1.Quality standard for Guishao Soft Capsules

AIM: To establish the quality standard for Guishao Soft Capsules(Radix Paeoniae Alba,Radix Angelicae Sinensis,Rhizoma Chuanxiong, etc.) METHODS: Radix Paeoniae Alba,Rhizoma Atractylodis,Rhizoma Chuanxiong,Rhizoma Alismatis and Radix Angelicae Sinensis were identified by TLC.The contents of albiflorin and paeoniflorin in Guishao Soft Capsules were determined by HPLC.HPLC analytical method was carried out(using) a C_(18) column and a mixture containing 17 volume of acetonitrile and 83 volume of 0.05% acetic acid as the mobile phase.The detection wavelength was set at 230 nm. RESULTS: Spots obtained from the test solutions had the same color in reference solution and medical material in the same location,and the blank solution had no interfernece.The linear range of albiflorin was from 0.53 ?g to 1.06 ?g(r=0.999 8),the average recovery was(100.3%.) The linear range of paeoniflorin was from 0.29 ?g to 2.00 ?g(r=0.999 9),the average recovery was(99.49%.) CONCLUSION: In TLC,the spots are very clear and specific to identify the herbal medicine in Guishao Soft Capsules.It is simple,quick,high precise and accurate for HPLC to control the quality of Guishao Soft Capsules.
2.Artificial ossicular chain reconstruction:biocompatibility, properties and laryngecartilage defect repair

BACKGROUND: Defected Laryngeal cartilage has many alternatives, including autologous cartilage, al ograft cartilage and metal stents. Although these materials can achieve desired outcomes in laryngeal cartilage defect repair, certain limitations exist. OBJECTIVE: To investigate the biocompatibility and properties of artificial ossicular chain reconstruction materials, and to explore the effect of artificial ossicular chain reconstruction materials on laryngeal cartilage defect repair. METHODS: Porous hydroxyapatite otosteon was prepared by high-temperature calcination of hydroxyapatite, fol owed by cultured in bone morphogenetic protein solution extracted from fresh human bone to construct bone morphogenetic protein-hydroxyapatite artificial ossicular chain reconstruction material. And then, the biocompatibility and characteristics of the material were analyzed. Forty adult male New Zealand white rabbits were randomly divided into porous hydroxyapatite group and artificial ossicular chain reconstruction material group (n=20 per group), and underwent repair with porous hydroxyapatite material and bone morphogenetic protein-hydroxyapatite artificial ossicular chain reconstruction material respectively after modeling of laryngeal cartilage defect. RESULTS AND CONCLUSION: There was a significant difference in compressive strength of artificial ossicular chain reconstruction materials with different porosities. No symmetry sphere formed in hol ows of the outer surface of the material, with polygonal appearance and with a pore size of 100-200 μm. There were no obvious adverse reactions in both two groups after implantation, but in the artificial ossicular chain reconstruction material group, numerous fibrous connective tissues and obvious bone nodules appeared, and the degradation rate of the material was faster. These results suggest that the bone morphogenetic protein-hydroxyapatite artificial ossicular chain reconstruction material exhibits good biocompatibility and properties, which wil obtain satisfactory outcomes for laryngeal cartilage defect repair. So, the material holds a great value of clinical application.

5.Signal transduction pathway for cardiac myocyte contraction induced by Substance K in rats

Objective To study the signal transduction pathway for myocyte contraction induced by Substance K (SK) in rats. Methods The fluorescent Ca 2+ indicator Fluo 3AM was used to quantitate the calcium signal directly in the primary cultured myocardial cells. Changes of free Ca 2+ concentration in cardiac myocytes were detected by flow cytometry. The phospholipase C (PLC) inhibitor, neomycin and IP3 receptor antagonist, heparin, were used to block signal system of phosphates Ca 2+ in order to investigate whether they took part in the SK induced changes of [Ca 2+ ]i in myocytes. Results SK(1.78?10 -5 mol/L) elevated [Ca 2+ ]i in myocytes. The effects could be blocked by neomycin and heparin. Conclusion SK can elevate [Ca 2+ ]i in myocyte, which may be mediated by a signal system of phosphates Ca 2+ .

A novel coronavirus (COVID-19) that broke out in December 2019 has been declared a public health emergency of international concern. Nucleic acid detection has an irreplaceable role in the diagnosis of 2019-novel coronavirus (2019-nCoV) infection. However, due to the high requirements of laboratories and technicians, cumbersome operations, and the possibility of omission, nucleic acid detection should be combined with specific antibodies to achieve large-scale screening of suspected patients and close contacts. Moreover, antibody detection can reduce the exposure risk of medical personnel during the collection of respiratory tract samples.
7.SEM observation of hair cell progenitors forced to express Hath1 from rat cochleae

OBJECTIVE To investigate the effect of Hath1 on epithelial ridge (GER) cells from postnatal rat cochlear. METHODS An experimental method was developed which allowed the isolation and culture of GER cells from P1 rat cochleae using a combinatorial approach of enzymatic digestion and mechanical separation. The dissociated GER cells were cultured in DMEM+10% FBS. The GER explants infected with ad-Hath1 for 3 hours were cultured for 10 days and observed under scanning electron microscope. RESULTS The GER cell cultures tended to attach to the substratum and grow in patches which assume a polygonal morphology similar to that of epithelial cells in medium containing 10% serum. Stereociliary bundle-like structures were observed in the boundary of GER cell patches forced to express Hath1. CONCLUSION That GER cells which are likely hair cell progenitors could be cultured in vitro and generate stereociliary bundle-like structure when forced to express Hath1 suggests that the misexpression of Hath1 probably can induce the predifferentiation of pure hair cell progenitors into hair cells in vitro.
8.Influencing factors for self-violent thoughts and behaviorsamong ninth-grade students

Objective:
To learn the influencing factors for self-violent thoughts and behaviors among the ninth-grade students in Jiangbei District of Ningbo, so as to provide the basis for carrying out psychological intervention.
Methods:
The ninth-grade students in a junior high school each of the six streets ( towns ) in Jiangbei District were recruited by multi-stage cluster sampling method from October to November 2018. A self-designed questionnaire was used to investigate the general information, life style, psychological condition, self-violent thoughts and behaviors in the latest year. Multivariate logistic regression model was used to analyze the influencing factors for self-violent thoughts and behaviors.
Results:
A total of 1 300 questionnaires were issued and 1 238 valid ones were recovered, with an effective rate of 95.23%. The prevalence rates of self-violent thoughts and behaviors among the ninth-grade students were 16.88% and 8.48%. TheResults of the multivariate logistic regression analysis showed that abnormal marital status of parents ( OR=2.074, 95%CI: 1.139-3.775 ) , no moderate physical activity ( OR=1.764, 95%CI: 1.101-2.826 ) , sleeping less than 8 hours ( OR=1.858, 95%CI: 1.273-2.712 ) , alcohol consumption ( OR=2.108, 95%CI: 1.448-3.070 ) , sense of despair ( OR=6.397, 95%CI: 4.195-9.757 ) , sense of anxiety ( OR=4.094, 95%CI: 2.725-6.151 ) , sense of loneliness ( OR=1.879, 95%CI: 1.241-2.854 ) , and cyber-bullying ( OR=5.155, 95%CI: 2.780-9.556 ) were risk factors for self-violent thoughts; not living with parents ( OR=2.057, 95%CI: 1.021-4.145 ) , alcohol consumption ( OR=1.756, 95%CI: 1.074-2.872 ) , sense of despair ( OR=7.197, 95%CI: 4.365-11.864 ) , sense of anxiety ( OR=5.832, 95%CI: 3.276-10.382 ) , fighting ( OR=2.272, 95%CI: 1.219-4.232 ) and physical bullying ( OR=3.462, 95%CI: 1.135-10.558 ) were risk factors for self-violent behaviors.
Conclusion
Drinking, family relationship, bullying, and mental health problems such as sense of despair and anxiety were associated with the self-violent thoughts and behaviors of ninth-grade students.
10.Breeding of High Streptolydigin-Producing Strain by Complex Mutagenesis and Resistance Selection

A high Streptolydigin-producing strain Streptomyces lydicus AS 4.2501-L8 with genetic stability was obtained from the original strain S. lydicus AS 4.2501-P28 by mutagenesis with UV and acridine orange, followed by resistance selection. Its yield of Streptolydigin was 177.0 ?g/mL, 96.2% higher than the original strain.
