Objective To investigate the anti-angiogenesis and tumor inhibitory effects of recombinant human endostatin(rhES)combined with paclitaxel-cisplatin chemotherapy(TP regimen)on xenograft tumors in nude mice of human breast carcinoma cell MCF-7.Methods Forty-eight xenograft nude mice were randomized into group A and group B,then every group were divided 4 subgroups,that is,TP group(cisplatin 20 mg/kg at days 1,7 and 14,and paclitaxel 5 mg/kg at days 1 and 7,i.p.),rhES group(rhES 10 mg?kg-1?d-1 for 14 d,i.v.),rhES+TP group(combined treatment as above-mentioned),and control group(normal saline,i.v.).Inhibitory rate of xenograft and tumor growth curve was calculated and plotted.Serum VEGF level was determined by ELISA,microvessel density(MVD)in tumors was measured by immunohistochemistry,and cell apoptosis was examined by TUNEL staining.Survival time was observed in group B.Results Serum VEGF and MVD was significantly lower in rhES+TP group than in other groups(P
2.Significance of total bilirubin/albumin in predicting bilirubin neurotoxicity

Objective To investigate the significance of total bilirubin/albumin in predicting bilirubin neurotoxicity.Methods Eighty-three cases with hyperbilirubinemia who treated from May 2007 to August 2010 were selected,the serum total bilirubin and albumin were detected and total bilirubin/albumin was calculated.According to brainstem auditory evoked potential(BAEP)results,the patients were divided into normal BAEP group and abnormal BAEP group and compared.Results There were 27 cases of abnormal BAEP group and 56 cases of normal BAEP group.Total bilirubin and total bilirubin/albumin in abnormal BAEP group were higher than those in normal BAEP group[(356.50±59.23)μmol/L vs.(318.70±55.12)μmol/L,(5.02±0.49)×10-3 vs.(4.56±0.43)×10-3],the differences were significant (P<0.05).Multiple regression analysis showed:abnormal BAEP was closely related to total bilirubin/albumin(r=0.72,P<0.05),whih abnormal BAEP was not obviously related to total bilirubin(r=0.19,P>0.05).Conclusion Total bilirubin/albumin can reflect serum unconjugated bilirubin level of neonatus with hyperbilirubinemia better than total bilirubin,and it can can be taken as one index to evaluate the risk factors of bilirubin neurotoxicity.
3.Reform of experimental course assessment methods of preventive medicine in undergraduates and its evaluation

ObjectiveTo aim at the deficiencies of existing assessment methods,and put forward ideas,methods and contents to reform experimental course assessment methods for undergraduates.MethodsAll of the fifth-grade preventive medicine undergraduates were divided into experimental group (n=34) and control group(n=31),reformed and conventional assessment methods were used in experimental group and control group respectively.The difference in attendance,experimental skill and reports were compared between two groups.ResultsAttendance,test scores and experimental skill scores of students in experimental group were significantly higher than that in control group and compared with the control group students (P<0,05),the experimental group students' understanding on the experimental principle,phenomena and data analysis were better.ConclusionReformed assessment methods of experimental course has an improved effect on teaching for undergraduates.
4.Establishment of a rapid method for synthesis and detection of gRNA

Objective The purpose of this study was to establish a rapid method for synthesis and detection of guild RNA (gRNA) which is an essential component in CRISPR/Cas9 knockout technology .Methods First, the Nkp46 gRNA core fragment was synthesized as amplification template .Second , the forward and reversed primers of the matched gRNA were designed using the Nkp46 gene as reference sequence .Third, the DNA fragment of Nkp46 gRNA was amplified by PCR technology using the synthesized gRNA core fragment as template .The gRNA was reversely transcribed in vitro u-sing amplified DNA fragment as template .The efficiency and specificity of gRNA and its interaction with Cas 9 were detec-ted in vitro.Results The specificity and activity of Nkp46 gRNA were high .The obtained gRNA interacted with Cas 9 en-zyme and successfully cut the target double-stranded DNA at the designed site .Conclusions The method for synthesis and detection of gRNA established in this study is faster than the original method , and the created gRNA is fully functional .
5.Study on Dermal Pharmacokinetic Difference of Triptolide in Normal and Diabetic Rats by Microdialysis

OBJECTIVE:To study the dermal pharmacokinetic difference of triptolide in normal and diabetic rats,and to pro-vide reference for rational drug use in the clinic. METHODS:12 Wistar rats were randomly divided into normal group and diabetic model group(0.1%streptozotocin intraperitoneally),with 6 rats in each group. Both group were given Triptolide cream 0.5 g to ab-dominal skin,and dialysate was collected by microdialysis every 30 min for consecutive 12 h. Subcutaneous concentration was de-tected by HPLC-MS,and subcutaneous concentration-time curves were analyzed and compared between two groups,and Winnon-lin 5.0.1 software was used to calculate pharmacokinetic parameters. RESULTS:The pharmacokinetic parameters of normal group and diabetic model group were that cmax were(1.54±0.37)and(5.12±1.34)μg/ml;tmax were(7.32±0.24)and(6.21±0.35)h;AUC0-12 h were (12.65 ± 4.64) and (37.43 ± 5.23)μg·h/ml,with statistical significance (P<0.05). CONCLUSIONS:The change of dermal structure caused by diabetes can increase percutaneous penetration amount of triptolide in rats,and drug dosage should be reduced according to circumstances so as to reduce side effects.

AIM: This study was conducted to evaluate the bone repairing ability of coral artificial bone(CAB) composite of human recombinant bone morphogenetic protein-2(rhBMP-2-CAB) implanted into extraction sockets. METHODS: 12 adult dogs served as the experimental animals. Immediately after extraction of the upper second and third incisors, rhBMP-2-CAB and CAB were respectively implanted into each site of extraction sockets. The bone repairing ability of the two grafts was analyzed with histology, image analysis system and radionuclide bone imagining at 4th, 8th and 12th week, respectively. Aim To evaluate the effects of Brucine of restraining transplanted tumor growth,prolonging their survival time and its toxicity on haematopoietic and immune systems as well as hepatic and renal function in mice transplanted with Hepatocarcinoma Heps cells.Methods ICR male mice of Hepatocarcinoma Heps tumor model(Heps mice)was made through Heps cells inoculation.Anti-tumor activity of Brucine was evaluated in terms of tumor inhibitory ratio and life elongational ratio of mice transplanted with Heps tumor.The toxicity of Brucine was measured the mice weight,immune organ weight,blood cell index(leukocyte,erythrocyte,platelet,and hemachrome protein),and hepatic function index(ALT,AST)as well as renal function index(BUN) of Heps mice.Results Brucine had significant restraining effect on the growth of Heps tumor in mice.Its inhibitory rate was 30.34%(1.61 mg?kg~(-1)),46.21%(3.23 mg?kg~(-1))and 42.07%(6.46 mg?kg~(-1))respectively,and 3.23 mg?kg~(-1)(1/20 LD_(50)) was the best dosage of Brucine for treating Heps tumor in vivo,while Brucine had no distinct effect of prolonging the life of Heps mice.Moreover,it enhanced the weight of body and thymus index as well as spleen index,and had no distinct influence on blood cell index as well as ALT,AST and BUN in Heps mice.Conclusion Brucine can inhibit the growth of transplanted Heps tumor,enhance the weight of immune organ index significantly,and has no toxicity on haematopoietic and immune systems as well as hepatic and renal function of mice transplanted with Heps tumor.It may be developed into a promising anticancer drug through further study
10.Changes of oxygenation index of severe acute respiratory syndrome treated with glucocorticosteroids

Objective To investigate the effect of glucocorticosteroids on pulmonary oxygenation function in severe acute respiratory syndrome(SARS)patients.Methods Two hundred and twenty-five SARS patients in 2003 were analyzed retrospectively.Oxygenation index(OI)was considered as the marker of glucocorticosteroids therapeutic effects.The criteria of effectiveness was that OI increased 20%or more than pre-treatment.The therapeutic effects of glueocorticosteroids were analyzed by analysis of variance and chi-square test.Results Glucocorticosteroids were used in 59.6%(134/225)of SARS cases.The average OI before and after glucocorticosteroids intravenous treatment was 237.08 mm Hg and 335.08 mm Hg,respectively.The average OI increased 110.26 mm Hg(46.4%)after treatment.In 96 SARS patients whose blood gas analysis results were available,85 cases(88.5%)had administrative indication.The average OI increased 103.14 mm Hg(44.4%),which was lower than 190.91 mm Hg(66.8 0A)in patients without administrative indication.There was no significant difference between these two groups of patients.The OI difference before and after glueocorticosteroids treatment was decreased with the increasing OI before treatment,the increase was minimum in patients with OI≥300 mm Hg.Conclusions The SARS patients could obtain pulmonary symptoms and function improvements with appropriate glucoconicosteroids treatment,especially for those who are in accord with acute lung injure or acute respiratory distress syndrome,and OI less than 300 mm Hg before treatment.
