1.Clinical research of Shenqichongcao capsule on oxidant/antioxidant imbalance in patients with chronic obstructive pulmonary disease

Objective To observe the effect of Shenqichongcao capsule on oxidant/antioxidant imbalance in patients with chronic obstructive pulmonary disease (COPD).Methods Selected 30 COPD patients from treatment group and control group respectively.Pre- and post treatment,the changes of points of Syndromes and their SOD,LPO and NO of patients were observed and compared with normal group.Results The level of SOD and LPO of COPD patients were lower than normal group with shenqichongcao capsule.There was significant difference before and after treatment (P<0.01).The control group had no significant difference.Conclusion The effect of Shenqichongcao capsule on the free radical injury in patients with COPD had some improving effect.
2.The impact of glucose-6-phosphate dehydrogenase deficiency in Chinese on neutrophil

Objective: The aim of this study is to evaluate the impact of glucose-6- phosphate dehydrogenase deficiency in Chinese on the intracellular glucose-6-phosphate dehydrogenase activity and reactive oxygen intermediates production of peripheral neutrophil. Methods: Glucose-6-phosphate dehydrogenase, its mRNA expression and reactive oxygen intermediates production(NBT test) in silent or stimulated neutrophil from health controls and patients with glucose-6-phosphate dehydrogenase deficiency were analysized. Results: Compared with health controls, the activities of glucose-6-phosphate dehydrogenase of peripheral neutrophil from patients with glucose-6 -phosphate dehydrogenase deficiency was decreased, and its mRNA expression increased. Reactive oxygen intermediate production increased(no statistical signification) in silence but decreased following stimulation by LPS in neutrophil from normal individuals than that from patients. Conclusion: Decreased activity of G-6-PD and production of reactive oxygen intermediates following stimulation in neutrophil could be caused by G-6-PD deficiency in Chinese.
3.Expression and clinical significance of ubiquitin-specific protease 9X in pancreatic carcinoma of elderly patients

Objective There is close relationship between ubiquitin-specific protease 9X(USP9X) and the biological behav-ior of some tumor.The aim of this study is to investigate the expression and clinical significance of USP 9X in pancreatic carcinoma of elderly patients. Methods The expression of USP9X was detected in 30 pieces of surgically resected primary pancreatic carcinoma tissue and adjacent nontumorous pancreatic tissue of elderly patients by streptavidin -perosidase immunohistochemical method . Results The rate of USP9X positive expression was 56.7%, there was not positive expression in adjacent nontumorous pancreatic tissue .There was no relation between the expression of USP 9X with gender, age, the tumor positin, the tumor size and degree of differentiation (P>0.05), while it was significantly correlated with lymph node metastasis and TNM stages (P<0.05).By using Cox proportional haz-ards analysis, multivariable analysis revealed that TNM stages , lymph node metastasis and USP9X expression were independent risk factor(P<0.05). Conclusion The results indicated that USP9X may play a role in the pathogenesis and prognosis of pancreatic cancer of elderly patients .
4.Evaluation of PTW Seven29 combining Octavius phantom for tomotherapy patient-specific quality assurance

Objective To investigate the feasibility of dose verification of helical tomotherapy (HT) using the PTW Seven29 2D-ARRAY with Octavius phantom.Methods The 12 patients HT plan were verified with the PTW Seven29 2D-ARRAY combining Octavius phantom.The detector array was guided and registered by MVCT imaging,and measured dose distributions on the corresponding planes were compared with those calculated by the treatment planning system (TPS).Based on several different Gamma criteria,the Gamma analysis method was utilized to evaluate the dose verification.Results According to the different acceptance criteria combination of dose difference/distance to agreement at different threshold level including 3%/2 mm,3%/3 mm,3%/4 mm,3%/5 mm at TH5,TH10,TH20,TH30,the mean passing rates with γparameter ≤ 1 were all above 91.7% and 93.9% when PTW Seven29 2D-ARRAY was horizontal and vertical.The dose distribution measured by the 2D-ARRAY combining Octavius phantom was well consistent with that calculated by the TPS.Conclusions 2D-ARRAY with Octavius phantom can get satisfying results when used in HT plan dose verification and is a viable tool to replace traditional film dosimetry system in clinical quality assurance work.
5.Studies on antioxidation of extract from Ruppia rostellata koch

Extract from Ruppia rostellata Koch was divided into three parts: petroleum ether part, CH2Cl2 and water-soluble part. Each part of them was studied upon it's antioxidation. The results indicated that the active compounds were only in the water-soluble part. In soybean oil, the antioxidation of water-soluble part was stronger than that of BHA and V. E. , In lard, the antioxidation of water-solubile part was a little weaker than that of BHA and V.E.
6.Verification of the linear range and the minimum detection limit in real-time fluorescence quantitative PCR for HBV DNA

Objective Use real-time fluorescence quantitative polymerase chain reaction(PCR)to determine HBV DNA,then calculate the linear range and the minimum detection limit,which are the main performance indicators in the laboratory verification. Methods According to the related documents,by serial dilution of high concentration samples,samples of serial concentrations were obtained which were out of the linear range mentioned in the instructions,then verifid the new linear range.By serial dilution of low concentration,samples were obtained,the concentrations of which were lower than the minimum detection limit of provide by the manufacturer,then the new minimum detection limit was verified.Results The linear range of HBV DNA detection was 8.58× 102 -8.41×107 IU/mL,and the minimum detection limit of HBV DNA detection was 4.07 ×102 IU/mL.Conclusion The linear range and the minimum detection limit of Real-time fluorescence quantitative PCR assessed reaches the expected requirement,and the method and validation scheme are simple and feasible.
7.Effect of amygdalin on end-plate chondrocytes apoptosis induced by IL-1βderived from rat intervertebral discs

Aim To observe the anti-apoptotic effect of different concentrations of amygdalin on the endplate chondrocytes induced by IL-1 βderived from rat inter-vertebral discs and explore the possible mechanism fur-ther.Methods Chondrocytes were obtained from endplate of one-month SD rat intervertebral discs and cultured primary chondrocytes.After identifying,they were divided into normal group,induced group and A-mygdalin 1 0 -2 mol·L -1 ,1 0 -3 mol·L -1 ,1 0 -4 mol· L -1 ,1 0 -5 mol · L -1 administration group.Then the apoptosis was detected by flow cytometry (FCM).Re-al-Time PCR was adopted to detect the mRNA expres-sion of Bax and Bcl-2.The protein expression of Bax and Bcl-2 was detected by Western blot.Results The apoptosis of the endplate chondrocytes induced by IL-1 βderived from rat intervertebral discs could be inhib-ited by amygdalin with different concentrations.Amyg-dalin could reduce the apoptotic rate analysed by FCM,down-regulate the Bax mRNA expression of Bax and up-regulate the Bcl-2 mRNA assayed by RT-PCR;compared with the induced group the differences were statistically significant (P <0.05).Besides,observa-tion of the protein expression of Bax and Bcl-2 by Western blot found that amygdalin 1 0 -4 mol · L -1 could inhibit the effect of IL-1 β,which could increase the protein expression of Bax and reduce the protein expression of Bcl-2.Conclusion Amygdalin has an effect on anti-apoptosis of the end-plate chondrocytes induced by IL-1 βderived from rat intervertebral discs and improve the degeneration of intervertebral discs.
8.The effects and mechanism of safflor yellow on myocardial ischemia

Aim To observe the effects and mechanism of safflor yellow on the experimental acute myocardial ischemia (AMI) model rats. Methods AMI model rats were induced by isoproterenol (10 mg?kg-1). The changes of indexes on hemodynamics such as MBP, LVSP, LVEDP and ?dp/dt_ max,the coronary artery flux(CF) and myocardia O_2 comption(MVO_2) in AMI model rats were observed respectively. Results The CF and MVO_2 in the heart muscle increased and decreased respectively the indexes of hemodynamics were improved in safflor yellow groups. Conclusion The results indicate that safflor yellow can inhibit the failure of heart function induced by AMI, increase the coronary artery flux and decrease MVO_2 significantly.
9.The preventive effects of safflor yellow on myocardium injury of mycardium ischemic reperfusion in rats

AIM The preventive effects of safflor yefflor (SY) on myocardium injury of myocardium ischemic reperfusion (I/R) was studied. METHODS The contents of MDA activity of SOD,as well as the activity of creatine phospho kinase(CK) and lactate dehydrogenase(LDH) were determined in rat models injured by I/R. RESULTS SY could reduce the activities of CK and LDH ,decrease the the content of MDA in plasmo, and increase the activities of SOD. CONCLUSION SY may obviously prevent the rat I/R model which may be related to its anti lipoperoxidation.
10.Posterior subtotal vertebrectomy for thoracolumbar vertebral refractures after vertebroplasty

Objective:To evaluate the efficacy of posterior subtotal vertebrectomy in the treatment of thoracolumbar vertebral refractures after vertebroplasty.Methods:A retrospective analysis was conducted in the 28 patients with refracture after percutaneous vertebroplasty(PVP) or percutaneous kyphoplasty(PKP) who had been treated at Department of Spinal Surgery, Henan Provincial People's Hospital from June 2017 to October 2019. They were 7 males and 21 females, with an average age of 62.4 years(from 61 to 76 years). A total of 28 vertebrae were involved, including 5 T11s, 9 T12s, 11 L1s and 3 L2s. Their previous operations were PKP in 17 cases and PVP in 11. After the spinous process, vertebral plate, articular process and transverse process were resected by posterior approach, the vertebral body, bone cement and upper and lower intervertebral discs were partially resected by trans-vertebral lateral approach. At the same time, nerve decompression was performed. Finally, the inter-vertebral support was fixated followed by the posterior screw-rod orthopedic fixation. The operation time and intraoperative bleeding volume were recorded. The cobb angles of kyphosis were compared on the X-ray films of the whole spine between preoperation and the last follow-up to evaluate correction. Functional improvement of the spine was evaluated by comparison of the visual analogue scale (VAS) and JOA(Japanese Orthopedics Association) scores between preoperation and the last follow-up.Results:The operation time averaged 182.1 min and intraoperative bleeding volume 996.2 mL. All the 28 patients were followed up for 8 to 29 months (mean, 19.8 months). No obvious neurological lesions or other serious complications were observed. The cobb angle was improved from preoperative 41.3°±10.3° to 6.4°±2.5° at the last follow-up, the VAS score from preoperative 7.3±1.8 to 2.5±1.0 at the last follow-up, and the JOA score from preoperative 8.4±2.3 to 21.3±2.5 at the last follow-up, showing a significant difference in all the comparisons ( P<0.05). Conclusion:The posterior subtotal vertebrectomy is effective for thoracolumbar vertebral refractures after vertebroplasty because it can remove bone cement, decompress the spinal canal, fuse the inter-vertebral graft and reconstruct the spinal stability in one stage.
