1.Construction of mammalian cell expression vector of human leukocyte antigen-C gene and expression in JAR cells

Objective:To construct mammalian cell expression vector of human leukocyte antigen C gene and express it in JAR cell and study its functions.Methods:Total cell RNA was extracted from peripheral blood mononuclear cells and the cDNA was amplified by RT PCR;the cDNA fragments were directly inserted into Pbluescript KS(+/-) and the recombinant PBS HLA C was identified by restriction endonuclease digestion and sequencing;then the plasmid of pcDNA HLA C was transfected into JAR cells.Lactate Dehydrogenase(LDH) release assay was employed to detect the cytotoxicities of NK cells proliferated in vitro against HLA C transfectants JAR cells.Results:The recombinant mammalian cell expression vectors of pcDNA3 HLA C was constructed,the sequences of the insert was identical to the published sequences encoding HLA Cw *0602 gene.Several transfected JAR clones were obtained,which expressed HLA Cw *0602 in high level and constitutively.The specific lysis was detected for NK cells stimulated by IL 2 against JAR cells and HLA C transfectants JAR cells.But both cells were resistant to freshly isolated PBL cells.Conclusion:The recombinant mammalian cell expression vectors of pcDNA3 HLA Cw *0602 was successfully constructed,JAR cells resistance to NK lysis could involve an HLA C independent mechanism.
2.Evaluation of tolerance and pharmacodynamics of nano-micelle irinotecan formulation

This study aimed to investigate the improvement of tolance and pharmacodynamics of nano-micelle irinotecan formulation compared with irinotecan hydrochloride injection(Campto). The toxic effects of the two formulations on colorectal cancer cells COLO205, HT-29, HCT-8 and SW480 were tested in vitro. COLO205 tumor-bearing mouse model was constructed. The two preparations were given via tail vein injection to investigate the maximum tolerance dose(MTD)of tumor-bearing mice to the two preparations, and then to explore the improvement of anti-tumor efficacy of nano-micelle irinotecan formulation near the MTD. The results showed that there was no significant difference in the inhibitory effect of the two formulations on the four colorectal cancer cells in vitro. The MTD of nano-micelle irinotecan formulation and Campto was 432. 0 and 276. 5 mg/m2 respectively. Both of the two formulations showed significant anti-tumor effect in vivo, and the relative tumor proliferation rate and tumor wet weight inhibition rate of nano-micelle irinotecan formulation at high dose(345. 6 mg/m2)were significantly better than those of Campto at two doses(177. 0 and 221. 2 mg/m2)(P< 0. 05).
3.Research progress of plants improving air quality in closed environment

Regarding the air quality problems,this article summarized the adverse factors such as trace harmful gases,small particulate matter,and high concentration carbon dioxide that existed in space closed environment,and compared the advantages and disadvantages of physical and chemical treatment methods to improve air quality.The focus was on exploring the improvement benefits of plants method on air quality,and summarizing the research progress in three aspects:absorption of trace harmful gases,release of negative oxygen ions,and absorption of carbon dioxide.The current research problems were pointed out,and the future research directions on using plants to improve the air quality in space closed environment were predicted.
4.Development and characteristics of the volume adjustable vegetable facilities

Objective To develop the volume adjustable vegetable facilities and meet the demands of research on the growing technique of vegetables.Methods Three series of adjustable volume vegetable facilities were developed with light and ventilation module,growth module,cultivation tray module and measurement module.Telescopic structure was adopted in the facilities and the performance testing and characteristics analyses were carried out.Results The facilities could be adjusted at the height range of 250-700 mm.Three kinds of LED lights and replaceable cultivation tray and matrix were installed in the facilities.The parameters including water content,electric conductivity and temperature in the matrix,CO2 concentration in the atmosphere of facilities were monitored.The test results showed that the performances and the indexes of facilities met the subsequent experimental demands.Conclusion Light weight,small volume,and adjustable volume vegetable facilities were developed.The production efficiency of lettuce(12 plants/0.12 m2)grown in the facilities was 49.7 g(FW)plant-1.The development of facilities laid the foundation of construction of on-orbit vegetable facility.
