1.Intratympanic Dexamethasone in the Treatment of Acute Low-Frequency Hearing Loss

Objective To explore the efficacy of intratympanic dexamethasone injection in treating acute low-frequency hearing loss(ALHL).Methods Thirty-seven ALHL cases not responsive to intravenous dexamethasone and vasodilator medications were enrolled in the study and divided into two groups.In the treatment group,dexamethasone was injected intratympanically once per day to 18 cases for a seven-day history with unilateral ALHL and 1 case with bilateral ALHL.In the control group,18 cases with unilateral ALHL were given intravenous vasodilator medications for 7 days.All patients were followed up for 1~ 2 years.Results In the treatment group with unilateral ALHL,8 cases achieved complete recovery,7 cases achieved partial recovery and no change was observed in the other 3 cases.The efficiency was 83.3 %.2 cases showed recurrent low-frequency hearing loss during the follow-up period.One case with bilateral ALHL did not respond to the treatment and no recurrence was observed.No side effect was observed in the study.In the control group,3 cases completely recovered,5 cases partially recovered and 10 cases showed no improvement.The efficiency was 44.4%.5 cases recurred and one of them progressed to Meniere's disease.There was a statistically significant difference in efficiency of patients with unilateral ALHL between two groups(P<0.05).Conclusion Intratympanic dexamethasone application was an effective and safe treatment option for ALHL.
2.APPLICATION OF ENDOSCOPIC ULTRASONOGRAPHY AND MICROVESSEL COUNTING IN PREOPERATIVE STAGING OF COLORECTAL CANCER

30 patients with colorectal carcinoma underwent endoscopic ultrasonography(EUS)and microvessel counting for staging before operation.The accuracy of EUS in assessing depth of tumor in- filtration was 76.7%,that in detecting lymph node involvement being 73.3%.The correctness of pre- operative Duke's staging remained 70%.Microvessel counting correlated with depth infiltration and lymph node invasion.The microvascularity in tumor with serosal involvement was obviously abundant than that without,so as in tumor with and without lymphatic invasion.The results revealed that both EUS and microvessel counting can be considered as an essential evidence for preoperative staging of col- orectal cancer and thereby for predicting prognosis.
3.The Long-term effect of augmentation rhinoplasty with silicone

Objective:To evaluate the long-term effect of augmentation rhinoplasty with silicone. Method:360 patients underwent augmentation rhinoplasty with silicone. Improved L shaped implants were used in 343patients. Result: Among 360 patients, 334 cases were successful enough (92.83 %0 ). Conclusion: The key to themaintenance of the long-term effect is to use properly improvedL-shaped implant,to choose a suitable tunnel,to prevent various complications and to ask patient's opinions for implant design.
4.Expression of human beta-defensin-3 in surrounding tissues of infected artificial prostheses

Objective To observe expression of human beta-defensin-3 (HBD-3) in tissues around the infected artificial prostheses and investigate its value in treatment and diagnosis of periprosthetic joint infection (PJI).Methods According to clinical diagnosis,periprosthetic tissues and normal synovial membrane excised in operation were collected and divided into the following four groups:PJI group (n =13),aseptic loosening group (loosening group,n =9),spacer treatment group (treatment group,n =12),and normal group (n =15).HE staining was used to observe infiltration of inflammatory cells.Immunofluorescence staining was used to detect positive cells number and fluorescence intensity.Image-pro plus (IPP) 7.0C software was used to measure the average value of absorbance.Preoperative peripheral white blood cell count,erythrocyte sedimentation rate (ESR),and C-reactive protein (CRP) results were documented.Then,differences of those parameters were analyzed and compared among groups.Results HE staining revealed that all groups had different degree of inflammatory cell infiltration except for normal group.Immunofluorescence staining revealed that the most number of positive cells and highest fluorescence intensity existed in PJI group.Value of absorbance in PJI group was 0.430 ± 0.013,followed by 0.308 ±0.005 in loosening group,0.234 ± 0.009 in treatment group,and 0.089 ± 0.019 in normal group.Preoperative peripheral white blood cell count,ESR and CRP were the highest in PFI group,but were not significantly different among the remaining three groups.Conclusion HBD-3 is highly expressed in tissues around the prostheses which had infection or aseptic loosening,but its expression in response to infection and loosening has difference.
5.BIODEGRADABLE POLY (D,L-LACTIDE-CO-GLYCOLIC ACID) MICROSPHERESUSED AS ADJUVANT IN DNA IMMUNIZATION OF HEPATITIS B VIRUS S GENE

Study the immunological adjuvant function of biodegradable microspheres for DNA immunization. Empty poly(D,L-lactide-co-glycolic acid) microspheres were prepared using the water-inoil-in-water(w-o-w)technique; A plasmid DNA pRc-CMV encoding hepatitis B virus S antigen was constructed;The mixture of the microspheres and the plasmid DNA was prepared by incubation method. The mixture was administered to Balb/c mice by intramuscular injection. Result: The high antibody titer (1:1600) of intramuscular injection of the mixture of microspheres and the plasmid DNA was obtained, similar to that of intramuscular injection of the mixture of AL(OH)3 and hepatitis B virus S antigen; while intramuscular injection the plasmid DNA elicited no serum antibody respones. Conclusion: biodegradable microspheres may be used as an good adjuvant for DNA immunization.
6.Progress in optimization of Cerenkov luminescence imaging

Cerenkov luminescence imaging ( CLI) , as an emerging molecular imaging method, has been extensively studied in tumor imaging, therapy monitoring and some other aspects. However, because of the weak penetration of Cerenkov radiation, CLI can not image the deep tissues. This review summarizes the modalities to overcome this problem.

8.Biomechanical performance and effect of static interlocking intramedullary nails on femoral fracture healing

BACKGROUND: Interlocking intramedullary nails(SliN)is frequently used to treat femoral fracture.Routine pulling and compression and postoperative motorization are important for fracture healing.To improve fracture healing rate has been focused using various factors.OBJECTIVE: To explore the biomechanical performance of static SliN in fixation of femoral fracture,and the effect on fracture healing.METHODS: A total of 8 pairs of femurs from adult cadavers immersed in formalin were used to prepare models of midpiece transverse fracture.The fracture was respectively fixed using static SliN,TiNi shape-memory sawtooth-arm embracing internal fixator(TiNi SMA),and dynamic compression plate(DCP).The anti-compression,anti-bending,and anti-torsion rigidity were measured,and the stress shielding rate was calculated.RESULTS AND CONCLUSION: SliN was similar to DCP group in anti-compression rigidity and anti-bending rigidity(P > 0.05),but the anti-torsion was lower than DCP group(P<0.05).Compared with TiNi SMA group,SliN displayed greater anti-compression rigidity and anti-bending rigidity(P<0.05),but similar anti-torsion(P > 0.05).SliN was similar to DCP group in stress shielding rate(P>0.05),but significantly greater than TiNi SMA group(P<0.01).The biomechanics of SliN was stable,but the stress shielding rate was high.Changing static interlocking into dynamic interlocking in time is necessary in clinic to promote fracture healing.
9.Preparation and in vitro evaluation of borneol and folic acid co-modified doxorubicin loaded PAMAM drug delivery system.

A novel targeting drug carrier (FA-BO-PAMAM) based on the PAMAM G5 dendrimer modified with borneol (BO) and folic acid (FA) molecules on the periphery and doxorubicin (DOX) loaded in the interior was designed and prepared to achieve the purposes of enhancing the blood-brain barrier (BBB) transportation and improving the drug accumulation in the glioma cells. 1H NMR was used to confirm the synthesis of FA-BO-PAMAM; its morphology and mean size were analyzed by dynamic light scattering (DLS) and transmission electron microscope (TEM). Based on the HBMEC and C6 cells, cytotoxicity assay, transport across the BBB, cellular uptake and anti-tumor activity in vitro were investigated to evaluate the properties of nanocarriers in vitro. The results showed that the nanocarrier of FA-BO-PAMAM was successfully synthesized, which was spherical in morphology with the average size of (22.28 ± 0.42) nm, and zeta potential of (7.6 ± 0.89) mV. Cytotoxicity and transport across the BBB assay showed that BO-modified conjugates decreased the cytotoxicity of PAMAM against both HBMEC and C6 cells and exhibited higher BBB transportation ability than BO-unmodified conjugates; moreover, modification with FA increased the total uptake of DOX by C6 cells and enhanced the cytotoxicity of DOX-polymer against C6 cells. Therefore, FA-BO-PAMAM is a promising nanodrug delivery system in employing PAMAM as a drug carrier and treatment for brain glioma.
10.The proliferation inhibition of colon cancer cells by agonistic CD40 monoclonal antibody in vitro

Objective To investigate the inhibitory effect of agonistic CD40 monoclonal antibody on the colon cancer cells (HCT116) proliferation in vitro.Methods The DCs (dendritic cells) loaded with tumor cells (HCT116) antigens were activated by different methods.According to the activation method,the cells were divided into three groups:agonistic CD40 monoclonal antibody group,blank control group and TNF-α positive control group.The cells were cultured for 7 days,and the expression rates of CD80,CD83,CD86 and HLA-DR on DC surface in each group were detected by flow cytometry.The concentration of cytokine IL-12(p70) in DCs culture supernatant was determined by ELISA kit.The proliferation activity of the T lymphocytes was evaluated by MTT (methyl thiazolyl tetrazolium).Then the inhibition rate of colon cancer HCT116 cells proliferation,which induced by the tumor-specific effector T lymphocytes,was assayed.Results Compared with the blank control group,the agonistic CD40 monoclonal antibody group had a significantly higher expression rates of CD80,CD83,CD86 and HLA-DR on DC surface (P<0.05).The concentration of IL-12 in the supernatant of DC was also much higher in the agonistic CD40 monoclonal antibody group (P<0.05,(716.80±53.43) pg/ml vs.(405.51 ±12.17) pg/ml).The DCs activated by CD40 monoclonal antibody had stronger ability to stimulate proliferation of T lymphocytes (P<0.05,the stimulation index was (2.006 2±0.438 3) to (1.365 0±0.209 8)).The tumor-specific CTLs induced by DCs in the agonistic CD40 monoclonal antibody group had stronger ability to inhibit colon cancer HCT116 cells (P<0.05,the inhibition rate was (66.08±0.41)% vs.(46.60± 1.10)%).However,there was no statistical significance between the agonistic CD40 monoclonal antibody group and the TNF-α positive control group (P>0.05).Conclusion Agonistic CD40 monoclonal antibody in vitro can promote activation and mature of DCs,then the activated DCs can induce the production of tumor-specific CTL,which can significantly inhibit the proliferation of colon cancer HCT116 cells.
