1.The impact of glucose-6-phosphate dehydrogenase deficiency in Chinese on neutrophil

Objective: The aim of this study is to evaluate the impact of glucose-6- phosphate dehydrogenase deficiency in Chinese on the intracellular glucose-6-phosphate dehydrogenase activity and reactive oxygen intermediates production of peripheral neutrophil. Methods: Glucose-6-phosphate dehydrogenase, its mRNA expression and reactive oxygen intermediates production(NBT test) in silent or stimulated neutrophil from health controls and patients with glucose-6-phosphate dehydrogenase deficiency were analysized. Results: Compared with health controls, the activities of glucose-6-phosphate dehydrogenase of peripheral neutrophil from patients with glucose-6 -phosphate dehydrogenase deficiency was decreased, and its mRNA expression increased. Reactive oxygen intermediate production increased(no statistical signification) in silence but decreased following stimulation by LPS in neutrophil from normal individuals than that from patients. Conclusion: Decreased activity of G-6-PD and production of reactive oxygen intermediates following stimulation in neutrophil could be caused by G-6-PD deficiency in Chinese.

Objective:To investigate the neuroprotective effect and possible mechanism on rats with low dose Lipopoly -saccharide ( LPS) preconditioning after spinal cord injury.Methods:120 female SD rats were randomly divided into the empty virus (EV) group,LPS+empty virus (LPS+EV) group,Nrf2 interference virus (NIV) group,LPS+Nrf2 interference virus (LPS+NIV) group.The model of traumatic spinal cord injury ( TSCI) was established by the modified Allen′s method,motor function of the rat hind limb was assessed by the Basso Beattie and Bresnahan (BBB) score at 1,3,7,14 and 28 d after the operation.The injured spinal cord tissue samples were harvested at each time ,and the pathological changes of rat spinal cord were observed by HE staining ,the Nissl body and neuron survival index were observed by Nissl staining ,the expressions of Nrf2 and GCLC protein level were detected by immunohis-tochemical staining and Western blot.Results:The rat BBB score of LPS+EV group increased significantly than EV group at 7,14,28 d after operation ( P<0.05 ,P<0.01 );The NIV group between LPS+NIV group have no statistical significance at each time.As compared with EV group:the Nrf2 protein of LPS+EV group was expression increased significantly and Nissl staining showed that the neurons survival index was increased at 1,3 and 7 d(P<0.05,P<0.01);The GCLC protein of LPS+EV group was expression increased significantly at 1-14 d( P<0.05 );HE staining showed that the injured spinal cord pathological changes of LPS +EV group was obviously improved.Conclusion:Low dose lipopolysaccharide preconditioning can accelerate the nerve function recovery on rats with traumatic spinal cord injury ,the mechanism may be regulated by activating the Nrf 2 antioxidant stress pathway.

AIM: To investigate the linkage between the polymorphism of -109 and Glu237 in the high-affinity IgE receptor?(Fc?RⅠ?) gene and susceptibilty of allergic asthma in adults in a Chinese population. METHODS: Allergic asthma sample in adult and age-and sex-matched control were studied. A-109C/T and a coding variant Glu237Gly in Fc?RⅠ? were detected with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). RESULTS: (1)The genotype frequencies were 0.415 for -109T/T, 0.491 for-109T/C and 0.094 for -109C/C in allergic asthma in adults. No significant difference in the distribution of-109C/T polymorphism was found between allergic asthma adult subjects and healthy control subjects. 4.Safety and efficacy of intraperitoneal hyperthermic perfusion chemotherapy following laparoscopic palliative resection for gastric cancer patients with peritoneal metastasis.

OBJECTIVETo investigate the safety and efficacy of postoperative intraperitoneal hyperthermic perfusion chemotherapy(IHPEC) following laparoscopic palliative resection for advanced gastric cancer patients with peritoneal metastasis.
METHODSBetween March 2010 and October 2013, 37 patients with advanced gastric cancer were treated by IHPEC(cisplatin 100 mg, 5-fluorouracil 1000 mg and saline 2000 mL) following laparoscopic palliative resection in our department between March 2010 and October 2013 were analyzed retrospectively. Short-term efficacy and adverse reactions were observed.
RESULTSComplete remission (CR), partial remission (PR), stable disease (SD) and progressive disease (PD) were found in 18, 4, 8 and 7 cases respectively, and the total progression-free rate was 59.5%(22/37). The significant improved, improved, stable and progressive cases of Karnofsky performance status were 6, 13, 10 and 8 respectively, and the rate of improved and stable cases was 78.4% (29/37). Serious adverse reactions (class III ( or IIII) were noted in 3 cases (8.1%), including 2 cases of abdominal pain (class III), 1 case of nausea and vomiting.
CONCLUSIONSThe modality of IHPEC adopting cisplatin plus 5-fluorouracil regimen following laparoscopic palliative resection for advanced gastric cancer patients with peritoneal metastasis is technically feasible and safe, which has certain effect on postponing the progression of gastric cancer.

5.Risk factors for early progression in patient with non-disabling ischemic cerebrovascular events

Objective To explore the risk factors for early deterioration in neurological function/progressive neurological deficit in patients with non-disabling ischemic cerebrovascular events (NICE). Methods We conducted a retrospective analysis consecutive patient with non-disabling ischemic cerebrovascular events from the Department of Neurology,the First Affiliated Hospital of Xiamen University between January 2016 and December 2019.Patients were divided into progressive group (n=70) and non-progressive group (n=254) and the term progressive were defined as patients had an increase in National Institutes of Health Stroke Scale(NIHSS) score by ≥2 points within 7-day. Patient demographics,clinical and radiological data were recorded and analyzed. Multinomial logistic regression analysis was performed to determine the independent risk factors for early progression in patients with NICE. Results On univariate analysis,the incidence of diabetes mellitus,stenosis or occlusion in the corresponding artery,acute infection and new onset ischemic stroke were significantly higher in progressive group than in non-progressive group (P<0.05).The proportion of higher Fazekas scores (PWMH 2~3,PWMH+DWNH 3~6) in progressive group was significantly higher than that in non-progressive group(P<0.05). Multivariate logistic regression analysis showed that diabetes mellitus (OR=2.355,95%CI 1.220~4.546,P=0.011),stenosis or occlusion in the corresponding artery (OR=2.542,95%CI 1.405~4.600,P=0.002),acute infection(OR=4.513,95%CI 1.699~11.986,P=0.002),new onset ischemic stroke (OR=2.820,95%CI 1.022~7.781,P=0.045),higher Fazekas scores(PWMH+DWNH 3~6,OR=2.061,95%CI 1.119~3.798,P=0.020;PWMH 2~3,OR=2.046,95%CI 1.096~3.817,P=0.024) were independently associated with early progression in patient with NICE. Conclusion Diabetes mellitus,stenosis or occlusion in the corresponding artery,acute infection,new onset ischemic stroke,higher Fazekas scores (PWMH+DWNH 3~ 6,PWMH 2~3) may increase the risk of early progression in patient with NICE.
6.Awake craniotomy of gliomas in eloquent areas: an analysis of 19 cases

Objective To discuss the techniques and methods of surgery for brain gliomas located in eloquent areas at awake anesthesia. Methods Nineteen patients with brain gliomas in eloquent areas, admitted to our hospital from December 2014 to May 2017, were operated under awake anesthesia with neuronavigation and intraoperative ultrasonography for locating the lesions and intraoperative direct electrical stimulation for functional mapping of the eloquent areas. All patients were followed up from 3 to 18 months; the surgical efficacies were analyzed. Results Of 19 patients, 18 (94.74%) were achieved awake and alert during brain mapping and resection of the tumors;17 (89.47%) were detected the motor areas by intraoperative direct electrical stimulation, 6 (31.58%) were detected the sensory cortex and 12 (63.16%) were detected language related cortex. Of 19 patients, MR imaging 2-3 months after surgery indicated that 5 (26.32%) received total resection of lesions, 9 (47.37%) subtotal resection of lesions and 5 (26.32%) partial resection of lesions. Seven patients (36.84%) had transitory postoperative aphasia, 4 (21.05%) were with transitory postoperative dyskinesia and one (5.26%) with permanent dyskinesia. Conclusion Comprehensive applications of awake anesthesia, neuronavigation, intraoperative ultrasonography and intraoperative direct electrical stimulation technologies allow maximum safe resection of gliomas in eloquent areas and protection of brain function.
7.Association between expression of plasma miRNA and the risk of childhood acute lymphocytic leukemia

Objective To investigate the characteristics of distribution and expression profiles of plasma miRNA in childhood acute lymphocytic leukemia (cALL) patients;the association between cALL incidence risk and plasma miRNA levels;the feasibility of plasma miRNA serving as cALL diagnostic biomarker.Methods A total of 111 pairs of newly diagnosed cALL patients and patients with fractures were collected from Shenzhen Children's Hospital,China,between January 2015 and November 2016.Age and sex of the cases and controls were 1 ∶ 1 matched and LNATM miRNA microarray was performed using 4 pairs of cALL and controls selected from the sample population.The expression level of miRNA was validated by real time quantitative PCR.Conditional logistic regression analysis was applied to evaluate the association between miRNA expression levels and the incidence risk of cALL.The receiver operating characteristic curve (ROC) and reclassification analysis were conducted to assess the feasibility of miRNAs serving as biomarkers for cALL.Results A total of 204 differentially expressed miRNA were screened out and let-7f-5p,miR-5100,miR-25-3p and miR-3654 were selected for validation identified according to the inclusion criteria.The expression levels of let-7f-5p,miR-5100 and miR-25-3p in the cALL patients were significantly lower than those of the controls (P<0.01).After adjusting for confounding factors,3 miRNAs remained significantly associated with the risk of cALL (OR and 95%CI were 0.84 (0.76-0.92),0.81 (0.73-0.90)and 0.81 (0.74-0.89),respectively.Results from both the ROC analysis and reclassification analysis showed that introduction of one or more miRNA to traditional risk factors improved the area under the curve (P<0.05) and provided additional values to diagnosis (P<0.01).Conclusion The expression levels of let-7f-5p,miR-5100 and miR-25-3p were significantly associated with the incidence rate of cALL,and these miRNAs might serve as promising biomarkers for cALL.
8. Association of formaldehyde exposure,genome-wide DNA methylation,and childhood acute lymphocytic leukemia

OBJECTIVE: To explore the relationship of formaldehyde exposure, genome-wide DNA methylation, and prevalence of childhood acute lymphocytic leukemia( cALL).METHODS: A case-control study was conducted.Fifty-nine newly diagnosed cALL patients were selected as case group,and 54 orthopedic patients were included in control group.Enzyme-linked immunosorbent assay was used to measure the level of formaldehyde-human serum albumin( FA-HSA) and immunofluorescence method was used to examine the genome-wide DNA methylation level in whole blood.RESULTS: The level of FA-HAS in the blood of the case group was higher than that in the control group( median: 59.61 vs 35.06 fg/L,P < 0.01).Genomic-wide DNA methylation level in the case group was lower than that in the controls[( 2.86 ± 0.31) vs( 3.00 ± 0.28),P < 0.05].Formaldehyde exposure level was not associated with genomic-wide DNA methylation( Spearman correlation coefficient =-0.18,P > 0.05).High FA-HAS level and hypomethylation of genomic-wide DNA were risk factors for cALL onset( P < 0.05).CONCLUSION: Patients with high level of formaldehyde exposure and hypomethylation of genomic-wide DNA have a high risk of cALL.
9.Cohort study on the relationship between women’s pre-pregnancy age, alanine aminotransferase, blood glucose level and premature birth

Objective To investigate the effect of pre-pregnancy age, alanine aminotransferase (ALT), blood glucose (GLU) level on premature birth. Methods Based on the free pre-marital health check-ups and pre-pregnancy eugenic health check-ups in Dongguan, the examination results of the subjects were obtained and the pregnancy outcomes were followed up. Logistic regression models were used to analyze the relationship between women’s pre-pregnancy age, ALT, and GLU levels and the incidence of premature birth. Results A total of 18 150 subjects were included in this study, and the overall incidence of premature birth was 6.06% (1,099/18,150). The women’s pre-pregnancy age 30 to 34 (RR=1.26, 95% CI:1.07~1.48), age>34 (RR=1.82, 95% CI:1.52~2.18), ALT≥40 U/L (RR=1.40, 95% CI: 1.09~1.80) and GLU>6.1 mmol/L(RR=1.74, 95% CI: 1.01~2.97) were the influencing factors of premature birth. Conclusion Older women, abnormal serum alanine aminotransferase level and hyperglycemia had an impact on the occurrence of premature birth, which has important guiding significance for health education of eugenics before pregnancy.

Taking chronic atrophic gastritis (CAG) as an example, the frontier technologies in data science have been introduced into the inheritance, innovation and development of traditional Chinese medicine (TCM), providing reference for conducting real-world clinical research on specialized diseases of TCM. This paper put forward the construction of CAG data science system by elaborating the connotation of data science and its application value in TCM, and discussed the path to build CAG data science system, namely through "data acquisition-knowledge expression-knowledge reasoning" to establish CAG database, knowledge base and develop diagnosis platform differentiating diseases and syndromes. Besides, this paper analyzed the prospects of CAG data science in improving data governance ability and knowledge discovery efficiency, deepening the level of knowledge sharing, promoting interdisciplinary integration, and strengthening the integration process of industry, academia and research.
