1.The effects of bone marrow mesenchymal stromal cells transplantation after mannitol pretreatment on behavioral performance and synaptophysin expression in the CA3 region in hippocampus of vascular dementia rats

Objective To investigate the effects of bone marrow mesenchymal stromal cells (BMSCs) transplantation after mannitol pretreatment on behavioral performance and synaptophysin expression in the CA3 region in hippocampus of vascular dementia (VD) rats.Methods The BMSCs of rats were isolated and purified by the whole bone marrow adherence method.The rats were subjected for permanent ligation of bilateral common carotid arteries at an interval of 3 days for each carotid artery.At the same time,Sham group was set in parallel.Four weeks after modeling,the VD rats were divided randomly into five groups:(1) VD control group; (2) culture media group; (3) mannitol group; (4) BMSCs group;(5) mannitol with BMSCs group.Morris water maze performance and synaptophysin expression in the CA3 region in hippocampus were observed at 4 weeks after transplantation.Results The morris water maze performance significantly improved in mannitol with BMSCs group when compared with BMSCs group,VD control group,culture media group,mannitol group.Moreover,the escape latency of fifth day decreased significantly ((9.3 ±2.9),(14.1 ±3.5),(23.5 ±4.4),(22.8 ±4.4),(23.2 ±2.8) s,F =43.900,P =0.000)),and the platform quadrant residence time increased significantly ((40.8 ± 6.3),(34.9 ±5.8),(26.4±4.8),(27.4 ±7.0),(28.5 ±6.2) s,F=13.000,P=0.000)).The synaptophysin expressions of the hippocampal CA3 region were significantly increased in the mannitol with BMSCs group (39 624 ± 7798) when compared with BMSCs group,VD control group,culture media group,mannitol group (27060 ±4668,18 294 ±6446,19 956 ±4244,18 946 ±4953,F =39.206,P =0.000).Conclusions Intravenous BMSCs transplantation after mannitol pretreatment improves the behavioral performance of VD rats and facilitates the synaptophysin expression of hippocampal CA3 region in VD rats than BMSCs transplantation alone.Mannitol pretreatment can amplify the therapeutic effect of intravenous BMSCs transplantation in VD rats.
2. Specimen processing and pathological evaluation of 113 samples of early colorectal cancer and precancerous lesion after endoscopic submucosal dissection

Objective:
To study the pathology and its significance of early colorectal cancer and precancerous lesions treated by endoscopic submucosal dissection (ESD).
Methods:
A total of consecutive 113 cases were collected at the endoscopy center from August 2012 to June 2016, which were diagnosed as early colorectal cancer and precancerous lesions and undergone ESD. According to Japanese colorectal cancer treatment guidelines, specimens were processed and pathologically evaluated for histological type, tumor diameter, depth of invasion, budding grading, vessel invasion, and horizontal and vertical margin, as well as curative resection.
Results:
There were 63 cases of adenoma (55.75%), including 29(25.66%) tubular adenoma, 2(1.77%) villous adenoma, and 32(28.32%) villioustublar adenoma. Thirty-four cases of serrated lesion were found, which included 19(16.81%) traditional serrated adenoma, 11(9.73%) sessile serrated adenoma, and 4(3.54%) hyperplasic polyp. There were also 16(14.16%) cases of early colorectal cancer with 7 cases of well-differentiated adenocarcinoma, 7 cases of moderately-differentiated adenocarcinoma, 1 case of poorly-differentiated adenocarcinoma, and 1 case of mucinous adenocarcinoma. Vessel invasion were observed in 2 of 16 cases of early colorectal cancer which were both moderately-differentiated adenocarcinoma in sigmoid colon. The vertical margins were negative in 108(95.58%) of 113 cases. Positive vertical margin were found in only 1 case (moderately-differentiated adenocarcinoma, pT1b2) and another case was suspected as positive. The rest 3 cases could not be precisely diagnosed. The horizontal margins were negative in 80(70.80%) of 113 cases and positive horizontal margin were found in 20(17.7%) cases (19 adenoma and 1 moderately-differentiated adenocarcinoma). Thirteen cases cannot be precisely diagnosed. Histologically, complete resection rate was 82.30%. The complete resection rate of invasive adenocarcinoma was 93.75%. Among 16 cases of invasive adenocarcinoma, 5 cases (curative rate: 31.25%) were judged as curative resection whereas 11 cases were considered as non-curative resection. Seven non-curative resection cases were treated with further surgery and did not relapse after the follow-up.
Conclusion
The standardized processing and precise histopathological evaluation are key factors for colorectal ESD technique, which play an important role in the success of endoscopic therapy.
3.Microembolic signal monitoring by trans-cranial Doppler in patients with carotid artery stenosis and its intervention therapy

Objective To investigate the microembolic signal (MES) in patients with carotid artery stenosis by transracial Doppler (TCD) diasonograph,and explore the influence of intervention treatment in MES of patients with carotid artery stenosis.Methods One hundred and fifty-eight patients with carotid artery stenosis,admitted to our hospital from March 2009 to November 2014,were enrolled as carotid artery stenosis group;and 158 workers from our hospital without carotid artery stenosis were used as control group.TCD was used to monitor the MES at the initiative internal carotid artery in the two groups;and then,10 d after aspirin enteric coated tablets (100 mg/d) on platelet aggregation and torvastatin (20 mg/d) treatment,MES was monitored again;the MES positive rate between the two groups or in carotid artery stenosis group between before and after medication was compared.Results As compared with the control group (1.9%),the carotid artery stenosis group (36.1%) had significantly increased MES positive rate (P<0.05).In the carotid artery stenosis group,MES positive rate before treatment (36.1%) was significantly higher than that after treatment (7.6%,P<0.05).Conclusion MES positive rate in the cerebral blood flow in patients with carotid artery stenosis is increased;treatment by drugs can reduce the microemboli positive rate.
4.Compositional Analysis of 11 Nucleosides and Bases in Fritillaria taipaiensis P.Y.Li from Different Origins and the Differences in Their Origin

Objective The contents of 11 nucleosides and base components in 10 batches of samples from 5 provinces(cities)including Chongqing,Yunnan and Shaanxi were determined,and the differences in nucleosides and base components in Fritillaria taipaiensis were compared by chemometric analysis,and the quality was comprehensively evaluated,so as to provide a reference for the cultivation of excellent varieties and the selection of medicinal materials.Methods Nucleoside and base components were extracted from Fritillaria taipaiensis by ultrasonication in aqueous solutions,and the content of each component was determined by HPLC-DAD method.The origin was classified by principal component analysis(PCA)and hierarchical cluster analysis(HCA).Partial least squares discriminant analysis(PLS-DA)was used to determine the differentiated index components in Fritillaria taipaiensis.Then the differences in the contents of the index components among samples from different origins were compared.Results It was found that 11 nucleoside and base components differed significantly among different origins of Fritillaria taipaiensis.Principal component analysis and hierarchical cluster analysis indicated that all samples could be clustered into 4 categories.Five characteristic components,including uracil,cytosine,uridine,inosine,and adenosine,were identified by PLS-DA.The nucleosides and bases in samples from Chongqing and Hubei were relatively high,and the quality of the samples was comparatively superior.Conclusion This method is simple,reproducible,accurate and reliable.It has screened out the index nucleoside and base components in the identification of Fritillaria taipaiensis of different origins,which can be used to initially elucidate the differences of samples between different origins.Additionally,it can better reflect the quality of Fritillaria taipaiensis,and can provide reference for the selection of procurement origin and the quality control for Fritillaria taipaiensis.
5.Effect and Mechanism of Steamed Allii Sativi Bulbus Polysaccharide Against Dysbiosis-induced Diarrhea in Young Rats

ObjectiveTo verify the efficacy of steamed Allii Sativi Bulbus polysaccharide in regulating the intestinal flora of young dysbiosis-induced diarrhea rats based on 16S rRNA gene sequencing. MethodThe young SD rats were randomly divided into blank group,model group,positive drug group (bifid triple viable capsules),and high-dose and low-dose steamed Allii Sativi Bulbus polysaccharide groups,six in each group. The dysbiosis-induced diarrhea rat model was established,and the blank group and the model group were given normal saline by gavage (each 10 mL·kg-1),and the high-dose and low-dose steamed Allii Sativi Bulbus polysaccharide groups were administered with corresponding drugs (500 mg·kg-1 and 250 mg·kg-1, respectively) ,once a day for seven consecutive days. The loose stool rate,loose stool grade,diarrhea index,small intestine propulsion rate and hematoxylin-eosin (HE) staining were used as indexes to investigate the effect of steamed Allii Sativi Bulbus polysaccharide on improving diarrhea symptoms in young rats. The feces of rats were collected for 16S rRNA gene high-throughput sequencing. ResultCompared with the model group, the positive drug group and the high-dose and low-dose steamed Allii Sativi Bulbus polysaccharide groups had alleviated symptoms, down-regulated loose stool rate and diarrhea index (P<0.01) and decreased small intestine advancement rate (P<0.05). HE staining showed that after the treatment with steamed Allii Sativi Bulbus polysaccharide,the inflammatory cell infiltration of the colon tissue was improved and the intestinal gland recovered to the normal condition,which indicated that steamed Allii Sativi Bulbus polysaccharide could significantly ameliorate the diarrhea in young rats. The sequencing results revealed that steamed Allii Sativi Bulbus polysaccharide had a moderating effect on the abundance of the intestinal flora of young dysbiosis-induced diarrhea rats,elevating the flora richness and diversity indexes. Specifically, the abundance of Bacteroidota was increased while that of Firmicutes and Proteobacteria was decreased. ConclusionSteamed Allii Sativi Bulbus polysaccharide can be used to treat dysbiosis-induced diarrhea in young rats by regulating the abundance of intestinal microbiota.
