells HaCaT. CHD1L expression in SiCHD1L/CAL27 cells was much lower than that in Scr/CAL27 cells. Results of EdU proliferation experiments showed the significant reduction in the cell proliferation ability of the SiCHD1L/CAL27 cells. Results of the wound healing experiments showed the reduction in the migration capacity of the SiCHD1L/CAL27 cells. The expression of E-cadherin increased, whereas that of Vimentin decreased, in SiCHD1L/CAL27 cells.
CONCLUSIONS
CHD1L promoted the EMT, proliferation, migration, and invasion ability of tongue squamous cell carcinoma cells.

8.Study on correlation between relative density and kinematical viscosity of concentration for Yuxianling granules.

OBJECTIVETo study the relation between relative density and kinematical viscosity of the concentration for Yuxianling granules.
METHODThe relative density and kinematical viscosity by alkali burette of the concentration were investigated. The connection between kinematical viscosity and spray drying, also and temperature of the concentration was respected. In the meantime, different mathematical models were simulated.
RESULTKinematical viscosity is susceptible than relative density to the effect for spray drying, and the mathematical model is also set up accordingly. The result can offer the proper inlet temperature and kinematical viscosity for spray drying. CONCLUSIon The method in the experiment is simple, practical and manipulate easily. The study will provide the basis for extracted solution of compression and spray drying.

9. A comparative study of simultaneous and delayed autologous lipofilling with rhytidectomy in facial rejuvenation

Objective:
To discuss and analyze the respective clinical effects of autologous fat filling for facial rejuvenation in the same period of three months after incisions.
Methods:
From June 2015 to January 2018, 117 patients with facial rejuvenation admitted to the Plastic Surgery Hospital of Weifang Medical College were randomly divided into A, B and C groups. Group A (39 cases, male 3 cases, female 36 cases, age 44-67 years old) received simple rhytidectomy. Group B (39 cases, male 4 cases, female 35 cases, age 45-67 years old) underwent rhytidectomy and autologous fat filling at the same time. Group C (39 cases, male 3 cases, female 36 cases, age 45-65 years old) received autologous fat filling 3 months after rhytidectomy. We compared the improvement of facial depression (nasolabial groove and mouth angle), the effect of facial rejuvenation after fat filling in three groups and the incidence of complications in group B and C.
Results:
15 cases in group A, 12 cases in group B and 2 cases in group C had no obvious improvement in facial atrophy, the remaining 88 cases had obvious improvement compared with before operation, and achieved the effect of facial rejuvenation. The incidence of complications in group C (7.69%) was significantly lower than that in group B (35.90%), the excellent or good rate of facial improvement and the nasolabial groove and peri-oral depression improvement in group C (87.18%, 92.31%) was significantly higher than that in group B (64.10%, 64.10%) and group A (33.33%, 17.95%) (
10. Biological characteristics of synovial-derived mesenchymal stem cells in serum-free culture media and in vitro construction of three-dimensional cartilage in vitro

Objective:
To explore the biological characteristics of synovial fluid-derived mesenchymal stem cells (SF-MSCs) cultured in serum-free medium and the ability of in vitro reconstruction of three-dimensional cartilage combined with scaffold material.
Methods:
Human SF-MSCs were cultured in serum medium and mesenchymal stem cells medium-serum free (MSCM-sf) respectively, then the proliferative ability and morphology of SF-MSCs were compared; The third passage SF-MSCs cultured in MSCM-sf were identified by flow cytometry, three-way(chondrogenic, osteogenic, adipogenic)differentiation assay and induced for chondrogenic differentiation when combined with polyglycolic acid/polylactic acid (PGA/PLA).
Results:
SF-MSCs cultured in MSCM-sf had better morphology and proliferative ability than that cultured in serum medium. The expression levels of positive markers of the third passage SF-MSCs cultured in MSCM-sf, such as CD73 (99.5%), CD90 (98.9%) and CD105 (96.5%), were more than 95%. However, the overall negative markers (CD34, HLA-DR and CD11b) expressed less than 2%. Three-way differentiation staining was positive. The combination of SF-MSCs and PGA / PLA can be induced into cartilage in vitro.
Conclusions
SF-MSCs cultured in MSCM-sf can be amplified under the condition of maintaining the stem cell characteristics, and can be combined with PGA/PLA scaffold to construct three-dimensional cartilage in vitro.
