Objective To study the mutants of pre C/C gene and the base core premotor (BCP) in chronic hepatitis B patients carrying or not carrying hepatitis B e antigen (HBeAg). Method The HBV DNA of BCP and PreC/C ware amplified by PCR from serum samples of patients with chronic hepatitis B, and the products were cloned into T vector and sequenced. Results Of BCP variants, the double mutations of T1762/ A1764 were found in 20 of 42 patients with chronic hepatitis, and the prevalence was significantly lower in HBeAg positive patients (7/20) than in HBeAg negative patients (13/22) (P
2.Mental health states of patients with recurrent chest pain after percutaneous coronary intervention and coronary artery bypass grafting

BACKGROUND:Many researchers appealed that coronary atherosclerotic heart disease patients were medical treated while special psychological intervention should be integrated into the rehabilitation plans (biology-psychology-social treatment) for coronary atherosclerotic heart disease patients.Pertinence psychological intervention would be processed after finding out mental health state of coronary atherosclerotic heart disease patients.OBJECTIVE:To make principal component analysis for mental health state of coronary atherosclerotic heart disease patients with relapsing pain in chest after percutaneous coronary intervention (PCI) and coronary artery bypass grafting (CABG) by questionnaire investigation,and to find out the primary factors.METHODS:A total of 40 coronary atherosclerotic heart disease patients with relapsing pain in chest after PCI and CABG were selected,(25 males and 15 females),mean age of (60±10) years old.It was used that the mental health section of the integrative system for diagnosing and assessing health state in Chinese population,including body symptoms,positive emotion,negative emotion,cognitive ability,behavior trouble,self-evaluation and social adapting problem.RESULTS AND CONCLUSION:Totally 4 principle component factors which the accumulative contribution rate reached to 83.89% were extracted.The first principle component factors which the contribution rate reached to 40% mainly demonstrated behavior trouble and social adapting problem.The second one mainly demonstrated positive emotion and negative emotion,the third for cognitive ability and self-evaluation,and the fourth for body symptoms.The behavior trouble and social adapting problem of coronary atheroscleroticheart disease patients with relapsing pain in chest after PCI and CABG must be paid great attention,and positive emotion would be leaded,negative emotion would be lowered.
3.Risk factors and prevention strategies of inguinal hernia after radical prostatectomy

Inguinal hernia (IH) is a common complication in prostate cancer patients undergoing radical prostatectomy (RP), which seriously affects their quality of life.The repair of IH is often challenging due to the adhesion in the inguinal area caused by previous surgery.This article focuses on the risk factors for IH after RP, including surgical approach, postoperative anatomical changes, low body mass index (BMI), muscle loss, advanced age, international prostate symptom score (IPSS), and patent sheath process, and prevention strategies for IH after RP, including robot assisted laparoscopic radical prostatectomy (RARP) with IH repair, preservation of the posterior pubic space in RARP, treatment of the spermatic cord, extraperitoneal RARP, and intraoperative management of the sheath like process, aiming to provide reference for urologists and to improve patients' quality of life.

5.Establishment of a novel co-cultured liver cancer model based on tumor microenvironment.

Objective: To construct a new co-cultured liver cancer research model composed of activated hepatic stellate cells (aHSC) and liver cancer cells, explore the efficacy difference between it and traditional model, so as to establish a liver cancer research model in vitro and in vivo that can reflect the real clinical efficacy. Methods: A new co-culture model of liver cancer consisting of aHSC and liver cancer cells was constructed. The differences in efficacy between the new co-culture model and the traditional single cell model were compared by cytotoxicity test, cell migration test, drug retention test and in vivo tumor inhibition test. Western blot was used to detect the drug-resistant protein P-gp and epithelial-mesenchymal transition-related proteins. Masson staining was used to observe the deposition of collagen fibers in tumor tissues of tumor-bearing mice. CD31 immunohistochemical staining was used to observe the microvessel density in tumor tissues of tumor-bearing mice. Results: The cytotoxicity of single cell model and co-culture model was dose-dependent. With the increase of curcumin (CUR) concentration, the cell viability decreased, but the cell viability of single cell model decreased faster than that of co-culture model. When the concentration of CUR was 10 μg/ml, the cell viability of the co-culture model was 62.3% and the migration rate was (28.05±3.68)%, which were higher than those of the single cell model [38.5% and (14.91±5.92)%, both P<0.05]. Western blot analysis showed that the expressions of P-gp and vimentin were up-regulated in the co-culture model, which were 1.55 and 2.04 fold changes of the single cell model, respectively. The expression of E-cadherin was down-regulated, and the expression level of E-cadherin in the single cell model was 1.17 fold changes of the co-culture model. Drug retention experiment showed that the co-culture model could promote drug efflux and reduce drug retention. In vivo tumor inhibition experiment showed that the m-HSC+ H22 co-transplantation model had faster tumor growth and larger tumor volume than those of the H22 single cell transplantation model. After CUR treatment, the tumor growths of m-HSC+ H22 co-transplantation model and H22 single cell transplantation model were inhibited. Masson staining showed that the deposition of collagen fibers in tumor tissues of m-HSC+ H22 co-transplantation model mice was more than that of H22 single cell transplantation model. CD31 immunohistochemical staining showed that the microvessel density in tumor tissue of m-HSC+ H22 co-transplantation model was higher than that of H22 single cell transplantation model. Conclusions: The aHSC+ liver cancer cell co-culture model has strong proliferation and metastasis ability and is easy to be resistant to drugs. It is a new type of liver cancer treatment research model superior to the traditional single cell model.

7.Expression of carbonic anhydrase IV in rabbit corneal endothelial cells.

OBJECTIVETo demonstrate the molecular expression of carbonic anhydrase IV (CA IV) in rabbit corneal endothelium.
METHODSReverse transcriptase polymerase chain reaction (RT-PCR) was performed using cultured and fresh rabbit corneal endothelial total RNA and specific primers for CA IV. The RT-PCR product was subcloned and sequenced. Immunoblotting and indirect immunofluorescence staining were performed to detect protein expression and distribution of CA IV using fresh and cultured rabbit corneal endothelium and rat anti-CA IV polyclonal antibody.
RESULTSRT-PCR screening gave positive bands at the predicted size for CA IV from fresh and cultured rabbit corneal endothelium. Sequencing further confirmed the identity of CA IV in corneal endothelium. Immunoblotting analysis showed a single band at 52 kDa for freshly isolated and cultured endothelial cells. Indirect immunofluorescence staining revealed an apparent positive staining in cultured endothelial cells.
CONCLUSIONCarbonic anhydrase IV is expressed in rabbit corneal endothelium, which could contribute to the transendothelial HCO(3)(-) flux that is necessary to maintain corneal hydration and transparency.

8.Effects of EGb761 on hippocamal synaptic plasticity of vascular dementia rats.

AIMTo investigate the effects of ginkgo biloba extract 761 (EGb761) on synaptic plasticity in hippocampus of vascular dementia (VD) rats.
METHODSThe escape latency (EL) of Morris water maze (MWM) task was measured at different time points (4 W, 8 W and 16 W), and the population spikes (PS) of granule cell layer in the dentate gyrus were induced by single pulse stimulation to perfo rate path fibers before and after high frequency stimulation (HFS) in vivo.
RESULTSMWM test showed that the escape latency (EL) of VD model group were highly longer than that of the sham-operated group, while the EL of EGb761-treated group was significantly shorter than that of model group, but still longer than that of the sham-operated group. The incidence rates of LTP induction in 1 m, 2 m and 4 m subgroups of model group were significantly lower than that of sham-operated group and EGb761-treated group at different time point. The relative amplitudes of PS after HFS in 1 m, 2 m and 4 m subgroups of model group were obviously reduced compared with that of the corresponding subgroups of sham-operated group and EGb761-treated group. There was no obvious significance in the peak latency of PS between different subgroups and different LTP-tested time point.
CONCLUSIONVD model rats had apparent and long-lasting dysfunction of learning and memory, EGb761 could accelerate the recovery of the pathological synaptic plasticity. This suggested that EGb761 played an important and improving role on learning and memory dysfunction of VD.

9.Antinociceptive Effect of Najanalgesin from Naja Naja Atra in a Neuropathic Pain Model via Inhibition of c-Jun NH2-terminal Kinase.

BACKGROUNDNajanalgesin, a toxin isolated from the venom of Naja naja atra, has been shown to exert significant analgesic effects in a neuropathic pain model in rats. However, the molecular mechanism underlying this protective effect of najanalgesin is poorly understood. The present study sought to evaluate the intracellular signaling pathways that are involved in the antinociceptive effect of najanalgesin on neuropathic pain.
METHODSThe antinociceptive properties of najanalgesin were tested in hind paw withdrawal thresholds in response to mechanical stimulation. We analyzed the participation of the mitogen-activated protein kinase p38, extracellular-regulated kinase (ERK), and c-Jun N-terminal kinase (JNK) by western blot analysis. This inhibition of JNK was confirmed by immunohistochemistry.
RESULTSThe phosphorylation levels of JNK (as well as its downstream molecule c-Jun), p38, and ERK were significantly increased after injury. Najanalgesin only inhibited JNK and c-Jun phosphorylation but had no effect on either ERK or p38. This inhibition of JNK was confirmed by immunohistochemistry, which suggested that the antinociceptive effect of najanalgesin on spinal nerve ligation-induced neuropathic pain in rats is associated with JNK activation in the spinal cord.
CONCLUSIONThe antinociceptive effect of najanalgesin functions by inhibiting the JNK in a neuropathic pain model.

10.Application of high fibular osteotomy in the treatment of knee osteoarthritis.

OBJECTIVETo evaluate the short-and mid-term curative effect of high fibular osteotomy on knee osteoarthritis.
METHODSFrom October 2014 to July 2016, 76 patients with knee osteoarthritis were treated with high fibular osteotomy. There were 22 males and 54 females, ranging in age from 47 to 82 years old, with an average of 61.62 years old. Internal tenderness of the knee joint was examined preoperatively. Preoperative examination showed medial knee pain and medial positive McMurray sign. Preoperative AP and lateral X-ray films of the weight-bearing side of the knee showed the narrowing of the medial space. The knee medial space, femoral and tibial angle, VAS pain score and the American Knee Society(AKS) score were evaluated after operation.
RESULTSCompared with the preoperative one, the postoperative knee medial gap was widened obviously. The femur and tibia angle after operation was bigger than that before surgery. The VAS was improved from preoperative 5.70±1.56, to 3.70±1.03 at the 1st week after operation, 3.20±0.95 at the 1st month after operation, 2.35±0.99 at the 3rd month after operation, and 2.10±0.97 at the 1st year after operation. According to the AKS score, the preoperative pain score of knee joint was 14.45±1.76, the activity score of knee joint was 12.60±1.98, and the stability score was 12.15±1.72; after operation, the above scores were improved to 42.60±2.28, 21.80±2.14, and 20.85±2.16 respectively.
CONCLUSIONSThe high fibular osteotomy with less complications is simple. It can effectively alleviate the pain of the knee joint and improve the function of knee joint, and the clinical effect is satisfactory.
