1.Research on single photon emission computed tomography imaging to brain gliomas with technetium~(99m) sestamibi

Objective To explore the diagnosis value of single photon emission computed tomography(SPECT) imaging to supratentorial brain gliomas with technetium 99m sestamibi.Methods In 53 patients with supratentorial brain glioma,31 cases for high grade malignant glioma, 22 cases for low grade glioma were examined by SPECT,(SPECT was performed after injection 740-930 MBq MIBI for 15 min),and the coronal,sagittal and transverse views were reconstructed. MIBI uptake index was computed as the ratio of counts in foci region and the counts in contralateral homologous region.Results In high-grade malignant gliomas,the MIBI uptake index was 1.9~6.6(mean 3.6?1.4),low grade malignant gliomas was 0.8~1.7 (1.1?0.2),there is a comparison between the two groups(P
2.Poly-beta-hydroxybutyrate as a scaffold for tissue-engineered cartilage

?Objective: To investigate the feasibility and of poly beta hydroxybutyrate (PBH) as a scaffold for chondrocytes to fabricate tissue engineered cartilage. Methods: PBH was manufactured into porous scaffold with 85% porosity and the diameter of the pore was 300 400 ?m, and then the scaffold was modified with 100 mg/L human fibronectin.Chondrcytes(4?10 7/ml) harvested from auricular cartilage of rabbits were seeded into the PHB scaffolds. The PBH/chondrocytes composites were translated in the dorsal subcutaneous tissue of 6 New Zealand rabbits . PBH scaffolds without cells were used as the controls. The specimens were harvested and examined histologically . Results: Four and eight weeks after transplantation, the PBH/chondrocytes composites showed the morphology of cartilage. New cartilage tissue was formed with a little matrix in four weeks. In eight weeks the newly formed cartilage tended to be mature with more matrix . Conclusion: PBH can be used as scaffold for tissue engineered cartilage.
3.Construction and Expression of Human pEGFP-GEF-H1 Vector

Background:Studies have shown that GEF-H1 can activate Rho protein, and is closely associated with tumorigenesis,tumor progression,infiltration,migration. Aims:To construct human pEGFP-GEF-H1 vector and stably transfect into colon cancer HCT116 cells,and determine the expression of GEF-H1. Methods:The gene sequence was designed according to the Online primers synthesis software for constructing recombinant plasmid pEGFP-GEF-H1. HCT116 cells were transfected with plasmid pEGFP-GEF-H1,empty plasmid,respectively,and HCT116 cells without transfection were served as blank control group. Expression of green fluorescence protein was observed under fluorescence microscope. mRNA and protein expressions of GEF-H1 were determined by RT-PCR and Western blotting,respectively. Results:Human pEGFP-GEF-H1 vector was successfully constructed and identified by sequence analysis. After transfection with pEGFP-GEF-H1,green fluorescence could be observed in HCT116 cells,and the mRNA and protein expressions of GEF-H1 were higher than those in empty plasmid group and blank control group. Conclusions:Recombinant plasmid pEGFP-GEF-H1 is successfully constructed in vitro,and transfection into HCT116 cells leads to high expression of GEF-H1, thereby can provide experimental materials needed for further study on correlation between colon cancer and GEF-H1.
4.Effect of sleep regulating technique of low resistance thought induction psychotherapy on primary insomnia

Objective To evaluate the efficacy of sleep regulation technique of thought imprint psychotherapy in lower resistant state (TIP) for the treatment of primary insomnia.Methods 45 hospitalized outpatients with primary insomnia from sleep medical clinic of Guang'anmen Hospital were randomized at a 2 ∶ 1 ratio into a therapy group and a control group using random table.In the therapy group,patients received sleep regudation technique of TIP twice per week.In the control group,patients were asked to take zopiclone 3.75mg per night 30 minutes before bedtime.And the maximum dose can be 7.5mg according to their condition.The treatment lasted one month.Pittsburgh sleep quality index (PSQI) was used to assess sleep quality.Results ① After 4 weeks treatment,the clinical efficacy showed significant difference between the two groups (therapy group was 80%,control group was 33.3%).② Inner group comparison:therapy group was significantly improved in PSQI sleep quality (1.20 ± 0.71),sleep latency(1.53 ±1.01),sleep time(1.23 ±0.82),sleep efficiency(0.90±1.21),sleep disorders (1.30±0.65),sleep medication(0.13±1.36),daytime function (1.67±0.99),total score(8.30±3.19),(P<0.05); the control group was significantly improved in the PSQI sleep quality(1.60±0.91),sleep latency(1.87±0.64),(P<0.05).③ Inter group comparison:after treatment,compared to the control group,the therapy group was superior in the sleep latency,sleep time,sleep efficiency,sleep medication,daytime function,and total score ofPSQI (P<0.05).Conclusion Sleep regulation techmique of thought imprint psychotherapy in lower resistant state can improve sleep quality of primary insomniacs.
5.Correlation study of Narcotrend values and patient-controlled sedation of dexmedetomidine under CSEA

Objective To explore the correlation of patient-controlled sedation of dexmedetomidine and Narcotrend values. Methods Forty patients with lower limb surgery were enrolled. Until CSEA block fixed , the electronic pump ran the patient-controlled sedation of dexmedetomidine. The parameter of electronic pump was set as follows: load dose 2 mL + background dose 1.5 mL/h + single dose 0.5 mL + locktime 20 s. The heart rate , mean arterial pressure, pressing times, effective times, OAA/S sedation scores and NI values were determined. Results At T4 point, the patients reached appropriate sedation. At T4 ~ T9 OAA/S scores kept 3 to 4. From T5 point, NI values showed significant decrease. After the T7 point. OAA/S scores and NI values reached the plateau time of (7.5 ± 1.8) min and (13.1 ± 3.4) min, OAA/S scores of 1, 2, 3, 4, respectively, corresponding roughly with NI values 95 to 100, 90 to 94, 65 to 89, 40 to 64. The correlation coefficient was 0.58. The time of NI values significant decreased in the younger group and in the elderly group, with (10.2 ± 1.6) min and (14.4 ± 2.2) min. In T5~ T9 point, NI values of the younger group were significantly lower than those in the elderly group. Conclusion Relevant relationships are observed between dexmedetomidine patient-controlled sedation depth and the narcotrend values under CSEA.
6.Influence of aging on motor function and striatum dopamine transporter of rats

Objective To investigate the influence of aging on motor function,binding activity and protein expression level of striatum dopamine transporter(DAT)of rats. Methods Rolling-bar test was performed to assess motor function.Western blot and 131I-FP-β-CIT up-take ratio were used to evaluate DAT protein 1evel and striatum DAT binding activity respectively. Results There were an age-related decline of rolling-bar latency and striatum 131I-FP-β-CIT up-take ratio in rats older than6 months.There was a significant difference between 6-month-old rats and older rats(12-month-old rats,16-month-old rats,20-month-old rats),and rolling-bar latency was correlated with striatum 131I-FP-β-CIT up-take ratio(r=0.656,P<0.01).There was no change in DAT protein 1evel during aging process. Conclusions The age-related decline of motor function of rats was correlated with the decreasing of striatum DAT binding activity.The synaptic membrane expression of striatum DAT may decrease in aged rats.

9.Effects of oscillating electric field on Wnt-3a expression and motor function in the injured rat spinal cord

BACKGROUND: Wnt signaling pathways can stimulate the proliferation and differentiation of neural stem cel s, and promote the repair of spinal cord injury. The electrical field stimulation can change the protein expression of Wnt signaling pathways.
QBJECTIVE: To explore the influence of oscil ating electric field on motor function recovery and the expression of Wnt-3a protein in rats with spinal cord injury.
METHODS: A total of 36 Sprague-Dawley rats models with spinal cord injury were induced by the Al en’s method. Al rats were randomly divided into oscil ating electric field stimulation group and spinal cord injury group. The stimulating electrodes were added in both groups. The oscil ating electric field stimulation group received oscil ating electric field intervention.
RESULTS AND CONCLUSION: At 3 days after model establishment, Basso, Beattie, and Bresnahan (BBB) locomotor rating scale scores and Wnt-3a expression were similar between the oscil ating electric field stimulation group and spinal cord injury group. At 7 and 14 days, significant differences in BBB scores and Wnt-3a expression were detected. These results indicate that oscil ating electric field stimulation can activate Wnt signaling protein in the early stage of spinal cord injury, which may be associated with the promoting effect of oscil ating electric field stimulation on spinal cord injury.
10.Effect of morphine exposure in late pregnancy on expression of NR2B in nucleus accumbens of off-spring rats

Objective To evaluate the effect of morphine exposure in the late pregnancy on the expression of 2B subunit-containing N-methyl-D-aspartate receptor (NR2B) in the nucleus accumbens of offspring rats.Methods Forty-eight Sprague-Dawley rats at 14 days of gestation,aged 16-20 weeks,weighing 400-600 g,were equally and randomly divided into control group (group C) and morphine group (group M).In group M,morphine was administered subcutaneously twice a day (9:00 a.m.and 5:00 p.m.) in the initial dose of 2 mg/kg followed by daily increment of 1 mg/kg until 6 mg/kg.The equal volume of normal saline was given instead of morphine in group C.Fifty offspring rats with similar body weight were selected from the two groups,and conventionally fed for 8 weeks.Thirty offspring rats were further selected,morphine 3 mg/kg was injected subcutaneously for 7 consecutive days,and then the rats underwent the conditioned place preference test to monitor the effect.The time spent in drug-paired side (gray area) was recorded at 24 h after the test.Ten offspring rats selected were sacrificed after the test,and the brain was removed.The nucleus accumbens was isolated for detection of the expression of NR2B by immunohistochemistry and Western blot.Results Compared with group M,the time spent in drug-paired side (gray area) was significantly prolonged,and the expression of NR2B in the nucleus accumbens was up-regulated in group C (P<0.05).Conclusion The mechanism by which morphine exposure in the late pregnancy causes susceptibility to morphine addiction is associated with up-regulation of NR2B expression in the nucleus accumbens of offspring rats.
