1.Study on the filter mating method of plasmid conjugation experiment for Corynebacterium

Objective To set up the filter mating method of plasmid conjugation experiment for Corynebacterium.Methods The first plasmid conjugation between different genus was performed by filter method.Three strains of highly erythromycin-resistance Enterococcus faecalis isolates[minimal inhibitory concentration(MIC)of erythromycin>256 mg/L,MIC of levofloxacin≤8 mg/L] which screened by agar dilution method were used as donators and 3 strains of Corynebacteria isolates(MIC of erythromycin≤32mg/L,MIC of levofloxacin=128 mg/L)were used as acceptors.In the second conjugation between the same genus but in different species,three Corynebacterium xerosis strains(MIC of erythromycin>256 mg/L,MIC of ceftazidime=16 mg/L)which were conjugants of the first conjugation were used as donators and 6 strains of Corynebacteria isolates(MIC of erythromycin≤32 mg/L,MIC of ceftazidime 128 mg/L)were used as acceptors.Results In the first conjugation between different genus,the plasimid conjugations were done for 9 times between 3 donators and 3 acceptors,4 of which gained the phenotype and genotype of resistance and the transfer rate was 44%.In the second conjugation experiment,the plasmid conjugations were done for 18 times between 3 donators and 6 acceptors,7 of which gained the resistant phenotype and the transfer rate was 39%.Erythromycin-resistance was transferred from the donators to the conjugants in varying degrees.The resistance was transferred from Enterococcus faecalis to Corynebacteria and then to other Corynebacteria which were of the salne genus but of different species.Conclusion Filter mating conjugation method could be used to study the mechanisms of resistance transmission of Corynebacterium.
2.Prevalence and postoperative changes of metabolic syndrome in patients with adrenocortical adenoma

Objective To investigate changes of metabolic syndrome (MS ) in patients with adrenocortical adenoma (ACA ). Methods Sixty‐five patients with aldosterone producing adenoma (APA) ,43 patients with cortisol‐secreting adenoma (CSA) and 24 patients with nonfunctional adrenal adenoma (NAA) were evaluated. Body Mass Index (BMI) ,blood pressure ,plasma lipid ,fasting and 2 h postprandial plasma glucose ,plasma and urinary aldosterone ,plasma potassium ,basic and upright plasma renin activity (PRA ) ,plasma and urinary cortisol ,plasma ACTH and plasma potassium ACA were examined and compared with those from 53 patients of essential hypertension (EH ) and 58 controls. Adrenalectomy was performed for all the patients with adrenocortical adenoma. The data after operation were compared with those before operation. Results (1) The prevalence of MS was 38.5% ,41.9% , 20.8% ,24.5% and 17.5% in the groups respectively. The prevalence of obesity/overweight was higher in CSA group than in the other groups. There were 44.6% patients with hyperglycemia in APA group ,and 48.8% in CSA group. The prevalence of hypertension was the highest in APA and EH group ,followed by CSA group ,NAA and control groups. There were 55.4% patients with lipid disturbance in APA group , and 46.5% in CSA group. (2 ) We divided all APA and CSA patients into groups with or without MS. The APA patients with MS had higher plasma aldosterone than those without MS. The CSA patients with MS had higher 08 :00 and 24 :00 plasma cortisol than those without MS. (3) After operation ,the plasma and urinary aldosterone were decreased and the plasma potassium was increased in APA patients. The 08 :00 ,24 :00 plasma cortisol and urinary cortisol were decreased and the 08 :00 plasma ACTH was increased in CSA patients. There were no significant differences in hormonal measurements of NAA patients before and after operation. (4 ) The prevalence of MS was 27.7% ,27.9% and 29.2% in three groups after operation. The systolic and diastolic blood pressures were decreased in APA patients after operation ,and the systolic blood pressure decreased in CSA patients. There was no significant difference in MS measurements of NAA patients before and after operation. Conclusions Our study indicates that patients with functional adrenocortical adenoma have a significantly higher prevalence of metabolic syndrome which is related to over secretion of adrenal cortex hormone. After operation ,accompanying normalization of hormonal secretion ,the condition of MS in functional adrenocortical adenoma patients is improved.
3.Study on the obtaining situation of osteoporotic fractures related knowledge and influencing factors of the nurses

Objective To know the situation of nurses obtaining osteoporotic fractures related knowledge ,and provid guiding basis for further education and relevant training on target people .Methods Questionnaire survey and semi -structured interviews methods were used to investigate 595 nurses ,9 clinical nurse workers and nurse managers who come from a tertiary comprehensive teaching hospital were interviewed personally .Results The scores of the related knowledge in nurses were 35 .70 ± 6 .36 ,score rate was 67 .36% ;Interview data analysis showed that lack of professional training ,training was not systematic ,the approach obtain knowledge was simple were result in lack of the related knowledge in nurses ;By the multiple liner stepwise regression ,The educa‐tion level and professional title were main factors which affect nurses acquired osteoporosis knowledge .Conclusion It is necessary to focus on improving the strengthen of related knowledge education for the nurses with low education background and low profes‐sional title ,At the same time ,related departments of the hospital should strengthen the training of professional knowledge .
4.Posterior interbody fusion with circular saw for treatment of lumbar spondylolisthesis

[Objective]To modify posterior lumbar interbody fusion for improvement of convenient manipulation and safety in the operation,and solid fusion. [Methods]After total decompression of the spinal nerve through posterior medline approach,discectomy of the slipping segment was performed with a circular saw with diameter of 1.0 centmeter.Under direct visualization through the hole of the circular saw,the disc tissue including cartilage of upper and lower end plates was removed with a long handle curette.After pedicle screw and rod system was placed,a bone block harvested from the posterosuperior iliac with a circular saw with diameter of 1.2 centmeter was implanted into the intervertebral space.Then compression of the intervertebral space to catch the bone lock firmly was conducted by regulating and fixing the the pedicle screw and rod system.[Results]Forty-eight patients in this report were followed up from 6 to 24 months.Based on JOA low back pain score,there were 36 patients was in excellent result,8 in good and 4 in fair,the satisfactory rate was 91.67% and effective rate was 100%.[Conclusion]Using circular saws in posterior lumbar interbody fusion has advantagas of good visualization profit removing disc tissue,preventing neurovascular injuries,and making good bone grafting bed.Convenient and secure bone harvesting decrease blood loss,time consume during the operation and prevent donor site complication.Futhermore,follow up results of the patients in this report demonstrate that posterior interbody fusion with circular saw for treatment of lumbar spondylolisthesis have satisfactory outcome and high fusion rate.
5.p16 gene methylation status of and its effect on mRNA expression in hepatocellular carcinoma

Purpose:To investigate the status of p16 gene and its influence in p16 mRNA expression in primary hepatocellular carcinoma (HCC). Methods:The methylation status of p16 gene and expression of p16 mRNA were examined by methylation-specific PCR (MSP) and RT-PCR in twenty-five primary HCC and corresponding paracancerous tissue specimens.Results:Methylation was detected in 12 of 25 (52.0%) tumor tissue specinmens, while 6 of 25 (24.0%) in corresponding paracancerous tissue specimens. Loss of p16 mRNA expression was found in 19 specimens with methylated p16 gene,while expression of p16 mRNA was detected in 27 of 31 specimens with unmethylated p16 gene. There was significant difference in mRNA expression between specimens with methylated p16 and unmethylated p16.Conclusions:p16 gene methylation is a frequent event in HCC. The methylation of p16 gene might lead to loss of p16 mRNA expression. Detection of p16 gene methylation may be a useful marker in the molecular diagnosis of hepatocellular carcinomas.
6.Effect of estradiol on expressions of transforming growth factor ? and smoothelin in human prostatic stromal cells

Objective To investigate the relationships among estrogen,transforming growth factor ?(TGF?) and smooth muscle cells in human prostatic tissue by cell experiment. Methods Eleven strains of prostatic stromal cells were cultured successfully from BPH patients,and stimulated by estradiol(E 2).Cell proliferations were observed by MTT,and mRNA expressions of TGF? 1,TGF? 2 and smoothelin were observed by RT-PCR. Results E 2 could not influence TGF? 1 expressions in prostatic stromal cells,but could upregulate expressions of TGF? 2 and smoothelin;and expression values of TGF? 2 and smoothelin had a positive correlation. Conclusions E 2 can influence growth and differentiation of prostatic smooth muscle cells through induction of TGF? 2 expression.

Objective To analyze the characteristics and influencing factors of coronary sinus(CS) dilation in patients with primary pulmonary hypertension(PPH) and persistent left superior vena cava(LSVC). Methods Control group included twenty-two normal subjects [mean age ( 40.6 ? 10.6 ) years],PPH group included twenty-six patients with PPH [mean age ( 41.9 ? 11.4 ) years] and LSVC group included thirty-three patients with LSVC [mean age ( 30.9 ? 15.8 ) years]. After the standard apical four-chamber view was obtained,the transducer was tilted backward to direct ultrasonic beam towards the left atrioventricular groove,then the distal part of the CS could be exposed. 9.Effect of combined cataract and glaucoma surgery on function of eye surface and tear film

Abstract?AIM:To evaluate the effect of combined glaucoma and cataract surgery on the function of eye surface and tear film.?METHODS:This clinical trial involved 75 patients ( 75 eyes ) with glaucoma complicated with cataract undergoing combined glaucoma and cataract surgery.All the eyes were divided into surgical group ( the eyes undergoing operation ) and control group ( the other eyes).The symptoms and signs of dry eye disease, Schirmer's I test, break -up time ( BUT ) , corneal fluorescein staining were tested and marked 1mo after operation.Comparison of the results was made between two groups.?RESULTS:The average intraocular pressure was 16.25± 0.46mmHg at 1mo after operation, compared with 45.29± 4.39mmHg at 3d before surgery, the difference was statistically significant ( P<0.05 ).Preoperative average visual acuity was 0.08±0.06, corrected visual acuity was<0.05 in 23 eyes,≥0.05 to<0.1 in 16 eyes,≥0.1 to<0.3 in 36 eyes.Postoperatively 1mo, corrected visual acuity:<0. 05 for 7 eyes (9%),≥0.05 to <0.1 for 11 eyes (15%), ≥0.1 to<0.3 for 38 eyes (51%), ≥0.3 to <0.5 for 11 eyes (15%) , ≥0.5 for 8 eyes (11%), postoperative visual acuity was 0.15 ±0.1, which was significantly higher than before surgeries ( P<0.05).The postoperative dry eye symptoms of surgical group was higher, compared with that of control group, and the differences of diagnosis of dry eye test indicators between the two groups were statistically significant(P<0.05).? CONCLUSION: Combined cataract and glaucoma surgery may affect postoperative ocular surface and tear film function, make the tear film stability damaged and lead to dry eye disease.
10.Screening of miRNAs in ovarian cancer patients and its correlation with clinicopathological features

Background and purpose:The prognosis of ovarian cancer is poor and the diagnosis is relatively late. It is needed to search for early diagnosis and treatment for ovarian cancer. This study investigated the expression of serum miRNAs in patients with malignancy or benign ovarian tumor preoperativey and analyzed its correlation with clinicopathological progress and prognostic features of epithelial ovarian cancer.Methods:Forty-eight miRNAs which have been reported to be related to ovarian cancer were ordered. The differential expression of 48 miRNAs in the serum of patients with malignant or benign epithelial ovarian tumors was detected by TaqMan low density array. The differentially expressed miRNAs were further confirmed by real-time fluorescent quantitative polymerase chain reaction (RTFQ-PCR). The relationship between the expression level of selected miRNA and clinical clinicopathological factors, progress and prognosis were analyzed.Results:TaqMan low density array and further RTFQ-PCR showed that only miR-125b was sig-nificantly increased in 135 ovarian cancer patients as compared with 38 individuals with benign tumor. The expression of miR-125b was higher in early stage patients than that in advanced stage patients (P=0.039). The patients without residual tumor expressed more miR-125b than patients with residual tumor (P=0.013). The high level of miR-125b was signifi-cantly correlated with longer progress free survival (PFS) (P=0.003), but not correlated with overall survival (OS) (P=0.069). Conclusion:MiR-125b may play an important role in the pathogenesis of epithelial ovarian cancer and prognosis. It may be a potential gene to predict the recurrence of epithelial ovarian cancer, but the change of gene expression at different stages and its underlying mechanism need further research.
