3.Effect of propofol on N-methyl-D-aspartate subtype of glutamate receptor in cultured rat hippocampal neurons

Objective To investigate the effect of propofol on N-methyl-D-aspartate (NMDA) receptor-mediated currents. Methods Hippocampal neurons were prepared from newborn Wistar rats and cultured for 8-12 days. Whole cell currents were recorded using patch-clamp technique and cells were voltaged-clamped at - 80 mV. 100 ?mol?L-1 NMDA and 3 or 48 ?g?ml-1 propofol were applied with a multi-pipe ejection system. GABAA receptor was then blocked with 100 ?mol?L-1 bicuculline to investigate the effect of propofol on NMDA receptor without the influence of GABAA receptor.Results Run-down of INMDA induced by 100 ?mol?L-1 NMDA applied to neurons which were cultured for 8-12 days was 15%?8%. Propofol 3 or 48 ?g?ml-1 significantly inhibited spontaneous excitatory postsynaptic currents and elicited a Cl- -mediated response by direct activation of GABAA receptor in the absence of GABA. therefore produced a reversible inhibition of whole cell currents activated by NMDA. After the GABAA receptor was blocked by 100 ?mol?L-1 bicuculine propofol still inhibited NMDA currents slightly. Conclusion Propofol inhibits the NMDA subtype of glutamate receptor mainly through activation of GABAA receptor. It can also directly suppress the NMDA receptor slightly.

Aim To investigate the protective effects of etomidate on cultured rat hippocampal neurons subjected to anoxia injury and its mechanism.Methods Hippocampal neurons of neonatal rat,which had been cultured in vitro for 10 days,were allocated to control groups and etomidate-treating groups.The neurons were exposed to oxygen-glucose deprivation for 24 h.The cell survival rate in each group was evaluated using MTT colorimetry.To explore the effect of etomidate on neuronal calcium overload evoked by anoxia or 50 mmol?L~(-1) KCl or 1 mmol?L~(-1) glutamate,fluo-3,a fluorescent probe,was used for imaging of intracellular calcium in laser scanning confocal microscope(LSCM)to measure real-time changes of [Ca~(2+)]_i in cultured rat hippocampal neurons.Results The hippocampal neurons developed acute neuronal swelling and widespread neuronal degeneration following anoxia for 24 h.Etomidate at concentrations of 1.2~4.8 mg?L~(-1) attenuated the neuronal injury in a dose-dependent manner(P

miRNA ( microRNA) is a small molecule, which is composed of about 20 nucleotides, and has been a hot research topic in recent years.Many studies indicated that the expression level of miRNA in tumor changed, which played a role in the differentiation and proliferation of tumor cells, as a function of oncogene or tumor suppressor gene.The occurrence and development of colon cancer is related to the abnormal expression of many genes, and the data shows that the abnormal expression of miRNA is closely related to the development of colon cancer.In this paper, the influence of miRNAs on the development of colon cancer and its relevant mechanism were collected, then the colon cancer occurrence, development and miRNA molecular mechanisms were studied, which provided a theoretical basis for the molecular target treatment of colon cancer.
6.The experimental study on the expression of Bcl-2 and Fas-L after trauma brain injury in rats

Objective To observe the alteration of Bcl-2 and Fas-L at various intervals after traumatic brain injury and study the relationship between the alteration and the post-injury interval. Method The rat brain contusion was incurred by falling impact injury, paraffin section was cut after the test group rats were killed after various survival interval and stained with antibody of Bcl-2 and Fas-L, the hemotoxylin and eosin (HE) staining was carried out meanwhile. The staining results were measured quantitatively with computer imaging analysis system. Results Positive staining nerve celis were observed around the contusion area. At 30min after injury, a few Bcl-2 could be found around the wound, the intensity and the quantity of Bcl-2 positive celis increased significantly as post-injury interval extended. At 4h, the intensity came to a peak. Then the staining decreased. Although some Fas-L positive staining celis could be found around the wound at 30min after injury, the staining increased insignificantly from Ih to 4h after the injury. After 4h, the Fas-L positive staining cell increased significantly both in intensity and in quantity as post-injury interval extended. Conclusion There is a rule that the expression of Bcl-2 and Fas-L alters along with post-injury interval extension, which will be of value in time estimation of brain injury.
7.Proton MR spectroscopy of hepatocellular carcinoma patients at 1.5 Tesla

Background and purpose: Magnetic resonance spectroscopy is currently the only non-invasive magnetic resonance imaging technique which can detect metabolite composition of live organ and tissue and also conduct semi-quantitative or quantitative analysis. We analyzed the characteristics of metabolic changes among normal liver, cirrhosis regenerative nodule and liver cancer on the background of cirrhosis by voxel proton MR spectroscopy at 1.5 Tesla. Methods: 20 healthy volunteers (control group), 25 cirrhosis (cirrhosis group) and 23 hepatocellular carcinoma after cirrhosis (HCC group) patients were our candidates. The characteristics of spectra of ~1H-MRS were observed. The height and ratio of choline complex (Cbo) and lipid methylene (Lip2) were recorded and the results were compared among the groups. Results: The success rate of ~1H-MRS was about 88.2% (60/68). There were statistically significant differences among the heights of Lip2 peak of the three groups, and also between that of the control group and the group of liver cancer. But there were no significant differences between that of control group and cirrhosis group, cirrhosis group and liver cancer group. The comparison of the height of Cho peak and the ratio of Cho/Lip2 showed statistically significances both among the three groups and between each other. Conclusion: The metabolic changes of the liver could be shown non-invasively by ~1H-MRS, which could be useful for differential diagnosis or diagnosis of cirrhosis regenerative nodule and HCC.
8.Retroperitoneal fibrosis: the clinical and radiological manifestation

Objective To analyze the clinical and radiological features of retroperitoneal fibrosis (RPF), and to deepen the understanding of this unusual disease and improve the diagnostic level at the early stage. Methods Fourteen cases (10 males and 4 females, mean age 45.8 years) of pathologically diagnosed RPF from January 1990 to June 2004 were summarized. The clinical and radiological performance of the cases were analyzed. All patients received non-contrast CT scanning, 10 of them underwent enhanced CT scanning as well. 8 patients received MRI, 10 patients received IVP examination, and 11 received B-ultrasound. Results (1)The very first symptoms usually included back pain, bellyache (10 cases), or urinary tract obstruction (3 cases), with increase of ESR, IgG, CRP value and abnormal renal function.(2)The result of radiological examination showed that 11 lesions of the 14 cases located at retroperitoneum. Ten cases were mass type and 4 cases were diffuse type. Non-contrast CT scanning revealed soft tissue mass at retroperitoneum with inhomogenous or homogenous density. After contrast medium injection the lesions were enhanced with different extent. MRI results showed that the lesions presented low signal in T_1WI, while in T_2WI the signals had no obvious coherence but were different from one case to another. Conclusion Radiological examination is one of the important methods for diagnosis of RPF. Based on the different characteristics of RPF in CT and MRI, together with the clinical findings, we will get valuable references for staging and follow-up of RPF.
9.CT and MRI appearances and the causes of misdiagnosis of hepatic adenoma

Objective To retrospectively analyze CT and MR appearances of hepatic adenoma,and to study the causes of misdiagnosis with review of relative literatures.Methods CT and MRI data of 8 patients with pathologically confirmed hepatic adenoma were retrospectively analyzed.Pre-and post-contrast tri-phase (arterial,portal venous and delayed) CT scans and routine MR .scan were performed in all 8 patients,and proton magnetic resonance spectroscopy (~1H-MRS) was performed in 2 patients.Results Typical demonstrations were found in 4 patients who were correctly diagnosed.Among the others with atypical findings,3 patients were misdiagnosed as liver cancer and 1 as focal nodular hyperplasia (FNH) .The ~1H-MRS curves of 2 adenoma tumor showed that the peak of choline of tumor tissue had no significant difference comparing with that of normal liver tissue around the tumor tissue.Conclusion CT and MRI findings of hepatic adenomas have some characteristics,but atypical manifestations lead to misdiagnose.~1H-MRS may be helpful for the differential diagnosis of hepatic adenoma.
