To counteract the immune system in parasitic hosts, some viruses encode proteins to suppress the RNA interference (RNAi) effect. In this report, we established two RNAi systems to be easily observed with strong and obvious effect. The function of the P19 of tomato bushy stunt virus, which suppresses RNAi in mammal cells, was then studied using these two systems. Short hairpin RNAs targeting green fluorescence protein (pshRNA-GFP) and firefly luciferase (pshRNA-luc) were designed and inserted into a eukaryotic transcriptional vector pTZU6+1, respectively. The shRNA expressing vectors were co-transfected with plasmids containing the target gene with or without P19. The GFP expression level was assayed by fluorescence microscopy, Western blotting and RT-PCR. The luciferase expression level was analyzed by the dual-luciferase assay system. pshRNA designed in this study down-regulated the target gene specifically and efficiently, with a decrease of expression of both genes of about 70%, respectively. When P19 was introduced into the RNAi systems, the expression of both GFP and the luciferase were mostly recovered compared with the control groups. The RNAi systems of GFP and luciferase were constructed successfully, demonstrating that P19 of tomato bushy stunt virus has the ability to counteract the RNAi effect induced by shRNA in mammal cells.

Objective To study the different ultrasonic features in patients of polycystic ovary syndrome (PCOS) with or without obesity based on body mass index (BMI), and to investigate whether certain hormonal factors correlate with ovarian morphology and blood flow, and to discuss the role of ultrasound combined with hormone test in the diagnosis of obese PCOS. Methods One hundred and five women with PCOS were recruited. Patients were divided into two groups according to BMI;obese PCOS group (OB-PCOS, n=32, BMI≥25 kg/m2) and non-obese PCOS (NOB-PCOS, n=73, BMI<25 kg/m2). The ultrasonic parameters of follicle number (FN), ovarian volume (Vol), resistance index (RI) of ovarian stromal blood, RI of uterine artery and serum levels of luteinizing hormone (LH), follicle stimulating hormone (FSH), the ratio of luteinizing hormone and follicle-stimulating hormone (LH/FSH), progesterone (P), estradiol (E2), free testosterone (FT), prolactin (PRL), sex hormoe binding globulin (SHBG), fasting plasma glucose (FPG), fasting insulin (FINS), the extent of insulin resistance and hyperandrogenism (HOMA-IR) were measured and compared. The correlation of the ultrasonic parameters and hormonal factors were analyzed. Results The Vol of OB-PCOS group was significantly higher than NOB-PCOS group [(12.25±4.89) ml vs (10.73±2.30) ml, t=2.20, P < 0.05]. FN and uterine artery RI of OB-PCOS group had a rising trend and RI of ovarian interstitial was on a reducing trend compared with NOB-PCOS group. But the differences were not statistically significant. The levels of FINS and HOMA-IR in OB-PCOS group [(14.82±6.45) mU/L and (3.91±3.30)] were significantly higher than those in NOB-PCOS group [(8.04±4.57) mU/L and (1.64±1.20)] (t=4.87, 3.47, respectively, both P < 0.01). And FSH in NOB-PCOS group was significantly higher than OB-PCOS group [(5.95±1.91) U/L vs (4.65±1.88) U/L, t=-2.77, P<0.01]. In POCS patients, FN was significantly associated with LH/FSH (r=0.35, P<0.01), and FT (r=0.38, P<0.01). Vol was significantly associated with LH/FSH, BMI, HOMA-IR and FPG (r=0.27, P<0.05;r=0.25, P<0.05;r=0.40, P<0.01;r=0.32, P<0.01). Objective To breed a guinea pig inbred strain and set up a method for detection of the microsatellite markers of genetic structure in guinea pigs. Method Using inbreeding methods we try to breed the Zmu-1:DHP inbred strain. With 15 pairs of polymorphism microsatellite primers, the genetic homozygosity of Zmu?1:DHP inbred strain,Zmu?1:DHP outbred strain and Zmu?2:DHP inbred strain ( as control) were examined by PCR. Results After breeding for 13 years, 8 sublines of Zmu?1:DHP inbred strain ( >20 generations) were bred. After identification, the gene frequency of the second subline of Zmu?1:DHP inbred strain was 86. 7%,higher than Zmu?1:DHP outbred strain (6. 7%) and Zmu?2:DHP inbred strain (66. 7%). The average number of loci of Zmu?1:DHP inbred strain was 1. 13,lower than that of Zmu?1:DHP outbred strain (2. 47%) and Zmu?2:DHP inbred strain (1. 33%). The genotypic frequency of Zmu?1:DHP in?bred strain was also higher than that of the other strains. The gene types of Zmu?1:DHP inbred strain were included in the genes of Zmu?1:DHP outbred strain, but Zmu?1:DHP inbred strain was short of 2 characteristic genes. The gene homozy?gous rates of 8 sublines of Zmu?1:DHP inbred strain were different with each other, among them, those of the 2nd and 8th sublines were higher than others. Conclusions There are both homozygosity and specificity in the Zum?1:DHP inbred strain and Zum?1:DHP outbred strain. The second Zum?1:DHP subline becomes a new inbred strain guinea pig. It is es?sential that the subline with the characteristic property is screened from these sublines. The guinea pigs of black Zmu?2:DHP inbred strain carrying microsatellite markers not present in the white strains, may carry optimal genes related with hair color properties.

Objective To investigate the application value and effect of lung vascular and lesion positioning guided by Doppler ultrasound in the operation of thoracoscopic lobectomy. Methods Thirty cases of experimental pigs were randomly divided into A, B and C three groups. The group A was operated by conventional thoracoscope for upper right lobectomy, and group B was operated by thoracoscope for upper right lobectomy which guided by Doppler ultrasound cavity prober. Then, the operation time, blood loss and materials costs were compared between the two groups. The group C was used to establish the model of lung cancer and CT scanning was used to indicate the lung lesions. During operation, lesions were positioned by Doppler ultrasound and wedge resections were performed under thoracoscope. Then, the pulmonary lobectomy guided by ultrasound and mediastinal lymph nodes dissection were performed. At last, the coincidence rate of intraoperative ultrasound and CT location was analyzed statistically. Results There were significant differences in operation time between Groups A and B ( < 0.01), while no significant differences in blood loss and material costs ( >0.05) . In group C, lesion location positioning guided by ultrasound during the operation was completely consistent with guided by CT before the operation. No significant differences in the lesion diameter were observed between ultrasound and CT methods ( > 0.05) . Conclusion With the application of Doppler ultrasound, the invisible and untouchable blood vessel can become visible and can be positioned in precise to shorten the time of thoracoscopic lobectomy. In addition, the accurate positioning of tiny lesions in the lung can solve the problem of lung small lesion positioning which is difficult in the operation under thoracoscope. Therefore, it can be promoted in clinical applications in a wide range.

According to the mean defect of visual field, they were divided into light ( observation Group 1 ) , moderate ( observation Group 2 ) and heavy group(observation Group 3). Another 65 cases of 130 eyes with cataract were taken as the control group in our hospital. The observation and expression of serum cytokines in these patients with glaucoma were taken. ·RESULTS:There was no significant difference in serum IL-2 and IFN- γlevels between the two groups (P>0. 05). The sIL-2R and IL-4 levels in the glaucoma group were higher than those in the control group, and the levels of IL-6 and IL-12 were lower than those of the control group (P<0. 05). The IOP and proportion of myopia in the glaucoma group was higher than that in the control group, the difference was not statistically significant ( P<0. 05) . The ratio of moderate myopia to severe myopia in each group was not statistically significant (P>0. 05). To observe the low myopia ratio in Group 3 of patients, it was less than observation Group 1 and observation Group2 ( P > 0. 05 ). There was no statistically significant difference between observation Group 1 and 2 of patients on IL-2, sIL-2R, IL-4, IL-6 and IFN-γlevels (P>0. 05). The level of sIL-2R in the Group 3 was higher than that in the Group 1, and the level of IL-12 was lower than that in the Group 1 and in the Group 2 (P<0. 05). There was no significant difference in IL-2, IL-4, IL-6 and IFN-γlevels between the glaucoma patients (P>0. 05). The IOP level and the proportion of myopia in the Group 3 were higher than those in the Group 1 and the Group 2 were observed, the difference was statistically significant (P<0. 05). The intraocular pressure was higher in Group 2 than in group 1 (P<0. 05). There was no statistically significant difference on proportion of myopia between observation Group 1 and observation Group 2 (P>0. 05). ·CONCLUSION: The levels of serum IL-12, sIL-2R and intraocular pressure in patients with primary open-angle glaucoma fluctuated significantly at different stages of the nerve injury, indicating that the immune response and intraocular pressure were involved in the process of optic nerve damage.
