1.Correlation of the expression of Noxa with hepatocellular carcinoma

Purpose To explore the correlation of Noxa gene with clinical pathology and its effect on proliferation and apoptosis of HepG2 cells.Methods Noxa protein in 100 hepatocellular carcinoma tissues and 100 paracancerous tissues were detected by imnunohistochemical technique,clinicopathological parameters and follow-up data were statistically analyzed.The recombinant eukaryotic expression plasmid pIRES2-EGFP-Noxa was transiently transfected into HepG2 cells with lipofectamine.Both Noxa mRNA and protein were detected by RT-PCR and Western blot respectively.The inhibition of cell proliferation was evaluated by MTT assay.The cell apoptosis was detected by flow cytometry (FCM).Results Immunohistochemistry showed that in tumor tissue of hepatocellular carcinona the positive rate of Noxa protein expression was 50%,while it was 78% in cancer adjacent normal mucosal tissue,with statistically significant difference (P < 0.05).The expression of Noxa was related to TNM staging and the tumor differentiation.Exotic Noxa gene was expressed successfully in HepG2 cells after transfected with lipofectamine.The expression of Noxa mRNA and protein of HepG2 cells was significantly up-regulated after transfected 24 h,48 h and 72 h (P < 0.05).MTT assay showed that the Noxa expression inhibited HepG2 cell proliferation in a time-dependent manner for 24 h,48 h and 72 h (P < 0.05),which was significantly different from that of the control group (P < 0.05).Apoptosis rate after transfected 24 h,48 h and 72 h was (15.5 ± 0.9)%,(24.6 ±0.8) % and (35.4 ±0.7) %,respectively.There were significant difference between Noxa and the control group (P < 0.05).Conclusion The expression of Noxa in hepatocellular carcinoma tissue is closely related to TNM stage and the tumor differentiation.Noxa gene overexpression is able to effectively inhibit the proliferation and promote the apoptosis of HepG2 cell line.
2.Advancement in researches of the effects of perioperative analgesia on immune function of cancer patients

Perioperative immunosuppression exists in cancer patients are as a result of their own disease,and postoperative pain inhibit immunological function.The effective postoperative analgesia can relieve the suppression of cell-mediated immunity,as well as reducing tumor recurrence and metastasis.However,diverse range of analgesic agents and techniques have a different impact on immune function.This article reviews the influence of various analgesic agents and techniques on the perioperative immune function of cancer patients so as to provide more suitable analgesia techniques,which is beneficial for regulating of immune balance,lessening tumor recurrence and improving prognosis.
4.Surgical treatment of severe acute cholangitis in aged patients

Objective To analyze the characteristics and therapeutic effects in the elderly patients with acute eholangitis of severe type(ACST) Methods The clinical data of 60 cases of elderly patients with ACST. Results The cured 44 cases(73.3% )and 16 deaths(26.7% ) ,the cause of death:septic shock four cases (25.0%) ,3 cases of acute renal failure(18.8%) ,ARDS four cases(25.0% ) ,multi - organ failure five cases(31.3% ). Incidence surgery within 24 h the deaths of three cases( 18.8% ) ,significantly less than 24 h after surgery incidence death in 13 cases (81.3%) ( X2= 7.72, P<0.01 );correct preoperative shock has been 40 cases, six cases of death ( 15.0% ) was significantly lower than the preoperative shock not corrected in 20 cases,death in 10 patients(50.0% ) ( X2=6.75 ,P <0.01 ). Conclusion The cure rate was improved from early diagnosis and perioperative management in the elderly patients with acute cholangitis of severe type.
5.Expression and implication of P-gp in colorectal cancer

Objective To evaluate the expression and significance of P-glycoprotein(P-gp) in colorectal carcinoma. Methods The expression patterns of P-gp in 48 patients with colorectal carcinoma were examined by immunohistochemistry SP mothod and were analyzed its correlation to clinic opathological features. Results The expression of P-gp was related to lympy node metastasis. Its expression was not related to sex and age of the patients, localization of the primary tumor, tumorsize, histological grade, localinvasion, distant metastasis and Duke's stage. Conclusion Detecting the expression of P-gp might play an important role in chemotherapy strategy of colorectal carcinoma.
6.THE SCAVENGING ROLE OF OXYGEN FREE RADICALS BY SALVIA MILTIORRHIZA

Using Xanthine-Xanthine Oxidase system to generate superoxide ani-on, using H2O2 -Fe2 + system to generate hydroxyl radical, the role of scavenging oxygen free radical by Miltiorrhiza was studied by electron paramagnetic resonance and spin trapping. The result suggested that the scavenging rate of hydroxyl radical by Salvia Miltiorrhiza injection is 65%, and that of superoxide anion is 100%. This result suggests that scavenging cytotoxic oxygen free radicals may be one of important pharmacological mechanism of Salvia Miltiorrhiza.
7.Usefulness of CT in Diagnosis of Acute Focal Bacterial Nephritis

ObjectiveTo evaluate the value of CT in diagnosis and discrimination of acute focal bacterial nephritis(AFBN). Methods 6 cases of AFBN were reported which were all performed with ultrasound and CT scan.4 of 6 cases were observed by delay contrast enhanced CT examination. ResultsCT scan showed wedge-shaped masses with thickened Gerota fascia.The delayed contrast enhanced CT revealed more clear shapes and verges of the lesions than immediate contrast enhanced CT. ConclusionCT scan is of significant value in diagnosis and discrimination of AFBN with overall review of documents and statistic analysis.
8.DNA methylation and hepatocellular carcinoma

Hepatocellular carcinoma is one of the most common cancer with a high incidence and mortality,representing a main type of primary liver cancer.However,the molecular and cellular mechanisms of hepatocellular carcinoma pathogenesis are still poorly understood.Traditionally,the development of hepatocellular carcinoma has been viewed as a process of transforming of normal cells into malignant driven by the genetic alterations in tumor-suppressor gene deactivation and pro-oncogene activation.In recent years,with the deeper understanding of tumor,it has been found that epigenetic alterations are closely related to the occurrence and the development of hepatocellular carcinoma.DNA methylation is one of the most common epigenetic events occurring in human genome,as well as the best studied of the epigenetic changes.This review focuses on the state-of-the-art advancements of DNA methylation in hepatocellular carcinoma.
9.Chimeric antigen receptor T cell based therapeutic modality in solid tumors

The CD19-targeted chimeric antigen receptor (CAR) T-cell treatment has achieved clinical success in treating B-cell lineage hematopoietic malignancies. This accomplishment involved the precise and efficient elimination of tumor cells by tumor-associated an-tigen-redirected immune cells. As a result, the reinitiation of several CAR T-cell (CART) based clinical trials in solid tumors was promot-ed. However, developing a feasible and efficient CAR T based therapeutic modality for solid tumors is urgently needed given the differ-ential properties between liquid and solid tumors. In this review, we discuss the advances in the management of cytotherapeutic mo-dality for solid tumors. The aspects considered include toxicity management, target selection, and primer or conditioning treatment.
